Objective To investigate the growth-inhibitory effect of sunitinib malate on human bladder transitional cell carcinoma (TCC) in vitro. Methods Human bladder TCC cell line T24 was cultured and exposed to graded concentrations of sunitinib malate for 72 hours in vitro to determine the sensitivities to drug. Cell viability was measured by MTT assay. Cell apoptotic morphology was observed by fluorescence microscope following DAPl staining. Band expressions of Fas, Fas ligand, poly (ADP-ribose) polyrnerase (PARP) and D-actin were analyzed by Western blot. Wound healing process of T24 cells exposed to sunitinib malate was assayed. Results Sunitinib malate exerted a concentration-dependent and time-dependent inhibitory effect on the T24 cell lines. Fluorescence microscopy showed that small vacuoles appeared in the nuclei of T24 cells and the vacuoles were bigger with higher drug concentrations. The expressions of Fas ligand and PARP in T24 cells treated with sunitinib malate exhibited a concentration-dependent increase. Moreover sunitinib malate suppressed the wound healing process in a concentration-dependent manner. Conclusion Sunitinib malate exerted marked inhibitory activity against bladder cancer cell line T24. 展开更多

Where are the limits of the effects of exercise intensity on cognitive control?

Purpose: This study aimed to investigate whether workload intensity modulates exercise-induced effect on reaction time (RT) performances, and more specifically to clarify whether cognitive control that plays a crucial role in rapid decision making is altered. Methods: Fourteen participants performed a Simon Task while cycling 20 min at a light (first ventilatory threshold, VT~ - 20%), moderate (VTI), or very hard (VTj + 20%) level of exercise. Results: After 15 min of cycling, RTs are faster than during the first 5 min of exercise. This benefit does not fluctuate with the intensity of exercise and enlarges as RT lengthens. Despite a numerical difference suggesting a greater facilitation during moderate exercise (-16 ms) than during a light exercise (-10 ms), the benefit is not statistically different. Interestingly, we did not observe any signs of worsening on RT or on accuracy during very hard exercise. Conclusion: Cognitive control is extremely robust and appears not to be affected by the intensity of exercise. The selective inhibition and the between-trials adjustments are effective from the beginning to the end of exercise, regardless of the workload output. 展开更多

The nature of Stokes shift in quantum dots(QD) can be used to improve the quality of CLI. However, the interaction mechanism of QD with Cerenkov light remains unclear. In this work, the interaction mechanism between QD and radionuclides emitting β rays, γ rays, and Cerenkov light was investigated. The 96-well plates were used to test the different levels of radioactivity of radionuclides with different QD concentrations. Transparent vials were used to determine the relationship between QD fluorescence intensity and the distance from QD to the radionuclide. In addition, black paper was used to block the transmission of Cerenkov light through the QD vials. A linear relationship was found between the number of photons and the radioactivity of radionuclides when the QD concentration was kept constant. Similarly, the number of photons was linearly related to the QD concentration when the radioactivity of radionuclides was kept constant. Furthermore, with the increases in the distance between radionuclides and quantum dots, the number of photons was exponentially decreased. Meanwhile, the number of photons emitted from QD excited by Cerenkov light accounted for 20% the total number of photons excited by 131 I radionuclide. The result proved that QD was not only excited by Cerenkov light but also by other rays. 展开更多

In this work, we fabricated a monodisperse nanocomposite by coating gold nanorods (AuNRs) with a layer of biocompatible, stable carbon, obtaining AuNR@Carbon core-shell nanocapsules, which without any functionalization could be used as a molecule loading material due to its high surface areas. In this system, the AuNR core had a high-absorption cross section for con- version of near-infrared light to heat, which could be ex- plored for local hyperthermia. The carbon shell, which was biocompatible and stable even under concentrated acidic and alkaline conditions, was able to adsorb molecules with n-n interactions or electrostatic interactions. In comparison with AuNR@SiO2, AuNR@Carbon nanocapsules demon- strate the following merits: (1) simple and green synthesis method, (2) far more stable with respect to high-tem- perature stability and (3) larger molecule loading capacity, which indicate great potential in the biomedical applications. 展开更多
