It aims to use genome-wide markers to estimate the effect...With marker and phenotype information from observed populations, genomic selection (GS) can be used to establish associations between markers and phenotypes. It aims to use genome-wide markers to estimate the effects of all loci and thereby predict the genetic values of untested populations, so as to achieve more comprehensive and reliable selection and to accelerate genetic progress in crop breeding. GS models usually face the problem that the number of markers is much higher than the number of phenotypic observations. To overcome this issue and improve prediction accuracy, many models and algorithms, including GBLUP, Bayes, and machine learning have been employed for GS. As hot issues in GS research, the estimation of non-additive genetic effects and the combined analysis of multiple traits or multiple environments are also important for improving the accuracy of prediction. In recent years, crop breeding has taken advantage of the development of GS. The principles and characteristics of current popular GS methods and research progress in hese methods for crop improvement are reviewed in this paper.展开更多
Polyploidy is common among agriculturally important crops. Popular genetic methods and their implementations cannot always be applied to polyploid genetic data. We give an overview about available tools and their limi...Polyploidy is common among agriculturally important crops. Popular genetic methods and their implementations cannot always be applied to polyploid genetic data. We give an overview about available tools and their limitations in terms of levels of ploidy, auto- and allo-ploidy. The main classes of tools are genotype calling, linkage mapping and haplotyping. The usability of the tools is discussed with a focus on their applicability to data sets produced by state of the art technologies. We show that many challenges remain until the toolset for polyploidy provides similar functionalities as those which are already available for diploids. Some tools have been developed over a decade ago and are now outdated. In addition, we discuss necessary steps to overcome this shortage in the future.展开更多

Methods: The human colorectal carcinoma cell line LoVo and HR-8348 w...Objective: The aim of the study was to observe the effects of Iobaplatin and oxaliplatin on biological behavior of colorectal carcinoma cell line. Methods: The human colorectal carcinoma cell line LoVo and HR-8348 were used in the present study, and different concentrations of Iobaplatin and oxaliplatin served as inhibitors. The invasion and metastasis potential were evaluated with Transwell chamber, and cell inhibition ratio was measured with MTT assay. Results: Lobaplatin and oxaliplatin could significantly decrease the abilities of invasion and metastasis of LoVo and HR-8348 as compared with the control group (P 〈 0.01), but no difference between Iobaplatin and oxaliplatin groups was observed (P 〉 0.05). No difference was found in the cell inhibition ratio between the Iobaplatin and oxaliplatin groups (P 〉 0.05). Lobaplatin could inhibit colorectal carcinoma cells that oxaliplatin couldn't do. Conclusion: Lobaplatin is a new antitumor platinum drug and can inhibit colorectal carcinoma cells. Moreover, Iobaplatin could also inhibit colorectal carcinoma cells which are resistant to oxaliplatin.展开更多
Soil microbiological and biochemical properties under various field crop rotations such as grains, pastures and vegetables have been studied intensively under short-term period. However, there is limited information a...Soil microbiological and biochemical properties under various field crop rotations such as grains, pastures and vegetables have been studied intensively under short-term period. However, there is limited information about the influence of banana-based rotations on soil organic C, total N(TN), microbial biomasses and enzyme activities under long-term crop rotations. A field experiment arranged in a randomized complete block design with three replicates was carried out at the Wanzhong Farm in Ledong(18?37′–18?38′N, 108?46′–108?48′E), Hainan Province, China, to compare the responses of these soil parameters to long-term(10-year) banana(Musa paradisiaca)-pineapple(Ananas) rotation(AB), banana-papaya(Carica) rotation(BB) and banana monoculture(CK) in a conventional tillage system in the Hainan Island. Soil p H, total organic C(TOC), dissolved organic C(DOC), TN, total P(TP) and available P(AP) were found to be significantly higher(P < 0.01) in AB and BB than CK at 0–30 cm soil depth. Microbial biomass C(MBC) and N(MBN) were observed 18.0%–35.2% higher in AB and 8.6%–40.5% higher in BB than CK at 0–30 cm. The activities of urease(UA), invertase(IA), dehydrogenase(DA) and acid phosphatase(APA) showed a mean of 21.5%–59.6% increase in AB and 26.7%–66.1% increase in BB compared with CK at 0–30 cm. Higher p H, TOC and DOC at 0–10 and 10–20 cm than at 20–30 cm were obtained despite of the rotations. Soil MBC and MBN and activities of UA, IA and DA decreased markedly(P < 0.01) with increasing soil depth in the different rotation soils as well as the monoculture soil. Ye...The Hedgehog (Hh) signaling pathway plays important roles in both embryonic development and adult tissue homeostasis. Such biological functions are mediated by the transcription factor Cubitus interruptus (Ci). Yet the transcriptional regulation of the effector Ci itself is poorly investigated. Through an RNAi-based genetic screen, we identified that female sterile (1) homeotic (Fsh), a transcription co-activator, directly activates Ci transcription. Biochemistry assays demonstrated physical interactions among Fsh, Sex combs extra (Sce), and Polycomb (Pc). Functional assays further showed that both Pc and Sce are required for Ci expression, which is not likely mediated by the derepression of Engraited (En), a repressor of Ci, in Pc or Sce mutant cells. Finally, we provide evidence showing that Pc/Sce facilitates the binding of Fsh at Ci locus and that the physical interaction between Fsh and Pc is essential for Fsh-mediated Ci transcription. Taken together, we not only uncover that Ci is transcriptionally regulated by Fsh-Pc-Sce complex but also provide evidence for the coordination between Fsh and PcG proteins in transcriptional regulation.展开更多
A series of new benzimidazole derivatives was synthesized and characterized by IR,1H NMR,13C NMR,MS,and HRMS spectra.All the new compounds were screened for their antimicrobial activities in vitro by a twofold serial ...A series of new benzimidazole derivatives was synthesized and characterized by IR,1H NMR,13C NMR,MS,and HRMS spectra.All the new compounds were screened for their antimicrobial activities in vitro by a twofold serial dilution technique.The bioactive evaluation showed that 3,5-bis(trifluoromethyl)phenyl benzimidazoles were comparably or even more strongly antibacterial and antifungal than the reference drugs Chloromycin,Norfloxacin,and Fluconazole.The combination of2,4-difluorobenzyl benzimidazole derivative 5l and its hydrochloride 7 respectively with the antibacterials Chloromycin,Norfloxacin,and the antifungal Fluconazole was more sensitive to methicillin-resistant MRSA and Fluconazole-insensitive A.flavus.In addition,the interaction of compound 5l with calf thymus DNA demonstrated that this compound could effectively intercalate into DNA to form a compound 5l-DNA complex that might block DNA replication and thereby exert good antimicrobial activity.展开更多

Recently, biochemists have discovered that multiple conformations and numerous parallel paths are involved during the processes catalyzed by enzymes. It is plausible that the entire macromolecular scaffold is involved in catalysis via cooperative motions that result in incredible catalytic efficiency. Moreover, some enzymes can very strongly bind the transition state with an association constant of up to 1024 M-1, suggesting that covalent bond formation is a possible process during the conversion of the transition state in enzyme catalysis, in addition to the concatenation of noncovalent interactions. Supramolecular chemistry provides fundamental knowledge about the relationships between the dynamic structures and functions of organized molecules. By tak-ing advantage of supramolecular concepts, numerous supramolecular enzyme mimics with complex and hierarchical structures have been designed and investigated. Through the study of supramolecular enzyme models, a great deal of information to aid our understanding of the mechanism of catalysis by natural enzymes has been acquired. With the development of supramolec-ular artificial enzymes, it is possible to replicate the features of natural enzymes with regards to their constitutional complexity and cooperative motions, and eventually decipher the conformation-based catalytic mystery of natural enzymes.展开更多
