Application value of ATP based bioluminescence tumor chemo-sensitivity assay in the chemotherapy for ascites caused by recurrent ovarian cancer 被引量:1

Methods: More than ... Objective: To investigate the clinical value and application of ATP based bioluminescence tumor chemo-sensitivity assay (ATP-TCA) in the chemotherapy for ascites caused by recurrent ovarian cancer. Methods: More than 10 kinds of chemotherapeutic drugs or combinations were applied and 35 ascites specimens from recurrent ovarian cancer were analyzed by ATP-TCA. Sensitivity of chemotherapeutic drugs was assessed. After 2-4 chemotherapeutic cycles, clinical outcomes were analyzed, which were compared with those of 40 cases by empirical regimens. Results: 32 of 35 specimens were evaluated with an overall evaluation rate of 91%. The assay results suggested that chemo-naive patients responded to chemotherapeutic drugs with individualized profiles. The sensitivity rates of GEM, EPI, OXA, DDP, CBP, ADM, VP-16, CTX, NVB, 5-FU, PTX and TXT were 40%, 30%, 33%, 29%, 33%, 38%, 25%, 33%, 38%, 33%, 25% and 20%, respectively. While the sensitivity rates of combinations GEM + EPI, GEM + CBP, GEM + DDP, NVB + DDP, CTX + ADM + DDP, CTX + ADM, DDP + VP-16, OXA + 5-FU, VP-16 + IFO, PTX + DDP, TXT + CBP, VCR + CTX + MTX, DDP + CPT-11, OXA + CPT-11, and DTIC + CTX were 47%, 50%, 36%, 44%, 30%, 33%, 27%, 33%, 40%, 27%, 23%, 14%, 28%, 30% and 17%, respectively. In vitro results correlated well with clinical outcomes. Objective response rate (RR) in chemo-sensitivity-guided group was of significance compared with that in empirical-regimen-guided group. Conclusion: ATP-TCA is a choice for the screening of chemotherapeutic drugs against ascites caused by recurrent ovarian cancer with excellent sensitivity and reliability. ATP-TCA assay results correlate well with clinical outcomes, suggesting its clinical value in the management of difficult-to-manage therapeutic situations such as ascites in recurrent ovarian cancer. 展开更多

Methods: The effects of CMNa combined with paclitaxel and carboplatin on elevated CA125 levels in 25 post-operation advanced ovarian cancer patients were retrospectively analyzed and com- pared with those in 20 control cases. Results: After 1 cycle of chemotherapy, CA125 levels had decreasing trend compared with control, but had no statistical significance. While after two cycles of chemotherapy, CA125 levels decreased more rapidly compared with those in control. Side effects in two groups were alike. Conclusion: CMNa combined with paclitaxel and car- boplaline has a stronger effect on the decrease of elevated CA125 levels than paclitaxel and carboplatin have in the treatment of post-operation advanced ovarian cancer, which indicates that CMNa has chemo-sensitizing effects on chemotherapy of paclitaxel and carboplatin. 展开更多

Objective:The aim of our study was to evaluate the clinical efficacy and tolerability of metronomic chemotherapy in patients with advanced ovarian carcinoma.Methods:Fifteen patients with advanced ovarian carcinoma and bad performance status were subjected to daily cyclophosphamide(CTX) after failure of 1st line chemotherapy which included paclitaxel and carboplatin.Evaluation of the cases during treatment as regard treatment side effects and progression free survival.Results:Patients could tolerate low dose oral cyclophosphamide treatment without considerable side effects with improvement of performance status.The mean progression free survival was 12 months.Conclusion:Low dose oral cyclophosphamide could be considered as a palliative treatment of pretreated ovarian carcinomas with poor performance status. 展开更多

In this study, a cytochemical method and transmission electron microscopy was used to examine acid phosphatase activities of yolk granules throughout the early developmental stages of the Pacific oyster Crassostrea gigas. This study aimed to investigate the dynamic change of yolk granule acid phosphatase, and the mechanisms underlying its involvement in yolk degradation during the early developmental stages of molluscs. Three types of yolk granules (YGI, YGⅡ, and YGⅢ) that differed in electron density and acid phosphatase reaction were identified in early cleavage, morula, blastula, gastrula, trochophore, and veliger stages. The morphological heterogeneities of the yolk granules were related to acid phosphatase activity and degrees of yolk degradation, indicating the association of acid phosphatase with yolk degradation in embryos and larvae of molluscs. Fusion of yolk granules was observed during embryogenesis and larval development of C. gigas. The fusion of YGI (free of acid phosphatase reaction) with YGII (rich in acid phosphatase reaction) could be the way by which yolk degradation is triggered. 展开更多
