[Objective]In order to improve expression vector of L.lactis at food grade and widen application range of original system.[Method] Taking pNZ8149 as basis,the promoter PnisA of pNZ8149 was inserted L.lactis MG1363 and...[Objective]In order to improve expression vector of L.lactis at food grade and widen application range of original system.[Method] Taking pNZ8149 as basis,the promoter PnisA of pNZ8149 was inserted L.lactis MG1363 and SPusp45 of unknown secretory protein in downstream.Through PCR technology,specific primers were used to delete restriction sites between promoter sequence and signal peptide gene sequence and ensure better distance between SD sequence and start codon to construct secreting expression vector pNZS.The reporter gene gus was recombined into multiple cloning site of pNZS to construct pNZS-gus and L.lactis was transformed by electroporation.10 ng/ml nisin was used for induction culture,then culture solution was conducted GUS staining test.[Result]The new constructed L.lactis N3900/pNZS-gus system could express active GUS protein and GUS protein could be secreted out of cell.[Conclusion]The successful construction of this system lays foundation for secretion expression study of protein and oral vaccine research.展开更多

According to the feature of the impurity, one image with twice processing is put forward based on blob analysis, which eliminates impurities that were brighter or darker than normal particles. With the image processing system developed by Matrox Imaging Library, black and white blobs are extracted for twice with one function, and then the size and the position of impurities are calculated quickly through mathematical morphologic and blob analysis. The method can be extended and applied in other fields due to its advantages of quick speed and high accuracy.展开更多
Three treatments including mechanical damage, Lymantria dispar attacking and daubing oral secretions of the in-sects on mechanically damaged cut were conducted on Populus simonii譖opulus pyramibalis c.v. in order to f...Three treatments including mechanical damage, Lymantria dispar attacking and daubing oral secretions of the in-sects on mechanically damaged cut were conducted on Populus simonii譖opulus pyramibalis c.v. in order to find the genuine reason leading to effective resistance response of tree to insects attacking. The release situation of the induced volatiles of the plant was analyzed by TCT-GC/MS at 24 hours after damages. The results indicated that some of the volatiles such as (Z)-3-hexenyl acetate, decanal, 3-hexenyl isovalerate, nonanal, ocimene, and 2-cyanobutane can be induced by both insects attack-ing and mechanical damage, while 2,6-dimethyl-1,3,5,7-octatetraene, 2-methyl-6-methylene-1,7-octadien-3-one, caryophyllene, Isovaleronitrile, diethyl-methyl-benzamide, and dicapryl phthalate were only induced by insects attacking. Such difference in volatiles was attributed to that there existed active components in oral sections of the larvae of Lymantria dispar展开更多

These two new species are similar to P. decipiens and P. ochracea as re...Two new species of the genus Pseudovolucella Shiraki are described from Sichuan in China: P. hengduanshanensis sp. nov. and P. dimorpha sp. nov. These two new species are similar to P. decipiens and P. ochracea as reviewed by Reemer and Hippa(2008). The males of these 4 species have black abdominal tergite 4, P. decipiens has the hind femur of male with a small apicoventral knob, but the new species and P. ochracea have the hind femora of males straight on ventral margins showing that they are more similar to P. ochracea. However, P. hengduanshanensis sp. nov. is distinguished from P. ochracea by face absent of dark brown median vitta, abdominal tergite 2 with a pair of broad yellow basal fasciae separated narrowly in the middle, tergite 3 in male and tergites 3 and 4 in female respectively with a pair of yellow narrow fasciae(P. ochracea has the face with a broad middle dark brown to black vitta diverging above and fading away the paler color, abdominal tergite 2 is wholly pale brownish yellow on basal 2/3, the remainder of tergites shining black, female unknown). P. dimorpha sp. nov. differs from P. ochracea with abdominal tergite 2 in male and tergites 2, 3 and 4 in female with a pair of yellow narrow fasciae respectively(abdominal characters of P. ochracea see above).
