AIM: To investigate apoptosis in human pancreatic cancer cells induced by Triptolide (TL), and the relationship between this apoptosis and expression of caspase-3' bcl-2 and bax. METHODS: Human pancreatic cancer cell line SW1990 was cultured in DMEM media for this study. MTT assay was used to determine the cell growth inhibitory rate in vitro. Flow cytometry and TUNEL assay were used to detect the apoptosis of human pancreatic cancer cells before and after TL treatment. RT-PCR was used to detect the expression of apoptosis-associated gene caspase-3' bcl-2 and bax. RESULTS: TL inhibited the growth of human pancreatic cancer cells in a dose-and time-dependent manner. TL induced human pancreatic cancer cells to undergo apoptosis with typically apoptotic characteristics. TUNEL assay showed that after the treatment of human pancreatic cancer cells with 40 ng/mL TL for 12 h and 24 h, the apoptotic rates of human pancreatic cancer cells increased significantly. RT-PCR demonstrated that caspase-3 and bax were significantly up-regulated in SW1990 cells treated with TL while bcl-2 mRNA was not. CONCLUSION: TL is able to induce the apoptosis in human pancreatic cancer cells. This apoptosis may be mediated by up-regulating the expression of apoptosis- associated caspase-3 and bax gene. 展开更多

Though they look very different,UOP SimulFlowTM,Koch-Glitsch Ultra-FracTM,Jaeger CoFloTM and Shell ConSepTM trays fall into the same category of trays using inertial separation technology. However,flooding mechanisms and the trends of entrainment and efficiency are different due to their different working principles. This paper provides a detailed analysis of these trays using available information from literature and U.S. Patents. Efforts are also made to interpret the observations reported. In terms of tray efficiency,it is found that SimulFlow,Ultra-Frac and CoFlo trays are typical point efficiency devices due to a completely mixed liquid pool on the tray deck,while ConSep trays can take advantage of liquid concentration gradient on the tray deck,which makes this tray at-tractive among all ultra high capacity trays. 展开更多

目的:探讨雷公藤甲素(triptolide,雷公藤甲素)致雌性大鼠生殖系统损害及防治方法。方法:采用体外卵巢颗粒细胞(granulosa cell,GCs)培养,分别给予不同浓度雷公藤甲素干预及不同浓度β-雌二醇与雷公藤甲素共同孵育,观察卵巢GCs的生长情况... 目的:探讨雷公藤甲素(triptolide,雷公藤甲素)致雌性大鼠生殖系统损害及防治方法。方法:采用体外卵巢颗粒细胞(granulosa cell,GCs)培养,分别给予不同浓度雷公藤甲素干预及不同浓度β-雌二醇与雷公藤甲素共同孵育,观察卵巢GCs的生长情况,采用MTT检测细胞增殖程度,TUNEL检测细胞凋亡程度。结果:细胞培养48小时后观察各组细胞形态,雷公藤甲素浓度大于100ug/L时,贴壁细胞数量明显减少;MTT检测显示,与对照组相比,当浓度>100ug/L的雷公藤甲素组均存在显著差异。TUNEL检测细胞凋亡提示β-雌二醇(10-7mol/L)与雷公藤甲素(2.78×10-7mol/L)共同孵育组,细胞凋亡程度明显减少,数量增多。结论:随着雷公藤甲素浓度增加,颗粒细胞生长及增殖受到显著抑制;β-雌二醇可以预防和修复雷公藤甲素对大鼠颗粒细胞造成的伤害,增强细胞活性,抑制细胞凋亡。 展开更多

目的:观察不同剂量京尼平苷对雷公藤甲素所致小鼠肝损伤的保护作用,并初步探讨其作用机制。方法:32只小鼠随机平均分成4组:空白对照组,每天灌胃生理盐水;雷公藤甲素组,每天灌胃雷公藤甲素400μg/kg;京尼平苷50mg/kg组:每天灌胃雷公藤甲... 目的:观察不同剂量京尼平苷对雷公藤甲素所致小鼠肝损伤的保护作用,并初步探讨其作用机制。方法:32只小鼠随机平均分成4组:空白对照组,每天灌胃生理盐水;雷公藤甲素组,每天灌胃雷公藤甲素400μg/kg;京尼平苷50mg/kg组:每天灌胃雷公藤甲素400ug/kg+京尼平苷50mg/kg;京尼平苷150mg/kg组:每天灌胃雷公藤甲素400ug/kg+京尼平苷150mg/kg。每天给药1次,连续28d后检测血清中ALT、AST、TBIL、ALP和LDH,测定肝组织中MDA、GST、GSH、SOD和UGT1含量。HE染色观察肝脏病理改变。结果:与雷公藤甲素组相比,联合给予京尼平苷150mg/kg的小鼠ALT、AST、TBIL和ALP有显著的下降,联合给予京尼平苷50mg/kg对ALT、AST、TBIL和LDH的改变没有显著性影响。京尼平苷150mg/kg和50mg/kg剂量均能不同程度的升高肝组织中GST、GSH、SOD和UGT1含量,但不能改善肝组织中MDA的含量。病理检查结果也显示京尼平苷的联合给予改善了雷公藤甲素引起的病理改变。结论:京尼平苷150mg/kg和50mg/kg剂量均能保护雷公藤甲素引起的小鼠肝损伤,其中150mg/kg剂量的保护效果更佳,其保护机制有待深入的研究。 展开更多
