The Influence of LHRH, EB and PGF2α on the Development of Ovarian Follicles in Guinea Pigs

Effects of luwinizing hormone releasing hormone ( LHRH), estradiot benzoate (EB), prostaglandin (PGF2α) and the age of animals on the development of ovarian follicles in immature and mature guinea pigs were studied. ... Effects of luwinizing hormone releasing hormone ( LHRH), estradiot benzoate (EB), prostaglandin (PGF2α) and the age of animals on the development of ovarian follicles in immature and mature guinea pigs were studied. Administration of 5ng LHRH / hr, 10 times a day, starting from 20-30 days Of age induced the flrst vaginal opening within 4 days, and 5 of the 10 animals had first ovulation. Tire results also show that appropriate doses of EB facilitated the development of ovary. The granulosa cells of guinea pigs at different ages were cocuttured with the pituitart, glands of mature ones in vitro. Results indicate that the responsiveness of granulosa cells of 20-day-guinea prgs to gonadotropins was similar to that of the matured ones. Effects of gonadotrophin on ovulation in hysterectomized and PGF2α or saline-treated animals were tested. Results demonstrate that when PGF2α was given simultaneously with hCG or FSH, 80% of the guinea pigs ovulawd and had fresh, healthy looking corpus tuteum. It is concluded that increased LHRH release from hypothatamus may be the key factor for tire onset of puberty in guinea prgs. EB and PGF2α can influence the development of the ovary. 展开更多

Association between Two Polymorphisms of Follicle Stimulating Hormone Receptor Gene and Susceptibility to Polycystic Ovary Syndrome: a Meta-analysis 被引量:3

Objective To investigate the association between two polymorphisms of follicle stimulating hormone receptor (FSHR) gene and polycystic ovary syndrome (PCOS) susceptibility. Methods Case-control studies on relationship of Thr307Ala and Asn680Ser polymorphisms in FSHR gene and PCOS susceptibility were searched from PubMed, ISI web of knowledge, EBSCO, and China National Knowledge Infrastructure (CNKI) databases up to March 21, 2013. The pooled odds ratio (OR) and 95% confidence interval (CO were calculated using fixed- or random-effect model based on heterogeneity test in 5 genotype models analyses. Results A total of 11 studies were included in the Meta-analysis. The random-effect analysis showed Asn680Ser was significantly associated with the reduced susceptibility to PCOS with dominant model (Asn/Asn+Asn/Ser vs. Ser/Ser, OR=0.83, 95% CI: 0.69-1.00), recessive model (Asn/Asn vs. Asn/Ser+ Ser/Ser, OR=0.84, 95% CI: 0.72-0.98), homozygote comparison (Ash/Ash vs. Ser/Ser, 0R=0.79, 95% CI: 0.63-0.98), and the allele contrast (Asn vs. Ser, OR=0.87, 95% CI: 0.79-0.97) respectively(P=0.02, I2=56.0%), being protective factors for PCOS. However, no significant associations were found between Thr307Ala and PCOS. Conclusion There might be a significant association between Asn680Ser polymorphism and PCOS. 展开更多

Objective: To observe whether the effect of electroacupuncture (EA) on improving sex hormone disorders and follicle development is by decreasing the expression of anti-Mullerian hormone (AMH) in rats with experimental polycystic ovarian syndrome (PCOS). Methods: Forty rats were randomly divided into four groups, a normal group (NG), a model group (MG), an EA at acupoints group (EAAG), and an EA at non-acupoints group (EANAG), with 10 rats in each group. The rats in the EAAG and EANAG were intervened by EA treatment for consecutive 14 d. Zhongji (CV 3) and Guanyuan (CV 4) were selected as the acupoints in the EAAG, and the tip of the tail and 1 cm up from the tail tip were selected as the non-acupoints in the EANAG. After treatment, the histomorphological changes of the ovary, the levels of aromatase P450 (P450arom)/ testosterone and estradiol in the ovarian tissues, and the expressions of follicle stimulating hormone (FSH) and AMH were observed. Results: After treatment, compared with the MG and EANAG, the expression of AMH decreased (P<0.05), the levels of P450arom and estradiol in creased sign ifica ntly, and the level of testostero ne decreased sign ifica ntly (all P<0.01) in the EAAG. Additionally, several normal follicles were present and the number of cystically dilated follicles decreased in the EAAG. Compared with the MG and EANAG, the EAAG obviously had more follicular granulosa cells. Con elusion: EA can dow n-regulate the abn ormally in creased expressi on of AMH to improve sex hormone disorders and follicle development in PCOS rats. 展开更多
