In the present paper, the author makes some comments on the holistic action of acupuncture from the regulative effect on the nerve-endocrine-immune network, and the close relationship between the nerve-endocrine-immune network and the meridian-collateral system of TCM. The wholism concept of TCM refers to the organism being an integrated entirety, and the regulatory effect of acupuncture on functional activities of the organism relies on the integral connection of the acupoint-meridian-collateral-zangfu-organ system. OBJECTIVE To investigate slow-release features of biodegradable anticancer 5-fluorouracil-loaded immunonanoparticles (5-FU INPs), and to assess their tumor cell killing activity in vitro.METHODS The method of vibrating dialysis at a constant temperature, and first-order derivative ultraviolet spectrophotometry were used to determine the drug-releasing character of 5-FU INPs. The methyl thiazolyl tetrazolium (MTT) colorimetric method was employed to assay the killing activity of 5-FU INPs on 5 tumor cell lines at different phases.RESULTS The 5-FU INPs had a favorable slow-release function, with a tl/2 release time of 10.4 days. The 5-FU INPs had a rather strong lethal effect on 5 tumor cell lines resulting in a positive correlativity between the killing activity and the action time and amount of the drug released.CON'CLUSION The drug disposition is uniform from the 5-FU INPs, and there is no impact on efficacy of the 5-FU during preparation and degradation of the 5-FU INPs. The 5-FU INPs have a favorable function for drug release, and can maintain an effective killing activity over a long period of time. 展开更多

Gonadotropin-releasing hormone (GnRH) is a crucial peptide for the regulation of reproduction. Using immunological techniques, we investigated the presence of GnRH in horseshoe crab Tachypleus tridentatus. Octopus GnRH-like immunoreactivity, tunicate GnRH-like immunoreactivity, and lamprey GnRH-I-like immunoreactivity were detected in the neurons and fibers of the protocerebrum. However, no mammal GnRH-like immunoreactivity or lamprey GnRH-LII-like immunoreactivity was observed. Our results suggest that a GnRH-like factor, an ancient peptide, existed in the brain of T. tridentatus and may be involved in the reproductive endocrine system. 展开更多

EAE (experimental autoimmune encephalomyelitis) is an established, inducible animal model employed in the study of MS (multiple sclerosis) characterized by inflammation, BBB (blood brain barrier) malfunction, demyelination and neuronal disruption. CRF (corticotropin releasing factor) is a neuropeptide critically associated with immune function, BBB permeability, and the hypothalamic-pituitary-adrenal axis. Potential CRF targets in the brain include astrocytes, as well as endothelial cells of cerebral microvessels, since they have been reported to express CRFR (CRF receptors). Further, both of these cell types function critically in regulating BBB permeability. CRF-BP (CRF binding protein) is also expressed in both neurons and glial cells. Changes in the cortical CRF system could be a contributing factor to the BBB disruption associated with MS/EAE and has been suggested to play a protective role against cytokine-induced inflammation. The current study assessed alterations associated with the C57BL/6 mouse model of EAE in the cortical CRF system and correlated these events with changes to the microvascular unit. Immunohistochemical confocal microscopy was used to analyze the distribution of CRF, CRF-BP, and CRFR in the mouse cerebral cortex. The authors observed a reduction in detectable CRF immunofluorescence in the EAE motor cortex, an increase in CRFBP immunoreactivity in EAE astrocytes and a concurrent reduction in astrocytic CRFR immunofluorescence. Staining techniques were used to visualize astrocytes/microvessels to document alterations in BBB integrity. Changes in the CRF system were associated with a modification of the blood brain barrier as manifested by a poorly defined astrocytic barrier in EAE microvessels. Evidence suggests that manipulation of CRF signaling pathways offers an intriguing target for interventional therapies designed to modify BBB permeability that may be beneficial for treating disease states such as MS. 展开更多

OBJECTIVE:To observe the effects of Fuxin decoction in rats with heart failure induced by doxorubicin injections.METHODS:We employed a doxorubicin-induced heart failure model to investigate the effect of Fuxin decoction.Rats were orally administered with Fuxin decoction from 10 to 40 mg/kg.Organization structure changes were assessed by histology,circulating levels of oxidative stress-related biomarkers and inflammatory cytokines were detected via enzyme-linked immunosorbent assay.RESULTS:Administration of Fuxin decoction improved the growth retardation in doxorubicin hydrochloride injection rats.Cardiac involvement was alleviated by Fuxin decoction,as well as changes of congestion systemic circulation in this rat models.Furthermore,Fuxin decoction regulates immune responses and suppresses oxidation reaction in doxorubicin hydrochloride-induced rats.CONCLUSION:Fuxin decoction might be an interesting candidate for congestive heart failure. 展开更多

The abnormal changes of immune-mediated inflammation interfere its pathogenesis and development directly. In recent years, various aspects of neuroendocrine responses, especially the regulatory effects of hypothalamic-pituitary-adrenal and sympathetico-adrenomedullary axes in inflammatory diseases have been the focus of research. Most importantly,corticotropin-releasing hormone (CRH) acts as a key player in the regulation of interactions between neuroendocrine and immunity both directly and indirectly.
