[Method] By employing SDS-PAGE and 2-demensional electrophoresis (2-DE) techniques,proteins in various heteromorphic leaves from the same adult tree of P. euphratica were isolated and separated to the electrophoresis technique suitable for the separation and analysis of proteins in leaves of P. euphratica tree. [Results] There were significant differences in the expressions of proteins in various heteromorphic leaves of P. euphratica tree. SDS-PAGE pattern showed that bands of proteins with molecular weight of 57.2,13.2,30.2,23.9 and 33.3 kDa were remarkably different. 2-D electrophoresis pattern presented that proteins in leaves of P. euphratica tree mainly belong to acidic proteins distributed at pH value of 5.0-6.5 and with molecular weight of 20-40 kDa; totally 73 different protein spots were observed,of which 51 were up expressed and other 22 were down expressed in the serrated ovate leaves. [Conclusion] Based on these results,we speculate that regulated gene expression in leaves of P. euphratica tree results in the generation of different shapes of leaves,in order to adapt to the surroundings better.展开更多

The stability of QCB under wave force action can be enhanced, and the rubble mound engineering can be le...The quarter-circular caisson breakwater (QCB) is a new type of breakwater, and it can be applied in deepwater. The stability of QCB under wave force action can be enhanced, and the rubble mound engineering can be less than that of semi-circular breakwaters in deepwater. In order to study the wave force distribution acting on the QCB, to find wave force formula for this type of breakwater, firstly in this paper, the distribution characteristics of the horizontal force, the downward vertical force and the uplift force on the breakwater were gotten based on physical model wave flume experiments and on the analysis of the wave pressure experimental data. Based on a series of physical model tests acted by irregular waves, a kind of calculation method, which was modified by Goda formula, was proposed to carry out the wave force on the QCB. Secondly, the reliability method with correlated variables was adopted to analyze the QCB, considering the high correlation between wave forces or moments. Utilizing the observed wave data in engineering field, the reliability index and failure probability of QCB were obtained. Finally, a factor Q=0.9 is given to modify the zero pressure height above SWL of QCB, and wave force partial coefficient 1.34 to the design expressions of QCB for anti-sliding, as well as 1.67 for anti-overturning, were presented.展开更多

A 3-D schema...This study quantifies the main characteristics of a terrain-following, G-coordinate through mathematical analyses of its covariant and contravariant basis vectors as well as the vertical coordinate of σ. A 3-D schematic of the σ-coordinate in a curvilinear coordinate system is provided in this study. The characteristics of the basis vectors were broken down into their "local vector charac- teristics" and "spatial distribution characteristics", and the exact expressions of the covariant; in addition, the con- travariant basis vectors of the G-coordinate used to eluci- date their detailed characteristics were properly solved. Through rewriting the expression of the vertical coordi- nate of G, a mathematical expression of all the cr-coor- dinate surfaces was found, thereby quantifying the so- called terrain-following characteristics and lack of flexi- bility to adjust the slope variation of G-coordinate sur- faces for the classic definition of G. Finally, an analysis on the range value of the vertical coordinate demonstrated that the general value range of G could be obtained by eliminating the G-coordinate surfaces below the Earth's surface. All these quantitative descriptions of the charac- teristics of G-coordinate were the foundation for improv- ing the G-coordinate or creating a new one.展开更多
Metalloproteinases have a critical role in a broad spectrum of cellular processesranging from the break-down of extracellular matrix to the processing of signaltransduction-related proteins.These hydrolytic functions ...Metalloproteinases have a critical role in a broad spectrum of cellular processesranging from the break-down of extracellular matrix to the processing of signaltransduction-related proteins.These hydrolytic functions underlie a variety of mechanisms related to developmental processes as well as disease states.Structural analysis of metalloproteinases from both invertebrate and vertebrate species indicates that these enzymes are highly conserved and arose early during metazoan evolution.In this regard,studies from vari-ous laboratories have reported that a number of classes of metalloproteinases are found in hydra,a member of Cnidaria,the second oldest of existing animal phyla.These studies demonstrate that the hydra genome contains at least three classes of metalloproteinases to include members of the 1)astacin class,2)matrix met-alloproteinase class,and 3)neprilysin class.Functional studies indicate that these metalloproteinases play diverse and important roles in hydra morphogenesis and cell differentiation as well as specialized functions in adult polyps.This article will review the structure,expression,and function of these metalloproteinases in hydra.展开更多
