[ Result] Two alleles, designated as A and B, were found at the locus 346 in the third intron of porcine H-FABP gene, and the mutation was caused by a A→G substitution. The ongoing need for better fuel economy and lower exhaust pollution of vehicles has increased the employment of electric power steering(EPS)in automotives.Optimal design of EPS for a product family reduces the develo...The ongoing need for better fuel economy and lower exhaust pollution of vehicles has increased the employment of electric power steering(EPS)in automotives.Optimal design of EPS for a product family reduces the development and fabrication costs significantly.In this paper,the TOPSIS method along with the NSGA-Ⅱis employed to find an optimum family of EPS for an automotive platform.A multi-objective optimization problem is defined considering road feel,steering portability,RMS of Ackerman error,and product family penalty function(PFPF)as the conflicting objective functions.The results for the single objective optimization problems and the ones for the multi-objective optimization problem,as well as two suggested trade-off design points are presented,compared and discussed.For the two suggested points,performance at one objective function is deteriorated by about 1%,while the commonality is increased by 20%–40%,which shows the effectiveness of the proposed method in first finding the non-dominated design points and then selecting the trade-off among the obtained points.The results indicate that the obtained trade-off points have superior performance within the product family with maximum number of common parts.展开更多

The aim of this study was to assess the effect of long-term in vitro sub-culturing on the varietal degeneration of three sweet potato varieties, namely, Monate, Mokone and Ndou which were sub-cultured for 32, 23 and 1...The aim of this study was to assess the effect of long-term in vitro sub-culturing on the varietal degeneration of three sweet potato varieties, namely, Monate, Mokone and Ndou which were sub-cultured for 32, 23 and 12 generations, respectively. Each generation was cultured in a media which is made from 4.43 g/L Murashige and Skoog (MS), 30 g/L sucrose and 2 g/L gelrite, respectively, and grown under 16 h light and 8 h dark photoperiod for 30 d. For each generation, 45 plantlets were acclimatized for two months in a glasshouse. Data on in vitro growth performance and 11 morphological characteristics during acclimatization were recorded. Early root and shoot formation was observed after the 27th and 21st sub-cultured generations of Monate and Mokone, respectively. During acclimatization, plantlets from the same variety showed differences in morphological traits such as leaf colour, abaxial leaf pigmentation, vine pigmentation, petiole pigmentation, leaf wrinkling and flowering. However, the rate of these morphological differences is random and irrespective to increase in sub-culturing. Therefore, to understand the genetic base of these morphological variability, two plantlets from each variety were subjected to genetic analysis by using five simple sequence repeat (SSR) primers (IB-242, IB-318, IB-255F, 1B-248 and IB-255). Although SSR loci IB-255F and IB-318 could distinguish between the three varieties, there were no allelic polymorphisms detected in plantlets from the same varieties. Therefore, long-term sub-culturing do not leads to quality degeneration in the three sweet potato varieties.展开更多
Objective: We aimed to evaluate the efficiency of serum testosterone suppression as well as the potential for agonistic stimulation of serum testosterone during chronic treatment with monthly (3.75 mg) depot formul...Objective: We aimed to evaluate the efficiency of serum testosterone suppression as well as the potential for agonistic stimulation of serum testosterone during chronic treatment with monthly (3.75 mg) depot formulation of domestic substitute of leuprorelin acetate microspheres for patients with metastatic prostate cancer. Methods: A total of 23 patients with metastatic prostate cancer were enrolled in the prospective study and received 6 monthly intramuscular depot injections of domestic substitute of leuprorelin acetate microspheres. Their levels and patterns of serum testosterone suppression and the potential for agonistic stimulation of serum testosterone were monitored following injection monthly (3.75 rag) depot formulation of domestic substitute of leuprorelin acetate microspheres for 24 weeks. Results: Mean testosterone was 431.4 ng/dL, 119.3 ng/dL, 28.2 ng/dL by week 1, 2, 3 and decreased to less than 15.6 ng/dL by week 4 where it remained throughout the treatment period. Median time to suppression of serum testosterone was 20.7 days. No transient minor "escape" from suppression occurred in all patients which was defined as a single testosterone value greater than 50 ng/dL once suppression was achieved. Assessment of agonistic stimulation following the second depot injection revealed no pattern of stimulation. Conclusion: We concluded that monthly (3.75 mg) depot formulation of domestic substitute of leuprorelin acetate microspheres could provide persistent, stable suppression of serum testosterone throughout the dosing intervals, and that the initial depot injection of this formulation also could provide sufficient pituitary desensitization to prevent agnostic stimulation of serum testosterone during chronic treatment.展开更多

In th...A gel microbead (GMD) cultivation method was employed to cultivate microorganisms from an amphioxus breeding zone in Qingdao, P. R. China. The culture results were compared with those by standard plating method. In the GMD-based method, the microcolony-forming GMDs were sorted by fluorescence-activated cell sorting (FACS). To further get pure cultures, a subsequent enrichment culture and a streaking purification procedure were conducted on marine R2A medium. Eighty bacterial strains isolated by the GMD-based method were randomly selected for sequencing. These isolates belonged to Alphaproteobacteria (33%), Gammaproteobacteria (44%), Bacteroidetes (11%), Actinobacteria (5%), Firmicutes (5%), Epsilonproteobacteria (1%), and Verrucomicrobia (1%), the last two groups being usually difficult to culture. The 16S rRNA gene sequences revealed a diverse community with 91.1%-100% of the bacterial rRNAs similarities. Thirteen strains were sharing 16S rRNA gene sequence which was less than 97% similar to any other rRNA genes currently deposited in TYP16S database. Seventy isolates derived from the standard plating method fell into 4 different taxonomic groups: Alphaproteobacteria (9%), Gammaproteobacteria (81%), Bacteroidetes (7%) and Firmicutes (3%) with a 16S rRNA gene sequence similarities between 95.8%-100%, in which only 3 strains were sharing 16S rRNA gene sequence of less than 97%. The results indicated that the GMD-based method with subenrichment culture yielded more taxonomic groups and more novel microbial strains, including members of previously rarely cultured groups, when compared with the standard plating method, and that this method markedly improved the bacterial cultivability.展开更多

In an operational context such as the NASA (National Aeronautics and Space Administration) information production based on the MODIS (Moderate Resolution Imaging Spectroradiometer) instrument on board Earth Observing System (EOS) Terra and Aqua satellites, efficient ways of detecting product anomaly, i.e., to discriminate between product artifacts and real changes in Earth processes being monitored, are extremely important to assist and inform the user communities about potential unreliability in the products. A technique for anomaly detection, known as MAD (the median of absolute deviate from the median), in MODIS land products via time series analysis is described, which can handle intra- and in-ter-annual variation in the data by using MAD statistics of the original data and their first-order difference. This method is shown to be robust and work across major land products, including NDVI, active fire, snow cover, and surface reflectance, and its applicabil-ity to multi-disciplinary products is anticipated.展开更多
Zhikong scallop(Chlamys farreri) is an important maricultured species in China.Many researches on this species,such as population genetics and QTL fine-mapping,need a large number of molecular markers.In this study,ba...Zhikong scallop(Chlamys farreri) is an important maricultured species in China.Many researches on this species,such as population genetics and QTL fine-mapping,need a large number of molecular markers.In this study,based on the expressed sequence tags(EST),a total of 300 putative single nucleotide polymorphisms(SNPs) were selected and validated using high resolution melting(HRM) technology with unlabeled probe.Of them,101(33.7%) were found to be polymorphic in 48 individuals from 4 populations.Further evaluation with 48 individuals from Qingdao population showed that all the polymorphic loci had two alleles with the minor allele frequency ranged from 0.046 to 0.500.The observed and expected heterozygosities ranged from 0.000 to 0.925 and from 0.089 to 0.505,respectively.Fifteen loci deviated significantly from Hardy-Weinberg equilibrium and significant linkage disequilibrate was detected in one pair of markers.BLASTx gave significant hits for 72 of the 101 polymorphic SNP-containing ESTs.Thirty four polymorphic SNP loci were predicted to be non-synonymous substitutions as they caused either the change of codons(33 SNPs) or pretermination of translation(1 SNP).The markers developed can be used for the population studies and genetic improvement on Zhikong scallop.展开更多
Experiments were conducted at Entomology division of Directorate of Rice research to study the qualitative and quantitative changes in biochemical constituents in the rice plant treated with different xenobiotics, viz...Experiments were conducted at Entomology division of Directorate of Rice research to study the qualitative and quantitative changes in biochemical constituents in the rice plant treated with different xenobiotics, viz. insecticides like deltamethrin, imidacloprid, herbicide like 2,4-Dichlorophenoxyacetic acid, fungicide like carbendazim, green manure Calotropis gigantea leaves, botanicals like Neem Seed Powder and fertilizer urea along with control plants of TN1, PTB33 and Swama varieties comprising of total 15 treatments. Application of butachlor, carbendazim, deltamethrin and excessive nitrogen fertilization resulted in increased population build up of brown plant hopper (BPH). Significantly, lower honeydew excretion (13 mme area) in the BPH nymphs was observed on resistant PTB33 variety compared to susceptible TN1 and Swarna varieties (116 and 110 mm2 respectively). In susceptible TN1 variety, the sucrose content was significantly higher (9.35 mg/g tissue) compared to BPH resistant PTB33 (5.32 mg/g tissue). The BPH resistant PTB33 variety exhibited significantly lower free amino acid content (1.23 mg/g tissue) as compared to TN1 (2.51 mg/g). The C/N ratio was comparatively high in the resistant PTB33 alone (4.32), but was low in PTB33 applied with 2,4-D + carbendazim (2.73), 2,4-D + imidacloprid ~ carbendazim (2.3), excess urea fertilizer (2.18). The total phenol contents measured in resistant PTB33 (7.5 mg/g dry wt), susceptible TN1 (5.865 mg/g dry wt) and the popular variety Swarna (5.57 mg/g dry wt) were almost same and no statistical difference was observed. Further, treatment of these varieties with different xenobiotics and botanicals also did not significantly alter their total phenol content. However, the phenol content of the resistant PTB 33 alone and treated with xenobiotics was higher compared to the susceptible TN 1 and Swarna varieties alone and treated with xenobiotics. The plant biochemical constituents such as free amino acids, sucrose were lower in untreated resistant PTB33 compared to susceptible TN1, and PTB33 treated with urea and deltamethrin. The high feeding rate and population build up on the susceptible variety TN1 is positively correlated with high quantities of sucrose, free amino acids and lower quantities of phenols and low C/N ratio.展开更多

Nosocomial infections caused by K. pneumoniae producing extended-spectrum β-lactamases (ESBLs) are very difficu...Klebsiella pneumoniae ( K. pneumoniae) is one of the main gmn-negative bacilli in clinical practice. Nosocomial infections caused by K. pneumoniae producing extended-spectrum β-lactamases (ESBLs) are very difficult to treat. This paper investigated the resistant characteristics of K. pneumoniae producing ESBLs and their aminoglycoside-modifying enzyme gene expressions including Nacetyltransferases and O-adenyltransferases. Bacteria identification and ESBLs confirmatory tests were performed by Phoenix^TM-100 system. And minimum inhibitory concentrations (MICs) of gentamicin, amikacin, kanamycin, tobranycin, netilmicin and neomycin in 53 K. pneumoniae isolates were detected by agar dilution. In addition, six aminoglycoside-modifying enzyme genes were amplified by polymerase chain reaction (PCR) and verified by DNA sequencer. It was found that imipenem and meropenem against 120 K. pneumoniae isolates produced powerful antimicrobial activities. The resistant rates of gentamicin and amikacin were 55.0% and 46.7%, respectively. Except neomycin, MIC50 and MIC90 of gentamicin, amikacin, kanamycin, tobramycin and netilmicin in 53 K. pneumoniae were all 〉 128 μg/ml, and the resistant rates were 83.0%, 52.3%, 75.5%, 81.1% and 69.8%, respectively. However, neomycin was only 39.6%. In addition, five modifying enzyme genes, including aac(3)-Ⅰ, aac(3)-Ⅱ, aac(6')-Ⅰb, ant(3")-Ⅰ, ant(2")-Ⅰ genes, were found in 53 isoaltes except aac (6')-Ⅱ, and their positive rates were 11.3%, 67.9%, 47.2%, 1.9% and 39.6%, respectively. It was also confirmed by nucleotide sequence analysis that the above resistant genes shared nearly 100% identities with GenBank published genes. The results obtained in the present study indicated that K. pneumoniae producing ESBLs strains are rapidly spreading in our hospital, and their resistance to aminoglycosides may be associated with aminoglycoside-modifying enzyme gene expressions.展开更多
To explore the prevalence of the plasmid-mediated quinolone resistance gene qnrA in Gramnegative bacteria and to investigate its molecular genetic background and resistance profile in isolates harboring this gene, a t...To explore the prevalence of the plasmid-mediated quinolone resistance gene qnrA in Gramnegative bacteria and to investigate its molecular genetic background and resistance profile in isolates harboring this gene, a total of 629 nalidixic acid-resistant isolates of non-repetitive Gram-negative bacteria were collected from clinical specimens between April 2004 and April 2006 and these isolates were screened for qnrA gene by PCR using specific primers combined with DNA sequencing. The extended spectrum β-1actamase (ESBL) or AmpC-producing isolates were distinguished by the phenotypic confirmatory test combined with DNA sequencing, and the antibiotics susceptibility test for qnrA-positive isolates was carried out by Kirby-Bauer and E-test method. To detect the location of the qnrA gene, plasmid conjugation and Southern hybridization were performed and the integron structure containing the qnrA gene was cloned by PCR strategy and sequenced by primer walking. It was demonstrated that the incidence of the qnrA-positive strains in nalidixic acid-resistant bacteria was 1.9% (12/629), in which the detection rates for Klebiesiella pneumoniae. Enterobacter cloacae, Enterobacter aerogenes, Citrobacterfreundii and Salmonella choeraesuis were 2.2% (3/138), 17. 1% (6/35), 9. 1% (1/11), 12.5% (1/8), and 14.3% (1/7), respectively. The qnrA gene was found to be embedded in the complex sull-type integron located on plasmids with varied size (80-180 kb). Among them, 4 qnrA-positive isolates carried integron In37 and 8 isolates carried a novel integron, temporarily desig- nated as InX. All the qnrA-positive isolates were ESBL-producing and transferable for the multi-drug resistance. It is concluded that the plasmid-mediated drug-resistance mechanism exists in the quinolone resistant strains of isolates from hospitals in Guangdong area, but the incidence was rather low. Nevertheless, it is still possible that the horizontal transfer of the resistant qnrA gene might lead to the spreading of drug-resistance.展开更多

This increase has been more pronounced in rural and remote areas especially among Aboriginal peoples. Substantial studies indicated that nearly half of the on-reserve Aboriginal households in British Columbia (BC) suffer from some degree of food insecurity. Despite the valuable research about obtaining traditional food, the issue of access to market food has been less discussed in the literature. The aim of this study was to assess the local food environment of on-reserve Aboriginal peoples by using a proximity-based approach tool in terms of access to healthy food stores such as supermarket and grocery stores. In the first stage, the addresses of all healthy food stores were geocoded into a map layer. Then the locations of Aboriginal reserves were geocoded to a separate map layer. In the second stage, using "Closest Facility Analysis" tool in ArcGIS Version 10.3, the distance based on a 15-minute driving time was measured from each reserve to the closest healthy food destination on CanMapRouteLogistics network. The results indicated that 25% of Aboriginal reserves did not have reasonable access to healthy food stores. We concluded that Aboriginal peoples in low access rural reserves with lower socioeconomic status are at potential risk of perpetuate food insecurity.展开更多

The chloroplast (cp) genome of M. grandiflora was sequenced using a 454 sequencing platform and the genome structure was compared with other related species. The complete cp genome ofM. grandiflora was 159623 bp in length and contained a pair of inverted repeats (IR) of 26563 bp separated by large and small single copy (LSC, SSC) regions of 87757 and 18740 bp, respectively. A total of 129 genes were successfully annotated, 18 of which included introns. The identity, number and GC content of M. grandiflora cp genes were similar to those of other Magnoliaceae species genomes. Analysis revealed 218 simple sequence repeat (SSR) loci, most composed of A or T, contributing to a bias in base composition. The types and abundances of repeat units in Magnoliaceae species were relatively conserved and these loci will be useful for developing M. grandiflora cp genome vectors. In addition, results indicated that the cp genome size in Magnoliaceae species and the position of the IR border were closely related to the length of the ycfl gene. Phylogenetic analyses based on 66 shared genes from 30 species using maximum parsimony (MP) and max- imum likelihood (ML) methods provided strong support for the phylogenetic position of Magnolia. The availability of the complete cp genome sequence of M. grandiflora provides valuable information for breeding of desirable varieties, cp genetic engineering, developing useful molecular markers and phylogenetic analyses in Magnoliaceae.展开更多

However, the rate of these morphological differences is random and irrespective to increase in sub-culturing. Therefore, to understand the genetic base of these morphological variability, two plantlets from each variety were subjected to genetic analysis by using five simple sequence repeat (SSR) primers (IB-242, IB-318, IB-255F, 1B-248 and IB-255). Although SSR loci IB-255F and IB-318 could distinguish between the three varieties, there were no allelic polymorphisms detected in plantlets from the same varieties. Therefore, long-term sub-culturing do not leads to quality degeneration in the three sweet potato varieties.

文摘Objective: We aimed to evaluate the efficiency of serum testosterone suppression as well as the potential for agonistic stimulation of serum testosterone during chronic treatment with monthly (3.75 mg) depot formulation of domestic substitute of leuprorelin acetate microspheres for patients with metastatic prostate cancer. Methods: A total of 23 patients with metastatic prostate cancer were enrolled in the prospective study and received 6 monthly intramuscular depot injections of domestic substitute of leuprorelin acetate microspheres. Their levels and patterns of serum testosterone suppression and the potential for agonistic stimulation of serum testosterone were monitored following injection monthly (3.75 rag) depot formulation of domestic substitute of leuprorelin acetate microspheres for 24 weeks. Results: Mean testosterone was 431.4 ng/dL, 119.3 ng/dL, 28.2 ng/dL by week 1, 2, 3 and decreased to less than 15.6 ng/dL by week 4 where it remained throughout the treatment period. Median time to suppression of serum testosterone was 20.7 days. No transient minor "escape" from suppression occurred in all patients which was defined as a single testosterone value greater than 50 ng/dL once suppression was achieved. Assessment of agonistic stimulation following the second depot injection revealed no pattern of stimulation. Conclusion: We concluded that monthly (3.75 mg) depot formulation of domestic substitute of leuprorelin acetate microspheres could provide persistent, stable suppression of serum testosterone throughout the dosing intervals, and that the initial depot injection of this formulation also could provide sufficient pituitary desensitization to prevent agnostic stimulation of serum testosterone during chronic treatment.

文摘A gel microbead (GMD) cultivation method was employed to cultivate microorganisms from an amphioxus breeding zone in Qingdao, P. R. China. The culture results were compared with those by standard plating method. In the GMD-based method, the microcolony-forming GMDs were sorted by fluorescence-activated cell sorting (FACS). To further get pure cultures, a subsequent enrichment culture and a streaking purification procedure were conducted on marine R2A medium. Eighty bacterial strains isolated by the GMD-based method were randomly selected for sequencing. These isolates belonged to Alphaproteobacteria (33%), Gammaproteobacteria (44%), Bacteroidetes (11%), Actinobacteria (5%), Firmicutes (5%), Epsilonproteobacteria (1%), and Verrucomicrobia (1%), the last two groups being usually difficult to culture. The 16S rRNA gene sequences revealed a diverse community with 91.1%-100% of the bacterial rRNAs similarities. Thirteen strains were sharing 16S rRNA gene sequence which was less than 97% similar to any other rRNA genes currently deposited in TYP16S database. Seventy isolates derived from the standard plating method fell into 4 different taxonomic groups: Alphaproteobacteria (9%), Gammaproteobacteria (81%), Bacteroidetes (7%) and Firmicutes (3%) with a 16S rRNA gene sequence similarities between 95.8%-100%, in which only 3 strains were sharing 16S rRNA gene sequence of less than 97%. The results indicated that the GMD-based method with subenrichment culture yielded more taxonomic groups and more novel microbial strains, including members of previously rarely cultured groups, when compared with the standard plating method, and that this method markedly improved the bacterial cultivability.

文摘With remote sensing information products becoming increasingly varied and arguably improved, scientific applications of such products rely on their quality assessment. In an operational context such as the NASA (National Aeronautics and Space Administration) information production based on the MODIS (Moderate Resolution Imaging Spectroradiometer) instrument on board Earth Observing System (EOS) Terra and Aqua satellites, efficient ways of detecting product anomaly, i.e., to discriminate between product artifacts and real changes in Earth processes being monitored, are extremely important to assist and inform the user communities about potential unreliability in the products. A technique for anomaly detection, known as MAD (the median of absolute deviate from the median), in MODIS land products via time series analysis is described, which can handle intra- and in-ter-annual variation in the data by using MAD statistics of the original data and their first-order difference. This method is shown to be robust and work across major land products, including NDVI, active fire, snow cover, and surface reflectance, and its applicabil-ity to multi-disciplinary products is anticipated.

文摘Zhikong scallop(Chlamys farreri) is an important maricultured species in China.Many researches on this species,such as population genetics and QTL fine-mapping,need a large number of molecular markers.In this study,based on the expressed sequence tags(EST),a total of 300 putative single nucleotide polymorphisms(SNPs) were selected and validated using high resolution melting(HRM) technology with unlabeled probe.Of them,101(33.7%) were found to be polymorphic in 48 individuals from 4 populations.Further evaluation with 48 individuals from Qingdao population showed that all the polymorphic loci had two alleles with the minor allele frequency ranged from 0.046 to 0.500.The observed and expected heterozygosities ranged from 0.000 to 0.925 and from 0.089 to 0.505,respectively.Fifteen loci deviated significantly from Hardy-Weinberg equilibrium and significant linkage disequilibrate was detected in one pair of markers.BLASTx gave significant hits for 72 of the 101 polymorphic SNP-containing ESTs.Thirty four polymorphic SNP loci were predicted to be non-synonymous substitutions as they caused either the change of codons(33 SNPs) or pretermination of translation(1 SNP).The markers developed can be used for the population studies and genetic improvement on Zhikong scallop.
文摘Experiments were conducted at Entomology division of Directorate of Rice research to study the qualitative and quantitative changes in biochemical constituents in the rice plant treated with different xenobiotics, viz. insecticides like deltamethrin, imidacloprid, herbicide like 2,4-Dichlorophenoxyacetic acid, fungicide like carbendazim, green manure Calotropis gigantea leaves, botanicals like Neem Seed Powder and fertilizer urea along with control plants of TN1, PTB33 and Swama varieties comprising of total 15 treatments. Application of butachlor, carbendazim, deltamethrin and excessive nitrogen fertilization resulted in increased population build up of brown plant hopper (BPH). Significantly, lower honeydew excretion (13 mme area) in the BPH nymphs was observed on resistant PTB33 variety compared to susceptible TN1 and Swarna varieties (116 and 110 mm2 respectively). In susceptible TN1 variety, the sucrose content was significantly higher (9.35 mg/g tissue) compared to BPH resistant PTB33 (5.32 mg/g tissue). The BPH resistant PTB33 variety exhibited significantly lower free amino acid content (1.23 mg/g tissue) as compared to TN1 (2.51 mg/g). The C/N ratio was comparatively high in the resistant PTB33 alone (4.32), but was low in PTB33 applied with 2,4-D + carbendazim (2.73), 2,4-D + imidacloprid ~ carbendazim (2.3), excess urea fertilizer (2.18). The total phenol contents measured in resistant PTB33 (7.5 mg/g dry wt), susceptible TN1 (5.865 mg/g dry wt) and the popular variety Swarna (5.57 mg/g dry wt) were almost same and no statistical difference was observed. Further, treatment of these varieties with different xenobiotics and botanicals also did not significantly alter their total phenol content. However, the phenol content of the resistant PTB 33 alone and treated with xenobiotics was higher compared to the susceptible TN 1 and Swarna varieties alone and treated with xenobiotics. The plant biochemical constituents such as free amino acids, sucrose were lower in untreated resistant PTB33 compared to susceptible TN1, and PTB33 treated with urea and deltamethrin. The high feeding rate and population build up on the susceptible variety TN1 is positively correlated with high quantities of sucrose, free amino acids and lower quantities of phenols and low C/N ratio.

文摘Klebsiella pneumoniae ( K. pneumoniae) is one of the main gmn-negative bacilli in clinical practice. Nosocomial infections caused by K. pneumoniae producing extended-spectrum β-lactamases (ESBLs) are very difficult to treat. This paper investigated the resistant characteristics of K. pneumoniae producing ESBLs and their aminoglycoside-modifying enzyme gene expressions including Nacetyltransferases and O-adenyltransferases. Bacteria identification and ESBLs confirmatory tests were performed by Phoenix^TM-100 system. And minimum inhibitory concentrations (MICs) of gentamicin, amikacin, kanamycin, tobranycin, netilmicin and neomycin in 53 K. pneumoniae isolates were detected by agar dilution. In addition, six aminoglycoside-modifying enzyme genes were amplified by polymerase chain reaction (PCR) and verified by DNA sequencer. It was found that imipenem and meropenem against 120 K. pneumoniae isolates produced powerful antimicrobial activities. The resistant rates of gentamicin and amikacin were 55.0% and 46.7%, respectively. Except neomycin, MIC50 and MIC90 of gentamicin, amikacin, kanamycin, tobramycin and netilmicin in 53 K. pneumoniae were all 〉 128 μg/ml, and the resistant rates were 83.0%, 52.3%, 75.5%, 81.1% and 69.8%, respectively. However, neomycin was only 39.6%. In addition, five modifying enzyme genes, including aac(3)-Ⅰ, aac(3)-Ⅱ, aac(6')-Ⅰb, ant(3")-Ⅰ, ant(2")-Ⅰ genes, were found in 53 isoaltes except aac (6')-Ⅱ, and their positive rates were 11.3%, 67.9%, 47.2%, 1.9% and 39.6%, respectively. It was also confirmed by nucleotide sequence analysis that the above resistant genes shared nearly 100% identities with GenBank published genes. The results obtained in the present study indicated that K. pneumoniae producing ESBLs strains are rapidly spreading in our hospital, and their resistance to aminoglycosides may be associated with aminoglycoside-modifying enzyme gene expressions.
文摘To explore the prevalence of the plasmid-mediated quinolone resistance gene qnrA in Gramnegative bacteria and to investigate its molecular genetic background and resistance profile in isolates harboring this gene, a total of 629 nalidixic acid-resistant isolates of non-repetitive Gram-negative bacteria were collected from clinical specimens between April 2004 and April 2006 and these isolates were screened for qnrA gene by PCR using specific primers combined with DNA sequencing. The extended spectrum β-1actamase (ESBL) or AmpC-producing isolates were distinguished by the phenotypic confirmatory test combined with DNA sequencing, and the antibiotics susceptibility test for qnrA-positive isolates was carried out by Kirby-Bauer and E-test method. To detect the location of the qnrA gene, plasmid conjugation and Southern hybridization were performed and the integron structure containing the qnrA gene was cloned by PCR strategy and sequenced by primer walking. It was demonstrated that the incidence of the qnrA-positive strains in nalidixic acid-resistant bacteria was 1.9% (12/629), in which the detection rates for Klebiesiella pneumoniae. Enterobacter cloacae, Enterobacter aerogenes, Citrobacterfreundii and Salmonella choeraesuis were 2.2% (3/138), 17. 1% (6/35), 9. 1% (1/11), 12.5% (1/8), and 14.3% (1/7), respectively. The qnrA gene was found to be embedded in the complex sull-type integron located on plasmids with varied size (80-180 kb). Among them, 4 qnrA-positive isolates carried integron In37 and 8 isolates carried a novel integron, temporarily desig- nated as InX. All the qnrA-positive isolates were ESBL-producing and transferable for the multi-drug resistance. It is concluded that the plasmid-mediated drug-resistance mechanism exists in the quinolone resistant strains of isolates from hospitals in Guangdong area, but the incidence was rather low. Nevertheless, it is still possible that the horizontal transfer of the resistant qnrA gene might lead to the spreading of drug-resistance.

文摘The rate of food insecurity has increased tremendously over the past decade in Canada. This increase has been more pronounced in rural and remote areas especially among Aboriginal peoples. Substantial studies indicated that nearly half of the on-reserve Aboriginal households in British Columbia (BC) suffer from some degree of food insecurity. Despite the valuable research about obtaining traditional food, the issue of access to market food has been less discussed in the literature. The aim of this study was to assess the local food environment of on-reserve Aboriginal peoples by using a proximity-based approach tool in terms of access to healthy food stores such as supermarket and grocery stores. In the first stage, the addresses of all healthy food stores were geocoded into a map layer. Then the locations of Aboriginal reserves were geocoded to a separate map layer. In the second stage, using "Closest Facility Analysis" tool in ArcGIS Version 10.3, the distance based on a 15-minute driving time was measured from each reserve to the closest healthy food destination on CanMapRouteLogistics network. The results indicated that 25% of Aboriginal reserves did not have reasonable access to healthy food stores. We concluded that Aboriginal peoples in low access rural reserves with lower socioeconomic status are at potential risk of perpetuate food insecurity.

文摘Magnolia grandiflora is an important medicinal, ornamental and horticultural plant species. The chloroplast (cp) genome of M. grandiflora was sequenced using a 454 sequencing platform and the genome structure was compared with other related species. The complete cp genome ofM. grandiflora was 159623 bp in length and contained a pair of inverted repeats (IR) of 26563 bp separated by large and small single copy (LSC, SSC) regions of 87757 and 18740 bp, respectively. A total of 129 genes were successfully annotated, 18 of which included introns. The identity, number and GC content of M. grandiflora cp genes were similar to those of other Magnoliaceae species genomes. Analysis revealed 218 simple sequence repeat (SSR) loci, most composed of A or T, contributing to a bias in base composition. The types and abundances of repeat units in Magnoliaceae species were relatively conserved and these loci will be useful for developing M. grandiflora cp genome vectors. In addition, results indicated that the cp genome size in Magnoliaceae species and the position of the IR border were closely related to the length of the ycfl gene. Phylogenetic analyses based on 66 shared genes from 30 species using maximum parsimony (MP) and max- imum likelihood (ML) methods provided strong support for the phylogenetic position of Magnolia. The availability of the complete cp genome sequence of M. grandiflora provides valuable information for breeding of desirable varieties, cp genetic engineering, developing useful molecular markers and phylogenetic analyses in Magnoliaceae.
文摘Lenin's theory of imperialism, which was based on a combination of Marxist theory with active reference to and dialectical critique of the theories of classical imperialism current at the time, provides an empirical analysis of the historical logic and laws of motion of capitalism. It realizes the sublation of theory and ideology by means of critical, explanatory and constructive logic, displaying a unique theoretical originality. Lenin's theory of imperialism offers a theoretical reflection on the development of capitalism and its historical trends; it is a practical response to the world structural contradictions arising from imperialism, an epochal reflection that vigorously explores and constructs the value claims of a new civilization and provides in-depth Marxist view of world history. The methodology and general principles of Lenin's study of imperialism are based on the materialist view of history. They are not out of date, nor are the propositions in his theory of imperialism, based as they are on research into monopoly. At the same time as it declared war on the values, systems and practices of the old world, Lenin's theory provided a completely new cognitive method, philosophical attitude and ideal values for the future society. This has become the precious wealth of mankind, wealth that transcends historical time and space.
