The effects of medium, pH, water potential and temperature on the culture for three pure strains (Lactarius deliciosus, Boletus edulis and Lactarius insulsus) of ectomycorrhizal fungi from plantation forests of Mong...The effects of medium, pH, water potential and temperature on the culture for three pure strains (Lactarius deliciosus, Boletus edulis and Lactarius insulsus) of ectomycorrhizal fungi from plantation forests of Mongolian pine (Pinus sylvestris var. mongolica) on sandy lands were observed to obtain the optimum conditions for the growth of ectomycorrhizal fungi. The results indicated that the three ectomycorrhizal fungi could grow well in the mediums containing natural components, such as vitamin, pine juice and yeast powder, pH had a slight effect on the growth of the three ectomycorrhizal fungi, and the optimum pH values were 6.0 for L. deliciosus, 5.0 for B. edulis, respectively. However, L. insulsus had a wide pH range, and it grew better than the other two strains in neutral and light alkalescent mediums. Water potential (produced by Polyethylene Glycol, PEG) had significant effects on the ecological adaptability for the tested three fungi strains. All of the three stains grow better at lower PEG concentration (100 g PEG.kg^-1 H2O). The best water potential was 10% PEG concentration for all of the three stains. Temperatures, especially high temperatures induced the fungi death. The optimum temperature for the growth of ectomycorrhizal fungi was 25-28℃ for all of the three stains.展开更多

[Method] PDA medium was used to isolate the fungus from C. abscondi...[Objective] This study aimed to uncover the biological characteristics of a parasitical fungus in Chrysosp/enium absconditicapsu/um J. T. Pan leaves. [Method] PDA medium was used to isolate the fungus from C. absconditicapsulum leaves; PDA medium, modified KB medium and Czapek medium were adopted to cultivate the isolated strain. [Result] Colonies of the strain were stretched, white, fedora- shaped with smooth and wavy edges, and showed diameter growth on PDA medi- um, modified KB medium and Czapek medium. At the late stage of culture, the colonies turned into cyanish brown on the above media. Spores were black and born on the surface of colonies on PDA medium with blackened medium. While on modified KB medium and Czapek medium, they were born at the edge of colonies with blackened medium. The spores varied in a wide range of shapes, mostly ob- clavate, sometimes spherical or ellipsoidal. The conidia were muriformly septate with transverse or longitudinal or oblique septations. The pseudo-beaks were short and cylindrical. [Conclusion] According to relevant literatures, the isolated strain is a fungus in Alternaria, Dematiaceae, Hylohomycetales, Hyphomycetes, Deuteromycotina.展开更多

Methods: Using the total RNA isolated from rat brain as template, the trkB gene was amplified ...Objective: To construct an eukaryotic expression vector carrying rat brain-derived neurotrophic factor receptor trkB gene. Methods: Using the total RNA isolated from rat brain as template, the trkB gene was amplified by reverse-transcription-polymerase chain reaction (RT-PCR) with a pair of specific primers which contained the restrictive sites of EcoR I and BamH I. The amplified fragment of trkB gene was digested with EcoR I and BamH I, and then subcloned into cloning vector pMD18-T and expression vector pEGFP-C2 respectively. The recombinant plasmids were identified by restriction endonuclease enzyme analysis and PCR. Results: The amplified DNA fragment was about 1461 bp in length. Enzyme digestion and PCR analysis showed that the gene of trkB had been successfully cloned into vector pMD18-T and pEGFP-C2. Conclusions: The trkB gene of rat has been amplified and cloned into the eukaryotic expression vector pEGFP-C2.展开更多
