[Method] By using 0.2 mg/ml ellagic acid standard solution, on the basis of single-factor experiment and orthogonal experiment, chromatographic conditions (mobile phase ratio, flow rate, col- umn temperature) for detecting ellagic acid using HPLC were optimized. Based on the optimal chromatographic conditions, the ellagic acid content in experimental pomegranate peels was determined. [Resull] The optimal chromatographic conditions for detecting ellagic acid in pomegranate peels using HPLC method are: 1.2% phos- phoric acid:acetonitrile=85:15, column temperature of 35 ℃, and flow rate of 1.0 ml/min. The linear regression equation of ellagic acid is: y=2.9e+0.6x+4.4e+5 (FF=9 999). Ac- cording to the standard addition recovery test, the average recovery rate of ellagic acid is 98.20%, and RSD is 0.60%. Under above optimized chromatographic condi- tions, ellagic acid can be well separated from other interfering components in pomegranate peels, with shorter peak time and ideal effect, which is convenient for the detection in production practices. [Conclusion] This study laid the foundation for detecting ellagic acid in pomegranate peels using HPLC method.展开更多

[Method] With ellagic acid yield as an indicator, ultrasound extraction method was adopted to extract the ellagic acid from pomegranate peels, and the concentration of ellagic acid was measured by using ul- traviolet spectrophotometry; L9(34) orthogonal experiment was designed with four fac- tors at three levels, to investigate the effect of extraction temperature, extraction du- ration, solid-liquid ratio and ultrasound power on extraction rate of ellagic acid. [Re- sult] The optimal extraction condition for ellagic acid is extraction temperature of 30 ~C, solid-liquid ratio of 1:200, extraction duration of 20 min, and ultrasound power of 50 W. The relationship between primary and secondary factors affecting experimental indicators was solid-liquid ratio 〉 extraction duration 〉 ultrasound power 〉 extraction temperature. [Conclusion] Solid-liquid ratio has the maximum effect on extraction rate of ellagic acid. The ellagic acid extraction technology identified in this study is rea- sonable and feasible.展开更多

(guava) were extracted by water and aqueous ethyl alcohol 50% (1:10) ratio, and the total phenolic content in the extracts was determined spectrophotometrically according to Folin- Ciocalteu’s phenol method and calculated as gallic acid equivalent (GAE). Remarkably high total phenolic content 575.3 ±15.5 and 511.6±6.2 mg of GAE/g of dried weight material (for ethanol guava leaf extracts and water guava leaf extracts, respectively) were obtained. The antioxidant activity of lyophilized extracts was determined at ambient temperature by means of a 2,2-diphenyl-1-picrylhydryzyl (DPPH˙) colorimetry with detection scheme at 515 nm. The activity was evalu- ated by the decrease in absorbance as the result of DPPH˙ color change from purple to yellow. The higher the sample concentration used, the stronger was the free radical-scavenging effect. The results obtained showed that ascorbic acid was a substantially more powerful antioxidant than the extracts from guava leaf. On the other hand, the commercial guava leaf extracts and ethanol guava leaf extracts showed almost the same antioxidant power whereas water guava leaf extracts showed lower antioxidant activity. The parameter EC50 and the time needed to reach the steady state to EC50 concentration ( TEC50 ) affected the antiradical capacity of the sample. The antioxidant efficiency (AE) has been shown to be a more ade- quate parameter for selecting antioxidants than the widely used EC50. This study revealed that guava leaf extracts comprise effective potential source of natural antioxidants.展开更多

In nuclear medicine, blood constituents (BC) are labelled with techne-tium-99m (99mTc) and used to image procedures. However, data have demonstrated that synthetic or natural drugs could modify the labelling of BC with 99mTc. The aim of this work was to evaluate the effects of aqueous extract of guava leaves on the labelling of BC with 99mTc. Blood samples of Wistar rats were incubated with different concentrations of guava extract and labelled with 99mTc after the percentage of incorporated radioactivity (%ATI) in BC was determined. The results suggest that aqueous guava extract could present antioxidant action and/or alters the membrane structures involved in ion transport into cells, thus decreasing the radiolabelling of BC with 99mTc. The data showed significant (P<0.05) alteration of ATI in BC from blood incubated with guava extract.展开更多

Antioxidative components from pomegranate peel were extracted by using different solvents like methanol, ethanol, acetone, chloroform and ethyl acetate. All the extracts were tested for Total phenolic content (TPC) and the methanol which proved to be best in extracting the phenolic compounds was used to assess the antioxidant property by using different methods like Diphenyl piridyl picryl hydrazyl (DPPH), Relative reducing power (RRP), Beta carotene bleaching and Ferric thiocyanate (FTC). The antioxidant activity (AA) of pomegranate peel extract (PPE) was compared with Butylated hydroxyl toluene (BHT) and Butylated hydroxyl anisole (BHA). The methanolic extract was examined by Liquid Chromatography and Mass Spectroscopy (LCMS) and the spectra showed the presence of different phenolic compounds. The extracts were tested for their efficiency in preventing rancidity of Refined Bleached Deodorized Soybean oil (RBDSBO) in comparison with BHT and control at 200 ppm concentration. The PPE proved to be best in preventing rancidity. Owing to the present results, the studies can further be extended to exploit the PPE for application into different food products.展开更多

The aim of this research was to investigate the cytotoxicity and antioxidant activities of Moroccan Pomegranate (peel, leaves, branches, flowers and corolla). Further, the biological activities were correlated with phytochemical contents of the plant extracts. Methanolic extract from different parts of Punica Granatum was assessed for its antiproliferative activity in two human cancer (breast and colon) cells lines (MBA-MD 231 and HT-29), through MTT (3-(4,5-dimethyl-2- thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide) bioassay using cell viability and cytotoxicity indices. DPPH (2,2-diphenyl-l-picrylhydrazyl) assay was conducted to screen the antioxidant property of the extracts together with its phenolic and flavonoids content were evaluated, as well. The methanolic extract ofPunica Granatum (peel, leaves, branches, flowers and corolla) showed the highest antiproliferative activity on MBA-MD 231 (IC50 was 133.53-233.32 μg/mL) and HT-29 (IC50 was 127.58-203.24 μg/mL) cells. Antioxidants contents are distributed as follows: peel 〉 leaf 〉 flower 〉 corolla 〉 branches. The inhibitory activities required for decreasing initial DPPH by 50% are 8.27, 9.9, 10.06, 11.67 and 13.28 μg/mL, respectively. These results are in correlation with polyphenols content from corolla, peel, leaves, flower and branches are 120.7, 115, 96.65, 90.73 and 64.67 mg GAE/g dw (mg gallic acid equivalents per g dry weight) and flavonoids are 188.8, 221.7, 180.2, 193.7 and 158.5 mg QE/g dw (mg quercetin equivalents per g dry weight). Our results show that the peel, flowers, corolla, leaves and branches of Moroccan Pomegranate may contain a lot of bioactive compounds which are responsible for strong antioxidant and cytotoxicity activities observed here. Our finding indicates the possibility of using the extracts of this plant as source of natural antioxidant and anticancer mainly for its abundant phenolic and flavonoid contents.展开更多
