Methods: ICCs were collected from the small intestines of miceand analyzed using an anti-c-Kit antibody. ICCs were divided into five groups: the blank group, the rapamycin group (anautophagy inducer), the 5% (rapamycin + 5% JQD-CS), the 20% (rapamycin + 20% JQD-CS) JQD-CS groups, and the3-Methyladenine (3-MA) group (rapamycin + 3-MA; positive control). Transmission electron microscopy was used toobserve the ultrastructure of ICCs. Western blotting was used to detect the expression of microtubule-associated protein1 light chain 3 (LC3-II), Beclin-1, phosphatidylinositol 3-kinase (PI3K), p-PI3K, protein kinase B (AKt), p-AKt,mammalian target of rapamycin (mTOR), and p-mTOR. Ca2+ current was examined by patch-clamp experiments. Cellcycle was detected by flow cytometry. Results: Unlike in the rapamycin group, the ICC structures were more integratedand a lower number of autophagic vacuoles were observed in the 20% JQD-CS group. Moreover, the expression ofLC3-II and Beclin-1 decreased, the expression of c-Kit, p-PI3K, p-AKt increased, the maximum current density valuedecreased, and the number of cells in the G1 phase increased while those in the G2/M phase decreased, with the additionof 20% JQD-CS. Conclusion: JQD-CS can antagonize rapamycin-induced autophagy in ICCs in vitro by promoting thephosphorylation of PI3K/AKt pathway, inhibiting Ca2 + inflow, and regulating the cell cycle.展开更多

Sixteen 60 day-old Germany Shepherd puppies[weight of (5.86±0.22) kg] were selected and randomly divided into 2 groups, one group fed basal diet was the control group, another group fed basal diet and supplemented with 1.2% glutamine was the experimental group. Trial period was 21 days. GPT and AKP activity were detected on 7th, 14th and 21 st day respectively. Dietary sup- plementation with 1.2% glutamine enhanced (P〈0.05) its activity of serum GPT and AKP compared with the control group. This experiment proved that glutamine could improve these two kinds of enzyme activity in serum, promotes protein synthesis, raise puppies anti-stress effect.展开更多

Moreover dependence between apoptosis and HBV-DNA or HCV-RNA levels was studied. METHODS:Eighty-six persons were included into study:34 healthy HBsAg carders,33 patients with chronic HBV infecl^on and 19 patients with chronic HCV infection.Serum levels of sFas/sFasL were measured by ELISA assay.HBV-DNA and HCV-RNA were measured by RT-PCR assay.Levels of sFas/sFasL were determined before and 2 and 12 wk after therapy in patients with chronic hepatitis B and C infection. HBV-DNA or HCV-RNA was detected before treatment and 6 mo after treatment. RESULTS:Twenty-four (71%) healthy HBsAg carders showed HBV-DNA over 10~5/mL,which was comparable to the patients with chronic hepatitis B.independently from HBV-DNA levels, the concentration of sFas among healthy HBsAg carders was comparable to healthy persons.Among patients with chronic hepatitis B and C,the concentration of sFas was significantly higher in comparison to healthy HBsAg carriers and healthy persons.In chronic hepatitis B patients the concentration of sFas was decreased during lamivudine treatment.Among chronic hepatitis C patients the concentration of sFas was increased during IFN alpha and ribavirin treatment,sFasL was not detected in control group.Furbhermore sFasL occurred more frequently in chronic hepatitis C patients in comparison to chronic hepatitis B patients. CONCLUSION:There are no correlations between apoptosis and HBV-DNA levels.However ther is an association between apoptosis and activity of inflammation in patients with chronic HBV infection.Apoptosis can be increased in patients with chronic hepatitis C by effective treatment which may be a result of apoptosis stimulation by IFN-α.展开更多
In this study, the effects of fermented whey (FW) in treating bacillary dysentery caused by Shigellaflexneri in albino rats and on the gastrointestinal (GIT) flora of apparently healthy albino rats (AHARs) were ...In this study, the effects of fermented whey (FW) in treating bacillary dysentery caused by Shigellaflexneri in albino rats and on the gastrointestinal (GIT) flora of apparently healthy albino rats (AHARs) were investigated. Prior the therapeutic assay, the growth inhibitory activity (GIA) of whey subjected to different fermentation durations at 30 ~ 2 ~C was first investigated using agar diffusion assay on the test organism, conventional antibiotics served as control. After this, the infectious dose of the organism was determined and used to infect another set of AHARs. The infected rats were grouped into two; group one was treated with 1.0 mL of the FW that exerted the highest GIA in the in vitro assay (FW1), once daily for 7 d while group two was left untreated. The rats were observed for signs of recovery while their large intestine was subjected to histopathological examinations. For the effects of whey on GIT flora of AHARs, another group of AHARs was fed with FW1 for 3months. At 7 d intervals, their faeces were examined for microbial types and load. The in vitro GIA of the FWs on the test organism was superior to that of most of the antibiotics used and the administration of FW1 to infected rats caused them to recover by 72 h while those not treated with FW1 started to recover by 168 h. FWl did not significantly (p 〈 0.05) affect the GIT microflora loads but only the types.展开更多
