[Objective]The aim was to isolate cellulose decomposing strains with high activity from straw-amended soil.[Method]Screening of CMC solid culture and shaking culture rescreening were carried out to obtain cellulose decomposing strains with high activity from the corn straw amended soil,which were analyzed by 16rDNA sequence analysis.[Result]A bacterium and a fungus with higher endonucleases activity were obtained through preliminary screening and secondary screening,a fungus with higher filter paper enzyme activity,and a bacterium(NO.5 strain) with higher of both filter paper enzyme activity and CMC enzyme activity were obtained.The result of 16S rDNA sequence analysis showed that the similarity was 100% between NO.5 strain and Bacillus subtilis.[Conclusion]NO.5 strain was identified as Bacillus subtilis. 展开更多

The isoIated stralns were preIiminariIy screened with Congo red stalning and the ceI uIases activities were determined with DNS method. The non-antagonis-tic highIy efficient ceI uIos-degrading stralns were seIected and cuItured combinedIy for deveIoping a ceI uIose-degrading compIex microbial system. The resuIts showed that the CMC enzyme activity of the mixed cuIture of the three fungi stralns was higher than those of the cuItures of the three singIe stralns. The morphoIogical and moIecuIar bioIogical identification indicated that F1 was Botryosphaeria, F2 was Rhi-zopus oryzae, and F5 was Fusarium oxysporum. When straw was used as carbon source, the CMC enzyme activities of F1, F2 and F5 were 39.2, 31.4 and 40.6 IU/mI, respectiveIy; whiIe the CMC enzyme activity of the mixed cuIture of F1+F2+F5 was 50.12 IU/mI, which was increased by 23% compared to that of the singIe straln of F5. The experimental resuIts indicated that the ceI uIose-degrading effect of the compIex microbial system was better than those of the singIe fungi stralns, and the singIe stralns of F1, F2 and F5 had certaln deveIopment potentials. 展开更多

Time-resolved electron spin resonance has been used to study quenching reactions between the antioxidant Vitamin C (VC) and the triplet excited states of 9,10-phenanthrenequinone (PAQ) in ethylene glycol-water (EG-H2O) homogeneous and inhomogeneous reversed micelle solutions. Reversed micelle solutions were used to be the models of physiological environment of biological cell and tissue. In PAQ/EG-H2O homogeneous solution, the excited triplet of PAQ (3PAQ*) abstracts hydrogen atom from solvent EG. In PAQ/VC/EG-H2O solution, 3pAQ* abstracts hydrogen atom not only from solvent EG but also from VC. The quenching rate constant of 3pAQ* by VC is close to the diffusion-controlled value of 1.41 × 108 L/(mol.s). In hexadecyltrimethylammonium bromide (CTAB)/EG-H2O and aerosol OT (AOT)/EG- H2O reversed micelle solutions, 3pAQ* and VC react around the water-oil interface of the reversed micelle. Exit of 3pAQ* from the lipid phase slows down the quenching reaction. For Triton X-100 (TX-100)/EG-H2O reversed micelle solution, PAQ and VC coexist inside the hydrophilic polyethylene glycol core, and the quenching rate constant of 3pAQ* by VC is larger than those in AOT/EG-H2O and CTAB/EG-H2O reversed micelle solutions, even a little larger than that in EG-H2O homogeneous solution. The strong emissive chemically induced dynamic electron polarization of As'- resulted from the effective TM spin polarization transfer in hydrogen abstraction of 3pAQ* from VC. 展开更多

The dynamic changes in the distribution of lignin and hemicelluloses (xylans and xyloglucans) in cell walls during the differentiation of secondary xylem in Eucommia ulmoides Oliv. were studied by means of ultraviolet light microscopy and transmission electron microscopy combined with immunogold labelling. In the cambial zone and cell expansion zone, xyloglucans were localized both in the tangential and radial walls, but no xylans or lignin were found in these regions. With the formation of secondary wall S-1 layer, lignin occurred in the cell corners and middle lamella, while xylans appeared in S-1 layer, and xyloglucans were localized in the primary walls and middle lamella. In pace with the formation of secondary wall S-2 and S-3 layer, lignification extended to S-1, S-2 and S-3 layer in sequence, showing a patchy style of lignin deposition. Concurrently, xylans distributed in the whole secondary walls and xyloglucans, on the other hand, still localized in the primary walls and middle lamella. The results indicated that along with the formation and lignification of the secondary wall, great changes had taken place in the cell walls. Different parts of cell walls, such as cell corners, middle lamella, primary walls and various layers of secondary walls, had different kinds of hemicelluloses, which formed various cell wall architecture combined with lignin and other cell wall components. 展开更多

[Result]The results showed that:BC1 and BC3 strains characterized higher carboxymethyl cellulose enzyme activity and higher filter paper activity,but their difference was small,then the 16S rDNA sequence of BC1 and BC3 strains were related to pseudomonas sp. In order to study the correlations among quality indexes of tobacco leaf and contents of cellulose and lignin in upper leaves of flue-cured tobacco, 48 B2F flue-cured tobacco samples in seven counties of Wulin mountain area in 2011 and 2012 were used as test materials to measure the contents of cellulose and lignin in tobacco leaves and analyze the differences among varieties and between years. Meanwhile, correlations among contents of cellulose and lignin and appearance quality, physical characteristics, general chemical components and indexes of senso- ry quality were studied. The results showed that the contents of cellulose and lignin had no significant difference among years and varieties. Cellulose content was sig- nificantly correlated with some indexes of appearance qualities, physical indexes, general chemical compositions and sensory qualities. Lignin content was significantly correlated with grayness and oil content but not other quality indexes of tobacco leaf. It could be seen that the differences of cellulose and lignin contents between different years and among varieties were not significant. Cellulose content was closely correlated with quality indexes of tobacco leaf while lignin content had a small effect on tobacco leaf quality. 展开更多

Carboxymethyl cellulose(CMC)was introduced as a depressant in reverse flotation separation of quartz from magnesite.The flotation behavior and surface properties of magnesite and quartz exposed to CMC were studied by zeta potential tests,atomic force microscopy imaging and contact angle measurements.The addition of CMC as the depressant in reverse flotation using dodecylamine(DDA)as the collector exhibited a selectively depressive performance towards magnesite and achieved an improved recovery of magnesite.The study of surface properties demonstrated that CMC and DDA exhibited different adsorption strengths on the surface of magnesite and quartz.It was found that the adsorption of CMC on magnesite surface was stronger than that of DDA,which hindered the subsequent adsorption of DDA on magnesite surface.On the contrary,the quartz surface was strongly adsorbed by DDA instead of CMC,which proved that the addition of CMC did not influence the flotation of quartz. 展开更多

The results show that the software can manage intelligently all aspects of the quality analysis and test of ascorbic acid products, uploading timely the data and results of the analysis and inspection to the network and saving it for users, enterprises and quality management, which set up a network of information sharing platform to ensure the authenticity and reliability of measurement results, improving greatly the speed of data processing, saving valuable time, reducing production costs with good economic efficiency and social benefit. A detailed mechanism analysis of cellulose pyrolysis was carried out according to the previous experimental results. On the basis of the Brodio-Shafizadeh model, a modified two-stage model was proposed to simulate the formation and decomposition of active cellulose (AC) and several main organic compounds, such as levoglucosan (LG), hydroxyl-acetaldehyde (HAA), acetol and furfural etc. During pryolysis, the temperature rise of cellulose can be divided into three stages. In the second stage, cellulose undergoes a main decomposition process in which the reaction temperature remains rather low because of the endothermic cracking of glucosidic bond of AC during the formation of LG. The components density of bio-oil, including LG and other competitive compounds, increased rapidly with the increase of temperature during the first stage. However, in the main decomposition process, LG density in bio-oil had an obvious decrease, while the competitive products appeared to increase gradually, which means the ring-opening and reforming reaction of pyranoid ring are superior to LG formation in high temperature.The secondary reaction of volatile components occurs largely in gaseous phase rather than in the solid phase. Short residence time of volatile materials in high temperature region will be advantageous to a high production of LG,which may otherwise decompose quickly under high temperature. An optimum yield of LG could be obtained when radiant source temperature is in the range of 730---920K and gas residence time is less than 1 s. In addition, the reaction temperature has a stronger effect than gas residence time on the formation of HAA, acetol, formaldehyde and furfural etc. 展开更多

Dispersion copolymerization of acryionitrile-vinyi acetate (AN-VAc) has been successfully performed in supercriticai carbon dioxide (ScCO2) with a series of iipophilic/CO2-philic diblock copolymers, such as poly( styrener-acrylonitrile)-b- poly ( 1, 1, 2, 2-tetrahydroperfluorooctyl methacrylate) (PSAN-b-PFOMA), as steric stabilizers. The structure and the particle morphology of the product were characterised by FT-IR and SEM. In addition, the effects of the stabilizer on the surface properties of the products were investigated in detail. Results indicate that the surface free energy of the poly (AN-r-VAc) (PAVAc) film decreases dramatically because of the existence of the stabilizer. And, when the initial concentration of the monomer was 10% (the mass (g) of monomer to the volume (mL) of ScCO2 ) the optimal concentration of the stabilizer is about 5% (w/w% to monomers). 展开更多

This study is aimed at identifying and determining the percentage of occurrence frequency of cellulose decomposing soil fungi. The soil samples were inoculated into culture plates prepared in Sabouraud medium under sterilized conditions and incubated at 30 ℃ for 4 to 7 d. The identified fungal species were incubated in self-designed cellulose medium for testing their cellulolytic ability. Forty-two species, including2 nova species, representing sixteen genera showed growth and sporulation in the cellulose medium. Most of the isolated species were from genus Aspergillus and Penicillium. Aspergillus niger and Mucor hiemalis showed highest occurrence frequency (45% and 36% respectively), as these species were collected from about 80% of soil samples. Being agar free and cheaper, the new fungal medium designed showed results equivalent to Sabouraud medium. 展开更多

A high-pressure slurry supplying system was developed to feed the solid material-water slurries. The apparatus features temperature up to 600℃, pressure up to 40MPa, residence time from 24s to 15min, maximum amount of slurry supply of 2.4 L·h-1, maximum solid content of slurry up to 10%(by mass) for cellulose from Merck, and resistance to corrosion. Long-time runs of decomposition of cellulose were carried out and steady runs were confirmed. Kinetics of cellulose decomposition was studied. The apparent activation energy evaluated was 147kJ·mol-1. In addition, a new three-step pathway for cellulose hydrolysis was proposed. The derived kinetic equation is in good agreement with the experimental data. 展开更多

[Objective]The energy productivity potential of Saccharum spontaneum L.germplasm resources as energy plants was discussed in this paper.[Method]The energy productivity potential of Saccharum spontaneum L.germplasm resources collected from the wild was assessed from the aspects of cellulose,semicellulose,lignin and crude ash contents and caloric value.[Result]The total content of cellulose and semicellulose ranged from 41.75%to 69.13%,the lignin content ranged from 2.16%to 11.75%,the crude ash content ranged from 4.79%to 9.34%and the caloric value ranged from 16.00 to 17.69 MJ/kg among the 30 Saccharum spontaneum L.germplasms.Compared to other plants,Saccharum spontaneum L.had higher contents of cellulose and semicellulose,a higher caloric value but lower contents of lignin and crude ash.[Conclusion]From the perspective of producing fuel ethanol with lignocellulose,Saccharum spontaneum L.has a great potential for development as an energy plant. 展开更多

Aldo-keto reductase 1D1(AKR1D1) deficiency,a rare but life-threatening form of bile acid deficiency,has not been previously described in China.Here,we describe the first two primary 4-3-oxosteroid 5β-reductase deficiency patients in China's Mainland diagnosed by fast atom bombardment-mass spectroscopy of urinary bile acids and confirmed by genetic analysis.A high proportion of atypical 3-oxo-4-bile acids in the urine indicated a deficiency in 4-3-oxosteroid 5β-reductase.All of the coding exons and adjacent intronic sequence of the AKR1D1 gene were sequenced using peripheral lymphocyte genomic DNA of two patients and one of the patient's parents.One patient exhibited compound heterozygous mutations:c.396C>A and c.722A>T,while the other was heterozygous for the mutation c.797G>A.Based on these mutations,a diagnosis of primary 4-3-oxosteroid 5β-reductase deficiency could be confirmed.With ursodeoxycholic acid treatment and fat-soluble vitamin supplements,liver function tests normalized rapidly,and the degree of hepatomegaly was markedly reduced in both patients. 展开更多

Gelidium amansii agar was fractionated on DEAE-Cellulose, and four fractions were obtained sequentially. The fractions were analyzed on physical and chemical properties, and IR and 13C-NMR spectroscopy applied for elucidating the chemical structure. Among the four fractions obtained, water fraction measured up to the standard of low EEO agarose. The sulfate content, ash content, electroendosmosis and gel strength (1%) of water fraction were 0.16%, 0.34%, 0.12 and 1 130g/cm2 respectively, similar to those of the Sigma products. 展开更多

The effects of reaction conditions such as pH of solution, temperature, oxidant concentration, oxidation time. the particle size of 2,3-dialdehyde cellulose and alkali treatment temperature on the dialdehyde concentration ot cellulose were investigated in detail, The results show that the aldehyde group content was created while reaction temperature and alkali treatment temperature increased. The most principal factors affecting the aldehyde group content of 2,3-dialdehyde cellulose were found out and the best oxidation conditions were as follows: the pH was 2. the reaction temperature was 45℃. the mass ratio of cellulose to NalO4 was 1/2, the reaction time was 4 h, the alkali treatment temperature was 70℃ and smaller particle size was 0.80 mm. 展开更多

Characterization of dominant and cellulolytic bacterial communities along the gut of silver carp Hypophthalmichthys molitrix during cyanobacterial blooms

Silver carp is one of the most important planktivorous fish in Chinese aquaculture and plays a significant role controlling cyanobacterial blooms. A balanced gut microbiota is crucial for growth and health of the host because of its important roles in immune defense, digestion of complex carbohydrates, and production of enterocytes. In our study, the dominant bacterial and cellulolytic bacterial (Clostridium 1, Clostridium 111, Clostridium XIVab, and Fibrobacter) communities in the contents and mucus of the silver carp gut (foregut, midgut, and hindgut) were analyzed by denaturing gradient gel electrophoresis and quantitative polymerase chain reaction (qPCR) analyses. The results revealed that the dominant and cellulolytic bacterial communities were significantly different among gut regions as well as in contents and mucus. Bacterial diversity and richness in contents and mucus increased along the gut and were higher in contents than those in local mucus. A sequence analysis of gut samples exhibited the conservative phylotypes of Proteobacteria, Actinobacteria, and Firmicutes. The gut of silver carp harbored an abundance of cellulolytic bacteria, particularly Clostridium XIVab. The foregut segment had the highest proportions of the four cellulolytic bacteria, followed by the midgut and hindgut. However, the proportions of cellulolytic species in the silver carp gut was much lower than those in the terrestrial vertebrate gastrointestinal tract. We conclude that gut bacteria could help silver carp obtain energy from cyanobacteria, which may be why silver carp can maintain high growth rates during cyanobacterial blooms. 展开更多

Conductive ionic hydrogels(CIH)have been widely studied for the development of stretchable electronic devices,such as sensors,electrodes,and actuators.Most of these CIH are made into 3D or 2D shape,while 1D CIH(hydrogel fibers)is often difficult to make because of the low mechanical robustness of common CIH.Herein,we use gel spinning method to prepare a robust CIH fiber with high strength,large stretchability,and good conductivity.The robust CIH fiber is drawn from the composite gel of sodium polyacrylate(PAAS)and sodium carboxymethyl cellulose(CMC).In the composite CIH fiber,the soft PAAS presents good conductivity and stretchability,while the rigid CMC significantly enhances the strength and toughness of the PAAS/CMC fiber.To protect the conductive PAAS/CMC fiber from damage by water,a thin layer of hydrophobic polymethyl acrylate(PMA)or polybutyl acrylate(PBA)is coated on the PAAS/CMC fiber as a water-resistant and insulating cover.The obtained PAAS/CMC-PMA and PAAS/CMC-PBA CIH fibers present high tensile strength(up to 28 MPa),high tensile toughness(up to 43 MJ/m~3),and good electrical conductivity(up to 0.35 S/m),which are useful for textile-based stretchable electronic devices. 展开更多

In this paper, in order to get the target microbe which has high cellulose bio-degradation ability, we collected soil samples from environment rich in cellulose. Carboxymethyl cellulose sodium gel plate and filter paper plate were used for microbe screening, and finally we got a strain named MI which has the highest cellulase producing ability, and it has been identified as Penicilium oxalicum by morphological and molecular biological identification. We have studied cellulose degradation ability of this strain under different conditions such as pH value, temperature and inoculating time. The results showed that CMCase which belongs to acid enzyme, could achieve its maximum enzyme activity 489.96 U/ml when the optimal pH value was 4. The FPAase could achieve its maximum enzyme activity 1,595.45 U/ml when the optimal pH value was 4, too. The optimal ferment temperature of CMCase is 50℃ when achieved the maximum of enzyme activity at the second day, whereas the FPAase could reach its tip top on the third day, So the optimal ferment temperature of FPAase is also 50℃. 展开更多
