A luminous bacterial strain D2 was isolated from the marine sediment samples collected near Donghai Island in Zhanjiang, China. The primary cultural and luminescent conditions of luminous bacterium D2 which was identified as Vibrio sp. were determined in liquid culture. The results showed that pH 7.0, 35 ℃, with 2.0 % NaCI, were the best growth conditions, and pH5 - 6, 20℃, OD600 0.08, with 3.0 % NaCI, were the optimal luminescent conditions.展开更多

Dynamical equation was established for the system with intelligent structure of piezoelectric...A new method was put forward to optimize the position of actuator/sensor of multi-body system with quick startup and brake. Dynamical equation was established for the system with intelligent structure of piezoelectric actuators. According to the property of the modes varying with time, the performance index function was developed based on the optimal configuration principle of energy maximal dissipation, and the relevant optimal model was obtained. According to its characteristic, a float-encoding genetic algorithm, which is efficient, simple and excellent for solving the global-optimal solution of this problem, was adopted. Taking the plane manipulator as an example, the result of numerical calculation shows that, after the actuator/sensor position being optimized, the vibration amplitude of the multi-body system is reduced by 35% compared with that without optimization.展开更多

Objective To reduce the toxicity and side effects of arsenic trioxide(ATO)and provide a new approach for the treatment of primary liver cancer,a folic acid-modified calcium arsenite liposomal“target-controlled”drug ...Objective To reduce the toxicity and side effects of arsenic trioxide(ATO)and provide a new approach for the treatment of primary liver cancer,a folic acid-modified calcium arsenite liposomal“target-controlled”drug delivery system(FA-LP-CaAs)was fabricated using the reverse microemulsion method.Methods A Malvern particle size analyzer and a transmission electron microscope were employed to determine the particle size,distribution,zeta potential and morphology of FA-LP-CaAs.Further,inductively coupled plasma emission spectrometry was employed to determine the drug loading capacity,entrapment efficiency,and in vitro release behavior of FA-LP-CaAs.To determine its toxicity in human hepatoma cells(HepG2)and human normal hepatocytes(LO2)and its effect on HepG2 cell cycle and apoptosis,the MTT method was used.Laser confocal and flow cytometry were also employed to determine the uptake of FA-LP-CaAs by cells.After establishing a mouse liver cancer model,the in vivo distribution of the drug included in the formulation was investigated using in vivo fluorescence.To evaluate the liver cancer targeting and anti-tumor effects of FALP-CaAs in vivo,the distribution of ATO in tissues and changes in tumor volume and body weight after liposomal administration were investigated using hematoxylin-eosin(HE)-stained tumor sections.Results The particle size,zeta potential and PDI of FA-LP-CaAs were(122.67±2.18)nm,(12.81±0.75)mV and 0.22±0.01,respectively,while its drug loading capacity was 18.49%±1.14%.In vitro experimental results revealed that FA-LP-CaAs had a strong killing effect on HepG2 cells.Further,the cell uptake capacity of this formulation was found to improve.Based on in vivo assessments,FA-LP-CaAs could significantly increase the distribution of ATO in tumor sites and inhibit tumor growth.Conclusions Herein,an FA-LP-CaAs formulation was successfully fabricated.This liposomal drug delivery system had a round appearance,uniform particle size,good polydispersity coefficient,evident“core-shell”structure,high drug loading capacity and pH response,tumor targeted drug delivery and sustained drug release.These findings support further research and the application of ATO as an anti-liver cancer prodrug and provide a new method for the treatment of liver cancer.展开更多

METHODS: The HBV-based vectors with green fluorescence protein (GFP) were packaged into the liver of immunodeficient mice through transfer and helper plasmid using hydrodynamic technology. Wild type HBV (wt HBV) was provided by plasmid MC2009. Primary human hepatocytes (PHH) were isolated and infected with recombinant HBV (rHBV) or wt HBV. GFP expression was monitored by confocal and flow cytometry. HBV DNA and HBV surface antigen (HBSAg) were analyzed by PCR and ELISA. RESULTS: 3 × 107 wt HBV copies/mL and 5 × 106 rHBV copies/mL were collected from mice serum. In the wt HBV infected group, HBV progeny was 2 × 107 copies/mL and HBSAg was 770 ng/mL. In the rHBV infected group, GFP fluorescence was detected on d 3 post-infection and over 85% of the parenchymal cells expressed green fluorescence on d 12 post-infection. Compared with wt HBV in the PHH infection system, no rHBV DNA or HBSAg were detected in PHH culture media. CONCLUSION: An effective HBV based vector was developed, which proved to be a useful HBV infection system. This vector and infection system can be applied to develop a therapeutic vector and study the HBV life cycle and viral pathogenesis.展开更多

No robust system has been obtained by empiric approaches, and HCV cell culture rem...Since the discovery of HCV in 1989, the lack of a cell culture system has hampered research progress on this important human pathogen. No robust system has been obtained by empiric approaches, and HCV cell culture remained hypothetical until 2005. The construction of functional molecular clones has served as a starting point to reconstitute a consensus infectious cDNA that was able to transcribe infectious HCV RNAs as shown by intrahepatic inoculation in a chimpanzee. Other consen- sus clones have been selected and established in a hu- man hepatoma cell line as replicons, i.e. self-replicating subgenomic or genomic viral RNAs. However, these repli- cons did not support production of infectious virus. Inter- estingly, some full-length replicons could be established without adaptive mutations and one of them was able to replicate at very high levels and to release virus particles that are infectious in cell culture and in vivo. This new cell culture system represents a major breakthrough in the HCV field and should enable a broad range of basic and applied studies to be achieved.展开更多
