目的川崎病(KD)大部分患者血清中存在抗内皮细胞受体抗体(AECA),本研究探讨KD患者血清中的AECA是否通过细胞内吸附分子-1(ICAM-1)的表达激活和损伤血管内皮细胞。方法检测35例KD患儿血清IgM-AECA水平,并用KD患儿血清及血清中纯化分离的... 目的川崎病(KD)大部分患者血清中存在抗内皮细胞受体抗体(AECA),本研究探讨KD患者血清中的AECA是否通过细胞内吸附分子-1(ICAM-1)的表达激活和损伤血管内皮细胞。方法检测35例KD患儿血清IgM-AECA水平,并用KD患儿血清及血清中纯化分离的IgM刺激人冠状动脉内皮细胞(HCAEC),检测ICAM-1的表达。结果 2/3的KD患儿急性期血清中存在IgM-AECA,IgM-AECA最高的2例患儿均发生冠状动脉瘤。急性期KD患儿血清刺激HCAEC后ICAM-1的表达较用恢复期血清刺激后ICAM-1的表达明显增高〔(120.1±48.5)与(90.5±17.8)〕,用IgM-AECA阳性组血清刺激HCAEC后ICAM-1的表达较用IgM-AECA阴性组血清刺激后ICAM-1的表达明显增高〔(146.3±41.5)与(75.3±14)〕,差异均有统计学意义(P均<0.05)。用AECA阳性IgM刺激后ICAM-1蛋白的表达较用AECA阴性IgM刺激后ICAM-1的表达明显增高〔(0.57±0.9)与(0.26±0.06)〕,差异有统计学意义(P<0.05)。结论 KD患者血清中的IgM-AECA通过诱导ICAM-1的表达激活内皮细胞,IgM-AECA在KD冠状动脉病变发病机制中可能发挥重要作用。 展开更多

本发明公开了一种回收分子筛吸附的特种气体的方法和装置,主要是采用氢气或氦气作为载气,在常温下向分子筛吸附柱内吹气,以撞击分子筛内的特种气体,从而实现常温下直接回收分子筛内的特种气体,回收成本低,容易操作,环保安全,回收率高达... 本发明公开了一种回收分子筛吸附的特种气体的方法和装置,主要是采用氢气或氦气作为载气,在常温下向分子筛吸附柱内吹气,以撞击分子筛内的特种气体,从而实现常温下直接回收分子筛内的特种气体,回收成本低,容易操作,环保安全,回收率高达90%以上,经济效益可观。由于本发明的回收方法无需加热,因此分子筛内吸附的水分等其它杂质不易解吸,使得回收的特种气体杂质含量低,利用价值度高。另外,本发明采用的载气不会引入杂质,且后续直接低温冷凝特种气体即可将特种气体与载气分离,且分离的载体可直接循环使用。[JP3]可见,本发明弥补了目前行业无法直接回收分子筛内吸附的特种气体的技术缺陷,也大大降低了特种气体的提纯成本。 展开更多

目的川崎病(Kawasaki Disease,KD)大部分病人血清中存在抗内皮细胞受体抗体-IgM(IgM An-ti-endothelial cell antibodies,IgM-AECA)。该研究旨在探讨抗内皮细胞受体抗体(AECA)通过何种途径诱导内皮细胞细胞内吸附分子-1(intercellular a... 目的川崎病(Kawasaki Disease,KD)大部分病人血清中存在抗内皮细胞受体抗体-IgM(IgM An-ti-endothelial cell antibodies,IgM-AECA)。该研究旨在探讨抗内皮细胞受体抗体(AECA)通过何种途径诱导内皮细胞细胞内吸附分子-1(intercellular adhesion molecule-1,ICAM-1)的表达以激活血管内皮细胞。方法检测30名KD患儿血清IgM-AECA浓度,并从17名AECA阳性KD患儿血清中提纯IgM-AECA,用IgM-AECA直接刺激人冠状动脉内皮细胞(human coronary artery endothelial cells,HCAEC)后,通过R T-PCR及ELISA方法检测ICAM-1 mR NA及蛋白的表达,同时应用了PD98059,SB203580,dimethylaminopurine(DMAP)和parthenolide,4种蛋白激酶阻断剂观察IgM-AECA通过何种途径激活HCAEC。结果 KD病人血清中的IgM-AECA可使HCAEC的ICAM-1 mRNA及蛋白表达明显增加,SB203580(p38阻断剂)、DMAP(JNK阻断剂)及parthenolide(NF-κB阻断剂)明显抑制ICAM-1的表达(均P<0.01),而PD98059(ERK1/2阻断剂)不能抑制ICAM-1的表达(P>0.05)。结论 KD病人血清中的IgM-AECA通过增强ICAM-1的表达来激活内皮细胞,IgM-AECA通过p38、JNK MAPK及NF-κB通路诱导ICAM-1的表达,IgM-AECA可能在KD冠状动脉病变的发生中发挥重要作用。 展开更多

目的研究活性炭盒法测氡检定过程,样品准备、流转等环节对测量结果的影像,为其他实验室开展类似工作提供经验。方法按照操作步骤准备活性炭盒并邮寄至中国原子能科学研究院,按照实验计划将活性炭盒放置在标准氡室内吸附48h,到达吸附时... 目的研究活性炭盒法测氡检定过程,样品准备、流转等环节对测量结果的影像,为其他实验室开展类似工作提供经验。方法按照操作步骤准备活性炭盒并邮寄至中国原子能科学研究院,按照实验计划将活性炭盒放置在标准氡室内吸附48h,到达吸附时间后密封活性炭盒并将其邮寄回实验室,对活性炭盒进行检测,并计算氡浓度。结果在衰变时间30.48h~50.40h内,氡浓度测量结果分别为533.6Bq/m^(3)、1147.8Bq/m^(3)、2512.4Bq/m^(3),与约定真值相对误差在1.56%~3.84%之间。结论在控制好活性炭盒制备、样品运输、氡室吸附、样品分析等各环节的情况下,可保证活性炭盒氡浓度测量系统检定工作顺利开展。 展开更多

It is hopeful to be used as adsorbent in hemoperfusion for clinical clearance of endotoxin. Gp96, a member of HSP90 family, is a versatile molecular chaperone with various newly-discovered functions, for example to serve as a low affinity, high capacity calcium binding protein, a natural adjuvant for therapeutic cancer vaccines, a tumor rejection antigen, an immune regulator to pathological cell death. Its multi-functional and structural characteristics make it also an interesting target to develop antibody-based therapeutics. However, its low immunogenicity to mice, because of its high-sequence similarity among different species, is an obstacle to obtain valuable monoclonal antibodies (MAbs). This is a common problem for any low immunogenic proteins, whose sequences share close identity between mice and other species. Here, a new strategy of priming was employed by swine endogenous full-length gp96 and then boosting by E. coli-system heterologously expressed gp96 N-terminal fragment (N-355) to generate MAbs. Twelve different highly-specific MAbs against swine/human endogenous gp96 were successfully obtained. The binding activities of these MAbs were confirmed by enzyme-linked immunosorbent assay (ELISA), Western blot (WB), immunofluorescence and flow cytometry analysis. This provides some important reagents for further research and potential therapeutics. The methods employed can be used for MAb production of any sequence-highly-conserved proteins between mice and swine/human (or any other species). 展开更多

Methods: Selec- tive COX-2 inhibitor NS-398 on the growth suppression was evaluated by MTT method. The apoptotic rate was quantified and cell cycle by flow cytometry (FCM). Invasive ability was detected by Transwell. The expression of vascular endothelial growth factor (VEGF) and COX-2 in cholangiocarcinoma line was determined by enzyme-linked immunosorbent assay (ELISA). Results: NS-398 could be time-dose dependently inhibited the growth, induced apoptosis, invasived ability, S phase was inhibited, down-regulate the expression of VEGF and COX-2 in cholangiocarcinoma line (QBC939). Conclusion: NS398 can inhibit proliferation of cholangiocarcinoma cell line QBC939 and inhibit the invasive ability and induce its apoptosis in vitro, which may contributed to the COX-2 dependent pathway to reduce the release of VEGF. 展开更多

Background Inhibition of aging of vascular endothelial cells (VECs) may delay aging and prolong life. The goal of this study was to prepare anti-CD31 monoclonal antibody conjugated PEG-modified liposomes containing the AU-rich region connecting factor 1 (AUF1) gene (CD31-PILs-AUF1) and to explore the effects of targeting CD31-PILs-AUF1 to aging VECs. Methods The mean particle sizes of various PEGylated immunoliposomes (PILs) were measured using a Zetasizer Nano ZS. Gel retardation assay was used to confirm whether PILs had encapsulated the AUF1 plasmid successfully. Fluorescence microscopy and flow cytometry were used to quantify binding of CD31-PILs-AUF1 to target cells. Flow cytometry was also used to analyze the cell cycles of aging bEnd3 cells treated with CD31-PILs-AUF1. We also developed an aging mouse model by treating mice with D-galactose. Enzyme-linked immunosorbent assay (ELISA) was used to evaluate the levels of interleuldn-6 (IL-6) and tumor necrosis factor-or (TNF-ct). The malondialdehyde (MDA) and the superoxide dismutase (SOD) levels were detected by commercial kits. Hematoxylin-eosin (HE) staining was used to determine whether treatment with CD31-PILs-AUF 1 was toxic to the mice. Results CD31-PILs-AUF 1 specifically could targeted bEnd3 VECs and increased the percentage of cells in the S and G2/M phases of aging bEnd3 cells. ELISA showed that content of the IL-6 and TNF-ct decreased in CD31-PILs-AUF1 group. The level of SOD increased, whereas MDA decreased in the CD31-PILs-AUF1 group. Additionally, CD31-PILs-AUF 1 was not toxic to the mice. Conclusion CD31-PILs-AUF 1 targets VECs and may delay their senescence. 展开更多

We prepared a novel pitch-based activated carbon microsphere(ACM)and studied its in vitro adsorption characteristics for biological molecules.The original spherical asphalt particles were prepared through bitumen emulsification.After oxidation and burning,ACMs with a range of parameters were prepared.In vitro adsorption experiments of ACMs for biological molecules were carried out.The prepared ACMs possessed a BET specific surface of 1566 m^2/g with a voidage of 0.653 cm^3/g and a volume of micropores of 0.478 cm^3/g.They showed high adsorption for glucose and creatintine.Compared with the medically used carbon powder,the prepared ACMs exhibited significantly lower adsorption for digestion enzymes.In conclusion,the prepared ACMs,as an oral adsorbent candidate,possessed higher BET specific surface area and larger volume of micropores;they also exhibited favorable selective adsorption features for biological molecules. 展开更多

OBJECTIVE: To explore the long-term effects and pain relief mechanism of acupotomy by observing changes in nitric oxide synthase (NOS) and beta-en- dorphin (~3-EP) in the hypothalamus, spinal cord, and peripheral blood of rats with third lumbar ver- tebrae (L3) transverse process syndrome. METHODS: Twenty-eight SD rats were randomly as- signed to normal, model, electroacupuncture (EA), and acupotomy group. The last three groups were put through an operation to emulate L3 transverse process syndrome. Fourteen days after the simulation operation, EA and acupotomy treatments were applied to the respective groups. Fifty-six days afterthe simulation operation, biochemistry tests and enzyme-linked immunosorbent assay were used to measure NOS and 13-EP in the hypothalamus, spinal cord, and peripheral blood. RESULTS: Rats with the simulation operation showed significantly higher levels of NOS and II3-EP in the hypothalamus, spinal cord, and peripheral blood than those in the normal group. The EA and acupotomy groups had significantly lower levels of NOS and β-EP than those in the model group. There was no statistical difference between the EA and acupotomy groups. CONCLUSION: EA and acupotomy treatments significantly lowered NOS and β-EP levels in the hypothalamus, spinal cord, and peripheral blood and alleviated L3 transverse process syndrome. 展开更多
