EphB4/ephrinB2逆向信号对RAW264.7破骨细胞分化中PDZ结构域蛋白表达变化的研究 被引量:2

Eph受体是酪氨酸蛋白激酶受体家族中最大的亚家族,ephrin(Eph受体相互作用蛋白)是其配体,它们是膜结合蛋白,相互依赖进行信号转导.内居蛋白(syntenin)与Pick1属于PDZ结构域(PSD-95/Dlg-/Zo-1 domain)蛋白,报道称能与ephrinB配体结合,但是否受Eph受体调控尚未见报道.以RAW264.7细胞株为研究对象,通过蛋白质印迹及/或免疫荧光分析显示RAW264.7细胞经RANKL诱导的破骨细胞表达ephrinB2、内居蛋白(syntenin)和Pick1三个蛋白质.将提前成簇的可溶性EphB4蛋白加入培养液,与ephrinB2配体结合,用来研究EphB4/ephrinB2逆向信号对syntenin和Pick1表达水平变化的影响.免疫印迹及Real-time RT-PCR分析结果显示,在EphB4-Fc实验组中Pick1的蛋白质及mRNA水平都有明显增加,然而在EphB4-Fc实验组与Fc对照组别间syntenin的蛋白质及mRNA水平未见明显变化.免疫共沉淀结果显示,syntenin和Pick1不能与ephrinB2共沉淀.以上结果初步探索了体外破骨细胞分化过程中,EphB4/ephrinB2逆向信号对PDZ结构域蛋白(ephrinB2配体潜在的下游信号分子)表达变化的调控. 展开更多

A case study is performed on the influence of the patio on the indoor environment of a traditional folk house group,named Zhangguying village in Hunan province,in the summer of 2007.Measurements include indoor and outdoor air temperature,indoor and outdoor illuminance,indoor and outdoor air speed,and carried out from August 5th to 10th.The results show that the patio,acting as a "buffer zone",can reduce the ambient impacts on the indoor thermal environment of rooms because the temperature of the patio is lower than that of the outdoor temperature but higher than that of the rooms.The patio can improve the indoor lighting environment because the illuminance of the patio is lower than that of outdoor and higher than that of the rooms.But the effect is too limited,because the illuminance of the rooms is lower than the national standards.This study shows that the shading design is the primary consideration in this kind of climate.The wind speed of the patio is stable and similar to that of the hall and the stack effect of the patio is not obvious.It shows that the patio is useful for natural ventilation,caused by wind pressure,in summer. 展开更多

A number of alterations have been found within the gutmicrobial profile of patients with inflammatory bowel diseases when compared with the healthy population; however, it is unclear whether such dysbiosis is the cause or simply the consequence of the disease state. In ulcerative colitis, the environment seems to play a crucial role in disease etiology since monozygotic twins show a concordance rate of only 8%-10%-though it is unclear whether it does so by acting through the microbiome. In this study, the authors investigated the influence of cohabitation on the gut microbial community in healthy partners of ulcerative colitis patients-with the intent of clarifying some of these issues. As expected, ulcerative colitis patients had a significant dysbiosis and alterations in microbial metabolism. Interestingly, these abnormal fecal microbial communities were relatively similar amongst patients and their spouses. Thus, this study shows that the microbial profile might be partially transferred from ulcerative colitis patients to healthy individuals. Whether this finding impacts on disease development or has any implication for the role of the microbiome in inflammatory bowel disease etiology remains to be determined. 展开更多

It is possible to identify distinct occupational stains within the neighborhood, specifically in the cell one in this article, as a pioneer construction, due to housing dimension that corresponds to different housing typologies, residents and daily use areas, equipment. The objective is therefore, to understand how these three factors are reflected inside the cell, managing occupational stains which differ in space appropriation. How those different occupational stains that derive from the housing and urban morphology, may result in the occupation and experience of its inhabitants, not invalidating a cohesive structure of the neighborhood? 展开更多

Remotely operated vehicles(ROVs) are unique tools for underwater industrial exploration and scientific research of offshore areas and the deep ocean. With broadening application of ROVs, the study of factors that affect their safe operation is important. Besides the technical skills to control ROV movement, the dynamical ocean environment may also play a vital role in the safe operation of ROVs. In this paper, we investigate the influence of large-amplitude internal solitary waves(ISWs), focusing on the forces exerted on ROVs by ISWs. We present a methodology for modeling ISW-induced currents based on Kd V model and calculate the ISW forces using the Morrison equation. Our results show that an extremely considerable load is exerted by the ISW on the ROV, resulting in a strong disturbance of the vehicle's stability, affecting the ROV control. The numerical results of this work emphasize the importance of considering dynamical conditions when operating underwater vessels, such as ROV. Further laboratory and field investigation are suggested to gain more understanding of this subject. 展开更多

In this paper, we present neighborhood-following algorithms for linear programming. When the neighborhood is a wide neighborhood, our algorithms are wide neighborhood primal-dual interior point algorithms. If the neighborhood degenerates into the central path, our algorithms also degenerate into path-following algorithms. We prove that our algorithms maintain the O9√nL) -iteration complexity still, while the classical wide neighborhood primal-dual interior point algorithms have only the O(nL-iteration complexity. We also proved that the algorithms are quadratic convergence if the optimal vertex is nondegenerate. Finally, we show some computational results of our algorithms. 展开更多
