Validation of Monomeric Anthocianin Determination Method for Bilberry Juice and Marc Extracts

The solid waste generated in industrial berry juice production was considered as a low cost raw material for the extraction of natural antioxidants. Berries contain phenolic compounds with high antioxidant potential, including anthocianin. Quantitative determination method for monomeric anthocianins in bilberry juice and marc was validated. An official method from Association of Analytical Communities was used to determine anthocianins in juice and marc extracts by measuring light absorption for solutions with pH values 1.0 and 4.5 at 520 nm and 700 rim. Results were expressed as cyanidin-3-glucoside equivalent, as it is the most common anthocianin pigment. Calibration curve was obtained, linearity was checked, working interval and accuracy was determined and samples were tested. Mentioned method was evaluated as appropriate for quantitative anthocianin analysis in bilberry juice and marc. Necessity of calibration curve was approved using extinction coefficient of cyanidin-3-glucoside instead. Method assures adequate precision and accuracy as well. 展开更多

To perform the mechanism study of special association for vancomycin and D-Ala-D-Ala-containing peptides on the interface of solution and self-assemble monolayer, the binding between vancomycin and pentapeptide (Lys-Lys-Gly-D-Ala-D-Ala) was investigated by flow injection surface plasmon resonance (FI-SPR) and flow injection quartz crystal microbalance (FI-QCM). To facilitate the formation of a compact vancomycin adsorbates layer with a uniform surface orientation, vancomycin molecules were attached onto a preformed alkanethiol self-assembled monolayer. By optimizing the conditions for the binding between Lys-Lys-Gly-D-Ala-D-Ala and vancomycin on the assembled chip, the detecting limit of Lys-Lys-Gly-D-Ala-D-Ala was greatly improved (reaching 0.5 ×10^- 6 mol/L or 7.5 × 10^-12 mol). The equilibrium constant of the association of Lys-Lys-Gly-D-Ala-D-Ala with vancomycin was also obtained (KAds=5.0×10^4 L/tool). 展开更多

Grafted Deproteinized Natural Rubber as an Impact Modifier in Styrene-Methyl Methacrylate Copolymer Sheet

This research was studied to prepare styrene-methyl methacrylate copolymer sheet (S-co-MMA sheet) by using DPNR (deproteinized natural rubber) as an impact modifier. The DPNR was prepared by adding SDS (sodium dodecyl sulfate) into the HANR (high-ammoniated natural rubber latex) and followed by centrifugation. It was found that DPNR was successfully prepared with the lowest nitrogeneous content about 0.034±0.01% w/w by adding SDS for 1.00 phr and centrifugation at 12,000 rpm. Then DPNR was grafted with styrene and methyl methacrylate (DPNR-g-S/MMA) obtained by emulsion polymerization. The effects of DPNR-g-S/MMA were studied by varied the amount of SDS and redox initiator. From this result the grafting efficiency DPNR-g-S/MMA was higher than that of NR-g-S/MMA. The DPNR-g-S/MMA was used as an impact modifier in S-co-MMA sheet. Furthermore, the appropriate quantities of styrene monomer and DPNR-g-S/MMA were also investigated. While DPNR-g-S/MMA was used as impact modifier in S-co-MMA sheet, the result was shown DPNR-g-S/MMA improve impact strength of S-co-MMA sheet. Scanning electron micrographs of S-co-MMA sheet with DPNR-g-S/MMA was found the smooth fracture surface. Thus impact strength of S-co-MMA/gDPNR sheet was high and physical properties of S-co-MMA/gDPNR sheet could be able to be accepted in industry. Concisely, DPNR-g-S/MMA can be used as an impact modifier in S-co-MMA sheet. 展开更多

Effect of electroacupuncture on expression of NR2B in prefrontal cortex in morphine-withdrawal rats

Objective To observe the effect of electroacupuncture (EA) on learning and memory abilities and expression of N-methyI-D-aspartate receptor subunit (NR2B) in prefrontal cortex in morphine-withdrawal rats and to investigate the molecular biological mechanisms. Methods Thirty-six male SD rats were randomly divided into four groups, namely control group (group A), model group (group B), model with acupuncture group (group C) and model with electroacupunture group (group D), with 9 in each group. All rats except those in group A were subcutaneously injected with morphine hydrochloride injectio on the back with daily dosage increased day by day. Naloxone was given 3 h after the last injection to establish the models of morphinewithdrawal rats. After the models were established, the rats were treated with acupuncture and electroacupuncture respectively at bilateral "Shenshu" (肾俞 BL 23) and "Zusanli" (足三里 ST 36) for 15 min per time, once daily for 6 days. Space learning and memory abilities of the rats were measured by Morris water maze, and protein and gene expression levels of NR2B in prefrontal cortex were measured by Western Blot and RT-PCR. Results In place navigation test, the escape latency in group B, group C and group D was significantly prolonged compared with that of group A (P〈0.01), the escape latency in group C and group D was significantly shortened compared with that of group B (P〈0.01) and the escape latency in group D was significantly shortened compared with that of group C (P〈0.05); during spatial probe test, the number of times crossing the platform of group B, group C and group D decreased compared with that of group A (P〈0.01), and compared with group B, the number of times crossing the platform of group C increased and the number of group D significantly increased (P〈0.01). Decreased protein expression level of NR2B was found in group B when compared with that of group A (P〈0.01), increased protein expression levels of NR2B were found in group C and group D when compared with that of group B (P〈0.01), however, the expression level in group D was higher than that in group C (P〈0.01). mRNA expression level of NR2B in prefrontal cortex in morphine-withdrawal rats decreased (P〈0.05), however, compared with that of group B, the expression level increased in group D (P〈0.05), and there was no statistical significance in increased expression level in group C (P〉0.05). Conclusion Acupuncture and eletroacupunture can improve space learning and memory abilities of merphine-withdrawal rats, with better efficacy of eletroacupuncture than that of acupuncture, the mechanisms of which may be associated with the regulation of NR2B expression in prefrontal cortex. 展开更多

Objective N-methyl-D-aspartate(NMDA)receptor has been indicated to be involved in the pathogenesis of Alzheimer’s disease(AD).The NMDA receptor subunit 2b(NR2B)has attracted more attention due to its characteristic distribution and selective reduction in AD brain.The present study aimed to explore the association between NMDA gene polymorphism and AD.Methods A total of 63 AD patients and 68 normal controls in Shanghai city were employed in this study.Genotype of C2664T variant(rs1806201)in the exon13 of GRIN2B gene was determined by gene sequencing. Results Among AD patients,15(23.6%)subjects were identified as C/C genotype,and 35(55.6%)were identified as C/T genotype.The left 13(20.6%)subjects were identified as T/T genotype.In normal controls,15(22.1%)subjects were identified as C/C genotype,39(57.4%)as C/T genotype and 14(20.6%)as T/T genotype.The distribution frequency of neither GRIN2B C2664T genotype(P=0.895)nor allele(P=0.790)was significantly different between AD patients and normal controls,even when the subjects were stratified by gender and age of disease onset in AD patients.Conclusion The results suggest that there is no relation between GRIN2B C2664T polymorphism and AD in Chinese Han population of Shanghai City. 展开更多
