The research progress in molecular chaperones, unfolded protein response (UPR) and ER-associated degradation (ERAD) involved in the protein quality control was summarized in this paper, and then the existing probl...The research progress in molecular chaperones, unfolded protein response (UPR) and ER-associated degradation (ERAD) involved in the protein quality control was summarized in this paper, and then the existing problems and the future devel- opment prospect were also discussed. It was pointed out that the life process of protein experienced four stages including synthesizing, folding, assembling and degradation, while each stage required strict quality control. In endoplasmic reticulum (ER), a variety of proteins had been synthesized, folded and modified to form func- tional proteins with certain conformation. When the folding was blocked in ER, the unfolded proteins would aggregate and induce the UPR, which up-regulated the level of modification enzymes folded by a series of molecular chaperones and proteins to help them accomplish folding and assembling. If these proteins were still folded incorrectly, they would enter into ERAD for being degraded.展开更多

Methods: We collected secretion specimens of geni-tourinary tract from sexual partners who attended ourclinic during the period from May 1, 1999 to May 31,2000 and then detected Ct and Uu in the specimens. Results: The inconsistent rate of Ct and Uu infectedbetween sexual partners was 59.22% and 38.92%respectively. There were no correlations between pa-tients' age and the inconsistent rate of Ct and Uu in-fected (P>0.05). The most common inconsistency be-tween regular partners was seen in man with Ct infec-tion while in female with Uu infection; and in manwith concurrent infection of Ct while in female onlywith Uu; or in man with Ct infection while in female noCt and Uu were detected. Conclusion: The sexual transmitting ability of Ctand Uu is not as active as we have expected.展开更多

HINT is a two-year project part funded Geoparks researching the uses and benefits of new technologies in by the European Leader Programme. The partners are Geopark Shetland (Scotland), Chablais Geopark (France), North Pennines Area of Outstanding Natural Beauty (England) and Geopark de Hondsrug (The Netherlands). Each partner has developed a pilot project to investigate the practicalities of interpreting and communicating heritage, especially geological heritage, using a particular technology. An overview of each of the four pilot projects is given. The overall aim of the pilot projects is to develop best practice in using new technologies for heritage interpretation and to share information and ideas within the project partnership and with geoparks and heritage organizations around the world. This paper, developed by the HINT partners, aims to give an insight into the practical process of developing interpretive tools using new technologies. Some common problems are highlighted and some ways in which these can be addressed are shown.展开更多

Objective To explore triple gene transfer of dopamine synthetic enzymes with separate adeno-associated virus (AAV) vectors.Methods The genes for dopamine synthetic enzymes, tyrosine hydroxylase (TH), aromatic L-amino ...Objective To explore triple gene transfer of dopamine synthetic enzymes with separate adeno-associated virus (AAV) vectors.Methods The genes for dopamine synthetic enzymes, tyrosine hydroxylase (TH), aromatic L-amino acid decarboxylase (AADC), and GTP cyclohydrolase Ⅰ (GCH, an enzyme critical for tetrahydrobiopterin synthesis) were cotransduced into 293 cells with separate AAV vectors. Expressions of TH, AADC and GCH were detected by Western blot analysis. Intracellular dopamine level was assayed by high-performance liquid chromatography.Results TH, AADC and GCH were effectively coexpressed in transduced cells with three separate AAV vectors, AAV-TH, AAV-AADC and AAV-GCH. Furthermore, the coexpression resulted in an effectively spontaneous dopamine production in cotransduced cells.Conclusion The triple transduction of TH, AADC and GCH genes with separate AAV vectors is effective, which might be important to gene therapy for Parkinson's disease.展开更多
文摘大疱性表皮松解症(epidermolysis bullosa,EB)是一种罕见的具有临床和遗传异质性的遗传性皮肤病,以皮肤、黏膜脆性和水疱形成为特征,常由微创诱发,在欧洲EB患儿的出生率为1.9/100000[1]。EB主要分为4种类型:单纯型(EBS)、交界型(JEB)、营养不良型(DEB)和Kindler综合征(KS)。EB的临床表型具有差异性,可从轻度皮肤脆性到严重皮肤受累。目前已报道与EB发病相关的致病基因至少有20种,这些基因编码的蛋白质主要影响细胞的完整性和黏附性[2-4]。作为JEB的亚型,EB伴幽门闭锁(EB with pyloric atresia,EB-PA)是指皮肤脆性伴有先天性胃肠道闭锁的综合征,包括幽门闭锁、十二指肠闭锁和食管闭锁。
