Objectives To identify the 5'untranslated region of Zika virus(ZIKV 5'UTR)RNA-binding proteins and to investigate the impact of the binding protein on the activity of internal ribosomal entry site(IRES)located...Objectives To identify the 5'untranslated region of Zika virus(ZIKV 5'UTR)RNA-binding proteins and to investigate the impact of the binding protein on the activity of internal ribosomal entry site(IRES)located in ZIKV 5'UTR and virus production.Methods Interacting proteins in U251 cells were captured using tRSA-tagged ZIKV 5'UTR RNA and tRSA-ZIKV 5'UTR RNA-binding proteins were visualized by SDS-PAGE silver staining,Subsequently,liquid chromatographytandem mass spectrometry(LC-MS/MS),bioinformatics analysis,and Western blot were used to identify the candidate proteins binding to ZIKV 5'UTR.Dicistronic expression assay and plaque forming assay were performed to analyze the effect of the binding protein on ZIKV IRES activity and ZIKV production,respecitvely.Results tRSA RNA pull-down assay,LC-MS/MS,and Western blot analysis showed that polypyrimidine tractbinding protein(PTB)bound to the ZIKV 5'UTR.Furthermore,dual luciferase reporter assay revealed that overexpression of PTB significantly enhanced the IRES activity of ZIKV(t=10.220,P<0.001),while PTB knockdown had the opposite effect(t=4.897,P<0.01).Additionally,virus plaque forming assay demonstrated that up-regulation of PTB expression significantly enhanced viral titer(t=6.400,P<0.01),whereas reducing PTB expression level weakened virus infectivity(t=5.055,P<0.01).Conclusion PTB positively interacts with the ZIKV 5'UTR and enhances IRES activity and virus production.展开更多

Methods The expression profile of microRNA-21 in human cholangiocarcinoma tissues and cholan- giocarcinoma cell line, QBC939, was evaluated by using real-time PCR analysis. The bona fide targets of microRNA-21 were analyzed and confirmed by dual luciferase reporter gene assay and western blot, respec- tively. The expressional correlation of microRNA-21 and its targets was probed in human cholangiocarci- noma tissues by using real-time PCR, locked nucleic acid in situ hybridization (LNA-ISH), and immunohis- tochemistry analysis. Results Real-time PCR analysis revealed that microRNA-21 expression depicted a significant up-regulation in human cholangiocarcinoma tissues about 5.6-fold as compared to the matched normal bileduct tissues (P〈0.05). The dual luciferase reporter gene assay revealed endogenous microRNA-21 in cholan- giocarcinoma cell line, QBC939, inhibited the luciferase reporter activities of wild-type PTEN (P〈0.01) and PDCD4 (P〈0.05) and had no this effect on mutated PTEN and PDCD4. Moreover, loss of microRNA-21 function led to a significant increase of PTEN and PDCD4 protein levels in QBC939 cells. [Method] Characters of temperature variation in solar greenhouse and minimal temper...[Objective] The aim was to improve meteorological service of protected agriculture and to reduce effects of meteorological disasters. [Method] Characters of temperature variation in solar greenhouse and minimal temperature forecast models in winter were analyzed based on meteorological data inside and outside of solar greenhouse in winter during 2008-2011, as per correlation and stepwise regression method. [Result] Temperature was of significant changes in solar greenhouse in sunny and cloudy days and the change was higher in sunny days. In overcast days, temperature in solar greenhouse was lower and plants were affected seriously. In addition, the minimal temperature was of good correlation with outside temperature and humidity, temperature and soil temperature in greenhouse. [Conclusion] The minimal temperature forecast model of solar greenhouse is established and the average absolute error of the forecasted minimums in different types of weather was less than 1 ℃ and the average relative error was lower than 10%.展开更多

METHODS: Expression of HO-1 mRNA was analyzed by ...AIM: To investigate the molecular mechanism and functional consequences of heme oxygenase-1 (HO-1) activation by lansoprazole in endothelial cells and macrophages. METHODS: Expression of HO-1 mRNA was analyzed by Northern blotting. Western blotting was used to determine the HO-1 and ferritin protein levels. NADPH-dependent reactive oxygen species (ROS) formation was measured with lucigenin-enhanced chemiluminescence. HO-1 promoter activity in mouse fibroblasts, stably transfected with a 15-kb HO-1 gene that drives expression of the reporter gene luciferase, was assessed usingin vivo bioluminescence imaging. RESULTS: Lansoprazole levels in endothelial cells increased HO-1 mRNA and HO-1 protein levels in macrophages. In addition, lansoprazole-induced ferritin protein levels in both cell systems. Moreover, induction of the antioxidant proteins HO-1 and ferritin by lansoprazole was followed by a decrease in NADPH- mediated ROS formation. The radical scavenging properties of lansoprazole were diminished in the presence of the HO inhibitor, chromium mesoporphyrin IX. Induction of HO-1 gene expression by lansoprazole was not related to oxidative stress or to the activation of the mitogen-activated protein kinase pathway. However, the phosphatidylinositol 3-kinase inhibitor LY294002 showed a concentration-dependent inhibition of HO-1 mRNA and promoter activity.CONCLUSION: Activation of HO-1 and ferritin may account for the gastric protection of lansoprazole and is dependent on a pathway blocked by LY294002.展开更多
AIM:To investigate the role of transforming growth factor(TGF)-β-inducible early gene 1(TIEG1) in TGF-β-induced growth inhibition in hepatocellular carcinoma(HCC) cells.METHODS:Human hepatocyte and HCC cell lines wi...AIM:To investigate the role of transforming growth factor(TGF)-β-inducible early gene 1(TIEG1) in TGF-β-induced growth inhibition in hepatocellular carcinoma(HCC) cells.METHODS:Human hepatocyte and HCC cell lines with varied susceptibilities to TGF-β1 were tested by methylthiazoletetrazolium(MTT) assay.The expression changes of Smad2,Smad3,Smad4,Smad7,TIEG1 and TIEG2 gene following treatment with TGF-β1 in a TGF-β-sensitive hepatocyte cell line(MIHA),a TGF-β-sensitive hepatoma cell line(Hep3B) and two TGF-β-insensitive hepatoma cell lines(HepG2 and Bel7404) were examined.SiRNA targeting TIEG1 was transfected into Hep3B cells and the sensitivity of cells to TGF-β1 was examined.Overexpression of TIEG1 was induced by lentiviral-mediated transduction in TGF-β1-resistant hepatoma cell lines(Bel7404 and HepG2).MTT assay and 4',6-Diamidino-2-phenylindole staining were used to identify cell viability and apoptosis,respectively.The expression level of stathmin was measured by reverse transcriptase polymerase chain reaction and Western-blotting analysis,and stathmin promoter activity by TIEG1 was monitored by a luciferase reporter gene system.RESULTS:TIEG1 was significantly upregulated by TGF-β1 in the TGF-β1-sensitive HCC cell line,Hep3B,but not in the resistant cell lines.The suppression of TIEG1 by siRNAs decreased the sensitivity of Hep3B cells to TGF-β1,whereas the overexpression of TIEG1 mediated growth inhibition and apoptosis in TGF-β1-resistant HCC cell lines,which resembled those of TGF-β1-sensitive HCC cells treated with TGF-β1.Our data further suggested that stathmin was a direct target of TIEG1,as stathmin was signif icantly downregulated by TIEG1 overexpression,and stathmin promoter activity was inhibited by TIEG1 in a dose-dependent manner.CONCLUSION:Our data suggest that transactivation of TIEG1 conferred growth inhibition of TGF-β-susceptible human HCC cells.展开更多

Coal as the primary energy consumption brings a large number of regional pollution in Huaibei area. Differential optical absorption spectroscopy (DOAS) as optical remote sensing technology has been applied to monitor regional average concen- trations and inventory of nitrogen dioxide, sulfur dioxide and ozone. DOAS system was set up and applied to monitor the main air pollutants in Huaibei area. Monitoring data were obtained from 7 to 28 August, 2011. Monitoring results show measurements in controlling pollution are effective, and emissions of pollutants are up to the national standard in Huaibei area. Prediction model was also created to track changing trend of pollutions. These will provide raw data support for effective evaluation of environmental quality in Huaibei area.展开更多
We explored the potential of the environment and disaster monitoring and forecasting small satellite constellations (HJ-1A/1B satellites) charge-coupled device (CCD) imagery (spatial resolution of 30 m, revisit time o...We explored the potential of the environment and disaster monitoring and forecasting small satellite constellations (HJ-1A/1B satellites) charge-coupled device (CCD) imagery (spatial resolution of 30 m, revisit time of 2 days) in the monitoring of total suspended sediment (TSS) concentrations in dynamic water bodies using Poyang Lake, the largest freshwater lake in China, as an example. Field surveys conducted during October 17-26, 2009 showed a wide range of TSS concentration (3-524 mg/L). Atmospheric correction was implemented using the Fast Line-of-sight Atmospheric Analysis of Spectral Hypercubes (FLAASH) module in ENVI with the aid of aerosol information retrieved from concurrent Terra/Moderate Resolution Imaging Spectroradiometer (MODIS) surveys, which worked well at the CCD bands with relatively high reflectance. A practical exponential retrieval algorithm was created between satellite remote sensing reflectance and in-situ measured TSS concentration. The retrieved results for the whole water area matched the in-situ data well at most stations. The retrieval errors may be related to the problem of scale matching and mixed pixel. In three selected subregions of Poyang Lake, the distribution trend of retrieved TSS was consistent with that of the field investigation. It was shown that HJ-1A/1B CCD imagery can be used to estimate TSS concentrations in Poyang Lake over synoptic scales after applying an appropriate atmospheric correction method and retrieval algorithm.展开更多

Our findings indicate a lack of modern science-based approaches to diagnosis, rehabilitation, and socio-medical examination of patients with ophthalmologic pathologies, thus requiring deeper study of this problem.展开更多

The results show that the main false alarm resourses of space- based laser warning system are sunlight and lightning. The sunlight should exposure the detector directly in one ninth of the satfllite orbital period, and the imaging spot of sun is similar to the attack laser. The lightning imaging spot is similar to the illumination laser. About 1.4 lightning events can occur in the field of view (FOV) of the warning system per second. It could not discriminate spots of sun, lightning and threaten laser by the frame subtraction technology.展开更多

HSPS has the two-mode characteris...With a heralded single photon source(HSPS), a measurement-device-independent quantum key distribution(MDIQKD) protocol is proposed, combined with a three-intensity decoy-state method. HSPS has the two-mode characteristic, one mode is used as signal mode, and the other is used as heralded mode to reduce the influence of the dark count. The lower bound of the yield and the upper bound of the error rate are deduced and the performance of the MDI-QKD protocol with an HSPS is analyzed. The simulation results show that the MDI-QKD protocol with an HSPS can achieve a key generation rate and a secure transmission distance which are close to the theoretical limits of the protocol with a single photon source(SPS). Moreover, the key generation rate will improve with the raise of the senders' detection efficiency. The key generation rate of the MDI-QKD protocol with an HSPS is a little less than that of the MDI-QKD protocol with a weak coherent source(WCS) in the close range, but will exceed the latter in the far range. Furthermore, a farther transmission distance is obtained due to the two-mode characteristic of HSPS.展开更多
We demonstrated an efficient solar photovoltaic-powered electrochemical CO_(2) reduction device with a high-pressure CO_(2)-captured liquid feed.In an“air-to-barrel”picture,this device holds promise to avoid both hi...We demonstrated an efficient solar photovoltaic-powered electrochemical CO_(2) reduction device with a high-pressure CO_(2)-captured liquid feed.In an“air-to-barrel”picture,this device holds promise to avoid both high-temperature gaseous CO_(2) regeneration and high energy-cost gas product separation steps,while these steps are necessary for devices with a gaseous CO_(2) feed.To date,solar fuel production with a CO_(2)-saturated liquid feed suffers from high over-potential to suppress the hydrogen evolution reaction and consequently,low solar-to-chemical(STC)energy conversion efficiency.Here,we presented a distinct high-pressure operando strategy,i.e.,we took extra advantage of the high pressure in catalyst synthesis besides in the period of the CO_(2) reduction reaction(CO_(2)RR).The power of this strategy was demonstrated by a proof-of-concept device in which a representative copper catalyst was first synthesized in operando in a high-pressure(50 bar)CO_(2)-saturated KHCO3 solution,and then this high-pressure CO_(2)-captured liquid was converted to solar fuel using the operando synthesized Cu catalyst.This Cu catalyst achieved 95%CO_(2)RR selectivity at the recorded low potential of−0.3 V vs.RHE enabled by the combination of operando facet engineering and oxide derivation.Furthermore,this device achieved a record-high STC efficiency of 21.6%under outdoor illumination,superior to other CO_(2)-saturated liquid-fed devices,and compared favorably to gaseous CO_(2)-fed devices.展开更多

文摘AIM:To investigate the role of transforming growth factor(TGF)-β-inducible early gene 1(TIEG1) in TGF-β-induced growth inhibition in hepatocellular carcinoma(HCC) cells.METHODS:Human hepatocyte and HCC cell lines with varied susceptibilities to TGF-β1 were tested by methylthiazoletetrazolium(MTT) assay.The expression changes of Smad2,Smad3,Smad4,Smad7,TIEG1 and TIEG2 gene following treatment with TGF-β1 in a TGF-β-sensitive hepatocyte cell line(MIHA),a TGF-β-sensitive hepatoma cell line(Hep3B) and two TGF-β-insensitive hepatoma cell lines(HepG2 and Bel7404) were examined.SiRNA targeting TIEG1 was transfected into Hep3B cells and the sensitivity of cells to TGF-β1 was examined.Overexpression of TIEG1 was induced by lentiviral-mediated transduction in TGF-β1-resistant hepatoma cell lines(Bel7404 and HepG2).MTT assay and 4',6-Diamidino-2-phenylindole staining were used to identify cell viability and apoptosis,respectively.The expression level of stathmin was measured by reverse transcriptase polymerase chain reaction and Western-blotting analysis,and stathmin promoter activity by TIEG1 was monitored by a luciferase reporter gene system.RESULTS:TIEG1 was significantly upregulated by TGF-β1 in the TGF-β1-sensitive HCC cell line,Hep3B,but not in the resistant cell lines.The suppression of TIEG1 by siRNAs decreased the sensitivity of Hep3B cells to TGF-β1,whereas the overexpression of TIEG1 mediated growth inhibition and apoptosis in TGF-β1-resistant HCC cell lines,which resembled those of TGF-β1-sensitive HCC cells treated with TGF-β1.Our data further suggested that stathmin was a direct target of TIEG1,as stathmin was signif icantly downregulated by TIEG1 overexpression,and stathmin promoter activity was inhibited by TIEG1 in a dose-dependent manner.CONCLUSION:Our data suggest that transactivation of TIEG1 conferred growth inhibition of TGF-β-susceptible human HCC cells.

文摘One kind of combustible gas alarms based on industrial Ethernet was designed to prevent the gas leakage in industrial production sites, The alarm adopted the high performance microprocessor LPC2214 as the main chip. The embedded operating system μC/OS-Ⅱand TCP/IP protocol stack uIP running on LPC2214 constitute a development platform of application of the combustible gas alarm, The test shows that it can automatically and continuously detect combustible gas in industrial production sites in several positions;it can give out sound-light alarm and take protective measures immediately against the gas leakage; and it can send the detected data to PC through the Ethernet interface to realize the remote detection. The designed project provides a reference to design industrial devices based on industrial Ethernet
文摘Huaibei is an energy city. Coal as the primary energy consumption brings a large number of regional pollution in Huaibei area. Differential optical absorption spectroscopy (DOAS) as optical remote sensing technology has been applied to monitor regional average concen- trations and inventory of nitrogen dioxide, sulfur dioxide and ozone. DOAS system was set up and applied to monitor the main air pollutants in Huaibei area. Monitoring data were obtained from 7 to 28 August, 2011. Monitoring results show measurements in controlling pollution are effective, and emissions of pollutants are up to the national standard in Huaibei area. Prediction model was also created to track changing trend of pollutions. These will provide raw data support for effective evaluation of environmental quality in Huaibei area.

文摘We explored the potential of the environment and disaster monitoring and forecasting small satellite constellations (HJ-1A/1B satellites) charge-coupled device (CCD) imagery (spatial resolution of 30 m, revisit time of 2 days) in the monitoring of total suspended sediment (TSS) concentrations in dynamic water bodies using Poyang Lake, the largest freshwater lake in China, as an example. Field surveys conducted during October 17-26, 2009 showed a wide range of TSS concentration (3-524 mg/L). Atmospheric correction was implemented using the Fast Line-of-sight Atmospheric Analysis of Spectral Hypercubes (FLAASH) module in ENVI with the aid of aerosol information retrieved from concurrent Terra/Moderate Resolution Imaging Spectroradiometer (MODIS) surveys, which worked well at the CCD bands with relatively high reflectance. A practical exponential retrieval algorithm was created between satellite remote sensing reflectance and in-situ measured TSS concentration. The retrieved results for the whole water area matched the in-situ data well at most stations. The retrieval errors may be related to the problem of scale matching and mixed pixel. In three selected subregions of Poyang Lake, the distribution trend of retrieved TSS was consistent with that of the field investigation. It was shown that HJ-1A/1B CCD imagery can be used to estimate TSS concentrations in Poyang Lake over synoptic scales after applying an appropriate atmospheric correction method and retrieval algorithm.
文摘The article represents the glaucoma disability rates in Ukraine and touches upon medical and social aspects of the problem We have screened a data from source medical documents and analyzed the MSEK (Physical Disability Board of Review) statistical reporting data, further used as a basis for calculation of live indices of general and primary disabilities caused by eye diseases, including the ones caused by primary open-angle glaucoma (7.05% and 9.99%, respectively), the extensive distribution indices of disability by gender (75.74% male), age (59.29% of II maturity patients), and a disability group (56% of patients recognized disabled persons of groups I and II). Our findings indicate a lack of modern science-based approaches to diagnosis, rehabilitation, and socio-medical examination of patients with ophthalmologic pathologies, thus requiring deeper study of this problem.
文摘In view of the problem of false alarm in imaging space-based laser warning system, the effects of sunlight and lightning on the threaten laser detection and attack event determination are studied by analyzing and calculating the radiant energy density and space-time feature of imaging spot, respectively. The results show that the main false alarm resourses of space- based laser warning system are sunlight and lightning. The sunlight should exposure the detector directly in one ninth of the satfllite orbital period, and the imaging spot of sun is similar to the attack laser. The lightning imaging spot is similar to the illumination laser. About 1.4 lightning events can occur in the field of view (FOV) of the warning system per second. It could not discriminate spots of sun, lightning and threaten laser by the frame subtraction technology.

文摘With a heralded single photon source(HSPS), a measurement-device-independent quantum key distribution(MDIQKD) protocol is proposed, combined with a three-intensity decoy-state method. HSPS has the two-mode characteristic, one mode is used as signal mode, and the other is used as heralded mode to reduce the influence of the dark count. The lower bound of the yield and the upper bound of the error rate are deduced and the performance of the MDI-QKD protocol with an HSPS is analyzed. The simulation results show that the MDI-QKD protocol with an HSPS can achieve a key generation rate and a secure transmission distance which are close to the theoretical limits of the protocol with a single photon source(SPS). Moreover, the key generation rate will improve with the raise of the senders' detection efficiency. The key generation rate of the MDI-QKD protocol with an HSPS is a little less than that of the MDI-QKD protocol with a weak coherent source(WCS) in the close range, but will exceed the latter in the far range. Furthermore, a farther transmission distance is obtained due to the two-mode characteristic of HSPS.

文摘We demonstrated an efficient solar photovoltaic-powered electrochemical CO_(2) reduction device with a high-pressure CO_(2)-captured liquid feed.In an“air-to-barrel”picture,this device holds promise to avoid both high-temperature gaseous CO_(2) regeneration and high energy-cost gas product separation steps,while these steps are necessary for devices with a gaseous CO_(2) feed.To date,solar fuel production with a CO_(2)-saturated liquid feed suffers from high over-potential to suppress the hydrogen evolution reaction and consequently,low solar-to-chemical(STC)energy conversion efficiency.Here,we presented a distinct high-pressure operando strategy,i.e.,we took extra advantage of the high pressure in catalyst synthesis besides in the period of the CO_(2) reduction reaction(CO_(2)RR).The power of this strategy was demonstrated by a proof-of-concept device in which a representative copper catalyst was first synthesized in operando in a high-pressure(50 bar)CO_(2)-saturated KHCO3 solution,and then this high-pressure CO_(2)-captured liquid was converted to solar fuel using the operando synthesized Cu catalyst.This Cu catalyst achieved 95%CO_(2)RR selectivity at the recorded low potential of−0.3 V vs.RHE enabled by the combination of operando facet engineering and oxide derivation.Furthermore,this device achieved a record-high STC efficiency of 21.6%under outdoor illumination,superior to other CO_(2)-saturated liquid-fed devices,and compared favorably to gaseous CO_(2)-fed devices.
