Objective To investigate effects of electroacupuncture (EA) on expression of intercellular adhesion molecule-1 (ICAM-1) in the rat of local cerebral ischemia-reperfusion. Methods Eighty SD rats were randomly divided into a normal control group, a sham operation group, a model group and an EA treatment group, 20 rats in each group. The thread-obstruction method was used for preparation of ischemia-reperfusion model. Zea-Longa rating criteria were used for evaluation of nervous function disorder; Immunohistochemical SABC method was used for detection of ICAM-1 expression in the microvascular endothelial cell of the ischemic brain region, and ELISA method for the soluble ICAM-1 (slCAM-1) content in peripheral blood. Re. suits After cerebral ischemia-reperfusion, both ICAM-1 expression level in the microvascular endethelium cell of the ischemic brain region and slCAM-1 content in the peripheral blood significantly increased in the model group as compared with the normal group and the sham operation group (P〈0.01); After EA treatment, the ICAM-1 expression level in the microvascular endothelial cell of the ischemic brain region and slCAM-1 content in the peripheral blood were significantly down-regulated in the EA treatment group as com- pared with the model group (P〈 0.05). Conclusion After cerebral ischemia-reperfusion, the microvascular endothelial cell of the ischemic brain region releases ICAM-1, which induces inflammatory injury of cerebral tissues; EA treatment can decease the expression of ICAM-1, so as to prevent the brain from the injury. 展开更多

AIM: To investigate the role of nuclear factor kappa B (NF-κB) in the pathogenesis of lung injury induced by intestinal ischemia/reperfusion (I/R), and its effect on intercellular adhesion molecule-1 (ICAM-1) ... AIM: To investigate the role of nuclear factor kappa B (NF-κB) in the pathogenesis of lung injury induced by intestinal ischemia/reperfusion (I/R), and its effect on intercellular adhesion molecule-1 (ICAM-1) expression and neutrophil infiltration. METHODS: Twenty-four Wistar rats were divided randomly into control, I/R and pyrrolidine dithiocarbamate (PDTC) treatment groups, n = 8 in each. I/R group and PDTC treatment group received superior mysenteric artery (SMA) occluding for 1 h and reperfusion for 2 h. PDTC group was administrated with intraperitoneal injection of 2% 100 mg/kg PDTC 1 h before surgery. Lung histology and bronchia alveolus lung fluid (BALF) protein were assayed. Serum IL-6, lung malondialdehyde (MDA) and myeloperoxidase (MPO) as well as the expression level of NF-κB and ICAM-1 were measured.RESULTS: Lung injury induced by intestinal I/R, was characterized by edema, hemorrhage and neutrophil infiltration as well as by the significant rising of BALF protein. Compared to control group, the levels of serum IL-6 and lung MDA and MPO increased significantly in I/R group (P=0.001). Strong positive expression of NF-κB p65 and ICAM-1 was observed. After the administration of PDTC, the level of serum IL-6, lung MDA and MPO as well as NF-κB and ICAM-1 decreased significantly (P〈 0.05) when compared to I/R group.CONCLUSION: The activation of NF-κB plays an important role in the pathogenesis of lung injury induced by intestinal I/R through upregulating the neutrophil infiltration and lung ICAM-1 expression. PDTC as an inhibitor of NF-κB can prevent lung injury induced by intestinal I/R through inhibiting the activity of NF-κB. 展开更多

目的探讨尿中性粒细胞明胶酶相关脂质运载蛋白(NGAL)和肾损伤分子-1(KIM-1)对非紫绀型先天性心脏病(先心病)患儿体外循环(CPB)术后急性肾损伤(AKI)的诊断价值。方法回顾性分析2015年6月至2017年5月天津市儿童医院收治的200例... 目的探讨尿中性粒细胞明胶酶相关脂质运载蛋白(NGAL)和肾损伤分子-1(KIM-1)对非紫绀型先天性心脏病(先心病)患儿体外循环(CPB)术后急性肾损伤(AKI)的诊断价值。方法回顾性分析2015年6月至2017年5月天津市儿童医院收治的200例在CPB下行心脏手术的非紫绀型先心病患儿的临床资料,将术后48 h内发生AKI的患儿纳入AKI组,同时选取其中与AKI患儿性别、年龄、体重、基础合并症、手术时间等因素相匹配的非AKI患儿纳入非AKI组。比较两组患者术前及术后各时间点血肌酐(SCr)和尿NGAL、KIM-1〔以尿肌酐(UCr)校正〕的差异,用受试者工作特征曲线(ROC)分析其对术后AKI的诊断价值。结果CPR术后48 h内有32例患儿发生不同程度的AKI,发生率为16.0%;非AKI对照组60例。与非AKI组比较,AKI组术后2 h尿NGAL、术后4 h尿KIM-1、术后10 h SCr即显著升高,分别于6、8、24 h达高峰后逐渐下降。诊断AKI的ROC曲线下面积(AUC)和95%可信区间(95%CI)分别为术后2 h尿NGAL 0.940(95%CI=0.890-0.990)、术后4 h尿KIM-1 0.939(95%CI=0.891-0.986)、术后10 h SCr 0.959(95%CI=0.916-1.000)。术后2 h尿NGAL截断值为588.0 μg/g时,诊断AKI的敏感度为87.5%,特异度为95.0%,准确性为93.5%;术后4 h尿KIM-1截断值为9.55 ng/mg时诊断AKI的敏感度为87.5%,特异度为91.7%,准确性为90.2%;术后10 h SCr截断值为61.90 μmol/L时诊断AKI的敏感度为90.6%,特异度为95.0%,准确性为95.7%。结论尿NGAL和KIM-1可以作为早期诊断非紫绀型先心病患儿CPB术后AKI的生物学标志物。 展开更多

Objective: To observe the changes of plasma concentrations of endotoxin, soluble intercellular adhesion molecule 1, tumor necrosis factor α, and urinary microalbumin in children undergoing cardiac procedure and to study the effects of cardiopulmonary bypass (CPB) on the injury or activation of endothelial cells and vascular permeability. Methods: Twenty children undergoing cardiac operation with CPB were selected in the study. Plasma concentrations of endotoxin, soluble intercellular adhesion molecule 1, tumor necrosis factor α, and urinary microalbumin were measured after anesthetic induction (baseline), bypass for 20 minutes, at the end of CPB, and at 2, 4, and 18 h after the end of CPB. Results: The plasma concentrations of endotoxin, soluble intercellular adhesion molecule 1, and urinary microalbumin began to increase at 2 h after the end of CPB, and remained higher than that of the baseline, while the concentration of tumor necrosis factor α increased only at the end CPB and at 2 h after the end of CPB. Conclusion: Cardiopulmonary bypass can induce inflammatory response, resulting in the activation or injury of vascular endothelial cells, and can increase the vascular permeability. 展开更多
