给-受体型窄带隙聚合物是一类新型可见-近红外电致变色材料,虽然可调性强、颜色丰富,但是其电致变色性能如对比度、稳定性等需要进一步提高。通过调节聚合物中构筑单元吡咯并吡咯二酮(DPP)、苯并噻二唑(BTZ)和噻吩(T)的比例(n(DPP)∶n(B... 给-受体型窄带隙聚合物是一类新型可见-近红外电致变色材料,虽然可调性强、颜色丰富,但是其电致变色性能如对比度、稳定性等需要进一步提高。通过调节聚合物中构筑单元吡咯并吡咯二酮(DPP)、苯并噻二唑(BTZ)和噻吩(T)的比例(n(DPP)∶n(BTZ)∶n(T)分别为1∶0∶1、1.5∶0.5∶1、2∶1∶1和3∶2∶1),合成了4种新型窄带隙电致变色聚合物,研究聚合物结构和电致变色性能的关系。研究发现,这类聚合物在近红外光谱区具有较高的对比度(ΔT:50%~60%)和变色效率(CE:300~600 cm2/C),尤其是在1550 nm处,聚合物P3的ΔT高达63%、P4的CE高达471 cm2/C。相比之下,含有BTZ基团的聚合物的吸收更长、对比度更高且更稳定。这为设计给-受体型高性能电致变色聚合物提供了新的思路。 展开更多

AIM: To establish a rapid and accurate method for the detection of lamivudine-resistant mutations in hepatitis B virus and monitor of lamivudine resistance during lamivudine treatment in patients with chronic hepatitis B virus infection. METHODS: We established a real-time PCR method using a universal template and TaqMan probe to detect YMDD mutants. Variants of YVDD and YIDD were tested by individual reactions (reaction Ⅴ and reaction Ⅰ) and total hepatitis B viruses were detected in another reaction for control (reaction C). Results were determined by △Ct〈3.5 (△Ct = Ct of reaction Ⅴ or Ⅰ - Ct of reaction C). Clones of the HBV polymerase gene containing different YMDD mutations were tested. Serum samples from 163 lamivudine-treated patients with chronic hepatitis B virus infection were detected using this method and the results were confirmed by DNA sequencing. RESULTS: As many as 1000 copies per milliliter of widetype plasmid were detected and nonspecific priming was excluded. In the 163 samples from patients treated with lamivudine, lamivudine-resistant mutations were detected in 51 samples. CONCLUSION: This universal real-time PCR is a rapid and accurate method for quantification of YMDD mutants of HBV virus in lamivudine-treated patients and can be used to monitor lamivudine-resistant mutations before and during lamivudine therapy. 展开更多

We study the kinetic behavior of a two-species aggregation-migration model in which an irreversible aggregation occurs between any two clusters of the same species and a reversible migration occurs simultaneously between two different species. For a simple model with constant aggregation rates and with the migration rates and , we find that the evolution behavior of the system depends crucially on the values of the indexes υ<SUB>1</SUB> and υ<SUB>2</SUB>. The aggregate size distribution of either species obeys a conventional scaling law for most cases. Moreover, we also generalize the two-species system to the multi-species case and analyze its kinetic behavior under the symmetrical conditions. 展开更多

Objective: The novel estrogen receptor-α (ER-α) variant ER-α36 is reported to be functional in the es-trogen signaling pathway and is related to tamoxifen resistance in breast cancer. However, ER-α36 tends to be a favorable factor for survival in patients without tamoxifen therapy. To investigate the mechanisms behind this paradox, we determined the differences between the transcriptional profiles of ER-α36 and full-length ER-α (ER-α66) in breast cancers and matched normal tissues. Methods: We analyzed ER-α36 and ER-α66 messenger RNA ( mRNA) levels in 74 pairs of breast cancers and matched normal tissues using a real-time quantitative polymerase chain reaction (PCR) assay, and correlated the results with their clinicopathological characteristics. Results: Breast cancers expressed lower ER-α36 mRNA levels than matched normal tissues regardless of their ER-α66 expression status. Down-regulation of ER-α36 mRNA was correlated with local progression, lymph node metastasis, and advanced cancer stage. The level of ER-α66 mRNA was lower in ER-α negative breast cancers compared with matched normal tissues. No differences in ER-α66 mRNA levels were observed during cancer progression. Conclusion: Down-regulation of ER-α36 is associated with carcinogenesis and progression of breast cancer. 展开更多

The intestinal epithelium is the main barrier to the oral delivery of poorly water-soluble drugs. Based on the specific transporters expressed on the apical membrane of the intestinal epithelium, novel polymer micelles targeting to the organic cation transporter 2(OCTN2) were constructed by combining carnitine conjugated poly(2-ethyl-2-oxazoline)-poly(D,L-lactide)(Car-PEOz-PLA) with monomethoxy poly(ethylene glycol)-poly(D,L-lactide)(mP EG-PLA). The structure of the synthesized Car-PEOz-PLA was confirmed by -1H NMR, TLC and ammonium reineckate precipitation reaction, and the number-average molecular weight determined by GPC was 7260 g/mol with a low PDI of 1.44. Coumarin 6-loaded carnitine modified polymeric micelles prepared by film hydration method were characterized to have a nano-scaled size of about 31 nm in diameter, uniform spherical morphology, high drug loading content of 0.098%±0.03% and encapsulation efficiency of 92.67%±2.80%. Moreover, the carnitine-modified micelles exhibited the similar in vitro release behavior in SGF and SIF, and evidently enhanced intestinal absorption of poorly water-soluble agent. Therefore, the designed OCTN2-targeted micelles might have a promising potential for oral delivery of poorly water-soluble drugs. 展开更多
