Enzyme-linked immuno-sorbent assay (ELISA) and coagulating time method were used to assess antigenic stability and coagulating stability, respectively. The change of antigenic activities and coagulating activities of Defibrase in the same buffer solutions (pH 6, 7 and 8, with the exception of pH 3.6) showed similar tendency to decline with the time. Concentrated Defi-brase was relatively stable at neutral pH 6~7, more than 95% of its initial activities (100BUmL-1) was kept after a 10-day storage at 40 oC, whereas in pH 3.6 and pH 9 buffer solutions, diluted Defibrase was very labile. Addition of Triton X-100 or bovine serum albumin could effectively prevent loss of Defibrase by minimizing adsorption of De-fibrase to plastic surface (P<0.005). Concentration of Defibrase could also affect its stability in aqueous solutions.展开更多

A bet...The Asian swamp eel (Monopterus albus) is one of the most economically important freshwater fish in East Asia, but data on the immune genes of M. albus are scarce compared to other commercially important fish. A better understanding of the eel's immune responses may help in developing strategies for disease management, potentially improving yields and mitigating losses. In mammals, interferon regulatory factors (IRFs) play a vital role in both the innate and adaptive immune system; though among teleosts IRF4 and IRFIO have seldom been studied. In this study, we characterized IRF4 and IRFIO from M. albus (malRF4 and malRFlO) and found that malRF4 cDNA consists of 1 716 nucleotides encoding a 451 amino acid (aa) protein, while malRFlO consists of 1 744 nucleotides including an open reading frame (ORF) of 1 236 nt encoding 411 aa. The malRFlO gene was constitutively expressed at high levels in a variety of tissues, while malRF4 showed a very limited expression pattern. Expression of malRF4 and malRFlO in head kidney, and spleen tissues was significantly up-regulated from 12 h to 48 h post-stimulation with polyinosinic: polycytidylic acid (poly I:C), lipopolysaccharide (LPS) and a common pathogenic bacteria Aeromonas hydrophila. These results suggest that IRF4 and IRF10 play roles in immune responses to both viral and bacterial infections in M. albus.展开更多

We describe the CS...In this study, the transmission capacity of VANETs in a highway scenario is analysed on the basis of a 1D line model and the carrier sense multiple access with collision avoidance(CSMA/CA) protocol. We describe the CSMA/CA protocol used in IEEE802.11 p from the perspective of the geometric relationship amongst simultaneous transmitters. The desired channel and interfering channels are assumed to experience the same amount of path-loss and Rayleigh fading. On the basis of the proposed model, we analyse the attempted transmission probability of each road segment and the maximum intensity of active transmitters, including their theoretical values. Then, we employ the physical model to obtain the outage probability and derive the upper bound of the transmission capacity of a VANET, which is defined as the average spatial density of successful transmissions in the network. Simulation results indicate that the theoretical value offers a good bound on network capacity.展开更多

Nowadays, it is expected that the active safety systems will aid avoiding co...In the near future, active safety systems will take more control over the vehicle driving, even up to introducing fully autonomous vehicles. Nowadays, it is expected that the active safety systems will aid avoiding collisions much more efficiently than human drivers. These systems can protect not only the passengers, but also other road users. To mitigate collision, certain maneuvers (e.g., sudden braking, lane change, etc.) need to be done in a reasonably quick time. However, this may lead to low-g energy pulses. The latter fact, may cause unexpected and, in some cases, unwanted occupant body motion resulting even in OOP (out of position) postures. New patterns of occupant reactions in such cases are, to some extent, confirmed experimentally [1-3]. This paper evaluates the limits of standard ATDs (anthropometric test devices) and chosen human models in well established maneuver scenarios. Obtained results are compared with experimental data available in the literature. Drawbacks identify new challenges for the near future simulation based safety engineering. One scenario with combined conditions of emergency braking during lane change has been used as an example of OOP posture after maneuver.展开更多
OBJECTIVE: To study glomerular expression of C-C chemokines, monocyte chemoattractant protein-1 (MCP-1), macrophage inflammatory protein-1alpha and beta (MIP-1alpha, MIP-1beta) and the effect of steroid and cyclophosp...OBJECTIVE: To study glomerular expression of C-C chemokines, monocyte chemoattractant protein-1 (MCP-1), macrophage inflammatory protein-1alpha and beta (MIP-1alpha, MIP-1beta) and the effect of steroid and cyclophosphamide (CTX) intermittent intravenous pulse therapy on expression in patients with crescentic glomerulonephritis (CGN) to further investigate the underlying mechanism of the treatment. METHODS: Twelve patients with initial biopsy-proven CGN(2), 6 with lupus nephritis (lupus-CGN, LN-CGN) and 6 with vasculitis, (vasculitis-CGN, V-CGN) were enrolled in this study. They underwent an initial biopsy before steroid and CTX intermittent intravenous pulse therapy and were biopsied again one to three months later. Expression of MCP-1, MIP-1alpha, MIP-1beta, and CD68 in glomeruli with cellular and fibrocellular crescents were examined by immunohistochemical analysis in serial sections of renal biopsies. The effect of the pulse therapy on histopathological changes was also observed. RESULTS: Although steroid and CTX intermittent intravenous pulse therapy markedly reduced the degree of glomerular crescent formation both in LN-CGN and V-CGN, the effect of the therapy on glomerular chemokine expression was significantly different between LN-CGN and V-CGN. It was found that steroid and CTX intermittent intravenous pulse therapy reduced the expression of CD68, MCP-1, and MIP-1alpha, but had no effect on MIP-1beta in glomeruli with cellular crescents of patients with LN-CGN. In patients with V-CGN, the therapy also reduced the expression of CD68, but had no effect on MCP-1, MIP-1alpha, and MIP-1beta in glomeruli with cellular crescents. It was noted that the degree of glomerulosclerosis and tubular interstitial fibrosis increased more significantly at the second biopsy in V-CGN as compared to LN-CGN. CONCLUSIONS: The efficacy of steroid and CTX intermittent intravenous pulse therapy in CGN might be affected by reduction of glomerular chemokine expression. The different changes in glomerular expression of MCP-1 and MIP-1alpha in patients with LN-CGN and V-CGN after pulse therapy may correlate to different responses to treatment and prognosis.展开更多
Paraquat(PQ),a highly effective herbicide,is widely used worldwide.PQ poisoning can cause multiple organ failure,in which the lung is the primary target organ.After PQ poisoning,the patient mortality rate is as high a...Paraquat(PQ),a highly effective herbicide,is widely used worldwide.PQ poisoning can cause multiple organ failure,in which the lung is the primary target organ.After PQ poisoning,the patient mortality rate is as high as 90%,and there is currently no specific antidote.The main clinical treatment is the use of glucocorticoids and cyclophosphamide for pulse therapy,but its effectiveness and safety are still uncertain.We investigated the effectiveness and safety of immunosuppressive pulse therapy with glucocorticoids and cyclophosphamide to evaluate the treatment value in patients with acute PQ poisoning.This meta-analysis,combined with seven trials that enrolled a total of 426 patients,showed that immunosuppressive pulse therapy with glucocorticoids and cyclophosphamide for PQ poisoning significantly reduced mortality of the study group(59.3%,134/226) compared with the control group(81.0%,162/200).There was no significant difference of hepatitis or renal failure between the control and study groups,indicating that immunosuppressive pulse therapy was relatively safe.Several patients were reported to have leukopenia and returned to normal after 1-2 weeks without any abnormalities.Two cases of non-fatal sepsis were reported and considered to be a side effect of the immunosuppressive pulse therapy.Thus,immunosuppressive pulse therapy can efficiently reduce the mortality of PQ poisoning and it is relatively safe.展开更多

DCs play a dual role by enhancing b...In this research paper, our main objective is to find out the meticulous role of activated dendritic cells (DCs) during the human immunodeficiency virus (HIV) infection process. DCs play a dual role by enhancing both HIV infection progression, as well as antiviral immune response. To explore the implications of these dual roles, we have formulated our mathematical model and analyzed the model by both analytical and numerical approaches. By using an impulsive differential equation, we have studied the effect of DC-based vaccination. Analytically we have determined the threshold value of drug dosage and dosing interval for optimum levels of injection. We have also investigated the effect of perfect adherence of drug dose on the immune cell count in extreme cases and observed that, systematic drug dose of the immune cells leads to its maximum level.展开更多
