In the present study, we developed a high performance liquid chromatography (HPLC) method for the determination of alantolactone in rat plasma, and pharmacokinetics of alantolactone was investigated after intravenous and oral administrations to Wistar rats. Separation was achieved on C18 column (4.6 mm×250 mm, 5.0 μm) using a mobile phase consisting of methanol-water (70:30, v/v) at a flow rate of 1.0 mL/min. The wavelength of the ultraviolet detector was set at 239 nm. The excellent linearity was found over a concentration range of 0.08-10 μg/mL (R2 = 0.9998). The intra- and inter-day precisions were good, and the RSD was lower than 2.27%. The mean absolute recovery of alantolactone in plasma ranged from 88.09% to 95.57%. After intravenous administration, alantolactone showed rapid systemic clearance (CL (0.11±0.014) L/h/kg) and small volume of distribution (Vd (0.71±0.14) L/kg). The biological half life (t1/2) was 56.24 min. After oral administration, alantolactone showed rapid oral absorption in rats, with a short Tmax of (45.02±0.88) and (45.13±0.39) min for 14 and 28 mg/kg, respectively. The bioavailability of alantolactone in rats was 50.88%, indicating that alantolactone was orally available.展开更多

However, the potential of endophytes within these bio-prospective medicinal plants remains unknown. The present study elucidates the endophytic diversity of medicinal plants(Caralluma acutangula, Rhazya stricta, and Moringa peregrina) and the endophyte role in seed growth and oxidative stress. Various organs of medicinal plants yielded ten endophytes, which were identified as Phoma sp.(6 isolates), Alternaria sp.(2), Bipolaris sp.(1), and Cladosporium sp.(1) based on 18 S rD NA sequencing and phylogenetic analysis. The culture filtrates(CFs; 25%, 50%, and 100% concentrations) from these endophytes were tested against the growth of normal and dwarf mutant rice lines. Endophytic CF exhibited dose-dependent growth stimulation and suppression effects. CF(100%) of Phoma sp. significantly increased rice seed germination and growth compared to controls and other endophytes. This growth-promoting effect was due to the presence of indole acetic acid in endophytic CF. The gas chromatography/mass spectrometry(GC/MS) analysis showed the highest indole acetic acid content((54.31±0.21) μmol/L) in Bipolaris sp. In addition, the isolate of Bipolaris sp. exhibited significantly higher radical scavenging and anti-lipid peroxidation activity than the other isolates. Bipolaris sp. and Phoma sp. also exhibited significantly higher flavonoid and phenolic contents. The medicinal plants exhibited the presence of bio-prospective endophytic strains, which could be used for the improvement of crop growth and the mitigation of oxidative stresses.展开更多

Methods: Seventy-eight patients were randomly divided into a combined acupuncture and herbs group...Objective: To compare the clinical efficacy of combined acupuncture and herbs with danazol in treating endometriosis. Methods: Seventy-eight patients were randomly divided into a combined acupuncture and herbs group (n= 40) and a Western medicine control group (n= 38). Observation was made on changes in clinical symptoms, signs, serum CA125,β-EP, PGE2 and PGF2α, and adverse reaction to the medicine in the two groups. Results: The overall efficacy was similar in the two groups, but the effects on lumbosacral pain, anal downbearing distention, irregular menstruation and infertility were significantly better in the treatment group than in the control group (P(0.05), and in the treatment group serum CA125, PGE2 and PGF2α were significantly lower and serum β-EP was significantly higher after treatment than before (P(0.05). Conclusion: Combined use of acupuncture and herbs has a marked effect on endometriosis and the adverse reaction is lower than the control group.展开更多
