There is a one-to-one relationship between the corresponding subunits of the two protein complexes in terms of their size and amino acid sequences. Eight groups of subunits from the COP9 signalosome and the proteasome lid complex of different organisms are collected from all the databases at the NCBI website. The corresponding subunits of COP9 signalosome and proteasome lid complex share at least 12% amino acid identity and some conserved regions, and the conserved sites spread evenly over the entire length of the subunits, suggesting that the two complexes have a common evolutionary ancestor. Phylogenetic analyses based on the amino acid sequences of the corresponding subunits of two protein complexes indicate that every tree consists of two clades. The subunits from one of the two protein complexes of different organisms are grouped into one of the two clades respectively. The sequences of single-cell organisms are always the basal groups to that of multi-cell animal and plant species. These results imply that the duplication/divergence events of COP9 signalosome and regulatory lid of the proteasome genes have occurred before the divergence of single-cell and multi-cell eukaryotes, and the genes of the two complexes are independently evolved. The analyses of dN/dS correlation show significant Pearson's correlations between 21 and 15 pairs of subunit-encoding sequences within the COP9 signalosome and the proteasome lid complex respectively, suggesting that those subunits pairs might have related functions and interacted with one another, and resulted in co-evolution.展开更多

Methods: After 24 h of treatment with 1 × 10-9 mol/L dihydrotestosterone (DHT), the expression of phosphorylated ERK proteins and cell cycle regulation molecules including CDK2, CDK4, CDK6 and P27kip1 in human androgen dependent prostate carcinoma cell line LNCaP was measured by Western blot analysis 0 h, 8 h and 24 h of after androgen withdrawal. Human androgen independent prostate carcinoma cell line PC-3 was also examined as control. Results: Down-regulation of phosphorylated ERK, CDK2, CDK4 and CDK6 and up-regulation of P27kip1 were found initially in LNCaP cell line 8 h after androgen withdrawal. The levels of phosphorylated ERK and CDKs decreased continuously and reached the lowest after 24 h, while continuous elevation of P27kip1 was detected thereafter to 24 h. No expression change of phosphorylated ERK, CDKs and P27kip1 were detected in PC-3 cell line. Conclusion: The androgen withdrawal can cause ERKs activation decrease and cell cycle regulation molecules changes, which may be one of the mechanisms for inhibited growth of androgen dependent prostate carcinoma after androgen ablation by either operative or medicine methods.展开更多

The observed concentration of OH radical showed obvious diurnal and seasonal variations, with maximum readings at noon or afternoon, similar to 80x10(6)OH/cm(3) in summer and similar to 20x10(6)-40x10(6)OH/cm(3) in fall. On the basis of measured data, the reaction rates related to the photochemical process of HO(x) (OH+HO(2)) were derived and characteristics of atmospheric chemical processes in the city were analyzed. The results showed that conversion rates of atmospheric OH and HO(2) in the summer of Beijing City were about 700x10(6) molecule/(cm(3) . s) and 600x10(6)molecule/(cm(3) . s), respectively. And the net production of OH in the air of the city mainly originated from the photolysis of the gaseous HNO(2), and the main sink of OH were the photochemical reactions with VOCs, NO(2), HCHO and CO. It was different from the clean area.展开更多

First, astochastic differential game model is proposed for non-cooperative decentralized uplink power control with a wisdom regulation factor over WA-BodyNets with a onehop star topology. This model aims to minimize the cost associated with the novel payoff function of a player, for which two cost functions are defined: functions of inherent power radiation and accumulated power radiation darmge. Second, the feedback Nash equilibrium solution of the proposed model and the constraint of the Quality of Service (QoS) requirement of the player based on the SIR threshold are derived by solving the Fleming-Bellman-Isaacs partial differential equations. Furthermore, the Markov property of the optimal feedback strategies in this model is verified. The simulation results show that the proposed game model is effective and feasible for controlling the transmitted power of WA-BodyNets.展开更多

It is thought that MyoD and Myf5 are required for myogenic determination, while myogenin is important for terminal differentiation and lineage maintenance. To better understand the function of myogenic regulatory factors in muscle development of flounder, an important economic fish in Asia, real-time quantitative RT-PCR was used to characterize the expression patterns of MyoD, Myf5, and myogenin at early stages of embryo development, and in different tissues of the adult flounder. The results show that, MyJ5 is the first gene to be expressed during the early stages of flounder development, followed by MyoD and myogenin. The expressions ofMyf5, MyoD, and myogenin at the early stages have a common characteristic: expression gradually increased to a peak level, and then gradually decreased to an extremely low level. In the adult flounder, the expression of the three genes in muscle is much higher than that in other tissues, indicating that they are important for muscle growth and maintenance of grown fish. During embryonic stages, the expression level of MyoD might serve an important role in the balance between muscle cell differentiation and proliferation. When the MyoD expression is over 30% of its highest level, the muscle cells enter the differentiation stage.展开更多

The optimal subcarrier, bit and power alloca- tion problems based on discrete adaptive modula- tion and coding scheme have been previously formulated for relay-enhanced networks, and have been reformulated into and solved by integer programming in optimization theory. If the system still has a surplus of subcarriers after resource allocation, we carry out Band- width Exchange (BE) to enable more subcar- riers to participate in transmission to save en- ergy. In addition, as the relay selection scheme is closely linked with heuristic energy saving resource allocation, a relay selection scheme is proposed. Simulation results indicate that the proposed algorithm consumes less energy when transmitting the same number of bits than greedy energy saving schemes, although its spectrum efficiency is worse.展开更多

The BEsproeiingsWAterbestuursprogram (BEWAB+) irrigation scheduling programme is based on the water balance equation and needs: (1) a crop production function; (2) a relative consumptive water demand curve and (3) an allowable depletion subroutine. The objective of this paper was to describe research aimed at obtaining information on (1) and (2) for pea and also to describe the effect of water application on yield and water use of pea. BEWAB+ uses this information to estimate the daily irrigation water requirements for a particular soil-crop-atmosphere system under irrigation. A field experiment, based on published line-source irrigation methodology, was conducted on a 3 m deep loamy fine sand Bainsvlei or Ustic Quartzipsamment soil near Bloemfontein (26°08′S; 29°01′E) in South Africa. Results showed that there is a linear relationship of the form Ys = 8.07ET - 249 (r2 = 0.91), where Ys is the seed yield of pea (kg/ha) and ET is evapotranspiration for the growing season (mm). The relative consumptive water demand curve is represented by the following third order polynomial function that describes the relationship between time and relative ET for a pea growing season of 120 days: ETrelx = 0.09419646 - 0.01302413x + 0.00059008x2 - 0.00000371x3. ETrelz denotes relative ET and x denotes time in days. A workable balance between practical problem solving and advanced irrigation science has been established with BEWAB+. Pre-plant irrigation schedules can be made for semi-arid areas with the BEWAB+ programme using easily obtainable inputs, like target yield, soil depth and soil particle size distribution information.展开更多

文摘Objective: To explore the possible mechanisms of growth regression of human androgen dependent prostate carcinoma cells caused by androgen withdrawal. Conclusion: The androgen withdrawal can cause ERKs activation decrease and cell cycle regulation molecules changes, which may be one of the mechanisms for inhibited growth of androgen dependent prostate carcinoma after androgen ablation by either operative or medicine methods.
文摘Catalytic water splitting potentially reduce the consumption of fossil fuels and has received intense research attention.Synergy effects in multi‐element transition metal‐based water splitting catalysts have evoked special interests.Studies on catalysts in interfacial structures are especially meaningful due to their pertinence in applications. In this study,we report the synergy effects in promoting catalytic power in the ternary transition metal Zn,Co,Ni alloy nanoparticles that embeds in the carbonized Ppy/CNT multilayered matrix.By comparison with a series of binary or single metal counterparts, the mechanism under the synergy effects are elucidated.Experimental and DFT calculation results indicate that the ternary transition metal catalysts in the N‐doped carbon matrix present special electronic structure, which benefits the reversible transition‐state adsorption in HER and OER and render the catalysts high conductivity in room temperature. We expect our findings inspire further development of efficient transition metal HER and OER catalysts.

文摘Surface OH radical concentration in Beijing City was measured by impregnated filter trapping technique-high performance liquid chromatography (IFT-HPLC). It was different from the clean area.

文摘In this study, aiming at the characteristics of randomness and dynamics in Wearable Audiooriented BodyNets (WA-BodyNets), stochastic differential game theory is applied to the investigation of the problem of transmitted power control inconsumer electronic devices. The simulation results show that the proposed game model is effective and feasible for controlling the transmitted power of WA-BodyNets.

文摘MyoD, Myf5, and myogenin are myogenic regulatory factors that play important roles during myogenesis. When the MyoD expression is over 30% of its highest level, the muscle cells enter the differentiation stage.

文摘Objective The objective of this study was to clarify the potential mechanism of quercetin against knee osteoarthritis(KOA)based on network pharmacology and molecular docking. Methods The targets of quercetin were predicted by PubChem and Swiss Target Prediction databases, and the targets of KOA were obtained by DisGeNET,OMIM,and GeneCards databases. Then, the targets of quercetin and KOA were intersected to find the potential targets of quercetin against KOA.The protein–protein interaction network was constructed through the STRING database, and the core targets were screened.Gene ontology(GO)functions enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed using DAVID database. The drug–target–pathway–disease network was constructed by Cytoscape software, and the molecular docking verification was performed by Vina. Results There were 49 potential targets for quercetin against KOA,including 10 core targets.GO functions enrichment analysis showed that the biological process of quercetin against KOA mainly involved the negative regulation of apoptotic process, collagen catabolic process, and extracellular matrix disassembly.KEGG pathway enrichment analysis showed that quercetin against KOA was closely related to PI3K-Akt signaling pathway,Rap 1 signaling pathway,FoxO signaling pathway,Ras signaling pathway,TNF signaling pathway, and ErbB signaling pathway. The results of molecular docking showed that the binding energies between ligand and receptors were less than−5 kcal·mol−1.Conclusions The molecular mechanism of quercetin against KOA involves many targets and pathways, which can regulate the proliferation and apoptosis of chondrocytes, degradation of extracellular matrix, and inflammatory reaction.Quercetin can stably bind to the active pockets of core target proteins, thereby exerting the effect against KOA.

文摘Factor analysis was used to investigate the changes of dry-wet climate in the dry season in Yunnan during 1961-2007 based on observed data from 15 stations.Three common factors were extracted from the 9 climatic factors. The results showed that the dry-wet climate has evidently changed since the early 1960s.The general trends in the changes of drywet climate were described as slight decrease in humidity and gradual enhancement in drought intensity. The climate during 1960s-1980s was under weak-medium drought. But since early 1990s,dry conditions have markedly strengthened and continued due to uneven temporal distribution of rainfall and climate warming.

文摘To reduce energy consumption while maintaining users' Quality of Service (QoS) in Orthogonal Frequency Division Mul- tiplex Access (OFDMA) relay-enhanced net- works, an adaptive energy saving subcarrier, bit and power allocation scheme is presented. Simulation results indicate that the proposed algorithm consumes less energy when transmitting the same number of bits than greedy energy saving schemes, although its spectrum efficiency is worse.

文摘A large portion of irrigation farmers make use of subjective (intuition) irrigation scheduling methods as supposed to objective or scientific irrigation scheduling methods, which need to be changed. Pre-plant irrigation schedules can be made for semi-arid areas with the BEWAB+ programme using easily obtainable inputs, like target yield, soil depth and soil particle size distribution information.

文摘Objective To screen forα-glucosidase inhibitor active compounds in the total saponins of Baibiandou(Lablab Semen Album)based on UHPLC-Q-Exactive Orbitrap MS technology and to evaluate its hypoglycemic activity in vivo. Methods Acarbose was used as the positive control, and the median inhibitory concentration(IC50)was used as the evaluation index ofα-glucosidase inhibitory activity to establish an in vitroα-glucosidase inhibition model.Further,UHPLC-Q-Exactive Orbitrap MS technology was used to screen and identify the active compounds ofα-glucosidase inhibitors in the total saponins of Baibiandou(Lablab Semen Album)in order to further verify the activity of the main active monomer and to perform homologous modeling and molecular docking of yeast-derivedα-glucosidase and human-derivedα-glucosidase,while the hypoglycemic activity was evaluated in diabetic mice. Results This study successfully identified 15 compounds with potentialα-glucosidase inhibitory activity, including Chikusetsusaponin IVa,from the total saponins of Baibiandou(Lablab Semen Album).Simultaneously,we verified the activity of the main active monomer Chikusetsusaponin IVa,and showed that it has strongα-glucosidase inhibitory activity.Theα-glucosidase inhibitory concentration IC50 was(565.2±1.026)μg/m L,and the IC50 of acarbose, which was lower than the positive control,was(706.6±1.058)μg/m L.The docking energies of Chikusetsusaponin IVa were–6.1 and–7.7 kcal/mol with yeast-derivedα-glucosidase and human-derivedα-glucosidase molecules, respectively. Both showed strong binding activity, and the levels of alanine aminotransaminase(ALT),aspartate aminotransaminase(AST),UREA,Creatinine(CREA),and cholesterol(CHO)were significantly decreased by Chikusetsusaponin IVa(P<0.05).In addition,it could repair damaged liver and pancreas cells of diabetic mice to some extent. Conclusion This study provides a basis for screeningα-glucosidase inhibitors and structural modifications of the total saponins of Baibiandou(Lablab Semen Album).
