益气清热活血方不同制剂对创伤性肌肉损伤兔肌肉组织中水通道蛋白-1及血清白细胞介素-8表达水平的影响

目的探讨益气清热活血方的浸渍剂与巴布膏剂对创伤性肌肉损伤兔肌肉组织中水通道蛋白-1(AQP-1)及血清中白细胞介素-8(IL-8)表达水平的影响。方法将40只新西兰雄兔在适宜环境下饲养1周后随机分为空白组、模型组、浸渍组和巴布膏组,每组1... 目的探讨益气清热活血方的浸渍剂与巴布膏剂对创伤性肌肉损伤兔肌肉组织中水通道蛋白-1(AQP-1)及血清中白细胞介素-8(IL-8)表达水平的影响。方法将40只新西兰雄兔在适宜环境下饲养1周后随机分为空白组、模型组、浸渍组和巴布膏组,每组10只。空白组兔不给予任何干预措施;模型组、浸渍组和巴布膏组兔制备创伤性肌肉损伤模型。造模前对各组兔进行脱毛,造模成功后40 min,模型组兔用不含药物的巴布膏贴剂贴于患处10 h,每日1次;浸渍组兔用棉签蘸取适量清热益气活血浸渍液涂擦于患处,每日3次;巴布膏组兔用清热益气活血巴布膏贴剂贴于患处10 h,每日1次。分别于脱毛后、造模后及治疗24、72 h时对各组兔进行损伤症状评分。治疗24、72 h时,采用酶联免疫吸附试验法检测各组兔血清中IL-8水平,采用荧光定量聚合酶链式反应法检测各组兔肌肉组织中AQP-1 mRNA的相对表达量。结果脱毛后4组兔损伤症状评分比较差异均无统计学意义(P>0.05)。造模后和治疗24、72 h时,模型组、浸渍组和巴布膏组兔损伤症状评分显著高于空白组(P<0.05);造模后、治疗24 h时,模型组、浸渍组和巴布膏组兔损伤症状评分比较差异均无统计学意义(P>0.05);治疗72 h时,浸渍组和巴布膏组兔损伤症状评分显著低于模型组(P<0.05),浸渍组与巴布膏组兔损伤症状评分比较差异无统计学意义(P>0.05)。空白组兔不同时间点损伤症状评分比较差异无统计学意义(P>0.05)。模型组、浸渍组、巴布膏组兔造模后及治疗24、72 h时的损伤症状评分均显著高于脱毛后(P<0.05)。模型组兔治疗24、72 h时的损伤症状评分与造模后比较差异无统计学意义(P>0.05);模型组兔治疗72 h时的损伤症状评分显著低于治疗24 h时(P<0.05)。浸渍组兔治疗24 h时的损伤症状评分与造模后比较差异无统计学意义(P>0.05);浸渍组兔治疗72 h时的损伤症状评分显著低于造模后(P<0.05);浸渍组兔治疗72 h时的损伤症状评分显著低于治疗24 h时(P<0.05)。巴布膏组兔治疗24、72 h时的损伤症状评分显著低于造模后(P<0.05);巴布膏组兔治疗72 h时的损伤症状评分显著低于治疗24 h时(P<0.05)。治疗24、72 h时,模型组、浸渍组、巴布膏组兔血清中IL-8水平均显著高于空白组(P<0.05)。治疗24 h时,浸渍组与模型组兔血清中IL-8水平比较差异无统计学意义(P>0.05),巴布膏组兔血清中IL-8水平显著低于模型组(P<0.05),巴布膏组与浸渍组兔血清中IL-8水平比较差异无统计学意义(P>0.05)。治疗72 h时,浸渍组、巴布膏组兔血清中IL-8水平显著低于模型组(P<0.05),巴布膏组与浸渍组兔血清中IL-8水平比较差异无统计学意义(P>0.05)。治疗24、72 h时,模型组、浸渍组、巴布膏组兔肌肉组织中AQP-1 mRNA相对表达量显著高于空白组(P<0.05)。治疗24 h时,模型组、浸渍组、巴布膏组兔肌肉组织中AQP-1 mRNA相对表达量比较差异均无统计学意义(P>0.05)。治疗72 h时,浸渍组和巴布膏组兔肌肉组织中AQP-1 mRNA相对表达量显著低于模型组(P<0.05),巴布膏组与浸渍组兔肌肉组织中AQP-1 mRNA相对表达量比较差异无统计学意义(P>0.05)。空白组、模型组兔治疗24 h时与治疗72 h时血清中IL-8水平和肌肉组织中AQP-1 mRNA相对表达量比较差异均无统计学意义(P>0.05)。浸渍组、巴布膏组兔治疗72 h时血清中IL-8水平和肌肉组织中AQP-1 mRNA相对表达量显著低于治疗24 h时(P<0.05)。结论创伤性肌肉损伤兔在自然条件下72 h后可缓慢修复,清热益气活血方的浸渍剂与巴布膏剂均可有效加快其修复,该作用可能与抑制AQP-1及IL-8的表达有关。 展开更多

A correlation study of the expression of resistin and glycometabolism in muscle tissue after traumatic brain injury in rats 被引量:5

Objective: To investigate the expression pattern of resistin (RSTN) in skeletal muscle tissue and its influence on glycometabolism in rats with traumatic brain injury (TBI). Methods: Seventy-eight SD rats were randomly divided into traumatic group (n=36), RSTN group (n=36) and sham operation group (n=6). Fluid percussion TBI model was developed in traumatic and RSTN groups and the latter received additional 1 mg RSTN antibody treatment for each rat. At respectively 12 h, 24 h, 72 h, 1 w, 2 w, and 4 w after operation, venous blood was collected and the right hind leg skeletal muscle tissue was sampled. We used real-time PCR to determine mRNA expression of RSTN in skeletal muscles, western blot to determine RSTN protein expression and ELISA to assess serum insulin as well as fasting blood glucose (FBG) levels. Calculation of the quantitative insulin sensitivity check index (Q value) was also conducted. The above mentioned indicators and their correction were statistically analyzed. Results: Compared with sham operation group, the RSTN expression in the skeletal muscle as well as serum insulin and FBG levels revealed significant elevation (P〈0.05), and reduced Q value (P〈0.05) in traumatic group. Single factor linear correlation analysis showed a significant negative correlation between RSTN expression and Q values (P〈0.001) in traumatic group. Conclusion: The expression of RSTN has been greatly increased in the muscular tissue of TBI rats and it was closely related to the index of glycometabolism. RSTN may play an important role in the process of insulin resistance after TBI. 展开更多

According to medical literature, fracture of the first rib is quite rare and the bilateral condition is especially rare. This type of fracture is usually associated with severe intrathoracic trauma and other bony or neurovascular injuries, thus can be considered as a harbinger of major trauma. However here we present three cases of low velocity first rib fractures without any major trauma or multisystem injuries. All the three patients were treated conservatively and did well on simple analgesics and rest and had no early or late complications. It can be seen that not all the first rib fractures are associated with major trauma or multisystem injuries. There is a variant of first rib fracture with low velocity injuries which is not associated with any major complications in contrast to majority of first rib fractures associated with high velocity injuries. Causative factor of such injuries may be violent muscular contraction of scalenus anterior or serratus anterior, but not direct trauma. 展开更多
