AIM: To evaluate the effects of adenovirus-mediated gene transfer of RhoA siRNA and RhoC siRNA on proliferation and invasion of SGC7901 cells by Rho/ PI3K/Akt pathway. METHODS: Plasmid of RhoA siRNA and RhoC siRNA were constructed and transfected into SGC7901 cells. siRNA and LY294002 (PI3K inhibitor) were designed as the control group. The mRNA and protein expressions of RhoA and RhoC were respectively detected with RT-PCR and western blotting. In order to fi nd out the changes of proliferation and invasion power of SGC7901 cell lines, we analyzed the data by MTT, Boyden chamber and evaluated apoptosis of cell with flow cytometry. We treated BALB /C nude mice with RhoA and RhoC-siRNA, and tumor control rate (%) in nude mice was calculated. RESULTS: RhoA and RhoC siRNA transfections specifically down-regulated the corresponding mRNA and protein levels in SGC7901 Cells.The experiment of permeated artificial basal membrane showed that the invasion power of SGC7901 cell lines are on the decline after treatment of Ad-RhoA and RhoC-siRNA (12.64 ± 3.27 vs 87.38 ± 17.38, P < 0.05). The values of 490 nm wavelength light absorption were different in the five groups. The number of alive cells in the group of RhoA and RhoC-siRNA was lower than others in the 6th d (0.71 ± 0.01 vs 3.82 ± 0,11 P < 0.05). The apoptosis rate of transfected RhoA and RhoC-siRNA group with FACS were 19.07% ± 1.78 and there were significant differences between treated and control groups (19.07 ± 1.78% vs 1.23 ± 0.11%, P < 0.01). The tumor transplantation experiment in BALB/C nude mice showed intratumoralinjection of RhoA or RhoC siRNA can inhibit tumor growth. CONCLUSION: RhoA and RhoC siRNA gene therapy mediated by adenovirus may be useful for inhibiting growth and invasion of SGC7901 through a PI3K/Akt pathway. These results provide a novel therapeutic target in preventing gastric cancer cell invasion and metastasis. 展开更多

[Objective] This research aimed to study the pre-treatment conditions of head space so as to establish a HS-GC determination method, which is suitable for China's conditions, for trace volatile organic compounds in drinking water. [Method]The preparation method of head space was adopted for the volatile organic compounds in drinking water. [Result] The 20 kinds of volatile organic compounds in drinking water all could be detected simultaneously by using HS-GC-FID method,and they all could be separated well. The HS-GC-FID method could analyze the detected substances qualitatively and quantitatively. In addition, this detection method was characterized by larger linear range of concentration, higher precision, higher detection limit and higher recovery rate. [Conclusion] Under certain conditions, HSGC can reduce the loss of volatile organic compound in drinking water and improve the sensitivity of detection. The traditional method to refine crude cottonseed oil is time-consuming and expensive.This study evaluates the effectiveness of coagulation–flocculation–sedimentation process using quaternary polyamine-based polymers in refining crude cottonseed oil.Flocculated by four commercial polyamine-based cationic polymers(SL2700,SL3000,SL4500 and SL5000)with varied molecular weight(MW)and charge density(CD)and followed by coagulation with sodium hydroxide,crude cottonseed oil can be effectively purified.Free fatty acids,gossypol,pigments and trace elements are all effectively and sufficiently removed by the four polymers in a MW-and CDdependent manner.Our results suggest that the use of polyamine-based cationic polymers may offer an effective and feasible alternative to the traditional method for crude cottonseed oil refining. 展开更多

The numerical method, which is based on heuristic algorithms, has been widely used. An approach that combines differential evolution (DE) algorithm and control vector parameteri- zation (CVP) is proposed in this paper. In the proposed CVP, control variables are approximated with polynomials based on state variables and time in the entire time interval. Region reduction strategy is used in DE to reduce the width of the search region, which improves the computing efficiency. The results of the case studies demonstrate the feasibility and efficiency of the oroposed methods. 展开更多

However, the user association and resource allocation mechanism has not been thoroughly studied under given content placement situation. In this paper, we formulate the joint optimization problem of user association and resource allocation as a mixed integer nonlinear programming(MINLP) problem aiming at deriving a balance between the total utility of data rates and the total data rates retrieved from caches. To solve this problem, we propose a distributed relaxing-rounding method. Simulation results demonstrate that the distributed relaxing-rounding method outperforms traditional max-SINR method and range-expansion method in terms of both total utility of data rates and total data rates retrieved from caches in practical scenarios. In addition, effects of storage and backhaul capacities on the performance are also studied. 展开更多

Level control in flotation columns is an important factor that influences the recovery and the grade of concentrate from the column. A flotation column is a nonlinear, multi-variable problem with changeable parameters that traditional methods have difficulty controlling. We have applied fuzzy control methods to the flotation column and tested the performance of the design by Matlab/Simulink simulation. The simulations show that level control in the flotation column becomes smoother and more rapid with the fuzzy controller. Compared to PID control methods the overshoot in valve position, the adjustment time, and the robustness of the controller are all improved. This indicates that it is suitable to model fuzzy controllers in applications for the study of automatic control of flotation column. 展开更多

In this part, developed method means a mixed method on the basis of optical system and magnetic system used mainly for the present method of motion capture. We lessen the weak points of each method and propoe solution of create motion for pupct by offering more advantages. However, the estimated BER is not precise enough if the practical value of BER is high. A weighted EEC estimation method is proposed to improve the accuracy performance of BER estimation by classifying the raw estimation results into intervals and multiplying them by different coefficients separately. The applications of weighted EEC in modulation selection scheme and distributed video coding are discussed. Simulation results show that the EEC-based modulation selection method can achieve better performance at a cost of little redundancy and computation, and the EEC-based rate estimation method in distributed video coding can save the decoding time. 展开更多

Methods: Osteosarcoma cells line MG-63 was cultured in vitro. MTT assay and flow cytometry were used to study the effect of LMWH with different concentration suppressed the prolifetation and induced apop- tosis in osteosarcoma cells line MG-63. The expression of Livin of osteosarcoma cells line MG-63 was analysed by the im- munohistochemistrical method and PT-PCR. Results: Low molecular weight heparin could inhibit the growth of osteosarcoma cell line MG-63. With the LMWH's increasing, the apoptosis rate was increased significantly. Immunohistochemistrical method and PT-PCR showed that the expression of Livin of osteosarcoma cells line MG-63 declined obviously than that before medi- cation. Conclusion: LMWH has very strong anti-tumor effect in vitro. The possible mechanisms of LMWH anti-tumor effect are associate with the effect of suppressing the expression of Livin and inducing cell apoptosis. 展开更多

A user-defined micromechanical model was developed to investigate the fracture mechanism of asphalt concrete (AC) using the discrete element method (DEM). A three-dimensional (3D) AC beam was built using the "Fish" language provided by PFC3D and was employed to simulate the three-point bending beam test at two temperature levels: -10 ℃ and 15℃. The AC beam was modeled with the consideration of the microstructural features of asphalt mixtures. Uniaxial complex modulus test and indirect tensile strength test were conducted to obtain material input parameters for numerical modeling. The 3D predictions were validated using laboratory experimental measurements of AC beams prepared by the same mixture design. Effects of mastic stiffness, cohesive and adhesive strength on AC fracture behavior were investigated using the DEM model. The results show that the 3D DEM fracture model can accurately predict the fracture patterns of asphalt concrete. The ratio of stress at interfaces to the stress in mastics increases as the mastic stiffness decreases; however, the increase in the cohesive strength or adhesive strength shows no significant influence on the tensile strength. 展开更多

This paper discusses a design method for the control system of a weigh feeder that supplies powder and granular material at a constant rate. Most weigh feeders employed in industry are controlled by proportional and integral (PI) compensation, and the control performance is decided by the selection of parameters. To attain advanced control performance by PI control, the PI parameters are designed on the basis of generalized minimum variance control (GMVC). In this study, to achieve user-specified control performance by GMVC-based PI control, the design parameters of GMVC are automatically adjusted using a performance-adaptive method. The control performance discussed in this study consists of the variance of the control error and that of the difference in the control input. In a conventional performance-adaptive method, the variance of the control error is reduced. In this study, to reduce energy consumption and to achieve user-specified control performance, the variance of the difference in the control input is specified and the design parameter is determined. To demonstrate its effectiveness, the proposed method is applied to an actual weigh feeder. 展开更多

Here, a simple technique is proposed with and without using fires to transform the useless natural or man-made rubbish things to be a superfiber as well as thin film with multitasking applications in human daily life. To get better tracking performance of attitude command over the reentry phase of vehicles, the use of state-dependent Riccati equation (SDRE) method for attitude controller design of reentry vehicles was investigated. Guidance commands are generated based on optimal guidance law. SDRE control method employs factorization of the nonlinear dynamics into a state vector and state dependent matrix valued function. State-dependent coefficients are derived based on reentry motion equations in pitch and yaw channels. Unlike constant weighting matrix Q, elements of Q are set as the functions of state error so as to get satisfactory feedback and eliminate state error rapidly, then formulation of SDRE is realized. Riccati equation is solved real-timely with Schur algorithm. State feedback control law u(x) is derived with linear quadratic regulator (LQR) method. Simulation results show that SDRE controller steadily tracks attitude command, and impact point error of reentry vehicle is acceptable. Compared with PID controller, tracking performance of attitude command using SDRE controller is better with smaller control surface deflection. The attitude tracking error with SDRE controller is within 5°, and the control deflection is within 30°. 展开更多

KCI), calibration curve was constructed in phosphate buffer (pH = 7.0), the relationship is linear within the concentration range 1.283 × 10.5 M - 3.66 × 10.5 M with the correlation (R = 0.9923). The serial addition ofCPM (chlorpheniramine maleate) leads to the decrease in the reduction current peak (Ip), quantitatively, the plot of Alp (Ip - Ip) where, Ip: Peak current of N, N-diethyl-p-nitroso aniline alone, lp: peak current of N,N-diethyl-p-nitroso aniline in the presence of CPM, versus concentration is linear within the concentration range 0.984 × 10-6 M - 9.756 × 10-6 M, the correlation coefficient was 0.9954. Extraction of AchE, relationship between substrate and enzyme concentration, and inhibition effects of the agrochemicals to AchE are discussed in this paper. Through the re-search, the proper AchE concentration for hydrolysis of 1 ml 1mmol/L substrate and I50 val-ues of the agrochemicals to AchE are decided. It is proved that Asch-DTNB method is a rapid test tool for agrochemical residues in fruits and vegetables.A rapid test card has been developed with sensitivity of 0.05mg/L. 展开更多
