利用RNA原位杂交和免疫组织化学定位技术分别检测了钙调素mRNA和钙调素蛋白在水稻(OryzasativaL.)花药和雌蕊发育过程中的时空分布特征。钙调素基因在绒毡层、柱头、花粉管生长途径、退化助细胞以及维管薄壁细胞... 利用RNA原位杂交和免疫组织化学定位技术分别检测了钙调素mRNA和钙调素蛋白在水稻(OryzasativaL.)花药和雌蕊发育过程中的时空分布特征。钙调素基因在绒毡层、柱头、花粉管生长途径、退化助细胞以及维管薄壁细胞中大量表达,也可在小孢子母细胞、小孢子、花粉、反足细胞、卵细胞以及中央细胞中检测到。钙调素基因的表达强度随不同的发育阶段而变化:花药发育早期表达强,以后逐渐减弱并向特定部位集中,如绒毡层和花粉萌发孔等。胚胎发育早期,钙调素基因在胚乳细胞中的表达比原胚中强,而后期则在分化胚中比胚乳细胞中强。推测在有性生殖过程中,钙调素可能通过Ca2+CaM信号途径调节小孢子发育、花粉萌发、花粉管生长。 展开更多

目的评价虹膜定位的波前引导个体化准分子激光原位角膜磨镶术(laser in situ keratomileusis,LASIK)后患者的视觉质量。方法选取2008年8月至12月在青岛明基眼科医院接受虹膜定位的波前引导个体化LASIK治疗的近视患者86例,其中男37例,女4... 目的评价虹膜定位的波前引导个体化准分子激光原位角膜磨镶术(laser in situ keratomileusis,LASIK)后患者的视觉质量。方法选取2008年8月至12月在青岛明基眼科医院接受虹膜定位的波前引导个体化LASIK治疗的近视患者86例,其中男37例,女49例。分别于术后1d、10d,1个月、3个月、6个月、12个月,通过回答问卷的方式,了解患者术后视觉质量下降症状的发生率和严重程度。结果术后裸眼视力明显提高。术后主要并发症为光晕、暗视力下降和眩光。随时间的推移,除单眼复视外各症状的发生率和严重程度逐渐下降,视觉质量明显改善,3个月时基本稳定,并且症状已不明显。术后6个月,98.8%的患者对手术的综合评价为满意。结论虹膜定位的波前引导个体化LASIK对患者的视觉质量影响轻微,患者满意度高。 展开更多

Comparative genetic studies have shown that there are widespread synteny and colinearity of the genes among different species within grass family. Rice ( Oryza sativa L.) is a model plant, and analysis of its genome allows us to reveal the common features and the evolutionary rules of the gramineous genomes and accumulate the data for establishment of a common genetic system in the Poaceae. In this study, a rice gene Pib ( 10.3 kb), a map-based cloned gene, and RFLP markers linked with it are used as the tested probes to investigate their homology and physical location among the tested species. Southern blotting analysis showed that there were sequences homologous to Pib in maize genome. Further, Pib was localized onto the chromosomes of O. sativa ssp. indica cv. Guangluai 4, O. officinalis Wall ex Watt and the inbred line of Zea mays cv. Huangzao 4. The results of fluorescence in situ hybridization (FISH) and double-color FISH indicated that a synteny of Pib and RFLP markers linked with Pib existed among the genomes of the three tested species. 展开更多

Recent molecular cytogenetic studies demonstrate that extensive centromere-telomere fusions are the main chromosomal rearrangements underlying the karyotypic evolution of extant muntjacs. Although the molecular mechanism of tandem fusions remains unknown, satellite DNA is believed to have facilitated chromosome fusions by non-allelic homologous recombination. Previous studies detected non-random hybridization signals of cloned satellite DNA at the postulated fusion sites on the chromosomes in Indian and Chinese muntjacs. But the genomic distribution and organization of satellite DNAs in other muntjacs have not been investigated. In this study, we have isolated four satellite DNA clones (BMCS, BM700, BM 1.1 k and FM700) from the black muntjac (Muntiacus crinifrons) and Fea's muntjac (M. feae), and hybridized these four clones onto chromosomes of four muntjac species (M. reevesi, M. crinifrons, M. gongshanenisis and M. feae). Besides the predominant centromeric signals, non-random interstitial hybridization signals from satellite I and II DNA clones (BMC5, BM700 and FM700) were also observed on the arms of chromosomes of these four muntjacs. Our results provide additional support for the notion that the karyotypes of M. crinifrons, M. feae and M. gongshanensis have evolved from a 2n = 70 ancestral karyotype by a series of chromosome fusions. 展开更多

In situ LAMP is a method that is generally more specific and sensitive than conventional techniques such as fluorescence in situ hybridization (FISH), primed in situ labeling (PRINS), and cycling primed in situ labeling (C-PRINS). Here, we describe the development and application of in situ LAMP to identify the chromosomal localization of DNA sequences. Actin filaments (AFs) in un-fixed pollen tubes of Amaryllis vittata Ait were visualized after TRITC-phalloidin staining with DMSO as a permeabilising agent. Typically, strands or hundles of microfilaments (Mfs) were distributed in the extreme tip as well as pollen tubes in a form of network.Fluorescent granules or circles of various sizes were frequently found that continued with the filamentous structures. In addition, a more brightly stained structure, possibly Mf organizing center, was observed. Treatment of pollen tubes with cytochalasin D(CD)for increasing time intervals (5-40 minutes) caused gradual reduction of strands until flurescent granules filled up the pollen tubes. Mcanwhile, cytoplasmie streaming was inhibited completely. Though closely associated with vegetative nuclei (VN) and generative cells (GC), AFs were not found in the cytoplasm of GC.Mg++concentration greatly affected the isolated Mfs. 展开更多

A full length amphioxus cDNA, encoding a novel phosducin-like protein (Amphi-PhLP), was identified for the first time from the gut cDNA library of Branchiostoma belcheri. It is comprised of 1 550 bp and an open reading frame (ORF) of 241 amino acids, with a predicted molecular mass of approximately 28 kDa. In situ hybridization histochemistry revealed a tissue-specific expression pattern of Amphi-PhLP with the high levels in the ovary, and at a lower level in the hind grit and testis, hepatic caecum, gill, endostyle, and epipbaryngeal groove, while it was absent in the muscle, neural tube and notochord. In the Chinese Hamster Ovary (CHO) cells transfected with the expression plasmid pEGFP-NI/Amphi-PhLP, the fusion protein was targeted in the cytoplasm of CHO cells, suggesting that Amphi-PhLP is a cytosolic protein. This work may provide a framework for further understanding of the physiological function of Amphi-PhLP in B. belcheri. 展开更多

Proteinase inhibitor (PI) mRNA was localized by in situ hybridization in tissue sections of root, stem and leaf of the resistant rice (B5) plant fed by brown planthopper nymphs. In the rice material without BPH feeding, PI gene was expressed in the root, stem and leaf, while the abundance of PI mRNA was low. In the rice material fed by BPH, PI gene was expressed substantially in the parenchyma of rice stem and leaf, but weakly in the root. The results indicated that the PI gene was up-regulated in the rice plant challenged by brown planthopper. For the first time, we reported the expression changes of proteinase inhibitor gene in plant which was infested by a piercing/sucking insect. 展开更多

The hafnium isotopic analysis using laser ablation has been widely conducted on Hf-rich minerals (zircon/baddeleyite/ calzirtite/eudialyte), however, little work has been reported on Hf-poor (〈100 ppm) minerals. This work presents a detailed procedure of in situ hafnium isotopic analysis from ruffle using laser ablation multiple collector inductively coupled plasma mass spectrometry (LA-MC-ICP-MS). The rutile U-Pb dating reference material JDX shows homogeneous hafnium isotopic ratios, with 176Hf/177Hf=0.281795±0.000015 (2SD, n=33) and176Lu/177Hf=0.000018±0.000004 (2SD, n=17) that suggest the possibility of using JDX as a new reference material hafnium isotopic measurement. We also measure hafnium isotopic compositions of other rutile U-Pb dating reference material (R10, Sugluk-4 and PCA-S207) and the 176Hf/177Hf values are similar to previously reported results, which confirms that we can acquire accurate and precise hafnium isotopic compositions using our developed analytical protocol. We analyzed hafnium isotopic compositions and U-Pb ages of rutile in high-temperature and ultrahigh-temperature granulites from various terrains of the Khondalite Belt from the North China Craton, combined with zircon results in the same area, suggesting that the metamorphic evolution history of the granulite is much more complicated than previously thought. 展开更多

Many genes involved in Wingless(wg) signal transduction pathway downstream of Wg, or it'' s vertebrate Wg homologue Wnt, have been identified.Transduction of the Wg signal downstream of Wg is mediated by nuclear TCF/LEF-1, through association with Ar-madillo (Arm)/β-catenin. Pygopus (pygo) is a new identified component in this pathway . Cellular localization experiment showed that pygo was expressed specifically in the nucleus. The expression profile of pygo in embryos was examined using in situ hybridization. Although pygo expressed ubiquitously in the embryos, it expressed at relatively high level in pre-blastoderm embryos which indicate a high degree of maternally provided message, fol-lowed by a low level of ubiquitous zygotic expression. This continues into larval tissues (including wing disc, eye disc and leg disc), where pygo appears to be expressed at low level. Comparison of pygo expression levels, in the wing disc, eye disc and leg disc, showed pygo expression level in the wing disc pouch and leg disc were rela-tive higher. 展开更多
