Taking the emergency braking of the 2×104 t combined train as example, the train longitudinal impulse, the coupler dynamic behaviors and wheel-rail interactions of vehicles distributing in the different positions are analyzed. The results indicate that under the coupler compressing forces, the couplers of middle locomotives may tilt to the free swing limits, which induces the unidirectional tilt of their connected wagon couplers. Consequently, the coupler longitudinal forces produce the lateral components, and then affect the wheel-rail dynamic interaction. The performance of the middle locomotive and their neighboring freight wagons deteriorate significantly, becoming the most dangerous parts in the combined train. The wagons disconnecting with the locomotives can basically keep their couplers to stabilize in the centering positions, even though the maximum coupler longitudinal force acts on it. And its corresponding running safety also has little changes.展开更多

Papaveraceae and has proved to be an- ti-arrhythmic. The purpose of our study is to investigate the effects of ALL on transmural repolarizing ionic ingredients of outward potassium current (Ito) and slow delayed rectifier potassium current (IKs). Methods The monophasic action potential (MAP) technique was used to record the MAP duration of the epicardium (Epi), myocardium (M) and endocardium (Endo) of the rabbit heart and the whole cell patch clamp was used to record/to and IKs in cardiomyocytes of Epi, M and Endo layers that were isolated from rabbit ventricles. Results The effects of ALL on MAP of Epi, M and Endo layers were disequilibrium. ALL could effectively reduce the transmural dispersion of repolarization (TDR) in rabbit transmural ventricular wall. ALL decreased the current densities of/to and IKs in a voltage and concentration dependent way and narrowed the repolarizing differences among three layers. The analysis of gating kinetics showed ALL accelerated the channel activation ofIto in M layers and partly inhibit the channel openings of/to in Epi, M and Endo cells. On the other hand, ALL mainly slowed channel deactivation of IKs channel in Epi and Endo layers without affecting its activation. Conclusions Our study gives partially explanation about the mechanisms of tmnsmural inhibition of/to and IKs channels by ALL in rabbit myocardium. These findings provide novel perspective regarding the anti-arrhythmogenesis application of ALL in clinical settings.展开更多

An example of the real RC building is used to demonstrate differences in the dynamic response results in the calculation of internal forces and displacements. Variant three options of the soil models were used as a building supporting structure. In the case of soil model A, the soil was modelled by using of equivalent stiffness values, stemming from the theory of a rigid circular disc on an elastic homogeneous half-space. Non-uniformly modelled vertical stiffness of the soil according to the Boussinesq model was used for model B. Both models A and B are characterised by the "averaged" soil model on the bases of spring constants. Model C was used for the soil better corresponding to its actual composition by the Winkler-Pasternak theory. Model C, where the actual layered soil is considered, is modelled more accurately than for the "averaged" soil of models A and B. The dynamic response of models operating with "averaged" values of rigid and soft soil layers is markedly shifted to the conservative smaller values of internal forces. The building response tbr model C in dynamic displacements is significantly higher than for the both models A and B.展开更多

In this study, a dataset containing 1969 compounds was compiled from literature and FDA-approved drugs. Using a support vector machine (SVM), two groups of computational models were built to distinguish whether a compound is a blocker or non-blocker of hERG potassium ion channel. These mod- els fit generally satisfactory. The 100 models built with MACCS fingerprints (Model Group A) showed an average accuracy of 90% and an average matthews correlation coefficient (MCC) value of 0.77 on the test sets. The 100 models built with selected MOE descriptors (Model Group B) showed an average accuracy of 89% and an average MCC value of 0.74 on the test sets. Molecular hydrophobicity and lipophilicity were found to be very important factors which lead to block the hERG potassium ion channel. Some other molecular properties such as electrostatic properties, features based on van der Waals surface area, the number of rigid bonds and molecular surface rugosity also played important roles in blocking bERG potassium ion channel.展开更多

Methods: Sixty Wistar rats were divided randomly into four groups: a control group (n=15), an SM group (n=15), a myocardial infarction (MI) group (n=15), and an MI+SM group (n=15). Chronic MI was obtained by ligating the left anterior descending branch (LAD) of rat hearts for eight weeks. The in vivo blockade of stretch-activated ion channels (SACs) was achieved by intramuscular injection of SM (180 mg/(kg·d)) for seven days after operation. The hearts were stretched for 5 s by occlusion of the aortic arch. Suction electrodes were placed on the anterior wall of left ventricle to record the monophasic action potential (MAP). The effect of stretching was examined by assessing the 90% monophasic action potential duration (MAPD90), premature ventricular beats (PVBs), and ventricular tachycardia (VT). Results: The MAPD90 decreased during stretching in both the control (from (50.27±5.61) ms to (46.27±4.51) ms, P〈0.05) and MI groups (from (65.47±6.38) ms to (57.47±5.76 ms), P〈0.01 ). SM inhibited the decrease in MAPD90 during inflation ((46.27±4.51) ms vs. (49.53±3.52) ms, P〈0.05 in normal hearts; (57.47±5.76) ms vs. (61.87±5.33) ms, P〈0.05 in MI hearts). The occurrence of PVBs and VT in the MI group increased compared with that in the control group (PVB: 7.93±1.66 vs. 1.80±0.86, P〈0.01; VT: 7 vs. 1, P〈0.05). SM decreased the occurrence of PVBs in both normal and MI hearts (0.93±0.59 vs. 1.80±0.86 in normal hearts, P〈0.05; 5.40±1.18 vs. 7.93±1.66 in MI hearts, P〈0.01). Conclusions: Stretch-induced MAPD90 changes and arrhythmias were observed in chronically infarcted myocardium. The use of SM in vivo decreased the incidence of PVBs but not of VT. This suggests that SACs may be involved in mechanoelectric feedback (MEF), but that there might be other mechanisms involved in causing VT in chronic MI.展开更多

In this paper, the Reynolds averaged Navier-Stokes equations (RANS) and the RNG k-epsilon turbulence model is employed to simulate the flow in a pump. The flow field in the centrifugal pump is computed for a range of flow rate. The simulation results have been compared with the experimental data and good agreement has been achieved. In order to study the interaction of the tongue with the impeller, fifteen monitor probes are evenly distributed circumferentially at three radii around the tongue. Pressure distribution is investigated at various blade positions while the blade approaches to and leaves the tongue region. Results show that pressure signal fluctuates largely around the tongue, and it is more intense near the tongue surface. At design condition, standard deviation of pressure fluctua- tion is the minimum. At large flow rate, the increased low pressure region at the blade trailing edge results in the increases of pressure fluctuation amplitude and pressure spectra at the monitor probes. Minimum pressure is obtained when the blade is facing to the tongue. It is found that the amplitude of pressure fluctuation strongly depends on the blade positions at large flow rate, and pressure fluctuation is caused by the relative movement between blades and tongue. At small flow rate, the rule of pressure fluctuation is mainly depending on the structure of vortex flow at blade passage exit besides the influence fTom the relative position between the blade and the tongue.展开更多

At the microscales, the DDD simulations are responsible for capturing the evolution of dislocation structures; at the nanoscales, the MD simulations are responsible for obtaining the GB energy and ISF energy which are then transferred hierarchically to the DDD level. In the present model, four kinds of dislocafion-GB interactions, i.e. transmission, absorption, re-emission and reflection, are all considered. By this methodology, the compression of a Cu mi- cro-sized bi-crystal pillar is studied. We investigate the characteristic mechanical behavior of the bi-crystal compared with that of the single-crystal. Moreover, the comparison between the present penetrable model of GB and the conventional impenetrable model also shows the accuracy and efficiency of the present model.展开更多
