In Silico Screening of Potential Drug with Antileishmanial Activty and Validation of their Activity by in Vitro and in Vivo Studies

The research on discovery and development of new treatments for cutaneous leishmaniasis has been declared as priority. Using bioinformatics approaches, this study aimed to identify antileishmanial activity in drugs that are currently used as anti-inflammatory and wound healing by such anti-Leishmania activity was validated by in vitro and in vivo assays. In silico analysis identified 153 compounds from which 87 were selected by data mining of DrugBank database, 22 and 44 were detected by PASS (http://pass.cribi.unipd.it) and BLAST (http://blast.ncbi.nlm.nih. gov/) alignment, respectively. The majority of identified drugs are used as skin protector, anti-acne, anti-ulcerative (wound healer) or anti-inflammatory and few of them had specific antileishmanial activity. The efficacy as antileishmanial was validated in vitro in 12/23 tested compounds and in all seven compounds that were evaluated in in vivo assays. Notably, this is the first report of antileishmanial activity for adapalene. In conclusion, bioinformatics tools not only can help to reduce time and cost of the drug discovery process but also may increase the chance that candidates identified in silico which have a validated antileishmanial activity by combining different biological properties. 展开更多

For athletes, a pair of good shoes is vital for comfort and may add to performance improvement. The main objective of this study is to study the performance of some commonly used shoes in the takraw games by using experimental tests and thermal measurements. The momentum and kinetic energies that were generated by the takraw shoes were also measured. The methods used in this study included free fall test, impact force analysis and surface thermal assessment. From the outcomes of the experiments, it was found that the prototype FA shoe was better in absorption of momentum and impact force than the other shoes and also with lower surface temperature after the trial game. These outcomes suggested that it was more suitable to be worn by the takraw players for better performance. All the information obtained from this study are particularly useful for improvement on the current design oftakraw shoe. 展开更多

In order to get a high flowrate gas-gas injector and its design methodology, the combustion flow field of a typical shear-coaxial injector was analyzed firstly. The dimensional analysis was applied in the phenomenon of gas-gas combustion, and design parameter optimization and a structure improvement were also carded out. A high flowrate single-element injector with high 1-12/O2 momentum ratio and tapered 02 post tip was obtained and validated by both numerical and experimental studies. This high flowrate injector has simple construction and it can effectively enhance the mixing, decrease the combustion completion length and also has a benign heat environment. Furthermore, based on the study of the single-element injector, numerical optimization and validation experiments were conducted successively on a multi-element injector equipped with high flowrate injection elements. The multi-element injector with the high flowrate elements started up and shut down smoothly, and operated steadily without any stability aids. In the combustor designed with nominal parameters, this injection element can obtain high combustion efficiency with a flowrate of 3.7 times that of SSME main injector element and shows a benign chamber wall heat compatibility. This injector design and the design methodology can become a reference for the design of other types of injectors for liquid rocket engines. 展开更多
