古诗词是中华民族文化的重要组成部分,积淀着我们民族的思想、文化、生活的起源、变化和发展的过程.然而目前中小学语文教学中古诗词教学的现状不甚理想,学生对古诗词普遍缺乏兴趣. 造成这种情况的原因很多,既有古诗词的内容比较深奥,... 古诗词是中华民族文化的重要组成部分,积淀着我们民族的思想、文化、生活的起源、变化和发展的过程.然而目前中小学语文教学中古诗词教学的现状不甚理想,学生对古诗词普遍缺乏兴趣. 造成这种情况的原因很多,既有古诗词的内容比较深奥,词句的意思不容易读懂的原因,也有一些教师和家长自身就没有学习古诗词的兴趣爱好,对学习古诗词的重要意义认识不清,更缺乏引导学生从小学习古诗词的意识,也和学生缺乏文学和历史知识,不了解诗人的生活简历及写作背景,很难真正理解诗意,只会死记硬背,缺乏感情有关系.作为中学语文教师,我们有责任和义务探讨摸索出一条古诗词教学的出路,让更多的学生爱上古诗词,让更多的青年能继承和发扬优秀的传统文化. 展开更多

Cordyceps ophioglossoides is a valuable traditional medicinal material.We have found that intracellular polysaccharide(IPS) is the major biologically active ingredient in Cordyceps ophioglossoides.This study is the first time to optimize the yield of IPS from Cordyceps ophioglossoides.The optimal medium for IPS production consists of glucose 54.50 g·L·1,yeast powder 25.50 g·L·1,NaH2PO4 0.4 g·L·1 and K2HPO4 0.4 g·L·1.The suggested culture conditions are 24 ℃,initial pH 4.5 with a rotary speed of 120 r·min·1 for 168 h.The yield of IPS is 737.93 mg·L·1,which is 50% higher than the yield under the conditions prior to optimization.The anti-oxidative activities of IPS in Cordyceps ophioglossoides L2 are also characterized using various in vitro assay.The anti-oxidative activity may explain the reason why IPS from Cordyceps ophioglossoides can be used to fight against neurodegenerative dis-eases and menopausal symptoms. 展开更多

Determination of Genistein in the Fermented By-product of Soybean Curd, an Indonesian Food, and Its in vivo Assay on Carrageenan-lnduced Mice

In our country, Indonesia, especially in West Java, people frequently consume fermented by-product of soybean curd for their main course. The by-product of curd, usually is discarded, can be fermented using either Neurospora (Neurospora sitophila) or Rhizopus (Rhizopus oligosporus) yeast to make oncom Genistein is a phytoestrogen, an estrogen-like chemical compound present in plants, that has anti-inflammatory property. Genistein can be determined by UV spectrophotometric method based on the UV absorbance of its complex with aluminum chloride. Addition of acid is a critical point in the procedure. The aim of this research was to know whether genistein is still contained in oncom This research also studied about oncom's activity on carrageenan-induced mice. Acetosal was used as drug control. Result showed that genistein was detected contained in oncom extract. The in vivo assay indicated that oncom extract (dose 1,000 mg/kg of body weight) reduced oedema in carragenan-induced paw of mice, which potencies were 066 times lower than that of acetosal's. Oncom might be useful as food and as a weak inflammation reducer as well. 展开更多

Though promoting ferroptosis can reduce cisplatin resistance in tumor cells,ferroptosis and cisplatin resistance in bladder urothelial carcinoma(BUC)following long non-coding RNAs(lncRNAs)is largely unknown.Here,we found the highly expressed lncRNA MAF transcription factor G antisense RNA 1(MAFG-AS1)in BUC,and its inhibition increased the sensitivity of BUC cells to cisplatin by promoting ferroptosis.Mechanically,binding to iron chaperone poly(rC)-binding protein 2(PCBP2)facilitated the recruitments of MAFG-AS1 to deubiquitinase ubiquitin carboxyl-terminal hydrolase isozyme L5(UCHL5),thus stabilizing PCBP2 protein itself.Then PCBP2 was confirmed to interact with ferroportin 1(FPN1),an iron export protein,leading to inhibition of ferroptosis.Moreover,the expression of MAFG-AS1 was regulated by the transcriptional factor MAFG.Interestingly,MAFG-AS1 stimulated MAFG transcription by recruiting histone acetyltransferase p300(EP300)to promote the histone 3 at lysine 27(H3K27ac)at genomic locus of MAFG,forming a MAFG-AS1/MAFG positive feedback loop.In patient samples,higher expression of MAFG-AS1 and MAFG in BUC tissues was significantly correlated with T status and N status,such that MAFG-AS1,MAFG,and the combination of the two were independent prognostic indicators and chemotherapy sensitivity predictive biomarkers for BUC patients.These findings suggest that inhibition of MAFG-AS1 and MAFG can increase the sensitivity of BUC cells to cisplatin through promoting ferroptosis,indicating the novel chemotherapy sensitivity biomarkers and therapeutic target for BUC. 展开更多
