目的以网络药理学结合分子对接为研究方法,分析养精种玉汤治疗多囊卵巢综合征(polycystic ovary syndrome,PCOS)的分子作用机制。方法通过TCMSP、TCMID、Swiss Target Prediction数据库预测及筛选养精种玉汤活性成分及靶点,利用GeneCard... 目的以网络药理学结合分子对接为研究方法,分析养精种玉汤治疗多囊卵巢综合征(polycystic ovary syndrome,PCOS)的分子作用机制。方法通过TCMSP、TCMID、Swiss Target Prediction数据库预测及筛选养精种玉汤活性成分及靶点,利用GeneCards、CTD数据库获取疾病靶点,两者取交集得到养精种玉汤-PCOS共同靶点。将交集靶点导入STRING平台构建蛋白质-蛋白质相互作用网络(protein-protein interaction,PPI),并进一步分析蛋白质功能子模块。利用Metascape网站对交集靶点进行基因本体(gene ontology,GO)和京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)通路富集分析。在Cytoscape 3.8.0拓扑分析相关结果并通过AltoDock-Vina软件进行分子对接验证。结果筛选得到养精种玉汤-PCOS共同靶点70个。拓扑分析显示养精种玉汤治疗PCOS的关键成分有山柰酚、β-谷甾醇、豆甾醇等,关键靶点有HMGCR、SHBG、SREBF2、CYP19A1、NR1H3、CYP17A1等,分子对接结果显示其对接活性皆良好。主要富集通路涉及类固醇激素生物合成、EGFR酪氨酸激酶抑制剂抗性、卵母细胞减数分裂、药物代谢、白介素-17信号通路、血清素能突触、白细胞跨内皮迁移、PPAR信号通路、NF-κB信号通路等。结论本研究初步分析了养精种玉汤治疗PCOS的分子作用机制,为后续研究提供思路。 展开更多

目的探索中西医结合治疗排卵障碍性不孕的方法。方法 130例排卵功能障碍性不孕患者随机分成治疗组(西医对症治疗和促排卵治疗基础上用养精种玉汤加味)65例,对照组(仅用西医对症治疗和促排卵治疗)65例,比较中西医结合与单纯使用西医药的... 目的探索中西医结合治疗排卵障碍性不孕的方法。方法 130例排卵功能障碍性不孕患者随机分成治疗组(西医对症治疗和促排卵治疗基础上用养精种玉汤加味)65例,对照组(仅用西医对症治疗和促排卵治疗)65例,比较中西医结合与单纯使用西医药的治疗效果,并用SPSS13.0对疗效做卡方检验。结果治疗组排卵率为92.31%,妊娠率为61.54%,优于对照组(66.15%,38.46%),排卵率和妊娠率的差异均有显著意义(χ2=13.510,P=0.000;χ2=6.923,P=0.009),诱发排卵和妊娠效果(OR值),治疗组分别是对照组的6.140倍和2.560倍,中西医结合与单纯使用西医药的总体疗效的差异亦有显著意义(χ2=14.271,P=0.001),中西医结合疗效更好。结论养精种玉汤加味具有促排卵功效,在西医对症治疗和促排卵治疗基础上,加中药养精种玉汤加味的中西医结合方法治疗排卵障碍性不孕具有明显优势,可以在临床推广。 展开更多

[Objective] The study aimed at reducing the economic cost in feeding Tenebrio molitor and improving the utilization ratio of corn stalks.[Method] The effects of different proportions of corn stalks on the weigh of Tenebrio molitor and the contents of crude protein,crude fat,Ca,P,dry matter and crude ash in the half-dry matter of Tenebrio molitor had been studied in the laboratory.[Result] Compared with the recipes of all wheat bran and pure fine fodder,feeding Tenebrio molitor by applying 30%-60% of corn stalks could significantly improve the content of crude protein in the half-dry materials of Tenebrio molitor,but it was detrimental to the improvement in weight of Tenebrio molitor and the accumulation of crude fat.To be specific,the recipe V(50% of corn stalks + 50% of fine fodder) was beneficial to the accumulation of dry matter and crude ash.The recipe VII(70% of corn stalks + 30% of fine fodder) and the recipe VIII(70% of corn stalks treated with Guangda feed leaven + 30% of fine fodder) were beneficial to the accumulation of crude protein,Ca and P.There were differences among the influences of corn stalks in different treatments on the crude protein and other nutrition indicators of Tenebrio molitor.[Conclusion] Appropriately applying corn stalks can improve the accumulation of the crude protein,crude ash,dry matter,Ca and P of Tenebrio molitor but was detrimental to the growth and development of Tenebrio molitor and the accumulation of crude fat. 展开更多

[Method] A field experiment was carried out to study the effects of different management practices (including nutrient management systems and cultivation methods) on crop yield and soil fertility in winter wheat/summer maize rotation system. [Result] The crop yield in the treatment of the high yield and high efficiency system was remarkably higher than farmer conventional management practice. After five crop seasons experiment, the contents of soil organic matter for high yield and high efficiency system increased 2.72-3.01 g/kg, and that of soil total nitrogen increased 0.12-0.16 g/kg, the soil Olsen-P increased 5.2 mg/kg and the soil available K (NH4OAC-K) increased about 37.8 mg/kg. [Conclusion] Considering the yield and soil fertility comprehensively, the management system of high yield and high efficiency could effectively increase the crop yield and improve the soil fertility. 展开更多

[Method] Taking the stem segments of E. tirucalli as explants,the effects of various mediums on germination rate,multiplication coefficient and rooting rate were studied. [Result] The optimum induction medium of germination culture was 1/2MS+NAA 0.02 mg/L+6-BA 1.0 mg/L,with differentiation rate of 89.7%; the best subculture medium was 1/2MS+NAA 0.02 mg/L+6-BA 0.60 mg/L+AD 3.0 mg/L,with multiplication coefficient of 5.70; the optimum rooting culture medium was 1/2MS+NAA 0.40 mg/L+IBA 0.4 mg/L,with rooting rate of 100% and transplanting survival rate of 80%. Various systems by using electric pulse, calcium, or polyethylene glycol have been developed in the past decade for the in vitro fusion of plant gametes. These in vitro systems provide a new way to study the fertilization mechanisms of plants. In this study, we developed a bovine serum albumin (BSA)-mediated fusion system for the in vitro fusion of maize gametes. The in vitro fusion of the isolated single egg cell and sperm cell of maize was observed microscopically in the BSA solution and the fertilized egg cell showed normal cell wall regeneration and nuclear division. The effects of the BSA concentration, pH value and calcium level on the efficiency of the maize gamete fusion were also assessed. BSA concentration and pH value did significantly affect the efficiency of the gamete fusion. Calcium was not necessary for the gamete fusion when BSA was present. The optimal solution for the gamete fusion contained 0.1% BSA, pH 6.0. The fusion frequency was as high as 96.7% in that optimal solution. This new in vitro fertilization system offers an alterna- tive tool for the in vitro study of fertilization mechanisms with much simpler manipulating procedure than PEG system, and it will be especially useful for the in vitro study of the calcium dynamics during plant fertilization. 展开更多
