Methods To assess the antinociceptive effects of TTX, ASA or TTX plus ASA, the acetic acid-induced abdominal constriction test and formalin pain test were used. Results TTX (0.5 - 4.0 μg· kg^-1 ) or ASA (25 - 200 mg· kg^-1 ) im produced a significant inhibition of acetic acid-induced abdominal constriction. The median inhibitory doses (ID508) were 2.1 μg· kg^-1 for TTX( and 64 mg· kg^-1 for ASA. TTX and ASA also showed a dose-dependent inhibition of the second phase response in the formalin pain model, the ID508, being 2.3μg·kg^-1 and 74.2 mg· kg^-1, respectively. The ihteraction between TTX and ASA was synergistic, as evidenced by the fact that (1) when ASA alone compared with the combination of TTX (0.79 μg · kg^-1 or 0.39μg· kg^-1 ) and ASA, the ID508, of ASA reduced from 64.0 mg· kg^-1 to 5.8 mg· kg^-1 or 12.6 mg· kg^-1, and from 74.2 mg· kg^-1 to 7.4 mg· kg^-1 or 13.0 mg· kg^-1 on tile two models of nociceptive tests, respectively; and that (2) synergism in the analgesic effects was shown by isobiolographic analysis. Conclusion TTX, ASA and the combination of the two drags produce analgesic effects in acetic acid-induced abdominal constriction test and formalin-induced pain test. The interactions between TTX and ASA may be useful in developing novel analgesic agents.展开更多

Opiate dependence involves physical and psychical dependence. Although many addicts can bedetoxified, the relapse ratio of 95% in 5 a demonstrates that opiate psychical dependence is aproblem more troublesome. It has been reported that acute and chronic administration of L- NNA canmarkedly retard the development of tolerance to physical dependence on morphine, and suppress theabstinence syndromes precipitated by naloxone in opiate dependent rodents, and even reverse theexisting morphine tolerance. However, the effect of L-NNA on the positive reinforcement ofpsychically active substances and its possible mechanism have not yet been reported. In presentstudy, the effect of L- NNA en the psychical dependence induced by opiates was evaluated on thebasis of conditioned place preference.展开更多

METHODS: A reporter gene system in HT-29 cells was used to measure levels of NF-KB activation after challenge with bifidobacteria or after bacterial pre-treatment following LPS challenge. IL-8 protein and pro-inflammatory gene expression was investigated using normal HT-29 cells. RESULTS: None of the bifidobacteria tested induced activation of nuclear factor κB (NF-κB) indicating that bifidobacteria themselves do not induce inflammatory events in IECs. However, six out of eight bifidobacteria tested inhibited lipopolysaccharide- (LPS-) induced NF-κB activation in a dose- and strain-dependent manner. In contrast, NF-κB activation in response to challenge with tumor necrosis factor-α (TNF-α) was affected by none of the tested bifidobacteria, indicating that the inhibitory effect of bifidobacteria is specific for LPS-induced inflammation in IECs. As shown with two of the six inhibitionpositive bifidobacteria, LPS-induced inhibition of NF-κB activation was accompanied by a dose-dependent decrease of interleukin 8 (IL-8) secretion and by lower mRNA levels for IL-8, TNF-α, cyclooxygenase 2 (Cox-2), and intercellular adhesion molecule 1(ICAM-1). CONCLUSION: Some strains of bifidobacteria are effective in inhibiting LPS-induced inflammation and thus might be appropriate candidates for probiotic intervention in chronic intestinal inflammation.展开更多

In group C, SMA was separated without occlusion. Plasma D-lactate levels were examined and histological changes were observed under a light microscope. The activity of p38 MAPK was determined by Western immunoblotting and apoptotic cells were detected by the terminal deoxynucleotidyl transferase (TdT)-mediated dUDP-biotin nick end labeling (TUNEL).RESULTS: Intestinal ischemia followed by reperfusion activated p38 MAPK, and the maximal level of activation (7.3-fold vs sham-operated group) was reached 30 min after I/R. Treatment with SB 203580, a p38 MAPK inhibitor, reduced intestinal apoptosis (26.72±3.39% vs62.50±3.08%in I/R vehicle, P<0.01) and decreased plasma D-lactate level (0.78±0.15 mmol/L in I/R vehicle vs0.42±0.17 mmol/L in SB-treated group) and improved post-ischemic intestinal histological damage.CONCLUSION: p38 MAPK plays a crucial role in the signal transduction pathway mediating post-ischemic intestinal apoptosis, and inhibition of p38 MAPK may attenuate ischemia-reperfusion injury.展开更多

The results showed that Cd^(2+) retarded microbial reduction of γ-Fe OOH and avoided formation of magnetite. The inhibitory effect on γ-Fe OOH transformation may not result from Cd^(2+) toxicity to the bacterium; it rather was probably due to competitive adsorption between Cd^(2+) and Fe^(2+) on γ-Fe OOH as its surface reduction catalyzed by adsorbed Fe^(2+) was eliminated by adsorption of Cd^(2+).展开更多

C. curvisetus was cultured with different amounts of dried G. lemaneiformis under controlled laboratory conditions. We measured the photosynthetic oxygen evolution rate and established the chlorophyll a fluorescence transient (OJIP) curve coupled with its specific parameters. We observed concentration-dependent and time-dependent relationships between dried G. lemaneiformis and inhibition of photosynthesis in C. curvisetus. Co-culture with dried G. lemaneiformis also resulted in a decrease in the light-saturated maximum photosynthetic oxygen evolution rate (P^ax) in C. curvisetus, and a decrease in the OJIP curve along with its specific parameters; the maximum photochemical efficiency of PSII (FJFm), the amount of active PSII reaction centers per excited cross section at t=0 and t=--tFM (RC/CS0 and RC/CSm, respectively), the absorption flux per excited cross section at t=0 (ABS/ CS0), and the efficiency with which a trapped exciton moves an electron into the electron transport chain (~u0). The dark respiration rate (Rd) increased in C. curvisetus co-cultured with dried G. lemaneiformis. The JIP-test and the oxygen evolution results indicated that dried G. lemaneiJbrmis decreased the number of active reaction centers, blocked the electron transport chain, and damaged the oxygen-evolving complex of C. curvisetus. This result indicated that dried fragments of G. lemaneiformis could effectively inhibit photosynthesis of C. curvisetus, and thus, could serve as a functional product to control and mitigate C. curvisetus blooms.展开更多

It has been found that the inclusion interaction between SDS and β-CD is much greater than the hydrophobic interaction between SDS and MC. As a result, in the coexistence of SDS and β-CD, the sol-gel transition temperature of MC solution keeps the same value, independent of the concentration of SDS in solution on con- dition that the concentration of SDS is less than β-CD. Our experimental results not only suggest that the effect of SDS upon the sol-gel transition of MC solution can be screened by β-CD completely but also indicate the inclusion ratio of SDS to β-CD can be determined quantitatively by using rheological measurement. The inclusion ratio of SDS to β-CD is 1:1, which is in good agreement with the inclusion ratio of SDS to β-CD in the presence of poly(vincyl pyrrolidone) determined by the viscosity measurement but is critically different from the inclusion ratio of SDS to β-CD in the presence of the oppositely charged polyelec-trolyte by using the rheological measurement, mainly due to the reason that the mechanism of the interaction between SDS and MC is critically different from the mechanism of the interaction between SDS and the oppositely charged polyelectrolyte.展开更多

Also, following administration of gossypol (12.5 mg/kg/day) for six weeks, the rate of fertilization in vitro by hamster sperm was significantly decreased. In the gossypol-treated group, extracts of testis sperm delayed dispersion of cumulus cells, suggesting inhibition of hyaluronidase and other acrosomal enzymes. Furthermore, the acrosin and arylsulfatase activities were shown to be markedly inhibited. Thus, a parallelism was displayed between the reduction of fertility and the decreasc in acrosin and arylsulfatase activities in epididymis sperms.Besides, the inhibition was reversible and was dosage-and durationdependent. In conclusion, the assay of acrosin activity might serve as a useful tool for monitoring the irreversible sterility induced by gossypol,展开更多

The inclusion ratio of SDS to β-CD is 1:1, which is in good agreement with the inclusion ratio of SDS to β-CD in the presence of poly(vincyl pyrrolidone) determined by the viscosity measurement but is critically different from the inclusion ratio of SDS to β-CD in the presence of the oppositely charged polyelec-trolyte by using the rheological measurement, mainly due to the reason that the mechanism of the interaction between SDS and MC is critically different from the mechanism of the interaction between SDS and the oppositely charged polyelectrolyte.

Methods: Germany Bipolar controlled hypothermia radio-frequency with varying tips was used for obstructive sleep apnea-hyponea syndrome, chronic hypertrophic rhinitis, allergic rhinitis, aural pseudocyst, phymatoid proliferation of tongue root, capillary hemangioma of nose and pharynx. Follow up was made up to six months. Results:All patients had curative effects. Conclusion: Hypothermia radio-frequency ablation technique was used effectively for many diseases in otolaryngology with features of accurate ablation, low complication, reliable effects as well as easyusing. It was a prospective method of micro invasive surgery.
文摘To investigate into the mechanisms underlying the irreversible sterility induced by gossypol, we studied the relationship between its inhibitory action on acrosomal enzymes and its antifertility effect. As shown by our result, after exposure to gossypol (l.25-60 μg/ml) for 15 min. in vitro, the sperms' ability to penetrate bovine cervical mucus and the fertility rate were significantly reduced. In conclusion, the assay of acrosin activity might serve as a useful tool for monitoring the irreversible sterility induced by gossypol,

文摘Glucocorticoid(GC) plays an important role in anti-inflammatory,anti-allergic effects and immunosuppression, and has become a widely used drug in clinical departments. However,GC also produces a number of serious side effects at the same time. After GC acting on human body, the syndrome change has some regular pattern and it can be treated on the basis of syndrome differentiation and stage to aim at further improving therapeutic efficacy. The Chinese medicine can reduce the side effects of GC when treating the primary disease, thus plays a role in Synergism and Detoxification.

文摘Using blind dock method,we find that thioflavin-T(ThT) can bind to both monomers and fibrils of the full-length β-amyloid peptide(Aβ1-42) and has a higher binding affinity to the fibrils. It is shown that the hydrophobic interaction between the ligand(ThT) and substrate(Aβ1-42) are stronger than hydrogen bonds.Furthermore,ThT tends to be located near the C-terminus of Aβ monomer through hydrophobic and electrostatic interactions, while it tends to contact the residues Met35 and Gly27 of the fibril surface mainly through hydrophobic interaction. Finally, according to the docking results and ThT fluorescence assay,a kinetic equation is proposed to deduce the aggregation rate coefficient of Aβ1-42.
