[Method] Various species of healthy Rosa chinensis were col- lected from Xi'an, Xianyang, Baoji and Weinan City in Shaanxi Province. Endophytic fungi was isolated from stems and leaves, and purified by 2-3 times of inoculation to screen endophytic fungi antagonistic to Marssonina rosae with modified punching method. [Result] Samples collected from Xianyang exhibited the highest colonization rate and isolation rate; endophytic fungi strains isolated from stems presented the highest colonization rate and isolation rate compared with leaves. A total of 67 en- dophytic fungi strains were isolated from Rosa chinensis, including 3 black spot-re- sistant strains which were all derived from Baoji. [Conclusion] This study laid the foundation for further screening candidate strains of biocontrol fungi and environ- ment-friendly fungal biological control.展开更多

[Meth...[Objective] This study was conducted to evaluate the effect of 1-methylcy- clopropene (1-MCP) on fruit firmness, and the activity of the enzymes involved in ethylene metabolism and membrane lipid peroxidation. [Method] The nearly ripe fruits of the papaya cultivar Risheng were randomly assigned to one of four groups. Two groups were treated under hypobaric and hypoxic (HH) atmosphere condition for six hours, and immediately soaked in deionized water (HH alone), or fumigated with 2.0 mg/L 1-MCP (HH+I-MCP) for 24 h. The other two groups untreated under HH condition were also soaked in deionized water (negative control), or fumigated with 2.0 mg/L 1-MCP (1-MCP alone) for 24 h. After that, the fruits of all the four treatments were stored at room temperature (23+1) ℃. Cell membrane permeability, fruit firmness, respiration rate, ethylene release rate, SOD activity, POD activity, CAT activity, MAD content and LOX activity were measured once every three days during storage. [Result] Treatment with 1-MCP delayed the occurrence of the peaks of respiration rate and ethylene release rate, significantly reduced the accumulation of malondialdehyde (MDA), and inhibited the decrease in papaya fruit firmness. Compared with the control, 1-MCP treatment significantly increased the SOD (su- peroxide dismutase), POD (peroxidase) and CAT (catalase) activity, reduced the ac- tivity of lipoxygenase (LOX), a product of lipid peroxidatlon in membranes, and in- hibited ethylene biosynthesis, thus delaying the aging process and prolonging the storage life of papaya fruits. [Conclusion] The results will provide a theoretical basis for analvzina the key factors controllinq postharvest maturity and aging of papaya fruits.展开更多

METHODS: Forty-two Sprague-Dawley rats were randomly dMded into model group (n ...AIM: To explore the relationship between changes of intestinal environment and pathogenesis of non-alcoholic steatohepatitis (NASH). METHODS: Forty-two Sprague-Dawley rats were randomly dMded into model group (n = 24), treatment group (n = 12), and control group (n = 6). The rats of model and treatment groups were given high-fat diet, and those of the control group were given normal diet. Furthermore, the rats of treatment group were given lactulose after 8 wk of high-fat diet. Twelve rats of the model group were killed at 8 wk of high-fat diet. At the 16 wk the rats of treatment group, control group, and the rest of the model group were killed. The serum levels of aminotransferase were measured and the histology of livers was observed by H&E staining. RESULTS: The livers of rats presented the pathological features of steatohepatitis with higher serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in the model group after 16 wk. Compared to the model group, the serum levels of ALT and AST in treatment group decreased significantly and were close to the normal group, and the hepatic inflammation scores also decreased markedly than those in the model group after 16 wk (5.83±2.02 vs 3.63±0.64, P〈0.05), but were still higher than those in the model group after 8 wk (3.63±0.64 vs 1.98±0.90, P〈0.05). However, the degree of hepatic steatosis had no changes in treatment group compared to the model group after 16 wk. CONCLUSION: Lactulose could ameliorate the hepatic inflammation of rats with steatohepatitis induced by fat- rich diet, but could not completely prevent the development of steatohepatitis. It is suggested that intestinal environmental changes such as intestinal bacteria overgrowth, are one of the important factors in the pathogenesis of NASH.展开更多

These membrane pr...The molecular mechanism of how hepatocytes maintain cholesterol homeostasis has become much more transparent with the discovery of sterol regulatory element binding proteins (SREBPs) in recent years. These membrane proteins aremembers of the basic helix-loop-helix-leucine zipper (bHLHZip) family of transcription factors. They activate the expression of at least 30 genes involved in the synthesis of cholesterol and lipids. SREBPs are synthesized as precursor proteins in the endoplasmic reticulum (ER), where they form a complex with another protein, SREBP cleavage activating protein (SCAP). The SCAP molecule contains a sterol sensory domain. In the presence of high cellular sterol concentrations SCAP confines SREBP to the ER. With low cellular concentrations, SCAP escorts SREBP to activation in the Golgi. There, SREBP undergoes two proteolytic cleavage steps to release the mature, biologically active transcription factor, nuclear SREBP (nSREBP). nSREBP translocates to the nucleus and binds to sterol response elements (SRE) in the promoter/enhancer regions of target genes. Additional transcription factors are required to activate transcription of these genes. Three different SREBPs are known, SREBPs-1a, -1c and -2. SREBP-1a and -1c are isoforms produced from a single gene by alternate splicing. SREBP-2 is encoded by a different gene and does not display any isoforms. It appears that SREBPs alone, in the sequence described above, can exert complete control over cholesterol synthesis, whereas many additional factors (hormones, cytokines, etc.) are required for complete control of lipid metabolism. Medicinal manipulation of the SREBP/SCAP system is expected to prove highly beneficial in the management of cholesterol-related disease.展开更多

A rim thickness parameter is defined as the flexibility of internal ring gear. Six evenly spaced linear springs are used to describe the fitting status between internal ring gear and the gearcase. The finite element model of the whole internal ring gear is established by means of Pro/E and ANSYS. The loads on meshing teeth of internal ring gear are applied according to the contact ratio and the load-sharing coefficient. With the finite element analysis (FEA), the influences of flexibility and fitting status on the stress and elastic deflection of internal ring gear are predicted. The simulation reveals that the principal stress and deflection increase with the decrease of rim thickness of internal ring gear. Moreover, larger spring stiffness helps to reduce the stress and deflection of internal ring gear. Therefore, the flexibility of internal ring gear must be considered during the design of high-speed planetary gear transmissions.展开更多

Indoor air quality will be improved by the ded...The hybrid system with radiant cooling and dedicated outdoor air not only possesses high energy efficiency, but also creates a healthy and comfortable indoor environment. Indoor air quality will be improved by the dedicated outdoor air system(DOAS) and indoor thermal comfort can be enhanced by the radiant cooling system(RCS). The optimal air-supply mode of the hybrid system and the corresponding design approach were investigated. A full-scale experimental chamber with various air outlets and the ceiling radiant cooling panels(CRCP) was designed and established. The performances of different air-supply modes along with CRCPs were analyzed by multi-index evaluations. Preliminary investigations were also conducted on the humidity stratification and the control effect of different airflow modes to prevent condensation on CRCP. The overhead supply air is recommended as the best combination mode for the hybrid system after comprehensive comparison of the experiment results. The optimal proportion of CRCP accounting for the total cooling capacities in accord with specific cooling loads is found, which may provide valuable reference for the design and operation of the hybrid system.展开更多

With the equivalent crack length Ieq of the wing crack introduced, the stress intensity factor Kl at the wing crack tip was as- sumed to the sum of two terms: on one hand a component K1^(1) for a single isolated straight wing crack of length 21, and subjected to hydraulic pressure in the wing crack and far-field stresses; on the other hand a component K1(2) due to the effective shear stress induced by the presence of the equivalent main crack. The lateral tensile stress and hydraulic high pressure are the key factors that induce crack propagation unsteadily. The new wing crack theoretical model proposed can supply references for the study on hydraulic fracture in fractured masses, hydraulic fracturing in rock masses.展开更多

Acetylspiramycin did not form the complexes, so potassium ferricyanide showed no effect on the PQT metabolism in microsomes from acetylspiramycin-treated rats. Triacetyloleandomycin,a specific inhibitor of cytochrome P-450ⅢAI, inhibited PQT metabolism by 53% in liver microsomes from dexamethasone-treated rats.These results indicate the cytochrome P-450ⅢA seems to be involved in metabolism of PQT in rat liver microsomes.展开更多

Most traditional technologies usual...Transceiver-free object localization can localize target through using Radio Frequency(RF) technologies without carrying any device, which attracts many researchers' attentions. Most traditional technologies usually first deploy a number of reference nodes which are able to communicate with each other, then select only some wireless links, whose signals are affected the most by the transceiver-free target, to estimate the target position. However, such traditional technologies adopt an ideal model for the target, the other link information and environment interference behavior are not considered comprehensively. In order to overcome this drawback, we propose a method which is able to precisely estimate the transceiver-free target position. It not only can leverage more link information, but also take environmental interference into account. Two algorithms are proposed in our system, one is Best K-Nearest Neighbor(KNN) algorithm, the other is Support Vector Regression(SVR) algorithm. Our experiments are based on Telos B sensor nodes and performed in different complex lab areas which have many different furniture and equipment. The experiment results show that the average localization error is round 1.1m. Compared with traditional methods, the localization accuracy is increased nearly two times.展开更多

We define a novel graphbased semantic region descriptions, which are estimated in a dynamically fashion. We propose a two-level update algorithm, namely, Symbols update level and Regions update level. The algorithm firstly adopts particle filter to update weights of the symbols, and then use the Viterbi algorithm to estimate the region the robot stays in based on those weights, optimally. Experimental results demonstrate that our proposed approach can solve problems of the long-term operation and kidnapped robot problem.展开更多

To evaluate the influence of the added external circulation on the development of granular sludge, the characteristics of the granular sludge taken from the two tested laboratory-scale reactors during start-up were studied. The results show that the added external circulation can enhance biomass granulation, accelerate granule development and improve sludge characteristics. At the end of start-up, the granular size of sludge in external circulation-added IC reactor greatly increases with a size distribution much better than that of sludge in IC reactor. The granular sludge originated from external circulationadded IC reactor contains more extracellular polymers and has a greater settling velocity than that from IC reactor. Methanogenic activity of the granular sludge from the external circulationadded IC reactor started 26 d ago reaches 358.23mL·g -1 ·d -1 , 1.66 and 1.20 times as great as that of the sludge from the IC reactor started 26 d and 58 d ago respectively.展开更多

The streamline and hydrodynamic parameters were measured in a 2-D airlift loop reactor(ALR)with different draft baffles. Three regimes were observed under different conditions. Particle image velocimetry(PIV)measurement showed that the liquid velocity distribution in horizontal direction presented different profiles in the three regimes. The length, the height and the spacing of draft baffles were applied in the experiments to optimize the ALR structure. It was found that the draft tube structure is of great importance in determining the hydrodynamics of ALRs. Additionally, the experimental results may serve as a step to the further optimization and design of ALR.展开更多

Methods Intracellular cholesterol accumulation was measured by Oil Red O staining and high performance liquid chromatography. The effects of rapamycin on interleukin-1β(1L-1β)-induced mRNA and protein changes of low-density lipoprotein receptor (LDLR) and ATP-binding cassette transporter Al (ABCAl) were assayed by quantitative real-time PCR and Western blot. Transient expressions of 3 types of mammalian target of rapamycin (mTOR), including mTOR-WT (wild type), mTOR-RR (rapamycin resistant, with kinase activity), and mTOR-RR-KD (rapamycin resistant, without kinase activity), were obtained by plasmid transfection. Results Rapamycin had no significant influence on intracellular cholesterol concentration trader normal condition, but it significantly decreased the intracellular cholesterol concentration in the presence of IL-1β. Rapamycin dose-dependently suppressed the increased expression of LDLR induced by IL-1β and up-regulated the suppressed expression of ABCAl caused by IL-1β Transient expression of 3 types of mTOR all reduced ABCAl mRNA expression significantly, which all could be overroded by rapamycin. Conclusions Rapamycin may contribute to the maintaining of glomerular mesangial cell intracellular cholesterol homeostasis under inflammatory state by both reducing cholesterol uptake and increasing cholesterol effiux. And the effect may be not completely mediiated by mTOR.展开更多
Energy performance assessment on central air-conditioning system is essential to optimize operating, reduce operating costs, improve indoor environmental quality, and determine whether the retrofitting of the equipmen...Energy performance assessment on central air-conditioning system is essential to optimize operating, reduce operating costs, improve indoor environmental quality, and determine whether the retrofitting of the equipment is necessary. But it is difficult to evaluate it reasonably and comprehensively due to its complexity. A "holistic" approach was discussed to evaluate the energy performance of central air-conditioning system for an extra-large commercial building in a subtropical city. All procedures were described in detail, including field investigation method, field measurement instruments, data processing and data analyzing. The main factors affecting energy consumption of air-conditioning system were analyzed and the annual cooling-energy use intensity of this building was calculated and also compared with other shopping malls and other types of buildings in Guangzhou. And COP(coefficient of performance) of chiller, water transfer factor of chilled water system and cooling water system were taken into consideration. At last, the thermal comfort and indoor air quality issues were addressed. The results show that the chilled water pumps are over-sized and the indoor environmental quality should be improved. The purpose of this work is to provide reference for energy performance assessment method for air-conditioning system.展开更多

METHODS: Forty-two Sprague-Dawley rats were randomly dMded into model group (n = 24), treatment group (n = 12), and control group (n = 6). The rats of model and treatment groups were given high-fat diet, and those of the control group were given normal diet. Furthermore, the rats of treatment group were given lactulose after 8 wk of high-fat diet. Twelve rats of the model group were killed at 8 wk of high-fat diet. At the 16 wk the rats of treatment group, control group, and the rest of the model group were killed. The serum levels of aminotransferase were measured and the histology of livers was observed by H&E staining. RESULTS: The livers of rats presented the pathological features of steatohepatitis with higher serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in the model group after 16 wk. Compared to the model group, the serum levels of ALT and AST in treatment group decreased significantly and were close to the normal group, and the hepatic inflammation scores also decreased markedly than those in the model group after 16 wk (5.83±2.02 vs 3.63±0.64, P〈0.05), but were still higher than those in the model group after 8 wk (3.63±0.64 vs 1.98±0.90, P〈0.05). However, the degree of hepatic steatosis had no changes in treatment group compared to the model group after 16 wk. CONCLUSION: Lactulose could ameliorate the hepatic inflammation of rats with steatohepatitis induced by fat- rich diet, but could not completely prevent the development of steatohepatitis. It is suggested that intestinal environmental changes such as intestinal bacteria overgrowth, are one of the important factors in the pathogenesis of NASH.
文摘The molecular mechanism of how hepatocytes maintain cholesterol homeostasis has become much more transparent with the discovery of sterol regulatory element binding proteins (SREBPs) in recent years. These membrane proteins aremembers of the basic helix-loop-helix-leucine zipper (bHLHZip) family of transcription factors. They activate the expression of at least 30 genes involved in the synthesis of cholesterol and lipids. SREBPs are synthesized as precursor proteins in the endoplasmic reticulum (ER), where they form a complex with another protein, SREBP cleavage activating protein (SCAP). The SCAP molecule contains a sterol sensory domain. In the presence of high cellular sterol concentrations SCAP confines SREBP to the ER. With low cellular concentrations, SCAP escorts SREBP to activation in the Golgi. There, SREBP undergoes two proteolytic cleavage steps to release the mature, biologically active transcription factor, nuclear SREBP (nSREBP). nSREBP translocates to the nucleus and binds to sterol response elements (SRE) in the promoter/enhancer regions of target genes. Additional transcription factors are required to activate transcription of these genes. Three different SREBPs are known, SREBPs-1a, -1c and -2. SREBP-1a and -1c are isoforms produced from a single gene by alternate splicing. SREBP-2 is encoded by a different gene and does not display any isoforms. It appears that SREBPs alone, in the sequence described above, can exert complete control over cholesterol synthesis, whereas many additional factors (hormones, cytokines, etc.) are required for complete control of lipid metabolism. Medicinal manipulation of the SREBP/SCAP system is expected to prove highly beneficial in the management of cholesterol-related disease.

文摘The stress and the elastic deflection of internal ring gear in high-speed spur planetary gear units are investigated. A rim thickness parameter is defined as the flexibility of internal ring gear. Six evenly spaced linear springs are used to describe the fitting status between internal ring gear and the gearcase. The finite element model of the whole internal ring gear is established by means of Pro/E and ANSYS. The loads on meshing teeth of internal ring gear are applied according to the contact ratio and the load-sharing coefficient. With the finite element analysis (FEA), the influences of flexibility and fitting status on the stress and elastic deflection of internal ring gear are predicted. The simulation reveals that the principal stress and deflection increase with the decrease of rim thickness of internal ring gear. Moreover, larger spring stiffness helps to reduce the stress and deflection of internal ring gear. Therefore, the flexibility of internal ring gear must be considered during the design of high-speed planetary gear transmissions.

文摘The hybrid system with radiant cooling and dedicated outdoor air not only possesses high energy efficiency, but also creates a healthy and comfortable indoor environment. Indoor air quality will be improved by the dedicated outdoor air system(DOAS) and indoor thermal comfort can be enhanced by the radiant cooling system(RCS). The optimal air-supply mode of the hybrid system and the corresponding design approach were investigated. A full-scale experimental chamber with various air outlets and the ceiling radiant cooling panels(CRCP) was designed and established. The performances of different air-supply modes along with CRCPs were analyzed by multi-index evaluations. Preliminary investigations were also conducted on the humidity stratification and the control effect of different airflow modes to prevent condensation on CRCP. The overhead supply air is recommended as the best combination mode for the hybrid system after comprehensive comparison of the experiment results. The optimal proportion of CRCP accounting for the total cooling capacities in accord with specific cooling loads is found, which may provide valuable reference for the design and operation of the hybrid system.

文摘A new wing crack model subjected to hydraulic pressure and far-field stresses was proposed considering the effect of hydraulic pressure in wing crack and the connected part of the main crack on the stress intensity factor at the wing crack tip. With the equivalent crack length Ieq of the wing crack introduced, the stress intensity factor Kl at the wing crack tip was as- sumed to the sum of two terms: on one hand a component K1^(1) for a single isolated straight wing crack of length 21, and subjected to hydraulic pressure in the wing crack and far-field stresses; on the other hand a component K1(2) due to the effective shear stress induced by the presence of the equivalent main crack. The lateral tensile stress and hydraulic high pressure are the key factors that induce crack propagation unsteadily. The new wing crack theoretical model proposed can supply references for the study on hydraulic fracture in fractured masses, hydraulic fracturing in rock masses.
文摘Our results show that in liver microsomes from erythromycin,acetylspiramycin and dexamethsone pretreated rats,the rate of praziquantel( PQT)disappearence was increased as compared with control rat When microsomes from erythromycin-treated rats were exposed to PQT in the presence of potassium ferricyanide which broke down the cytochrome P-450 Fe(Ⅱ)-metabolite complexes and restored the functional cytochrome P-450,PQT metabolism was further increased. Acetylspiramycin did not form the complexes, so potassium ferricyanide showed no effect on the PQT metabolism in microsomes from acetylspiramycin-treated rats. Triacetyloleandomycin,a specific inhibitor of cytochrome P-450ⅢAI, inhibited PQT metabolism by 53% in liver microsomes from dexamethasone-treated rats.These results indicate the cytochrome P-450ⅢA seems to be involved in metabolism of PQT in rat liver microsomes.

文摘Transceiver-free object localization can localize target through using Radio Frequency(RF) technologies without carrying any device, which attracts many researchers' attentions. Most traditional technologies usually first deploy a number of reference nodes which are able to communicate with each other, then select only some wireless links, whose signals are affected the most by the transceiver-free target, to estimate the target position. However, such traditional technologies adopt an ideal model for the target, the other link information and environment interference behavior are not considered comprehensively. In order to overcome this drawback, we propose a method which is able to precisely estimate the transceiver-free target position. It not only can leverage more link information, but also take environmental interference into account. Two algorithms are proposed in our system, one is Best K-Nearest Neighbor(KNN) algorithm, the other is Support Vector Regression(SVR) algorithm. Our experiments are based on Telos B sensor nodes and performed in different complex lab areas which have many different furniture and equipment. The experiment results show that the average localization error is round 1.1m. Compared with traditional methods, the localization accuracy is increased nearly two times.

文摘In this work, in order to improve spatial recognition abilities for the long-term operation tasks of the assistant robot for the elderly, a novel approach of semantic region estimation is proposed. We define a novel graphbased semantic region descriptions, which are estimated in a dynamically fashion. We propose a two-level update algorithm, namely, Symbols update level and Regions update level. The algorithm firstly adopts particle filter to update weights of the symbols, and then use the Viterbi algorithm to estimate the region the robot stays in based on those weights, optimally. Experimental results demonstrate that our proposed approach can solve problems of the long-term operation and kidnapped robot problem.
文摘The start-up of external circulationadded internal circulation(IC) reactor was finished in 26 d, 32 d fewer than that of IC reactor. To evaluate the influence of the added external circulation on the development of granular sludge, the characteristics of the granular sludge taken from the two tested laboratory-scale reactors during start-up were studied. The results show that the added external circulation can enhance biomass granulation, accelerate granule development and improve sludge characteristics. At the end of start-up, the granular size of sludge in external circulation-added IC reactor greatly increases with a size distribution much better than that of sludge in IC reactor. The granular sludge originated from external circulationadded IC reactor contains more extracellular polymers and has a greater settling velocity than that from IC reactor. Methanogenic activity of the granular sludge from the external circulationadded IC reactor started 26 d ago reaches 358.23mL·g -1 ·d -1 , 1.66 and 1.20 times as great as that of the sludge from the IC reactor started 26 d and 58 d ago respectively.

文摘In this paper, a 2-D airlift reactor was developed. The streamline and hydrodynamic parameters were measured in a 2-D airlift loop reactor(ALR)with different draft baffles. Three regimes were observed under different conditions. Particle image velocimetry(PIV)measurement showed that the liquid velocity distribution in horizontal direction presented different profiles in the three regimes. The length, the height and the spacing of draft baffles were applied in the experiments to optimize the ALR structure. It was found that the draft tube structure is of great importance in determining the hydrodynamics of ALRs. Additionally, the experimental results may serve as a step to the further optimization and design of ALR.
文摘Objective To investigate the effects of rapamycin on cholesterol homeostasis of glomerular mesangial cells and the underlying mechanisms. Methods Intracellular cholesterol accumulation was measured by Oil Red O staining and high performance liquid chromatography. The effects of rapamycin on interleukin-1β(1L-1β)-induced mRNA and protein changes of low-density lipoprotein receptor (LDLR) and ATP-binding cassette transporter Al (ABCAl) were assayed by quantitative real-time PCR and Western blot. Transient expressions of 3 types of mammalian target of rapamycin (mTOR), including mTOR-WT (wild type), mTOR-RR (rapamycin resistant, with kinase activity), and mTOR-RR-KD (rapamycin resistant, without kinase activity), were obtained by plasmid transfection. Results Rapamycin had no significant influence on intracellular cholesterol concentration trader normal condition, but it significantly decreased the intracellular cholesterol concentration in the presence of IL-1β. Rapamycin dose-dependently suppressed the increased expression of LDLR induced by IL-1β and up-regulated the suppressed expression of ABCAl caused by IL-1β Transient expression of 3 types of mTOR all reduced ABCAl mRNA expression significantly, which all could be overroded by rapamycin. Conclusions Rapamycin may contribute to the maintaining of glomerular mesangial cell intracellular cholesterol homeostasis under inflammatory state by both reducing cholesterol uptake and increasing cholesterol effiux. And the effect may be not completely mediiated by mTOR.

文摘Energy performance assessment on central air-conditioning system is essential to optimize operating, reduce operating costs, improve indoor environmental quality, and determine whether the retrofitting of the equipment is necessary. But it is difficult to evaluate it reasonably and comprehensively due to its complexity. A "holistic" approach was discussed to evaluate the energy performance of central air-conditioning system for an extra-large commercial building in a subtropical city. All procedures were described in detail, including field investigation method, field measurement instruments, data processing and data analyzing. The main factors affecting energy consumption of air-conditioning system were analyzed and the annual cooling-energy use intensity of this building was calculated and also compared with other shopping malls and other types of buildings in Guangzhou. And COP(coefficient of performance) of chiller, water transfer factor of chilled water system and cooling water system were taken into consideration. At last, the thermal comfort and indoor air quality issues were addressed. The results show that the chilled water pumps are over-sized and the indoor environmental quality should be improved. The purpose of this work is to provide reference for energy performance assessment method for air-conditioning system.
