Progress has bee...This paper looks back to the development of plant tissue culture in China in the last century. Since 1934, tissue culture studies in China has kept up with the international development in the fields. Progress has been made by Chinese in nearly every branches of tissue culture, including in vitro organogenesis, shoot tip culture, anther culture, ovary culture, endosperm culture, protoplast culture as well as mass cell culture. On the basis of reviewing the articles written by Chinese on plant tissue culture, the internationally recognized contributions are specially mentioned. The applications of plant tissue culture to agriculture and industry in China are also introduced.展开更多

The authors observed the ultrastructure of isolated vegetative cells cultured on semi-solid media and compared them with those of monospores and isolated cells cultured in liquid media. The results showed that subcellular structures were quite different among cells in different conditions. In their development, isolated cells on semi-solid media did not show the characteristic subcellular feature of monospore formation, such as production of fibrous vesicles. Callus-like cells formed on semi-solid media underwent a distinctive modification in cellular organization. They developed characteristic cell inclusions and a special 2-layer cell covering. Golgi bodies, ER, starch grains, mitochondria. Vacuoles were not commonly found in them.展开更多
After deregulating the purine and riboflavin synthesis in the Gram-positive bacterium Bacillus subtilis,it is critical to amplify riboflavin operon with appropriate dosage in the host strain for remarkable increase of...After deregulating the purine and riboflavin synthesis in the Gram-positive bacterium Bacillus subtilis,it is critical to amplify riboflavin operon with appropriate dosage in the host strain for remarkable increase of riboflavin production.Bacillus subtilis RH13, a riboflavin-producing strain, was selected as host strain in the construction of engineering strains by protoplast fusion. The integrative plasmid pRB63 and autonomous plasmid pRB49, pRB62 containing riboflavin operon of B.subtilis 24 were constructed and transformed into the host strain respectively. Increasing one operon copy in B.subtilis RH13 results in about 0.4 g/L improvement in riboflavin yield and the appropriate number of operon copies was about 7—8. Amplifying more riboflavin operons is of no use for further improvement of yield of riboflavin. Furthermore, excessive operon dosage results in metabolic unbalance and is fatal to the host cells producing riboflavin.展开更多

Systematic study was carricd out on PEG-mediatcd transformation of cotyledon protoplast u...Transgenic plants were obtained by PEG-mediated tranfer of foreign gene into cotyledon protoplasts of Orychophragums violaceus. Systematic study was carricd out on PEG-mediatcd transformation of cotyledon protoplast using transient expression system, which showed 25-30 μg of pasmid, 15% PEG and a pH value of 8.0 as the optimal parameters contributing to the highest expression level. Using these parameters, cotyledon protoplasts were isolated, treated with bacterial plasmid DNA (pBI222 with HPT as selective marker) and PEG, and cultured at a density of 5×10 4/ml.After 10-15 days,they were selected by adding 25 μg/ml hygromycine. One month later, a few calli were observed, which were then transferred onto a solid medium with 50-100 μg/ml hygromycine for proliferation. Later they were transferred successively onto differentiation and rooting media and finally hygromycineresistant whole plants were obtaincd. The plants grew well in pots and a regeneration rate of 5 ×10(-5) was achieved. Then,excised leaves of the transgenic plants were used as explants for Southern blot analysis, which confirmed the stable integration of HPT gene into the chromosomal genome of Orychophragmus violaceus The transformation frequency was 10-5.展开更多

We collected eleven bench samples of No. 6 coal from the Heidaigou Surface Mine, Jungar Coalfield, Inner Mongolia, China, and four samples from the affiliated coal preparation plant. Based on these samples, we used in...We collected eleven bench samples of No. 6 coal from the Heidaigou Surface Mine, Jungar Coalfield, Inner Mongolia, China, and four samples from the affiliated coal preparation plant. Based on these samples, we used inductively coupled-plasma mass spectroscopy, X-ray diffraction, scanning electron microscope with an energy-dispersive X-ray spectrometer techniques, and borehole exploration data, to investigate the distribution, occurrence and enrichment causes of gallium (Ga) in the coals. Our results show: (1) Gallium is significantly enriched in the coal seams from the study area, with an average content of 18.8-26.0 ppm. Gallium is distributed heterogeneously in the coals, and reaches ore-forming scales only in No. 6 coal of Heidaigou Surface Mine, not in the other mining districts of Jungar Coalfield. (2) On the horizontal plane, Ga is enriched in the main minable coals from the northern and middle part of the coalfield. In the vertical profile, Ga content in the coal seams is higher at the base of Taiyuan Formation (Nos. 8 and 9) and Shanxi Formation (Nos. 3 and 4) than at the top of the Taiyuan Formation. Within the identical coal seam, Ga content is higher in the benches near the roof and floor than in the middle section. (3) Gallium in the coals is associated mainly with kaolinite and boehmite. Additionally, Ga may be adsorbed to some extent by humic acid, resulting in a high level in weathering coal. (4) Geological factors affect Ga enrichment in coal, such as the prop- erty of parent rocks in the source area, the sedimentary environment, organic matter, structure, and past magmatic hydrothermal activity. Especially, Ga content in parent rocks plays a leading role. (5) The mobility and precipitation of trace elements like Ga are controlled principally by the geochemical behavior of the major element A1. Terrestrial and transgressive environments can cause the precipitation of bauxite, whereas marine-continental depositional environments may cause the separation of Ga from A1. In addition, Ga may migrate in the form of gas tively enriched in high-volatile coal. and may be affected by the ground temperature. Thus, it is relatively enriched in high-volatile coal.展开更多

The silicene monolayers epitaxially grown on Ag(111) surfaces demonstrate various reconstructions with different electronic structures. Yet t...The progressive stacking of chalcogenide single layers gives rise to two- dimensional semiconducting materials with tunable properties that can be exploited for new field-effect transistors and photonic devices. Yet the properties of some members of the chalcogenide family remain unexplored. Indium selenide (InSe) is attractive for applications due to its direct bandgap in the near infrared, controllable p- and n-type doping and high chemical stability. Here, we reveal the lattice dynamics, optical and electronic properties of atomically thin InSe flakes prepared by micromechanical cleavage. Raman active modes stiffen or soften in the flakes depending on which electronic bonds are excited. A progressive blue-shift of the photoluminescence peaks is observed for decreasing flake thickness (as large as 0.2 eV for three single layers). First-principles calculations predict an even larger increase in the bandgap, 0.40 eV, for three single layers, and as much as 1.1 eV for a single layer. These results are promising from the point of view of the versatility of this material for optoelectronic applications at the nanometer scale and compatible with Si and III-V technologies.展开更多

The results show signi...In this study, the Pb/U fractionation between zircon and uraninite during femtosecond Laser Ablation Inductively Coupled Plasma Mass Spectrometry (fs-LA-ICP-MS) analysis was studied in detail. The results show significant Pb/U fractionation between zircon and uraninite during fs-LA-ICP-MS analysis that when calibrated against the zircon standard M257, the obtained U-Pb age of the Chinese national uraninite standard GBW04420 is 17% older than the recommended value. Thus, the accurate in-situ U-Pb dating of uraninite by LA-ICP-MS requires matrix-matched external standards for calibration. Uraninite in thin sections of two U-mineralized leucogranite from the Gaudeanmus in Namibia was analyzed by a fs-LA-ICP-MS equipped with a Signal Smooth Device (SSD), using laser spot and frequency of 10 μm and 1 Hz, respectively. When calibrated using GBW04420 as the external standard, two samples give weighted mean 2066pb/238U ages of 504±3 Ma (2σ, n=21) and 503±3 Ma (2σ, n=22), and only one of two samples yields a concordia U-Pb age of 507±1 Ma (2or, n=21). These results are consistent with ID-TIMS U-Pb ages of 509±1 and 508±12 Ma and are also indistinguishable from zircon U-Pb upper intercept ages of 506±33 Ma (2σ, n=29) and 501±51 Ma (2σ, n=29). The present study shows that in-situ U-Pb dating of uraninite can deliver more reliable formation ages of the deposit than dating coeval high-U zircon because the latter commonly suffer significant Pb loss after formation. Our results confirm that GBW04420 is an ideal matrix matching standard for in-situ U-Pb dating of uraninite.展开更多
The structural,electronic and elastic properties of solid nitromethane are investigated under pressure by performing first-principles density functional theory(DFT)calculations within the generalized gradient approxim...The structural,electronic and elastic properties of solid nitromethane are investigated under pressure by performing first-principles density functional theory(DFT)calculations within the generalized gradient approximation(GGA)and the local density approximation(LDA).The obtained ground state structure properties are found to be consistent with existing experimental and theoretical results.The pressure-induced variations of structure parameters(a,b,c and V)indicate that the solid nitromethane has an anisotropic compressibility,and the compression along the c direction is more difficult than along a and b directions.From the vibration curves of intermolecular bond length and bond angle,we find that the C—N bond is the most sensitive among these bonds under pressure,suggesting that the C—N bonds may be broken first under external loading.The influence of pressure on the electronic properties of solid NM has been studied,indicating that solid NM is an insulating compound with a large indirect band gap and tends to be a semiconductor with increasing pressure.Finally,we predict the elastic constants and their pressure dependence for the solid NM with the bulk modulus,Young’s modulus,shear modulus and the Poisson’s ratio derived.展开更多
Large roughness and structure disorder in ferroelectric ultrathin Langmuir-Blodgett(LB)film results in severe space scatter in electrical,ferroelectric and piezoelectric characteristics,thus limiting the nanoscale res...Large roughness and structure disorder in ferroelectric ultrathin Langmuir-Blodgett(LB)film results in severe space scatter in electrical,ferroelectric and piezoelectric characteristics,thus limiting the nanoscale research and reliability of nano-devices.However,no effective method aiming at large-area uniform organic ferroelectric LB film has ever been reported to date.Herein,we present a facile hot-pressing strategy to prepare relatively large-area poly(vinylidene fluoride)(PVDF)LB film with ultra-smooth surface root mean square(RMS)roughness is 0.3 nm in a 30μm×30μm area comparable to that of metal substrate,which maximized the potential of LB technique to control thickness distribution.More importantly,compared with traditionally annealed LB film,the hot-pressed LB film manifests significantly improved structure uniformity,less fluctuation in ferroelectric characteristics and higher dielectric and piezoelectric responses,owing to the uniform dipole orientation and higher crystalline quality.Besides,different surface charge relaxation behaviors are investigated and the underlying mechanisms are explained in the light of the interplay of surface charge and polarization charge in the case of nanoscale non-uniform switching.We believe that our work not only presents a novel strategy to endow PVDF LB film with unprecedented reliability and improved performance as a competitive candidate for future ferroelectric tunnel junctions(FTJs)and nano electro mechanical systems(NEMS),but also reveals an attracting coupling effect between the surface potential distribution and nanoscale non-uniform switching behavior,which is crucial for the understanding of local transport characterization modulated by band structure,bit signal stability for data-storage application and the related surface charge research,such as charge gradient microscopy(CGM)based on the collection of surface charge on the biased ferroelectric domains.展开更多

文摘Large roughness and structure disorder in ferroelectric ultrathin Langmuir-Blodgett(LB)film results in severe space scatter in electrical,ferroelectric and piezoelectric characteristics,thus limiting the nanoscale research and reliability of nano-devices.However,no effective method aiming at large-area uniform organic ferroelectric LB film has ever been reported to date.Herein,we present a facile hot-pressing strategy to prepare relatively large-area poly(vinylidene fluoride)(PVDF)LB film with ultra-smooth surface root mean square(RMS)roughness is 0.3 nm in a 30μm×30μm area comparable to that of metal substrate,which maximized the potential of LB technique to control thickness distribution.More importantly,compared with traditionally annealed LB film,the hot-pressed LB film manifests significantly improved structure uniformity,less fluctuation in ferroelectric characteristics and higher dielectric and piezoelectric responses,owing to the uniform dipole orientation and higher crystalline quality.Besides,different surface charge relaxation behaviors are investigated and the underlying mechanisms are explained in the light of the interplay of surface charge and polarization charge in the case of nanoscale non-uniform switching.We believe that our work not only presents a novel strategy to endow PVDF LB film with unprecedented reliability and improved performance as a competitive candidate for future ferroelectric tunnel junctions(FTJs)and nano electro mechanical systems(NEMS),but also reveals an attracting coupling effect between the surface potential distribution and nanoscale non-uniform switching behavior,which is crucial for the understanding of local transport characterization modulated by band structure,bit signal stability for data-storage application and the related surface charge research,such as charge gradient microscopy(CGM)based on the collection of surface charge on the biased ferroelectric domains.
