目的探讨转化生长因子β1(TGFβ1)诱导人视网膜色素上皮(RPE)细胞凋亡过程中Survivin基因的表达及意义。方法用免疫组织化学染色法检测人RPE细胞Survivin蛋白的表达;用AnnexinVFITC标记法测定TGFβ1作用下的人RPE细胞凋亡发生率;用RTPC... 目的探讨转化生长因子β1(TGFβ1)诱导人视网膜色素上皮(RPE)细胞凋亡过程中Survivin基因的表达及意义。方法用免疫组织化学染色法检测人RPE细胞Survivin蛋白的表达;用AnnexinVFITC标记法测定TGFβ1作用下的人RPE细胞凋亡发生率;用RTPCR方法检测不同质量浓度TGFβ1作用下人RPE细胞Survivin基因的表达。结果免疫化学染色法显示:人RPE细胞表达Survivin基因;AnnexinVFITC标记法检测结果显示:人RPE细胞的凋亡率随着TGFβ1质量浓度的升高而增高;RTPCR结果显示:随着TGFβ1质量浓度的升高,Survivin基因的表达逐渐降低。结论Survivin基因在人RPE细胞的凋亡过程中起着非常重要的作用。 展开更多

AIM: To investigate the hepatoprotective activity of tea polyphenols (TP) and its relation with cytochrome P450 (CYP450) expression in mice. METHODS: Hepatic CYP450 and CYPbs levels were measured by UV-spectrophotometry in mice 2 d after intraperitoneal TP (25, 50 and 100 mg/kg per day). Then the mice were intragastricly pre-treated with TP (100, 200 and 400 mg/kg per day) for six days before paracetamol (1000 mg/kg) was given. Their acute mortality was compared with that of control mice. The mice were pre-treated with TP (100, 200, and 400 mg/kg per day) for five days before paracetamol (500 mg/kg) was given. Hepatic CYP2E1 and CYPIA2 protein and mRNA expression levels were evaluated by Western blotting, immunohistochemical staining and transcriptase-polymerase chain reaction. RESULTS: The hepatic CYP450 and CYPb5 levels in mice of TP-treated groups (100, 200 and 400 mg/kg per day) were decreased in a dose-dependent manner compared with those in the negative control mice.TP significantly attenuated the paracetamol-induced hepatic injury and dramatically reduced the mortality of paracetamol-treated mice. Furthermore, TP reduced CYP2E1 and CYPIA2 expression at both protein and mRNA levels in a dose-dependent manner. CONCLUSION: TP possess potential hepatoprotective properties and can suppress CYP450 expression. 展开更多

During embryonic development, pluripotent endoderm tissue in the developing foregut may adopt pancreatic fate or hepatic fate depending on the activation of key developmental regulators. Transdifferentiation occurs between hepato- cytes and pancreatic cells under specific conditions. Hepatocytes and pancreatic cells have the common endodermal progenitor cells. In this study we isolated hepatic stem/progenitor cells from embryonic day (ED) 12-14 Kun-Ming mice with fluorescence-activated cell sorting (FACS). The cells were cultured under specific conditions. The cultured cells deploy dithizone staining and immunocytochemical staining at the 15th, 30th and 40th day after isolation. The results indicated the presence of insulin-producing cells. When the insulin-producing cells were transplanted into alloxan- induced diabetic mice, the nonfasting blood glucose level was reduced. These results suggested that fetal liver stem/ progenitor cells could be converted into insulin-producing cells under specific culture conditions. Fetal liver stem/ progenitor cells could become the potential source of insulin-producing cells for successful cell transplantation therapy strategies of diabetes. 展开更多

Some authors have reported that meningioma is more prevalent among adult males, but there is no gender prevalence. The accepted origin of meningiomas is from the arachnoid ceils lining the meninges, or the choroid plexuses. Since Beckwith and Palmer introduced the term 'rhabdoid tumor' in 1978 in reference to a subgroup of childhood malignant renal tumors, many tumors with a rhabdoid morphology have been reported in various sites, including the central nervous system.
