CT、MRI对卵巢癌定性、分期、侵袭及转移的诊断敏感性研究

目的研究CT、磁共振成像(MRI)对卵巢癌定性、分期、侵袭及转移的诊断敏感性。方法选取80例经病理诊断为卵巢癌患者,分别开展CT、MRI检查,对比两种方式用于卵巢癌定性、分期、侵袭及转移诊断的敏感性。结果MRI在卵巢癌定性诊断中的准确率... 目的研究CT、磁共振成像(MRI)对卵巢癌定性、分期、侵袭及转移的诊断敏感性。方法选取80例经病理诊断为卵巢癌患者,分别开展CT、MRI检查,对比两种方式用于卵巢癌定性、分期、侵袭及转移诊断的敏感性。结果MRI在卵巢癌定性诊断中的准确率97.50%明显高于CT 87.50%,差异有统计学意义(P<0.05)。MRI在卵巢癌分期诊断中的准确率明显高于CT,差异有统计学意义(P<0.05)。MRI肠道、子宫、输卵管、肝脏等远处、淋巴结侵袭及转移诊断敏感性明显高于CT,但诊断腹膜、网膜侵袭及转移诊断敏感性则低于CT,差异有统计学意义(P<0.05)。结论相比CT,MRI用于卵巢癌定性、分期、侵袭及转移诊断中准确率、敏感性均明显更优,若进一步提升疾病诊断准确率,可将两种方式联合应用。 展开更多

目的探究卡铂联合杨梅素对人卵巢癌HO-8910 PM细胞侵袭、转移及丝裂原活化蛋白激酶(mitogen activated protein kinase,MAPK)/细胞外信号调节激酶(extracellular signal-regulated protein kinase,ERK)通路的影响。方法体外培养人卵巢癌... 目的探究卡铂联合杨梅素对人卵巢癌HO-8910 PM细胞侵袭、转移及丝裂原活化蛋白激酶(mitogen activated protein kinase,MAPK)/细胞外信号调节激酶(extracellular signal-regulated protein kinase,ERK)通路的影响。方法体外培养人卵巢癌HO-8910 PM细胞,以不进行处理的细胞作为正常组,分别用5、20、50μmol/L卡铂,20、40、60 mg/L杨梅素以及两者联合处理,MTT法筛选联合处理最佳浓度,Transwell侵袭实验检测HO-8910 PM细胞侵袭能力变化情况,划痕实验检测细胞转移能力,RT-PCR检测HO-8910 PM细胞伤害性信息与即刻早期基因c-fos、细胞增殖相关基因反式作用因子激活蛋白c-Jun、基质金属蛋白酶(matrix metallopeptidase 9,MMP-9)表达,Western blot检测细胞外调节蛋白激酶(ERK 1/2)、p-ERK 1/2、cfos、c-jun、活化蛋白转录因子-1(activator protein-1,AP-1)、MMP-9蛋白的表达。结果卡铂、杨梅素单独处理时,细胞抑制率呈剂量依赖性升高,卡铂20μmol/L联合杨梅素60 mg/L处理细胞时(联合组),细胞抑制率最高为(84.69±14.19)%(P <0.05)。与正常组、卡铂20μmol/L、杨梅素60 mg/L组相比,联合组穿膜细胞数量显著降低,迁移抑制率显著升高(P <0.05)。RT-PCR检测结果显示联合组c-fos、c-jun、MMP-9mRNA表达量均显著低于正常组、卡铂20μmol/L、杨梅素60 mg/L处理组(P <0.05)。联合组p-ERK 1/2、c-fos、c-jun、AP-1、MMP-9蛋白表达量均显著低于正常组、卡铂20μmol/L、杨梅素60 mg/L处理组(P <0.05)。结论卡铂联合杨梅素可明显抑制人卵巢癌HO-8910 PM细胞侵袭、转移,其机制可能与抑制MAPK/ERK通路有关。 展开更多

Objective: To investigate the relationship between matrix metalloproteinases-9 (MMP-9) and sialyl Lewis X (CD15s) antigen in invasion and metastasis of gastric carcinoma. Methods: Expression of CD105, MMP-9 and CD15s in 47 cases of gastric carcinoma undergone radical surgery were evaluated by SP immunohistochemical staining using the respective monoclonal antibody. The microvessel density (MVD) marked with CD105 was detected. Correlation between MVD and MMP-9, CD15s was also statistically analyzed. Results: The MVD in both MMP-9 and CD15s positive expression group was higher significantly than that in MMP-9 or CD15s positive expression alone group, and it was also significantly higher than that in both MMP-9 and CD15s negative expression group. Conclusion: MMP-9 is a marker of invasion and CD15s is a marker of metastasis in gastric carcinoma. Combining detection of MMP-9 and CD15s has certain clinical significance for diagnosis, treatment and assessing the prognosis for gastric caner. 展开更多

Objective To explore the role and regulation of guanine nucleotide-binding protein G(i), a-1 subunit (GNAI1) in hepatocellular carcinoma (HCC). Methods Expression of GNAI1 in HCC samples was determined by qRT-PCR and immunohistochemical (IHC) staining. Huh-7 and SNU-387 cells stably expressing GNAI1 were established by the infection of lentivirus transducing unit containing GNAI1. siRNA against GNAI1 was transfected into SMMC-7721 cells to knock down the GNAI1 expression in HCC cells. Mir-320a/c/d mimics were transfected into SMMC-7721 and SK-Hep-1 cells and the expression of GNAll was determined by Western blot. The migration and invasion of Huh-7, SNU-387, SK-Hep-1 and SMMC-7721 cells were investigated by Transwell assays. Results The GNAI1 protein was significantly downregulated in HCC samples without changes in its mRNA levels. GNAI1 could inhibit the migration and invasion of HCC cells in vitro. Further investigations indicated that GNAI1 was a target of miR-320a/c/d in HCC cells. Transwell assays demonstrated that these microRNAs could promote the migratory ability and invasivesess of HCC cells in vitro. Conclusions GNAII is downregulated in HCC and inhibits the migration and invasion of HCC cells. This study is the first to investigate the role of GNAI1 in cancer. Regulation of GNAI1 by miR-320a/c/d indicates new therapeutic avenues for targeting HCC metastasis. 展开更多

Objective:To detect the expression of RECK gene in the highly and low metastatic cell sublines of human osteosarcoma cell HOS and explore its possible roles on the occurrence and metastasis of osteosarcoma.Methods:RT-PCR, gelatin zymography, and matrigel invasion assay were respectively used to evaluate the endogenous expression of RECK mRNA, MMP-2 activation ratio and invasive capacity in the two osteosarcoma cell sublines.Results:The highly metastatic cell group expressed significantly lower mRNA level of RECK than the low metastatic cell group(P < 0.05), but showed higher MMP-2 activation ratio and invasive capacity(P < 0.05 and P < 0.01, respectively).Conclusion:The abnormal low expression of RECK may participate in osteosarcoma invasion and metastasis, and may be a new therapeutic target for osteosarcoma. 展开更多

Investigations on the relationship between some tumor cell properties and their metastatic potential using different clones from two tumor cell lines are reported. CNE2L2(highly metastatic) and CNE2L4 (low metastatic) clones were isolated from human nasopharyngeal carcinoma CNE-2Z cell line. LA1. LAD and LA5 clones were isolated from mouse pulmonary adenocarcinoma LA795 cell line with different metastatic potentials. The results from both cell lines showed that the metastatic potential of tumor cell positively correlated with elecirophoretic mobility rate. adhesion with endothelial cells and in vitro and in vivo invasiveness. and negatively correlated with self-adhesion and adhesion to fibroblast cells. The data further confirmed the relationship between these factors and the metastatic potential of tumor cells.The results provide a theoretical basis for elucidating the mechanism of metastasis. 展开更多
