The relationship between ENSO and Indian Ocean Dipole was discussed by using the data set of sea temperature from Scripps Institute of Oceanography, the air temperature at 1000hPa from the NCEP reanalysis data and the...The relationship between ENSO and Indian Ocean Dipole was discussed by using the data set of sea temperature from Scripps Institute of Oceanography, the air temperature at 1000hPa from the NCEP reanalysis data and the Nino3 index from the Climate Prediction Center (CPC) of U.S.A. during the period from 1955 to 2001. The results show that there exists a Dipole on the maximum temperature anomalous level (MTAL) in the Indian Ocean, which close relates to ENSO in the Pacific Ocean. During El Nino periods there are good relationships between ENSO and Indian Ocean Dipole which maximum correlation occurring when ENSO leads by one month, but in La Nina periods the relationship is not so good. The distribution of Dipole in Indian Ocean is from northeast to southwest, which one (west) pole in 65°E - 75°E, 6°S - 10°S and the other in 85°E - 95°E, 2°N - 6°N, which is different from that defined by Saij. The correlation coefficients of Nino3 index with temperature anomalies in the west/east poles on the MTAL are over 0.4 - 0.15, respectively. It is a main sea temperature system in the tropical Indian Ocean. However, in the surface layer from sea surface to the depth of 20 m - 30 m there is no such a dipole with opposite sea temperature anomalies in the NE and SW of tropical Indian Ocean. The SSTA in the NE might be influenced by the sensible exchange process because the evolution of sea and 1 000 hPa air temperature anomaly time series of the NE of tropical Indian Ocean is quite similar except those during 1962 - 1963 and 1986. The periods of Indian Ocean Dipole are shorter than that of ENSO, and about 1 to 6-year.展开更多
AIM:To investigate the correlation of hyperlipemia(HL) and acute cerebral ischemia/reperfusion(I/R) injury on liver damage and its mechanism.METHODS:Rats were divided into 4 groups:control,HL,I/R and HL+I/R.After the ...AIM:To investigate the correlation of hyperlipemia(HL) and acute cerebral ischemia/reperfusion(I/R) injury on liver damage and its mechanism.METHODS:Rats were divided into 4 groups:control,HL,I/R and HL+I/R.After the induction of HL via a high-fat diet for 18 wk,middle cerebral artery occlusion was followed by 24 h of reperfusion to capture I/R.Serum alanine transaminase(ALT) and aspartate aminotransferase(AST) were analyzed as part of liver function tests and liver damage was further assessed by histological examination.Hepatocyte apoptosis was evaluated by terminal deoxynucleotidyl transferase dUTP nick-end labeling(TUNEL) assay.The expression of genes related to apoptosis(caspase-3,bcl-2) was assayed by immunohistochemistry and Western blotting.Serum tumor necrosis factor-(TNF-),interleukin-1(IL-1) and liver mitochondrial superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),malondialdehyde(MDA) and Ca 2+ levels were measured to determine inflammatory and oxidative/antioxidative status respectively.Microsomal hydroxylase activity of the cytochrome P450 2E1(CYP2E1)-containing enzyme was measured with aniline as the substrate,and CYP2E1 expression in the liver tissue and microsome was determined by immunohistochemistry and Western blotting respectively.RESULTS:HL alone induced by high-fat diet for 18 wk resulted in liver damage,indicated by histopathological analysis,and a considerable increase in serum ALT(25.13 ± 16.90 vs 9.56 ± 1.99,P < 0.01) and AST levels(18.01 ± 10.00 vs 11.33 ± 4.17,P < 0.05) compared with control.Moreover,HL alone induced hepatocyte apoptosis,which was determined by increased TUNEL-positive cells(4.47 ± 0.45 vs 1.5 ± 0.22,P < 0.01),higher caspase-3 and lower bcl-2 expression.Interestingly,compared with those in control,HL or I/R groups,massive increases of serum ALT(93.62 ± 24.00 vs 9.56 ± 1.99,25.13 ± 16.90 or 12.93 ± 6.14,P < 0.01) and AST(82.32 ± 26.92 vs 11.33 ± 4.17,18.01 ± 10.00 or 14.00 ± 6.19,P < 0.01) levels in HL+I/R group were observed suggesting severe liver damage,which was confirmed by liver histology.In addition,HL combined with I/R also caused significantly increased hepatocyte apoptosis,as evidenced by increased TUNEL-positive cells(6.20 ± 0.29 vs 1.5 ± 0.22,4.47 ± 0.45 or 1.97 ± 0.47,P < 0.01),elevated expression of caspase-3 and lower expression of bcl-2.Furthermore,when compared to HL or I/R alone,HL plus I/R enhanced serum TNF-,IL-1,liver mitochondrial MDA and Ca 2+ levels,suppressed SOD and GSH-Px in liver mitochondria,and markedly up-regulated the activity(11.76 ± 2.36 vs 4.77 ± 2.31 or 3.11 ± 1.35,P < 0.01) and expression(3.24 ± 0.38 vs 1.98 ± 0.88 or 1.72 ± 0.58,P < 0.01) of CYP2E1 in liver.CONCLUSION:The coexistence of HL and acute cerebral I/R induces severe liver damage,suggesting that cerebral ischemic stroke would exaggerate the damage of liver caused by HL.This effect is possibly due to en-hanced CYP2E1 induction which further promotes oxidative damage,inflammation and hepatocyte apoptosis.展开更多

In this study, the authors investigated circulation anomalies in the extratropics, in particular for the zonal teleconnection, under different combinations of subtropical convection anomalies over the northern Indian continent (IND) and the westem North Pacific (WNP). The outof-phase configuration (i.e., stronger (weaker) IND convection and weaker (stronger) WNP convection) was found to be more common than the in-phase configuration (i.e., stronger (weaker) IND convection and stronger (weaker) WNP convection), which is consistent with previous results. Composite results indicated that circulation anomalies for out-of-phase configurations of 30-60-day convection oscillations are much stronger in the middle latitudes than those for in-phase configurations. In addition, zonal teleconnection patterns are predominant for the out-of-phase configurations, particularly for the configuration of strong IND convection and weak WNP convec- tion; however, they are either weak or obscure for the in-phase configurations. These results suggest that the zonal teleconnection pattem along the Asian jet is dependent on different combinations of the 1ND and WNP subtropical convection anomalies.展开更多
The precision modeling of dam break floods can lead to formulation of proper emergency action plan to minimize flood impacts within the economic lifetime of the assets.Application of GIS techniques in integration with...The precision modeling of dam break floods can lead to formulation of proper emergency action plan to minimize flood impacts within the economic lifetime of the assets.Application of GIS techniques in integration with hydrological modeling for mapping of the flood inundated areas can play a momentous role in further minimizing the risk and likely damages.In the present study,dam break analysis using DAMBRK model was performed under various likely scenarios.Probable Maximum Flood (PMF)calculated for a return period of 1000 years using deterministic approach was adopted for dam break analysis of the proposed dam under various combinations of breach dimensions.The available downstream river cross-sections data sets were used as input in the model to generate the downstream flood profile.Dam break flow depths generated by the DAMBRK model under various combinations of structural failure are subsequently plotted on Digital Elevation Model(DEM)of the downstream of dam site to map the likely affected area.The simulation results reveals that in one particular case the flood without dam may be more intense if a rainfall of significant intensity takes place.展开更多

Results show that these models are abl...Performances of 5 models from the Coupled Model Intercomparison Project Phase 5 (CMIP5) in simulating the chloro-phyll concentration over the tropical Indian Ocean are evaluated. Results show that these models are able to capture the dominant spatial distribution of observed chlorophyll concentration and reproduce the maximum chlorophyll concentration over the western part of the Arabian Sea, around the tip of the Indian subcontinent, and in the southeast tropical Indian Ocean. The seasonal evolution of chlorophyll concentration over these regions is also reproduced with significant amplitude diversity among models. All of 5 mod-els is able to simulate the interannual variability of chlorophyll concentration. The maximum interannual variation occurs at the same regions where the maximum climatological chlorophyll concentration is located. Further analysis also reveals that the Indian Ocean Dipole events have great impact on chlorophyll concentration in the tropical Indian Ocean. In the general successful simulation of chlorophyll concentration, most of the CMIP5 models present higher than normal chlorophyll concentration in the eastern equatorial Indian Ocean.展开更多

To investigate the function and evolution of this gene, the cDNA of full length Mest gene was obtained using 5'- and 3'-RACE from the Chinese Large Toad (Bufo gargarizans). The transcript is 1 325bp in length which contains a complete open reading frame (ORF) encoding a polypeptide of 326 amino acids (GenBank accession number: ABQ10905). There is a typical 0./13 hydrolase fold domain in the putative gene product, and it shows high similarity to sequence of homologous protein of Xenopus tropicali (86%), mammlian (70% - 80%). RT-PCR (reverse transcriptase-polymerase chain reaction) analysis demonstrated that the Bufo gargarizans Mest (BgMest) gene is expressed widely in testis, ovary, liver, kidney, spleen, brain, stomach and lung. The conservation of the BgMest gene sequences, protein secondary structure of the BgMest protein, in addition to the expression pattern of the BgMest gene, suggested that the function of BgMest was conserved in amphibians. However, the phylogenetic tree of the imprinting gene of the mammals and other vertebrates examined in this study indicated their divergent origins.展开更多
Using the monthly wind and sea surface temperature (SST) data, southern meridional atmospheric circulation cells associated with the Indian Ocean Dipole Mode (IOD) events in the Indian Ocean are for the first time...Using the monthly wind and sea surface temperature (SST) data, southern meridional atmospheric circulation cells associated with the Indian Ocean Dipole Mode (IOD) events in the Indian Ocean are for the first time described and examineS. The divergent wind and pressure vertical velocity are employed for the identification of atmospheric circulation cells. During the four different phases of the positive IOD events, the anomalous meridional Hadley circulation over the western Indian Ocean shows that the air rises in the tropics, flows poleward in the upper troposphere, sinks in the subtropics, and returns back to the tropics in the lower troposphere. The anomalous Hadley circulation over the eastern Indian Ocean is opposite to that over the western Indian Ocean. During positive IOD events, the meridional Hadley circulation over the eastern Indian Ocean is weakened while it is strengthened over the western Indian Ocean. Correlation analysis between the IOD index and the indices of the Hadley cells also proves that, the atmospheric circulation patterns are evident in every IOD event over the period of record.展开更多

The results indicate that warmer SSTA contributes to the increasing of the temperature over the Plateau in early summer, resulting in the intensification of tropical easterly jet on 100 hPa and northward shift of Northern Hemisphere subtropical westerly jet in May. It is obviously favorable for the subtropical high enhancement over western Pacific Ocean in May and subtropical westerly jet maintaining at 35~40 °N in June, making the Mei-Yu come earlier and stay over the Changjiang basin in 1991. Furthermore, warmer SSTA is also advantageous to averaged temperature rise in East Asia land region and Nanhai monsoon development. These roles are helpful in accelerating the seasonal transition for East Asia in early summer.展开更多

In the South Indian and Atlantic Oceans, there is a subtropical dipole pattern slanted in the southwest-northeast direction. In the South Pacific Ocean, a meridional tripole structure emerges, whose middle pole co-varies with the dipoles in the South Indian and Atlantic Oceans and is used in this study to track subtropical Pacific variability. The South Indian and Atlantic Ocean dipoles and the subtropical Pacific variability are phase-locked in austral summer. On the inter-decadal time scales, the dipoles in the South Indian and Atlantic Oceans weaken in amplitude after 1979/1980. No such weakening is found in the subtropical South Pacific Ocean. Interestingly, despite the reduced amplitude, the correlation of the Indian Ocean and Atlantic dipoles with E1 Nino and Southern Oscillation (ENSO) are enhanced after 1979/1980. The same increase in correlation is found for subtropical South Pacific variability after 1979/1980. These inter-decadal modulations imply that the Southern Hemisphere participates in part of the climate shift in the late 1970s. The correlation between Southern Hemisphere SST and ENSO reduces after 2000.展开更多
AIM: To evaluate the effect of mitochondrial tumor ne- crosis factor receptor-associated protein-1 (TRAP-l) on the lymph node metastasis (LNM) in Chinese colorectal cancer (CRC) patients, and develop potential ...AIM: To evaluate the effect of mitochondrial tumor ne- crosis factor receptor-associated protein-1 (TRAP-l) on the lymph node metastasis (LNM) in Chinese colorectal cancer (CRC) patients, and develop potential LNM- associated biomarkers for CRC using quantitative real- time polymerase chain reaction (RT-PCR) analysis. METHODS: Differences in mitochondrial TRAP-1 gene expression between primary CRC with LNM (LNM CRC) and without LNM (non-LNM CRC) were assessed in 96 Chinese colorectal carcinoma samples using quantita- tive RT-PCR analysis, Western blotting, and confirmed with immunohistochemical assay. The relationship between clinicopathological parameters and potential diaclnostic biomarkers was also examined.RESULTS: TRAP-1 was significantly upregulated in LNM CRC compared with non-LNM CRC, which was confirmed by RT-PCR, Western blotting and immuno- histochemical assay. The expression of TRAP-1 in two different metastatic potential human colorectal cancer cell lines, LoVo and HT29, was analyzed with Western blotting. The expression level of TRAP-1 was dramati- cally higher in LoVo than in HT29. Overexpression of TRAP-1 was significantly associated with LNM (90.2% in LNM group vs 22% in non-LNM group, P 〈 0.001), the advanced tumor node metastasis stage (89.1% in LNM group vs 26.9% in non-LNM group, P 〈 0.001), the increased 5-year recurrence rate (82.7% in LNM group vs 22.6% in non-LNM group, P 〈 0.001) and the decreased 5-year overall survival rate (48.4% in LNM vs 83.2% in non-LNM group, P 〈 0.001). Univariate and multivariate analyses indicated that TRAP-1 expression was an independent prognostic factor for recurrence and survival of CRC patients (Hazard ratio of 2.445 in recurrence, P = 0.017; 2.867 in survival, P = 0.028). CONCLUSION: Mitochondria TRAP-1 affects the lymph node metastasis in CRC, and may be a potential bio- marker for LNM and a prognostic factor in CRC. Methods Neonatal rat cardiomyocytes were cultured from the ventricles of l-day-old Wistar rats. Cells were exposed to different concentrations of salubrinal (10, 20, and 40 gmol/L) for 30 min followed by TM treatment or hypoxia for 36 h. Apoptosis was measured by a multiparameter HCS (high content screening) apoptosis assay, TUNEL assay and flow cytometry. The phosphorylation of eukaryotic translation initiation factor 2 subunit alpha (eIF2c0 and the expression of cleaved caspase-12 were determined by Western blotting. C/EBP homologous protein (CHOP) was detected by immunocytochemistry. Results HCS, TUNEL assays and flow cytometry showed that salubrinal protected cardiomyocytes against apoptosis induced by TM or hypoxia. Western blotting showed that salubrinal protected cardiomyocytes against apoptosis by inducing eIF2ct phosphorylation and down-regulating the expression of the endoplasmic reticulum stress-mediated apoptotic proteins, CHOP and cleaved caspase-12. Conclusions Our study suggests that salubrinal protects rat cardiomyocytes against TM- or hypoxia-associated apoptosis via a mechanism involving the inhibition of ER stress-mediated apoptosis.展开更多

METHODS: Colons from Sprague-Dawley rat fetuses, young and adult (8 wk old) animals were used in this study. Expression levels of AFP in colons of different development stage were detected by reversetranscriptase PCR (RT-PCR) and Western blotting. To identify the cell location of AFP in the developing rat colons, double-immunofluorescent staining was performed using antibodies to specific cell markers and AFP, respectively. RESULTS: The highest levels of AFP mRNA were detected in colons of rats at embryonic day 18.5 (e18.5). Compared to e18.5 d, the AFP expression was significantly decreased during rat development (85% for e20.5, P 〈 0.05, 58% for postnatal day 0.5 (P〈0.5), P 〈 0.05, 37% for P7, P 〈 0.05, 24% for P14, P 〈 0.05, and 11% for P21, P 〈 0.05) and undetected in adult rats. Only the 72-kDa isoform of AFP was detected by Western blotting, the expression pattern was similar to AFP mRNA and conformed to the results of mRNA expression. The AFP positive staining was identical to different distribution patterns in fetuses, young and adult animals and positive staining for both AFP and vimentin was overlapped in mesenchymal cells at each stage tested. CONCLUSION: This study has for the first time demonstrated that AFP is localized in the mesenchyme of rat colon from the embryo to the weaning stage by immunofluorescence and presents 72-kDa isoform in the developing rat colons by Western blotting. The dynamic expression of AFP in the various developmental stages of the colon indicates that AFP might be involved in many aspects of colon development.展开更多

METHODS: Sixty formalin-fixed and paraffin-...AIM: To assess the significance of phosphatidylinositol 3-kinase (PI3K) in colorectal cancer (CRC) and toxicity of LY294002 in CRC cells with different metastatic abilities. METHODS: Sixty formalin-fixed and paraffin-embedded CRC tumor specimens were investigated. Adjacent normal colonic mucosa specimens from 10 of these cases were selected as controls. PI3K protein was detected by immunohistochemistry and PIK3CA mutations were investigated by gene sequencing analysis. A flowcytometry-based apoptosis detection kit was used to determine PI3K inhibitor-induced apoptosis in CRC cell lines SW480 and SW620. Expression of phosphorylated protein kinase B in CRC cell lines was detected by Western blotting. RESULTS: There was a significant difference in the proportion of primary lesions (30%, 18/60) vs metastatic lesions (46.7%, 28/60) that were positive for PI3K (P < 0.05). Mutations were detected in exon 9 (13.3%) and exon 20 (8.3%). Out of 60 cases, seven mutations were identified: two hotspot mutations, C.1633G>A resulting in E545A, and C.3140A>G resulting in H1047R; two novel missense mutations C.1624G>A and C.3079G>A; and three synonymous mutations (C.1641G>A, C.1581C>T and C.3027T>A). Exposure of SW480 cells to PI3K inhibitor for 48 h resulted in a significant increase of apoptotic cells in a dose-dependent manner [3.2% apoptotic cells in 0 μmol/L, 4.3% in 5 μmol/L, 6.3% in 10 μmol/L (P < 0.05), and 6.7% in 20 μmol/L (P < 0.05)]. Moreover, PI3K inhibitor induced a similar significant increase of apoptotic cells in the SW620 cell line for 48 h [3.3% apoptotic cells in 0 μmol/L, 13.3% in 5 μmol/L (P < 0.01), 19.2% in 10 μmol/L (P < 0.01), and 21.3% in 20 μmol/L (P < 0.01)]. CONCLUSION: High PI3K expression is associated with CRC metastasis. PI3K inhibitor induced apoptosis in CRC cells and displayed strong cytotoxicity for highly metastatic cells. PI3K inhibition may be an effective treatment for CRC.展开更多

A destructive extreme heat attacked Northeast Asia(NEA)in the midsummer of 2018,characterized by the average midsummer Tmax(daily maximum air temperature at 2 m)ranking first during the study period.The current study ...A destructive extreme heat attacked Northeast Asia(NEA)in the midsummer of 2018,characterized by the average midsummer Tmax(daily maximum air temperature at 2 m)ranking first during the study period.The current study indicates that the cyclonic anomaly over the western North Pacific(WNP)was an important cause,which presents an anomaly of two standard deviations.The cyclonic anomaly over the WNP was accompanied by anomalous convection,which favored descending and anticyclonic anomalies over NEA through a local meridional cell.The anticyclonic anomaly over NEA corresponds to the northwestward extension of the WNP subtropical high and facilitated the occurrence of extreme heat.The tropical sea surface temperature anomaly(SSTA)presents a La Ni?a decaying episode,but the SSTA over the tropical Pacific and North Indian Ocean was weak in the summer.In contrast,the southeastern tropical Indian Ocean(SETIO)was obviously cool,which was the coolest after detrending.The SETIO cooling triggered a low-level southeasterly anomaly,which turned into a southwesterly after crossing the equator,due to the Coriolis force.The southwesterly anomaly extended eastwards and favored the cyclonic anomaly over the WNP.Meanwhile,the circulation anomalies over the SETIO and WNP were connected via a local meridional cell,with the ascending branch over the WNP.Moreover,the above mechanism also operates for the climate statistics,verifying the robust in?uence of the SETIO SSTA.Considering the consistency of the SETIO SSTA,it could be a potential predictor for the climate over the WNP and NEA.展开更多

Our results are shown as follows: (1) CAM3.0, driven by the combined SSTAs over the four oceanic regions, can simulate well the features of anomalous atmospheric circulations over Eurasia in January 2008, indicating that the effects of the SSTAs over these four regions were one of the key causes of the anomalous systems over Eurasia. (2) The SSTAs over each key region contributed to the intensification of blocking over the Urals Mountains and a main East Asian trough. However, the influence of the SSTAs over individual oceanic regions differed from one another in other aspects. The SSTAs over the North Atlantic had an impact on the 500-hPa anomalous height (Z500A) over the middle-high latitudes and had a somewhat smaller effect over the low latitudes. For the warm SSTAs over Kuroshio, the subtropical high was much stronger, spread farther north than usual, and had an anomalous easterly that dominated the northwest Pacific Ocean. The warm SSTAs over the tropical Indian Ocean could have caused a negative Z500A from West Asia to Middle Asia, a remarkably anomalous southwesterly from the Indian Ocean to the south of China and an anomalous anticyclone circulation over the South China Sea-Philippine Sea region. Because of the La Nifia event, the winter monsoon was stronger than normal, with an anomalously cooler northerly over the southeastern coastal areas of China. (3) The combined effects of the SSTAs over the four key regions were likely more important to the atmospheric circulation anomalies of January 2008 over Eurasia than the effects of individual or partly combined SSTAS. This unique SSTA distribution possibly led to the circulation anomalies over Eurasia in January 2008, especially the atmospheric circulation anomalies over the subtropics, which were more similar to those of the winter E1 Nifio events than to the circulation anomalies following La Nifia.展开更多

The mature virion comprises 11 dsRNA genomes enclosed by two concentric icosahedral proteins shells that is comprised of five core proteins and two outer capsid proteins. The genome sequence and 3D structure demonstrate there is a higher level of sequence homology in structural proteins between GCRV and mammalian orthoreoviruses (MRV) compared to other members of the family. To understand the pathogenesis of GCRV infection, the outer capsid protein VP5, a homology of the μ1 protein of MRV, was expressed in E.coli. It was found that the recombinant VP5 was highly expressed, and the expressed His-tag fusion protein was involved in the formation of the inclusion body. Additionally, specific anti-VP5 serum was prepared from purified protein and western blot demonstrated that the expressed protein was able to bind immunologically to rabbit anti GCRV particle serum and the immunogenicity was determined by ELISA assay. Additional experiments in investigating the functional properties of VP5 will further elucidate the role of the GCRV outer capsid protein VP5 during entry into host cells, and its interaction among viral proteins and host cells during the infection process.展开更多

文摘The relationship between ENSO and Indian Ocean Dipole was discussed by using the data set of sea temperature from Scripps Institute of Oceanography, the air temperature at 1000hPa from the NCEP reanalysis data and the Nino3 index from the Climate Prediction Center (CPC) of U.S.A. during the period from 1955 to 2001. The results show that there exists a Dipole on the maximum temperature anomalous level (MTAL) in the Indian Ocean, which close relates to ENSO in the Pacific Ocean. During El Nino periods there are good relationships between ENSO and Indian Ocean Dipole which maximum correlation occurring when ENSO leads by one month, but in La Nina periods the relationship is not so good. The distribution of Dipole in Indian Ocean is from northeast to southwest, which one (west) pole in 65°E - 75°E, 6°S - 10°S and the other in 85°E - 95°E, 2°N - 6°N, which is different from that defined by Saij. The correlation coefficients of Nino3 index with temperature anomalies in the west/east poles on the MTAL are over 0.4 - 0.15, respectively. It is a main sea temperature system in the tropical Indian Ocean. However, in the surface layer from sea surface to the depth of 20 m - 30 m there is no such a dipole with opposite sea temperature anomalies in the NE and SW of tropical Indian Ocean. The SSTA in the NE might be influenced by the sensible exchange process because the evolution of sea and 1 000 hPa air temperature anomaly time series of the NE of tropical Indian Ocean is quite similar except those during 1962 - 1963 and 1986. The periods of Indian Ocean Dipole are shorter than that of ENSO, and about 1 to 6-year.
文摘AIM:To investigate the correlation of hyperlipemia(HL) and acute cerebral ischemia/reperfusion(I/R) injury on liver damage and its mechanism.METHODS:Rats were divided into 4 groups:control,HL,I/R and HL+I/R.After the induction of HL via a high-fat diet for 18 wk,middle cerebral artery occlusion was followed by 24 h of reperfusion to capture I/R.Serum alanine transaminase(ALT) and aspartate aminotransferase(AST) were analyzed as part of liver function tests and liver damage was further assessed by histological examination.Hepatocyte apoptosis was evaluated by terminal deoxynucleotidyl transferase dUTP nick-end labeling(TUNEL) assay.The expression of genes related to apoptosis(caspase-3,bcl-2) was assayed by immunohistochemistry and Western blotting.Serum tumor necrosis factor-(TNF-),interleukin-1(IL-1) and liver mitochondrial superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),malondialdehyde(MDA) and Ca 2+ levels were measured to determine inflammatory and oxidative/antioxidative status respectively.Microsomal hydroxylase activity of the cytochrome P450 2E1(CYP2E1)-containing enzyme was measured with aniline as the substrate,and CYP2E1 expression in the liver tissue and microsome was determined by immunohistochemistry and Western blotting respectively.RESULTS:HL alone induced by high-fat diet for 18 wk resulted in liver damage,indicated by histopathological analysis,and a considerable increase in serum ALT(25.13 ± 16.90 vs 9.56 ± 1.99,P < 0.01) and AST levels(18.01 ± 10.00 vs 11.33 ± 4.17,P < 0.05) compared with control.Moreover,HL alone induced hepatocyte apoptosis,which was determined by increased TUNEL-positive cells(4.47 ± 0.45 vs 1.5 ± 0.22,P < 0.01),higher caspase-3 and lower bcl-2 expression.Interestingly,compared with those in control,HL or I/R groups,massive increases of serum ALT(93.62 ± 24.00 vs 9.56 ± 1.99,25.13 ± 16.90 or 12.93 ± 6.14,P < 0.01) and AST(82.32 ± 26.92 vs 11.33 ± 4.17,18.01 ± 10.00 or 14.00 ± 6.19,P < 0.01) levels in HL+I/R group were observed suggesting severe liver damage,which was confirmed by liver histology.In addition,HL combined with I/R also caused significantly increased hepatocyte apoptosis,as evidenced by increased TUNEL-positive cells(6.20 ± 0.29 vs 1.5 ± 0.22,4.47 ± 0.45 or 1.97 ± 0.47,P < 0.01),elevated expression of caspase-3 and lower expression of bcl-2.Furthermore,when compared to HL or I/R alone,HL plus I/R enhanced serum TNF-,IL-1,liver mitochondrial MDA and Ca 2+ levels,suppressed SOD and GSH-Px in liver mitochondria,and markedly up-regulated the activity(11.76 ± 2.36 vs 4.77 ± 2.31 or 3.11 ± 1.35,P < 0.01) and expression(3.24 ± 0.38 vs 1.98 ± 0.88 or 1.72 ± 0.58,P < 0.01) of CYP2E1 in liver.CONCLUSION:The coexistence of HL and acute cerebral I/R induces severe liver damage,suggesting that cerebral ischemic stroke would exaggerate the damage of liver caused by HL.This effect is possibly due to en-hanced CYP2E1 induction which further promotes oxidative damage,inflammation and hepatocyte apoptosis.

文摘A zonal teleconnection has been found along the Asian jet over the Eurasian continent during summer. In this study, the authors investigated circulation anomalies in the extratropics, in particular for the zonal teleconnection, under different combinations of subtropical convection anomalies over the northern Indian continent (IND) and the westem North Pacific (WNP). The outof-phase configuration (i.e., stronger (weaker) IND convection and weaker (stronger) WNP convection) was found to be more common than the in-phase configuration (i.e., stronger (weaker) IND convection and stronger (weaker) WNP convection), which is consistent with previous results. Composite results indicated that circulation anomalies for out-of-phase configurations of 30-60-day convection oscillations are much stronger in the middle latitudes than those for in-phase configurations. In addition, zonal teleconnection patterns are predominant for the out-of-phase configurations, particularly for the configuration of strong IND convection and weak WNP convec- tion; however, they are either weak or obscure for the in-phase configurations. These results suggest that the zonal teleconnection pattem along the Asian jet is dependent on different combinations of the 1ND and WNP subtropical convection anomalies.
文摘The precision modeling of dam break floods can lead to formulation of proper emergency action plan to minimize flood impacts within the economic lifetime of the assets.Application of GIS techniques in integration with hydrological modeling for mapping of the flood inundated areas can play a momentous role in further minimizing the risk and likely damages.In the present study,dam break analysis using DAMBRK model was performed under various likely scenarios.Probable Maximum Flood (PMF)calculated for a return period of 1000 years using deterministic approach was adopted for dam break analysis of the proposed dam under various combinations of breach dimensions.The available downstream river cross-sections data sets were used as input in the model to generate the downstream flood profile.Dam break flow depths generated by the DAMBRK model under various combinations of structural failure are subsequently plotted on Digital Elevation Model(DEM)of the downstream of dam site to map the likely affected area.The simulation results reveals that in one particular case the flood without dam may be more intense if a rainfall of significant intensity takes place.

文摘Performances of 5 models from the Coupled Model Intercomparison Project Phase 5 (CMIP5) in simulating the chloro-phyll concentration over the tropical Indian Ocean are evaluated. Results show that these models are able to capture the dominant spatial distribution of observed chlorophyll concentration and reproduce the maximum chlorophyll concentration over the western part of the Arabian Sea, around the tip of the Indian subcontinent, and in the southeast tropical Indian Ocean. The seasonal evolution of chlorophyll concentration over these regions is also reproduced with significant amplitude diversity among models. All of 5 mod-els is able to simulate the interannual variability of chlorophyll concentration. The maximum interannual variation occurs at the same regions where the maximum climatological chlorophyll concentration is located. Further analysis also reveals that the Indian Ocean Dipole events have great impact on chlorophyll concentration in the tropical Indian Ocean. In the general successful simulation of chlorophyll concentration, most of the CMIP5 models present higher than normal chlorophyll concentration in the eastern equatorial Indian Ocean.

文摘The Mest (mesoderm-specific transcript) gene has been considered an imprinting gene in human and mouse, and was also confirmed in other mammals and flowering plants. To investigate the function and evolution of this gene, the cDNA of full length Mest gene was obtained using 5'- and 3'-RACE from the Chinese Large Toad (Bufo gargarizans). The transcript is 1 325bp in length which contains a complete open reading frame (ORF) encoding a polypeptide of 326 amino acids (GenBank accession number: ABQ10905). There is a typical 0./13 hydrolase fold domain in the putative gene product, and it shows high similarity to sequence of homologous protein of Xenopus tropicali (86%), mammlian (70% - 80%). RT-PCR (reverse transcriptase-polymerase chain reaction) analysis demonstrated that the Bufo gargarizans Mest (BgMest) gene is expressed widely in testis, ovary, liver, kidney, spleen, brain, stomach and lung. The conservation of the BgMest gene sequences, protein secondary structure of the BgMest protein, in addition to the expression pattern of the BgMest gene, suggested that the function of BgMest was conserved in amphibians. However, the phylogenetic tree of the imprinting gene of the mammals and other vertebrates examined in this study indicated their divergent origins.

文摘Using the monthly wind and sea surface temperature (SST) data, southern meridional atmospheric circulation cells associated with the Indian Ocean Dipole Mode (IOD) events in the Indian Ocean are for the first time described and examineS. The divergent wind and pressure vertical velocity are employed for the identification of atmospheric circulation cells. During the four different phases of the positive IOD events, the anomalous meridional Hadley circulation over the western Indian Ocean shows that the air rises in the tropics, flows poleward in the upper troposphere, sinks in the subtropics, and returns back to the tropics in the lower troposphere. The anomalous Hadley circulation over the eastern Indian Ocean is opposite to that over the western Indian Ocean. During positive IOD events, the meridional Hadley circulation over the eastern Indian Ocean is weakened while it is strengthened over the western Indian Ocean. Correlation analysis between the IOD index and the indices of the Hadley cells also proves that, the atmospheric circulation patterns are evident in every IOD event over the period of record.

文摘By employing the CCM1(R15L12)long-range spectral model, study is undertaken of the effects of sea surface temperature anomaly(SSTA) for tropical Indian ocean on circulation transformation in the early summer in East Asia in 1991. The results indicate that warmer SSTA contributes to the increasing of the temperature over the Plateau in early summer, resulting in the intensification of tropical easterly jet on 100 hPa and northward shift of Northern Hemisphere subtropical westerly jet in May. It is obviously favorable for the subtropical high enhancement over western Pacific Ocean in May and subtropical westerly jet maintaining at 35~40 °N in June, making the Mei-Yu come earlier and stay over the Changjiang basin in 1991. Furthermore, warmer SSTA is also advantageous to averaged temperature rise in East Asia land region and Nanhai monsoon development. These roles are helpful in accelerating the seasonal transition for East Asia in early summer.

文摘Empirical orthogonal function (EOF) analysis reveals a co-variability of Sea surface temperatures (SSTs) in the Southern Hemisphere (0°-60°S). In the South Indian and Atlantic Oceans, there is a subtropical dipole pattern slanted in the southwest-northeast direction. In the South Pacific Ocean, a meridional tripole structure emerges, whose middle pole co-varies with the dipoles in the South Indian and Atlantic Oceans and is used in this study to track subtropical Pacific variability. The South Indian and Atlantic Ocean dipoles and the subtropical Pacific variability are phase-locked in austral summer. On the inter-decadal time scales, the dipoles in the South Indian and Atlantic Oceans weaken in amplitude after 1979/1980. No such weakening is found in the subtropical South Pacific Ocean. Interestingly, despite the reduced amplitude, the correlation of the Indian Ocean and Atlantic dipoles with E1 Nino and Southern Oscillation (ENSO) are enhanced after 1979/1980. The same increase in correlation is found for subtropical South Pacific variability after 1979/1980. These inter-decadal modulations imply that the Southern Hemisphere participates in part of the climate shift in the late 1970s. The correlation between Southern Hemisphere SST and ENSO reduces after 2000.

文摘AIM: To evaluate the effect of mitochondrial tumor ne- crosis factor receptor-associated protein-1 (TRAP-l) on the lymph node metastasis (LNM) in Chinese colorectal cancer (CRC) patients, and develop potential LNM- associated biomarkers for CRC using quantitative real- time polymerase chain reaction (RT-PCR) analysis. METHODS: Differences in mitochondrial TRAP-1 gene expression between primary CRC with LNM (LNM CRC) and without LNM (non-LNM CRC) were assessed in 96 Chinese colorectal carcinoma samples using quantita- tive RT-PCR analysis, Western blotting, and confirmed with immunohistochemical assay. The relationship between clinicopathological parameters and potential diaclnostic biomarkers was also examined.RESULTS: TRAP-1 was significantly upregulated in LNM CRC compared with non-LNM CRC, which was confirmed by RT-PCR, Western blotting and immuno- histochemical assay. The expression of TRAP-1 in two different metastatic potential human colorectal cancer cell lines, LoVo and HT29, was analyzed with Western blotting. The expression level of TRAP-1 was dramati- cally higher in LoVo than in HT29. Overexpression of TRAP-1 was significantly associated with LNM (90.2% in LNM group vs 22% in non-LNM group, P 〈 0.001), the advanced tumor node metastasis stage (89.1% in LNM group vs 26.9% in non-LNM group, P 〈 0.001), the increased 5-year recurrence rate (82.7% in LNM group vs 22.6% in non-LNM group, P 〈 0.001) and the decreased 5-year overall survival rate (48.4% in LNM vs 83.2% in non-LNM group, P 〈 0.001). Univariate and multivariate analyses indicated that TRAP-1 expression was an independent prognostic factor for recurrence and survival of CRC patients (Hazard ratio of 2.445 in recurrence, P = 0.017; 2.867 in survival, P = 0.028). CONCLUSION: Mitochondria TRAP-1 affects the lymph node metastasis in CRC, and may be a potential bio- marker for LNM and a prognostic factor in CRC. 文摘Objectives This study examined the protective effect of salubrinal and the mechanism underlying this protection against tunicamycin (TM)- and hypoxia-induced apoptosis in rat cardiomyocytes. Methods Neonatal rat cardiomyocytes were cultured from the ventricles of l-day-old Wistar rats. Cells were exposed to different concentrations of salubrinal (10, 20, and 40 gmol/L) for 30 min followed by TM treatment or hypoxia for 36 h. Apoptosis was measured by a multiparameter HCS (high content screening) apoptosis assay, TUNEL assay and flow cytometry. The phosphorylation of eukaryotic translation initiation factor 2 subunit alpha (eIF2c0 and the expression of cleaved caspase-12 were determined by Western blotting. C/EBP homologous protein (CHOP) was detected by immunocytochemistry. Results HCS, TUNEL assays and flow cytometry showed that salubrinal protected cardiomyocytes against apoptosis induced by TM or hypoxia. Western blotting showed that salubrinal protected cardiomyocytes against apoptosis by inducing eIF2ct phosphorylation and down-regulating the expression of the endoplasmic reticulum stress-mediated apoptotic proteins, CHOP and cleaved caspase-12. Conclusions Our study suggests that salubrinal protects rat cardiomyocytes against TM- or hypoxia-associated apoptosis via a mechanism involving the inhibition of ER stress-mediated apoptosis.
文摘AIM: To investigate the expression of α-fetoprotein (AFP), a cancer-associated fetal glycoprotein, and its involvement during rat colon development. METHODS: Colons from Sprague-Dawley rat fetuses, young and adult (8 wk old) animals were used in this study. Expression levels of AFP in colons of different development stage were detected by reversetranscriptase PCR (RT-PCR) and Western blotting. To identify the cell location of AFP in the developing rat colons, double-immunofluorescent staining was performed using antibodies to specific cell markers and AFP, respectively. RESULTS: The highest levels of AFP mRNA were detected in colons of rats at embryonic day 18.5 (e18.5). Compared to e18.5 d, the AFP expression was significantly decreased during rat development (85% for e20.5, P 〈 0.05, 58% for postnatal day 0.5 (P〈0.5), P 〈 0.05, 37% for P7, P 〈 0.05, 24% for P14, P 〈 0.05, and 11% for P21, P 〈 0.05) and undetected in adult rats. Only the 72-kDa isoform of AFP was detected by Western blotting, the expression pattern was similar to AFP mRNA and conformed to the results of mRNA expression. The AFP positive staining was identical to different distribution patterns in fetuses, young and adult animals and positive staining for both AFP and vimentin was overlapped in mesenchymal cells at each stage tested. CONCLUSION: This study has for the first time demonstrated that AFP is localized in the mesenchyme of rat colon from the embryo to the weaning stage by immunofluorescence and presents 72-kDa isoform in the developing rat colons by Western blotting. The dynamic expression of AFP in the various developmental stages of the colon indicates that AFP might be involved in many aspects of colon development.

文摘AIM: To assess the significance of phosphatidylinositol 3-kinase (PI3K) in colorectal cancer (CRC) and toxicity of LY294002 in CRC cells with different metastatic abilities. METHODS: Sixty formalin-fixed and paraffin-embedded CRC tumor specimens were investigated. Adjacent normal colonic mucosa specimens from 10 of these cases were selected as controls. PI3K protein was detected by immunohistochemistry and PIK3CA mutations were investigated by gene sequencing analysis. A flowcytometry-based apoptosis detection kit was used to determine PI3K inhibitor-induced apoptosis in CRC cell lines SW480 and SW620. Expression of phosphorylated protein kinase B in CRC cell lines was detected by Western blotting. RESULTS: There was a significant difference in the proportion of primary lesions (30%, 18/60) vs metastatic lesions (46.7%, 28/60) that were positive for PI3K (P < 0.05). Mutations were detected in exon 9 (13.3%) and exon 20 (8.3%). Out of 60 cases, seven mutations were identified: two hotspot mutations, C.1633G>A resulting in E545A, and C.3140A>G resulting in H1047R; two novel missense mutations C.1624G>A and C.3079G>A; and three synonymous mutations (C.1641G>A, C.1581C>T and C.3027T>A). Exposure of SW480 cells to PI3K inhibitor for 48 h resulted in a significant increase of apoptotic cells in a dose-dependent manner [3.2% apoptotic cells in 0 μmol/L, 4.3% in 5 μmol/L, 6.3% in 10 μmol/L (P < 0.05), and 6.7% in 20 μmol/L (P < 0.05)]. Moreover, PI3K inhibitor induced a similar significant increase of apoptotic cells in the SW620 cell line for 48 h [3.3% apoptotic cells in 0 μmol/L, 13.3% in 5 μmol/L (P < 0.01), 19.2% in 10 μmol/L (P < 0.01), and 21.3% in 20 μmol/L (P < 0.01)]. CONCLUSION: High PI3K expression is associated with CRC metastasis. PI3K inhibitor induced apoptosis in CRC cells and displayed strong cytotoxicity for highly metastatic cells. PI3K inhibition may be an effective treatment for CRC.

A detailed description of Phycocaris simulans Kemp, 1916 based on the specimen collected from Hainan Island is presented. Specific features and the differences between the Indian Ocean and the present material are described.

文摘A destructive extreme heat attacked Northeast Asia(NEA)in the midsummer of 2018,characterized by the average midsummer Tmax(daily maximum air temperature at 2 m)ranking first during the study period.The current study indicates that the cyclonic anomaly over the western North Pacific(WNP)was an important cause,which presents an anomaly of two standard deviations.The cyclonic anomaly over the WNP was accompanied by anomalous convection,which favored descending and anticyclonic anomalies over NEA through a local meridional cell.The anticyclonic anomaly over NEA corresponds to the northwestward extension of the WNP subtropical high and facilitated the occurrence of extreme heat.The tropical sea surface temperature anomaly(SSTA)presents a La Ni?a decaying episode,but the SSTA over the tropical Pacific and North Indian Ocean was weak in the summer.In contrast,the southeastern tropical Indian Ocean(SETIO)was obviously cool,which was the coolest after detrending.The SETIO cooling triggered a low-level southeasterly anomaly,which turned into a southwesterly after crossing the equator,due to the Coriolis force.The southwesterly anomaly extended eastwards and favored the cyclonic anomaly over the WNP.Meanwhile,the circulation anomalies over the SETIO and WNP were connected via a local meridional cell,with the ascending branch over the WNP.Moreover,the above mechanism also operates for the climate statistics,verifying the robust in?uence of the SETIO SSTA.Considering the consistency of the SETIO SSTA,it could be a potential predictor for the climate over the WNP and NEA.

文摘In this paper, we discussed the features of atmospheric circulations over Eurasia as a response to sea surface temperature anomalies (SSTAs) over the tropical Indian Ocean, the equatorial Pacific, Kuroshio and the North Atlantic. Our results are shown as follows: (1) CAM3.0, driven by the combined SSTAs over the four oceanic regions, can simulate well the features of anomalous atmospheric circulations over Eurasia in January 2008, indicating that the effects of the SSTAs over these four regions were one of the key causes of the anomalous systems over Eurasia. (2) The SSTAs over each key region contributed to the intensification of blocking over the Urals Mountains and a main East Asian trough. However, the influence of the SSTAs over individual oceanic regions differed from one another in other aspects. The SSTAs over the North Atlantic had an impact on the 500-hPa anomalous height (Z500A) over the middle-high latitudes and had a somewhat smaller effect over the low latitudes. For the warm SSTAs over Kuroshio, the subtropical high was much stronger, spread farther north than usual, and had an anomalous easterly that dominated the northwest Pacific Ocean. The warm SSTAs over the tropical Indian Ocean could have caused a negative Z500A from West Asia to Middle Asia, a remarkably anomalous southwesterly from the Indian Ocean to the south of China and an anomalous anticyclone circulation over the South China Sea-Philippine Sea region. Because of the La Nifia event, the winter monsoon was stronger than normal, with an anomalously cooler northerly over the southeastern coastal areas of China. (3) The combined effects of the SSTAs over the four key regions were likely more important to the atmospheric circulation anomalies of January 2008 over Eurasia than the effects of individual or partly combined SSTAS. This unique SSTA distribution possibly led to the circulation anomalies over Eurasia in January 2008, especially the atmospheric circulation anomalies over the subtropics, which were more similar to those of the winter E1 Nifio events than to the circulation anomalies following La Nifia.

文摘Grass carp reovirus (GCRV) is a tentative member of the Aquareovirus genus in the family Reoviridae. The mature virion comprises 11 dsRNA genomes enclosed by two concentric icosahedral proteins shells that is comprised of five core proteins and two outer capsid proteins. The genome sequence and 3D structure demonstrate there is a higher level of sequence homology in structural proteins between GCRV and mammalian orthoreoviruses (MRV) compared to other members of the family. To understand the pathogenesis of GCRV infection, the outer capsid protein VP5, a homology of the μ1 protein of MRV, was expressed in E.coli. It was found that the recombinant VP5 was highly expressed, and the expressed His-tag fusion protein was involved in the formation of the inclusion body. Additionally, specific anti-VP5 serum was prepared from purified protein and western blot demonstrated that the expressed protein was able to bind immunologically to rabbit anti GCRV particle serum and the immunogenicity was determined by ELISA assay. Additional experiments in investigating the functional properties of VP5 will further elucidate the role of the GCRV outer capsid protein VP5 during entry into host cells, and its interaction among viral proteins and host cells during the infection process.

文摘This paper reports the occurrence of a marine brachyuran crab species Eucrate alcocki Serene, in Serene et al., 1973, of the family Euryplacidae Stimpson, 1871, for first time from India, based on a male specimen from Parangipettai fish landing centre in Bay of Bengal, Southeast Coast of India. Although morphologically corresponding with what is currently defined as E. alcocki, the color pattern of the carapace of the present specimen is rather different from that of the Chinese material-only the anterior fifth of the carapace is marked with scattered red spots, the rest of the surface is yellowish, with four unusually shaped red blotches which almost look like Sanskrit characters.
