云南省2021年猪蓝耳病流行情况调查及基因序列分析 被引量:1

为全面准确掌握云南省猪蓝耳病(PRRS)病原分布、免疫效果及流行情况,根据2021年云南省动物疫病监测与流调计划,每季度在各州(市)下辖各县(市)区按照配额抽样方法,分别在规模场、散养户和屠宰场采集棉拭子、全血和脾肺淋巴共6457份病原... 为全面准确掌握云南省猪蓝耳病(PRRS)病原分布、免疫效果及流行情况,根据2021年云南省动物疫病监测与流调计划,每季度在各州(市)下辖各县(市)区按照配额抽样方法,分别在规模场、散养户和屠宰场采集棉拭子、全血和脾肺淋巴共6457份病原学检测样品以及37912份血清学检测样品,尽量保证全年采集样品覆盖全省所有县(市)区。采用ELISA和qPCR方法进行抗体和病原检测,采用克隆测序方法对病原学阳性样品中的ORF7基因和Nsp2基因进行测序分析。结果显示,2021年云南省PRRS抗体阳性率77.29%;病原学样品阳性率2.91%,场阳性率19.86%。所选毒株ORF7基因序列同源性为98.5%,与国外参考毒株同源性为88.6%~92.9%;所选毒株Nsp2基因序列同源性为97.7%~100%,与国外参考毒株同源性为93.9%~97.3%,分布于美洲株JXA1分支。虽PRRS已退出强免,但养殖场户仍在进行主动免疫,PRRS抗体水平维持在70%以上,后续应根据不同防控阶段,制定实施科学免疫方案,逐步实现从免疫控制到免疫退出。病原学阳性分布污染面广,毒株复杂但变异相对保守,后续应以疫病净化为防治重点,不断完善养殖场生物安全体系,逐步实现净化目标。 展开更多

To investigate molecular epidemiology of DuCV in Cherry Valley ducks in China, the complete genomes of six DuCV strains, which were detected from Cherry Valley ducks in China between 2007 and 2008, were sequenced. Sequence and phylogenetic analysis were carried out to compare these six strains with another 27 DuCV strains from Mulard duck, Muscovy duck, Pekin ducks and Mule duck. The analysis showed that the six DuCV strains exhibited typical genetic features of the family of DuCV, such as a stem-loop structure, three major open reading frames (Rep, Cap and ORF3), four intergenic repeats and the conserved motifs for rolling circle replication and for the dNTP binding domain located in the Rep protein. Phylogenetic analysis of the nucleotide sequences of the complete genome and Cap gene of these strains together with those that have been previously published demonstrated two distinct DuCV genotypes. The DuCV strains with complete genomes containing 1988 and 1989 nucleotides clustered in genotype A, whereas the strains with complete genomes containing 1991, 1992, 1995 and 1996 nucleotides lay in genotype B. The six DuCV strains from Cherry Valley ducks were divided into the two groups. The results of the study provides some insight into the variation of DuCVs in Cherry Valley ducks. 展开更多

Despite being a promising photoanode material for water splitting,WO_(3) has low conductivity,high onset potential,and sluggish water oxidation kinetics.In this study,we designed Ti-doped WO_(3) nanoplate arrays on fluoride-doped tin oxide by a seed-free hydrothermal method,and the effects of doping on the photoelectrochemical performance were investigated.The optimal Ti-doped WO_(3) electrode achieved a photocurrent density of 0.53 mA/cm^(2) at 0.6 V(vs Ag/AgCl),110%higher than that of pure WO_(3) nanoplate arrays.Moreover,a significant cathodic shift in the onset potential was observed after doping.X-ray photoelectron spectroscopy valence band and ultraviolet–visible spectra revealed that the band positions of Ti-doped WO_(3) photoanodes moved upward,yielding a lower onset potential.Furthermore,electrochemical impedance spectroscopy measurements revealed that the conductivities of the WO_(3) photoanodes improved after doping,because of the rapid separation of photo-generated charge carriers.Thus,we report a new design route toward efficient and low-cost photoanodes for photoelectrochemical applications. 展开更多
