Cheng by alcohol extraction,n-hexane,ethyl acetate and n-utanol phase.Furthermore antialgal activities of all components were determined.[Result] The active substances of M.grandiflora mainly existed in n-hexane and n-butanol phases.The active substances of S.japonica mainly existed in n-hexane phase,while the active substances of B.sinica mainly existed in ethyl acetate and aqueous phase.All the active substances showed significant antialgal activity (50% inhibition) after culture for 1 d except aqueous phase of B.sinica.In addition,inhibition rates of the tested phases increased with the increasing of culture duration and testing concentrations.The inhibition rates were over 90% against the growth of M.aeruginosa after culture for 7 d.The n-hexane and n-butanol phases of M.grandiflora exhibited the best antialgal activity,over 96% inhibition rate at the concentration of 25 μg/ml,which was significantly higher than those of the other components,furthermore it could cause shrinkage and obvious surface inanition of Microcystis aeruginosa.[Conclusion] The n-hexane and n-butanol phases of M.grandiflora have the best inhibition activities on the growth of Microcystis aeruginosa,so they are worthy of further study.展开更多

METHODS: The effect of dexamethasone (DEX) on growth and chemosensitivity was assessed in 14 carc...AIM: To determine how glucocorticoids (GCs) may affect the growth and chemosensitivity of common carcinoma cells. METHODS: The effect of dexamethasone (DEX) on growth and chemosensitivity was assessed in 14 carcinoma cell lines. The function of GC receptors (GR) was assessed by MMTV reporter assay. Overexpression of GR was done by in vitro transfection and expression of a GR-expressing vector. Immunohistochemical stain of tissues and ceils were done by PA1-511A, an anti-GR monodonal antibody. RESULTS: DEX inhibited cell growth of four (MCF-7, MCF- 7/MXR1, MCF-7/TPT300, and HeLa), increased cisplatin cytoxicity of one (SiHa), and decreased dsplatin cytotoxicity of two (H460 and Hep3B) cell lines. The GR content of the seven cell lines affected by DEX was significantly higher than those of the seven cell lines unaffected by DEX (5.2±2.5×10^4 sites/cell vs1.3±1.4×10^4 sites/cell, P= 0.005). Only two DEX-unresponsive cell lines {NPC-TW01 and NPC- TW04) oontained high GR amounts in the range (1.9-8.1×10^4 sites/cell) of the seven DEX-responsive cell lines. The GR function of NPC-TW01 and NPC-TW04, however, was foundto be impaired. The importance of high cellular amount of GR in mediating DEX susceptibility of the cells was further exemplified by GR dose-dependent drug resistance to cisplatin of AGS, a cell line with low GR content and was unaffected by DEX before transfection of GR-expressing vector. Immunohistochemical studies of human cancer tissues showed that 5 of the 45 (11.1%) breast cancer and 43 of the 85 (50.6%) non-small cell lung cancer had high GR contents at the ranges of the GC-responsive carcinoma cell lines. CONCLUSION: The growth and chemosensitivity of human carcinomas with high GR contents may be affected by GC. However, in light of the heterogeneous and even contradictive effects of GC on these cells, routine examination of GR contents of human carcinoma tissues may not be clinically useful until other markers that help predict the ultimate effect of GC on individual patients are identified.展开更多

We investigated the allelopathic interactions between Karenia mikimotoi and Dunaliella salina in laboratory cultures based on different temperature (15℃, 20℃, and 25℃) and lighting (40,80, and 160 umol/(m2·s)) conditions. The growth of D. salina in bi-algae culture (1:1 size/density) was significantly restrained. The results of cell-free filtrate culture indicate that direct cell-to- cell contact was not necessary in interspecific competition. Further experimental results demonstrated that allelochemicals released from K. miMmotoi were markedly influenced by both temperature (P=0.013) and irradiance (P=0.003), resulting in different growth characteristics olD. salina in filtrate mediums. Compared with the plateau period, K. mikimotoi exudates in the exponential phase had a stronger short-term inhibition effect on D. salina in normal conditions. A clear concentration-dependent relationship was observed in the effect of allelochemicals released from K. mikimotoi with low-promoting and high-repressing effects on D. Salina in a short time-scale. In addition, allelopathic substances remain stable and effective under high temperature and pressure stress. Many flocculent sediments adhering with D. salina cells were observed in all filtrate mediums, while the quantity and color depended on the original culture conditions.展开更多

Three types of palm diacylglycerol olein (PDAGOL) at different degree of unsaturation (PDAGOL1V56, PDAGOLIV62 and PDAGOLIV64) were used as main raw materials blended with palm stearin (PS). The blending compositions ranged from 30% to 70% of PDAGOLIV56/PS, PDAGOLIV62/PS and PDAGOLIV64/PS, respectively. The physicochemical properties of all binary blend systems were characterized for fatty acid composition (FAC), slip melting point (SMP) and solid fat content (SFC). The selected bakery shortening formulations were further characterised for polymorphic form of fat crystal and thermal behavior, using X-ray diffractometer (XRD) and differential scanning calorimetry (DSC). Bakery shortening enriched with diacylglycerol that were produced from 40DS56 (40% PDAGOLIV56/60% PS), 40DS62 (40% PDAGOL1V62/60% PS) and 40DS64 (40% PDAGOLIV64/60% PS) had 45%-50% unsaturated fatty acid and crystallized in β + β polymorphs; thus they were suitable for shortening system. Based on product's baking performance, it could be found that all Madeira cakes prepared from bakery shortening enriched with diacylglycerol had higher specific cake volume as compared to commercial shortening (CS). In customer acceptance test, Madeira cake made from 40DS56 shortening scored the highest rating for all sensory attributes, including overall customer acceptability. It had given an indication that 40DS56 shortening formulation was the most suitable fat blends to be used as bakery shortening.展开更多
To explore the polysaccharides from selected seaweeds of Atlantic Canada and to evaluate their potential anti-influenza virus activities, polysaccharides were isolated from several Atlantic Canadian seaweeds, includin...To explore the polysaccharides from selected seaweeds of Atlantic Canada and to evaluate their potential anti-influenza virus activities, polysaccharides were isolated from several Atlantic Canadian seaweeds, including three red algae (Polysiphonia lanosa, Furcellaria lumbricalis, and Palmaria palmata), two brown algae (Ascophyllum nodosum and Fucus vesiculosus), and one green alga (Ulva lactuca) by sequential extraction with cold water, hot water, and alkali solutions. These polysaccharides were ana-lyzed for monosaccharide composition and other general chemical properties, and they were evaluated for anti-influenza virus activities. Total sugar contents in these polysaccharides ranged from 15.4% (in U. lactuca) to 91.4% (in F. lumbricalis); sulfation level was as high as 17.6% in a polysaccharide from U. lactuca, whereas it could not be detected in an alikali-extract from P. palmaria. For polysaccharides from red seaweeds, the main sugar units were sulfated galactans (agar or carrageenan) for P. lanosa, F. lumbricalis, and xylans for P. palmata. In brown seaweeds, the polysaccharides largely contained sulfated fucans, whereas the polysaccharides in green seaweed were mainly composed of heteroglycuronans. Screening for antiviral activity against influenza A/PR/8/34 (H1N1) virus revealed that brown algal polysaccharides were particularly effective. Based on a series of aqua-planet and air–sea coupled experiments,the influence of unrealistic treatment of water substance in the Flexible Global Ocean–Atmosphere–Land System Model,spectral version 2(FGOALS-s2),o...Based on a series of aqua-planet and air–sea coupled experiments,the influence of unrealistic treatment of water substance in the Flexible Global Ocean–Atmosphere–Land System Model,spectral version 2(FGOALS-s2),on the model's climate sensitivity is investigated in this paper.Because the model does not adopt an explicit microphysics scheme,the detrained water substance from the convection scheme is converted back to the humidity.This procedure could lead to an additional increase of water vapor in the atmosphere,which could strengthen the model's climate sensitivity.Further sensitivity experiments confirm this deduction.After removing the water vapor converted from the detrained water substance,the water vapor reduced significantly in the upper troposphere and the high clouds also reduced.Quantitative calculations show that the water vapor reduced almost 10% of the total water vapor,and 50% at 150 h Pa,when the detrained water substance was removed,contributing to the 30% atmospheric surface temperature increase.This study calls for an explicit microphysics scheme to be introduced into the model in order to handle the detrained water vapor and thus improve the model's simulation skill.展开更多

Reference materials with two concentration levels of heavy metals were prepared by spiking Pb and Cd compounds to epoxy molding compounds made by mixing silica powders and epoxy resin. The concentration changes of the reference materials during stability test for 1 a were not observed. In the homogeneity assessment, the as-prepared reference materials were studied by using three different analytical tools, inductively coupled plasma atomic emission spectrometry (ICP-AES), X-ray fluoroescence spectrometry (XRF) and laser ablation ICP mass. In order to define the combined effects of spinosad and plant allelochemicals on Helicoverpa armigera(Lepidoptera:Noctuvidae), the research fed them on artificial diets with different doses of spinosad, gossypol, quer...In order to define the combined effects of spinosad and plant allelochemicals on Helicoverpa armigera(Lepidoptera:Noctuvidae), the research fed them on artificial diets with different doses of spinosad, gossypol, quercetin and tannicacid, and the changes of pupal and imaginal biological characteristics were observed and recorded. The results showed that spinosad at sublethal concentrations decreased pupation ratio and pupal weight, prolonged prepupal and pupal periods, and decreased emergence ratio, fecundity and longevity of adults. Combined with plant allelochemicals, spinosad significantly prolonged pupal period and shorten adult longevity. Other life-cycle parameters such as pupation ratio, pupal weights, emergence ratio, eggs laid per female and hatch ratio did not change significantly. The results clearly indicated that plant allelochemicals did not change spinosad efficiency on H. armigera, but the combination of spinosad and plant allelochemicals might affect pest population dynamics significantly by changing its development.展开更多

Methods: Eukaryotic expression plasmid pcDNA3.1(±)-ssRKIP containing human overall length RKIPcDNA was transfected into cervical cancer Hela cell by lipofectin assay, establishing a stable cell line containing a target gene by G418. Expression of RKIP in Hela cells was measured by Western blot analysis. After treatment with cisplatin of different concentrations and intervals of time, the effect of RKIP on the proliferation of Hela cells was evaluated by MTT method. The flow cytometry was used to investigate whether the RKIP could inhibit apoptosis in Hela cells induced by cisplatin. Results: The expression of RKIP in Hela cells transfected with pcDNA3.1-ssRKIP was increased obviously. After different concentrations of cisplatin treatment cells for 24, 48 and 72 h, the growth inhibition rate in Hela cells transfected with pcDNA3.1-ssRKIP was significantly higher than in control cells (P 〈 0.05). With 5 pg/mL cisplatin treatment for 24 h, pcDNA3.1-ssRKIP-transfected Hela cells had an obviously higher percentage of apoptosis (23.2 ± 0.24)% than non-transfected cells (12.4 ± 0.31)% and empty vector-transfected cells (13.4 ± 0.47)%. Without treatment of cisplatin, the percentage of apoptosis for Hela cells transfected with pcDNA3.1-ssRKIP was (5.7 ± 0.12)%, which was still higher than those of the non-transfected cells (2.9 ± 0.21)% and empty vector-transfected cells (3 ± 0.08)%. Conclusion: Higher expres- sion of RKIP gene can improve chemosensitivitv of cervical cancer Hela cells to cisplatin.展开更多

In this study, a simple one-pot synthesis method was proposed for the preparation of fluorescent probes based on ovalbumin-stabilized gold nanoclusters. This strategy allowed the generation of water-soluble gold nanoclusters within 5 min. The as-prepared fluorescent probe exhibited a red fluorescence emission at 625 nm, and good thermostability. The fluorescent probe was applied to measure glucose concentrations based on the hydrogen peroxide-induced fluorescence quenching principle, and showed favorable biocompatibility, high sensitivity and good selectivity. As a result of the advantageous properties and performance of this fluorescent probe, the present assay allowed for the selective determination of glucose in the range of 5.0×10-6 to 10.0×10-3 mol/L with a detection limit of 1.0×10?6 mol/L. Moreover, the glucose content in urinary samples was analyzed using the constructed fluorescent probe: this indicated the potential of the fluorescent gold nanoclusters for applications in biological and clinical diagnosis and therapy.展开更多

文摘Based on a series of aqua-planet and air–sea coupled experiments,the influence of unrealistic treatment of water substance in the Flexible Global Ocean–Atmosphere–Land System Model,spectral version 2(FGOALS-s2),on the model's climate sensitivity is investigated in this paper.Because the model does not adopt an explicit microphysics scheme,the detrained water substance from the convection scheme is converted back to the humidity.This procedure could lead to an additional increase of water vapor in the atmosphere,which could strengthen the model's climate sensitivity.Further sensitivity experiments confirm this deduction.After removing the water vapor converted from the detrained water substance,the water vapor reduced significantly in the upper troposphere and the high clouds also reduced.Quantitative calculations show that the water vapor reduced almost 10% of the total water vapor,and 50% at 150 h Pa,when the detrained water substance was removed,contributing to the 30% atmospheric surface temperature increase.This study calls for an explicit microphysics scheme to be introduced into the model in order to handle the detrained water vapor and thus improve the model's simulation skill.

文摘Reference materials for quantitative determination of regulated heavy metals, such as Pb and Cd in electronic components, were designed and investigated in terms of stability and homogeneity. Reference materials with two concentration levels of heavy metals were prepared by spiking Pb and Cd compounds to epoxy molding compounds made by mixing silica powders and epoxy resin. The concentration changes of the reference materials during stability test for 1 a were not observed. In the homogeneity assessment, the as-prepared reference materials were studied by using three different analytical tools, inductively coupled plasma atomic emission spectrometry (ICP-AES), X-ray fluoroescence spectrometry (XRF) and laser ablation ICP mass. The results show different homogeneities by the characteristics of analytical tools and the materials.

文摘In order to define the combined effects of spinosad and plant allelochemicals on Helicoverpa armigera(Lepidoptera:Noctuvidae), the research fed them on artificial diets with different doses of spinosad, gossypol, quercetin and tannicacid, and the changes of pupal and imaginal biological characteristics were observed and recorded. The results showed that spinosad at sublethal concentrations decreased pupation ratio and pupal weight, prolonged prepupal and pupal periods, and decreased emergence ratio, fecundity and longevity of adults. Combined with plant allelochemicals, spinosad significantly prolonged pupal period and shorten adult longevity. Other life-cycle parameters such as pupation ratio, pupal weights, emergence ratio, eggs laid per female and hatch ratio did not change significantly. The results clearly indicated that plant allelochemicals did not change spinosad efficiency on H. armigera, but the combination of spinosad and plant allelochemicals might affect pest population dynamics significantly by changing its development.

文摘Objective: The purpose of the study is to investigate the effects of up-regulation of Raf kinase inhibitor protein (RKIP) on the chemosensitivity of cervical cancer Hela cells. Methods: Eukaryotic expression plasmid pcDNA3.1(±)-ssRKIP containing human overall length RKIPcDNA was transfected into cervical cancer Hela cell by lipofectin assay, establishing a stable cell line containing a target gene by G418. Expression of RKIP in Hela cells was measured by Western blot analysis. After treatment with cisplatin of different concentrations and intervals of time, the effect of RKIP on the proliferation of Hela cells was evaluated by MTT method. The flow cytometry was used to investigate whether the RKIP could inhibit apoptosis in Hela cells induced by cisplatin. Results: The expression of RKIP in Hela cells transfected with pcDNA3.1-ssRKIP was increased obviously. After different concentrations of cisplatin treatment cells for 24, 48 and 72 h, the growth inhibition rate in Hela cells transfected with pcDNA3.1-ssRKIP was significantly higher than in control cells (P 〈 0.05). With 5 pg/mL cisplatin treatment for 24 h, pcDNA3.1-ssRKIP-transfected Hela cells had an obviously higher percentage of apoptosis (23.2 ± 0.24)% than non-transfected cells (12.4 ± 0.31)% and empty vector-transfected cells (13.4 ± 0.47)%. Without treatment of cisplatin, the percentage of apoptosis for Hela cells transfected with pcDNA3.1-ssRKIP was (5.7 ± 0.12)%, which was still higher than those of the non-transfected cells (2.9 ± 0.21)% and empty vector-transfected cells (3 ± 0.08)%. Conclusion: Higher expres- sion of RKIP gene can improve chemosensitivitv of cervical cancer Hela cells to cisplatin.

文摘Protein protected gold nanoclusters have outstanding physical and chemical properties that make them excellent scaffolds for the construction of novel chemical and biological probes. In this study, a simple one-pot synthesis method was proposed for the preparation of fluorescent probes based on ovalbumin-stabilized gold nanoclusters. This strategy allowed the generation of water-soluble gold nanoclusters within 5 min. The as-prepared fluorescent probe exhibited a red fluorescence emission at 625 nm, and good thermostability. The fluorescent probe was applied to measure glucose concentrations based on the hydrogen peroxide-induced fluorescence quenching principle, and showed favorable biocompatibility, high sensitivity and good selectivity. As a result of the advantageous properties and performance of this fluorescent probe, the present assay allowed for the selective determination of glucose in the range of 5.0×10-6 to 10.0×10-3 mol/L with a detection limit of 1.0×10?6 mol/L. Moreover, the glucose content in urinary samples was analyzed using the constructed fluorescent probe: this indicated the potential of the fluorescent gold nanoclusters for applications in biological and clinical diagnosis and therapy.
