[Method] With mature seeds of C. dayanum as explants, different media were selected to establish asymbiotic germination technique system. With protocorms as materials, conservation, resumptive proliferation and plant regeneration conditions were selected to establish low-temperature in vitro conservation technique system preliminarily. [Result] Mature seeds of C. dayanum could germinate after cultured 90 days on MS media as well as "Hyponex 1" media. The germination rate reached more than 98%. Protocorms inoculated in "Hyponex 1" media could be conserved continuously at 5 ℃ in dark for more than 18 months and the survival rate could reach 90%. Conserved protocorms could realize resumptive preliferation culture both on 1/2 MS and "Hyponex 1" media. The seed- ling-strengthening and rooting media were 1/2 MS media. [Conclusion] This research provided practical basis for in vitro conservation and rapid propagation of C. dayanum germplasm resource.展开更多

Of the 402 nucleotides, twenty-seven (6.72%) are polymorphic and all are synonymous substitutions. At the third positions of genetic condon of cytochrome b gene, the two species show an extreme anti-G bias (<4%) and a pronounced bias towards A and C (>68%). There is no amino acid sequence divergence between the partial cytochrome b genes of the two species, indicating a close genetic relationship between them. The k-2p genetic distance of partial cytochrome b segment of the two species is 0.072, suggesting that the species were separated 3.6 Ma ago, in the middle Pliocene. Our result reveals that the cytochrome b gene is an appropriate marker for studies of population genetic structures and phylogeographic pat- terns of the two species.展开更多

Using biological methods to control algal growth, especially based on allelopathic inhibitory effects of aquatic macrophytes on phytoplankton growth, have been received world-wide attention. In this study, the allelopathic activity of the invasive macrophyte, Eichhornia crassipes (water hyacinth), on blue-green algae, Microcystis aeruginosa (PCC7806) was investigated using coexistence assay. Our results showed that water hyacinth had disparate effects on the growth ofM~ aeruginosa (PCC7806) under different initial algal densities. Under lower initial algal density (OD650 = 0.10 and OD650 = 0.05), the algal growth was significantly inhibited by water hyacinth (inhibition ratio was 95.6% and 97.3%, respectively). While it was stimulated at higher initial algal densities (OD650 = 0.20). Water hyacinth inhibited the growth of algae mainly through its root system. Culture water from water hyacinth and aqueous methanol extracts from dry roots samples also showed inhibition effects on algal growth. The inhibition effects increased as the increase of crude extract concentration, suggest that water hyacinth may excrete inhibitory substances from root system and show allelopathic inhibitory potential to the growth ofM. aeruginosa.展开更多
