AIM:To investigate screening makers for gastric cancer,we assessed the association between gastric cancer and serum pepsinogens(PGs).METHODS:The subjects comprised 450 patients with gastric cancer,111 individuals with...AIM:To investigate screening makers for gastric cancer,we assessed the association between gastric cancer and serum pepsinogens(PGs).METHODS:The subjects comprised 450 patients with gastric cancer,111 individuals with gastric atrophy,and 961 healthy controls.Serum anti-Helicobacter pylori(H.pylori) immunoglobulin G(IgG),PGⅠand PG Ⅱ were detected by enzyme-linked immunosorbent assay.Gastric atrophy and gastric cancer were diagnosed by endoscopy and histopathological examinations.Odds ratios and 95%CIs were calculated using multivariate logistic regression.RESULTS:Rates of H.pylori infection remained high in Northeastern China.Rates of H.pylori IgG positivity were greater in the gastric cancer and gastric atrophy groups compared to the control group(69.1% and 75.7% vs 49.7%,P < 0.001).Higher levels of PG Ⅱ(15.9 μg/L and 13.9 μg/L vs 11.5 μg/L,P < 0.001) and lower PGⅠ/PG Ⅱ ratio(5.4 and 4.6 vs 8.4,P < 0.001) were found in patients with gastric cancer or gastric atrophy compared to healthy controls,whereas no correlation was found between the plasma PGⅠconcentration and risk of gastric cancer(P = 0.537).In addition,multivariate logistic analysis indicated that H.pylori infection and atrophic gastritis were independent risk factors for gastric cancer.Lower plasma PGⅠ/PG Ⅱ ratio was associated with higher risks of atrophy and gastric cancer.Furthermore,plasma PG Ⅱ?level?significantly?correlated?with?H.pyloriinfected gastric cancer.CONCLUSION:Serum PG Ⅱ concentration and PG Ⅰ/PG Ⅱ ratio are potential biomarkers for H.pyloriinfected gastric disease.PG Ⅱ is independently associated with risk of gastric cancer.展开更多

METHODS:Seventy seven patients with recurrence after pancreatic cancer surgery were retros...AIM:To investigate predilection sites of recurrence of pancreatic cancer by computed tomography(CT)in follow-up after surgery. METHODS:Seventy seven patients with recurrence after pancreatic cancer surgery were retrospectively identified.The operative technique,R-status,T-stage and development of tumor markers were evaluated. Two radiologists analyzed CT scans with consensus readings.Location of local recurrence,lymph node recurrence and organ metastases were noted.Surgery and progression of findings on follow-up CT were con-sidered as reference standard. RESULTS:The mean follow-up interval was 3.9± 1.8 mo,with a mean relapse-free interval of 12.9± 10.4 mo.The predominant site of recurrence was local (65%),followed by lymph node(17%),liver metastasis (11%)and peritoneal carcinosis(7%).Local recurrence emerged at the superior mesenteric artery(n=28),the hepatic artery(n=8),in an area defined by the surrounding vessels:celiac trunk,portal vein,inferior vena cava(n=22),and in a space limited by the mesenteric artery,portal vein and inferior vena cava(n=17). Lymph node recurrence occurred in the mesenteric root and left lateral to the aorta.Recurrence was confirmed by surgery(n=22)and follow-up CT(n=55).Tumor markers[carbohydrate antigen 19-9(CA19-9),carcinoembryonic antigen(CEA)]increased in accordance with signs of recurrence in most cases(86%CA19-9;79.2% CEA). CONCLUSION:Specific changes of local and lymph node recurrence can be found in the course of the cardinal peripancreatic vessels.The superior mesenteric artery is the leading structure for recurrence.展开更多

METHODS: A retrospective analysis of dat...AIM: To investigate the usefulness of tumor markers and adenosine deaminase in differentiating between tuberculous peritonitis (TBP) and peritoneal carcinoma- tosis (PC). METHODS: A retrospective analysis of data was per- formed on consecutive patients who underwent perito- neoscopic and abdominal computed tomography (CT) evaluations. Among 75 patients at the Seoul National University Hospital from January 2000 to June 2010 who underwent both tests, 27 patients (36.0%) and 25 patients (33.3%) were diagnosed with TBP and PC, re- spectively. Diagnosis was confirmed by peritoneoscopic biopsy. RESULTS: Serum c-reactive protein (7.88:1:6.62 mg/ dL vs 3.12 + 2.69 mg/dL, P = 0.01), ascites adenos- ine deaminase (66.76:1:32.09 IU/L vs 13.89 :l: 8.95 IU/L, P 〈 0.01), ascites lymphocyte proportion (67.77 :1: 23.41% vs 48.36 + 18.78%, P 〈 0.01), and serum- ascites albumin gradient (0.72 + 0.49 g/dL vs 1.05 + 0.50 g/dL, P = 0.03) were significantly different be- tween the two groups. Among tumor markers, serum and ascites carcinoembryonic antigen, serum carbohy- drate antigen 19-9 showed significant difference be- tween two groups. Abdominal CT examinations showed that smooth involvement of the parietal peritoneum was more common in the TBP group (77.8% vs 40.7%) whereas nodular involvement was more common in the PC group (14.8% vs 40.7%, P = 0.04). From receiver operating characteristic (ROC) curves ascites adeno- sines deaminase (ADA) showed better discriminative capability than tumor markers. An ADA cut-off level of 21 IU/L was found to yield the best results of differ- ential diagnosis; sensitivity, specificity, positive predic- tive value, and negative predictive value were 92.0%, 85.0%, 88.5% and 89.5%, respectively. CONCLUSION: Besides clinical and radiologic findings, ascitic fluid ADA measurement is helpful in the differen- tial diagnosis of TBP and PC.展开更多

By coupling these indexes and instru...This paper presents the results of determining trace elements and δ 13C, δ 18O in Gyraulus sibirica shell continuously preserved in Xingcuo Lake sediments in the recent 50 years. By coupling these indexes and instrumental meteorological data on its basin to determine the relations among them, we probed quantitatively the climatic signals recorded in Xingcuo Lake sediments. The results showed that gastropod shells in Xingcuo Lake were formed in warm seasons; that trace elements, and stable isotope in shells just recorded the climatic signals of the warm seasons instead of that of the whole year; that Mg/Ca ratio and δ 18O proxy had certain correlation with air temperature in warm seasons, when the δ 18O proxy was more sensitive; with the average ratio of them being 0.28×10 -3/℃ and the variation rate dδ 18O/dT being 1.64×10 -3/℃; that the Sr/Ca ratio and δ 13C proxy, especially Sr/Ca ratio, had close correlation; and that dSr/Ca/dP was -0.045/mm.展开更多

Methods We enrolled all patients tested for HBV serological markers, such as HBV surface antigen (HBsAg), HBV surface antibody (HBsAb), hepatitis B e antigen (HBeAg), hepatitis B e antibody (HBeAb), HBV core antibody (HBcAb), and HBV-DNA from July 2008 to July 2009 in Peking Union Medical College Hospital. The positive rate of each HBV serological marker was calculated according to gender, age, and department, respectively. The positive rates of HBV-DNA among patients with positive HBsAg were also analyzed. Results Among 27 409 samples included, 2681 (9.8%) were HBsAg positive. When patients were divided into 9 age groups, the age-specific positive rate of HBsAg was 1.2%, 9.6%, 12.3%, 10.9%, 10.3%, 9.7%, 8.0%, 5.8%, and 4.3%, respectively. The positive rate of HBsAg in non-surgical department, surgical department, and health examination center was 16.2%, 5.8%, and 4.7%, respectively. The positive rate of HBsAg of males (13.3%) was higher than that of females (7.3%, P=0.000). Among the 2681 HBsAg (+) patients, 1230 (45.9%) had HBV-DNA test, of whom 564 (45.9%) were positive. Patients with HBsAg (+), HBeAg (+), and HBcAg (+) result usually had high positive rate of HBV-DNA results (71.8%, P=0.000). Conclusions Among this group of patients in our hospital, the positive rate of HBsAg was relatively high. Age group of 20-29, males, and patients in non-surgical departments were factors associated with high positive rate of HBsAg.展开更多

We used two-dimensional electrophoresis (2-DE)-based proteomic strategies to identify potential serum markers predicting hepatitis B e antigen (HBeAg) seroconversion in these patients during IFN therapy. Two groups of patients were enrolled: training and validation. In the training group, 2-DE experiments and subsequent identification of altered levels of proteins showed that α-2-HS-glycoprotein, leucine-rich α-2-glycoprotein, and haptoglobin were significantly upregulated as compared with baseline levels in the HBeAg seroconversion group, whereas apolipoprotein C-III precursor, leucine-rich α-2-glycoprotein, and α-albumin were downregulated in the non-seroconversion group. For patients with HBeAg seroconversion in the training group, Western blot analyses showed that α-2-HS-glycoprotein levels in 75% of patients were significantly upregulated at the end of the treatment as compared with baseline levels. Subsequent experiments in the validation group showed that α-2-HS-glycoprotein levels were significantly increased at week 4 in 83.33% of patients in the HBeAg seroconversion group. Dynamic changes in the serum level of α-2-HS-glycoprotein may be a potential early marker for predicting HBeAg seroconversion during IFN treatment for CHB.展开更多

Here, we performed a study to investigate the differences of TrxR activity between normal people and breast tumor patients, with the expectation that TrxR activity can be considered as a novel biomarker involved in breast cancer diagnosis and treatment. In this study, we have demonstrated for the first time that TrxR levels in breast cancer patients before therapy were significantly higher than those of normal people (P〈0.05). No significant difference of TrxR activity was found among the patients at different stages of breast tumor progression. In the patient group of TrxR 〈10 U/mL, evaluation of TrxR activity has shown a high consistency with the results from cancer diagnostic imaging. Overall, these observations suggest TrxR as an effective biomarker to monitor and evaluate the clinical outcome of breast cancer therapeutics, and identify TrxR activity as a novel approach for breast cancer early-stage detection.展开更多
The study and characterization of biomolecules involved in the interaction between mycobacteria and their hosts are crucial to determine their roles in the invasion process and provide basic knowledge about the biolog...The study and characterization of biomolecules involved in the interaction between mycobacteria and their hosts are crucial to determine their roles in the invasion process and provide basic knowledge about the biology and pathogenesis of disease.Promising new biomarkers for diagnosis and immunotherapy have emerged recently.Mycobacterium is an ancient pathogen that has developed complex strategies for its persistence in the host and environment,likely based on the complexity of the network of interactions between the molecules involved in infection.Several biomarkers have received recent attention in the process of developing rapid and reliable detection techniques for tuberculosis.Among the most widely investigated antigens are CFP-10(10-kDa culture filtrate protein),ESAT-6(6-kD a early secretory antigenic target),Ag85 A,Ag85 B,CFP-7,and PPE18.Some of these antigens have been proposed as biomarkers to assess the key elements of the response to infection of both the pathogen and host.The design of novel and accurate diagnostic methods is essential for the control of tuberculosis worldwide.Presently,the diagnostic methods are based on the identification of molecules in the humoral response in infected individuals.Therefore,these tests depend on the capacity of the host to develop an immune response,which usually is heterogeneous.In the last 20 years,special attention has been given to the design of multiantigenic diagnostic methods to improve the levels of sensitivity and specificity.In this review,we summarize the state of the art in the study and use of mycobacterium biomolecules with the potential to support novel tuberculosis control strategies.展开更多
Two comparative simulation experiments(a normal atmospheric-pressure opening system and a 20 MPa closed system)were conducted to study the geochemical evolution of n-alkane,sterane,and terpane biomarkers in the proces...Two comparative simulation experiments(a normal atmospheric-pressure opening system and a 20 MPa closed system)were conducted to study the geochemical evolution of n-alkane,sterane,and terpane biomarkers in the process of oil cracking into gas under different pressures.With an initial experimental temperature set at 300°C,the temperature was increased to 650°C at a heating rate of 30°C/h.The products were tested every 50°C starting at 300°C,and a pressure of 20 MPa was achieved using a water column.The low-maturity crude oil sample was from the Paleogene system in the Dongying sag in eastern China.The threshold temperature obtained for the primary oil cracking process in both pressure systems was 450°C.Before the oil was cracked into gas,some components,including macromolecular n-alkanes,were cracked into medium-or small-sized n-alkanes.The secondary oil cracking of heavy hydrocarbon gases of C2–5to methane mainly occurred between 550°C to 650°C,and the parameters Ln(C1/C2)and Ln(C1/C3),as well as the dry coefficients,increased.Overpressure inhibited the oil cracking process.In the 20 MPa system,the oil conversion rate decreased,the temperature threshold for gas generation rose,and oil cracking was inhibited.Compared with the normal pressure system,high-carbon n-alkanes and other compounds in the 20 MPa pressure system were reserved.Furthermore,the parameters∑C21-/∑22+,Ln(C1/C2),and Ln(C1/C3),as well as the dry coefficients,decreased within the main temperature range.During secondary oil cracking(550°C to 600°C),the Ph/nC18and Pr/nC17decreased.High pressure influenced the evolution of the biomarkers Ts and Tm,C31homohopane,C29sterane,and their related maturity parameters to different extents during oil cracking under different temperature ranges.展开更多
OBJECTIVE:To analyze the distinctive pathological characteristics in the spectrums of spontaneous infrared radiation at the Taiyuan(LU 9) acupoint in patients with asthma.METHODS:A highly sensitive infrared spectrum d...OBJECTIVE:To analyze the distinctive pathological characteristics in the spectrums of spontaneous infrared radiation at the Taiyuan(LU 9) acupoint in patients with asthma.METHODS:A highly sensitive infrared spectrum detecting device was used to detect the spectrums of spontaneous infrared radiation at Taiyuan(LU 9) in 37 asthma patients and 34 healthy volunteers.RESULTS:Asthma patients had significantly lower infrared intensity than that of the healthy volunteers(P>0.01).Asthma patients had significantly lower overall infrared radiation intensity at the left Taiyuan(LU 9) than that of healthy volunteers(P>0.05),but there was no significant difference between healthy volunteers and asthma patients at the right Taiyuan(LU 9)(P>0.05).The infrared radiation intensity of 17 wavelength spots at the left Taiyuan(LU 9) and 4 wavelength spots at the right Taiyuan(LU 9) in asthma patients were significantly lower than those of healthy volunteers(P>0.05).At 2 μm,the infrared radiation intensity of asthma patients was significantly stronger than that of healthy volunteers(P>0.05).At 19 wavelength spots in the healthy volunteers and at 4 wavelength spots in the asthma patients,the left Taiyuan(LU 9) showed a significantly stronger intensity than that of the right Taiyuan(LU 9)(P>0.05).By Pearson's χ2 test,healthy volunteers had more wavelength spots that were significantly different between the left and right Taiyuan(LU 9) than the asthma patients(P>0.01).CONCLUSION:Changes in the infrared spectrum at the Taiyuan(LU 9) acupoint in asthma patients may reflect distinct pathological changes.Certain acupuncture points may be related to specific organs.展开更多

But application of iPSCs in regenerative medicine requires rapid and accurate ch...Induced pluripotent stem cells (iPSCs) are an attractive cell source for regenerative medicine through cell therapy or drug screening. But application of iPSCs in regenerative medicine requires rapid and accurate charac- terization of iPSCs. Here, we demonstrate the detection of multiple antigens present in iPSC lysate using rapid, label- free surface plasmon resonance imaging (SPRi) assay. Validation of pluripotency is an important aspect of iPSC research. In this study, we fabricated antibody array against pluripotency biomarkers and found that our array suc- cessfully detect corresponding antigens in stem cell lysate. Each antibody recognized its specific antigens presented in iPSC lysates and a certain degree of variability was observed in comparison with other cell lysates. The results suggested that SPRi is a versatile technology feasible for the detection of multiple antigens presented in iPSC lysate. Further extension of this method may be applied in the characterization and high-throughput biomarker profiling of iPSCs.展开更多

Patients with HBsAg (+), HBeAg (+), and HBcAg (+) result usually had high positive rate of HBV-DNA results (71.8%, P=0.000). Conclusions Among this group of patients in our hospital, the positive rate of HBsAg was relatively high. Age group of 20-29, males, and patients in non-surgical departments were factors associated with high positive rate of HBsAg.

文摘The efficacy of interferon (IFN) is limited in about 1/3 of patients with chronic hepatitis B (CHB). We used two-dimensional electrophoresis (2-DE)-based proteomic strategies to identify potential serum markers predicting hepatitis B e antigen (HBeAg) seroconversion in these patients during IFN therapy. Two groups of patients were enrolled: training and validation. In the training group, 2-DE experiments and subsequent identification of altered levels of proteins showed that α-2-HS-glycoprotein, leucine-rich α-2-glycoprotein, and haptoglobin were significantly upregulated as compared with baseline levels in the HBeAg seroconversion group, whereas apolipoprotein C-III precursor, leucine-rich α-2-glycoprotein, and α-albumin were downregulated in the non-seroconversion group. For patients with HBeAg seroconversion in the training group, Western blot analyses showed that α-2-HS-glycoprotein levels in 75% of patients were significantly upregulated at the end of the treatment as compared with baseline levels. Subsequent experiments in the validation group showed that α-2-HS-glycoprotein levels were significantly increased at week 4 in 83.33% of patients in the HBeAg seroconversion group. Dynamic changes in the serum level of α-2-HS-glycoprotein may be a potential early marker for predicting HBeAg seroconversion during IFN treatment for CHB.

文摘Overexpression of thioredoxin reductase has been identified in a variety of primary tumors, suggesting the levels of TrxR activity between tumor patients and normal people are potentially different. Here, we performed a study to investigate the differences of TrxR activity between normal people and breast tumor patients, with the expectation that TrxR activity can be considered as a novel biomarker involved in breast cancer diagnosis and treatment. In this study, we have demonstrated for the first time that TrxR levels in breast cancer patients before therapy were significantly higher than those of normal people (P〈0.05). No significant difference of TrxR activity was found among the patients at different stages of breast tumor progression. In the patient group of TrxR 〈10 U/mL, evaluation of TrxR activity has shown a high consistency with the results from cancer diagnostic imaging. Overall, these observations suggest TrxR as an effective biomarker to monitor and evaluate the clinical outcome of breast cancer therapeutics, and identify TrxR activity as a novel approach for breast cancer early-stage detection.

文摘The study and characterization of biomolecules involved in the interaction between mycobacteria and their hosts are crucial to determine their roles in the invasion process and provide basic knowledge about the biology and pathogenesis of disease.Promising new biomarkers for diagnosis and immunotherapy have emerged recently.Mycobacterium is an ancient pathogen that has developed complex strategies for its persistence in the host and environment,likely based on the complexity of the network of interactions between the molecules involved in infection.Several biomarkers have received recent attention in the process of developing rapid and reliable detection techniques for tuberculosis.Among the most widely investigated antigens are CFP-10(10-kDa culture filtrate protein),ESAT-6(6-kD a early secretory antigenic target),Ag85 A,Ag85 B,CFP-7,and PPE18.Some of these antigens have been proposed as biomarkers to assess the key elements of the response to infection of both the pathogen and host.The design of novel and accurate diagnostic methods is essential for the control of tuberculosis worldwide.Presently,the diagnostic methods are based on the identification of molecules in the humoral response in infected individuals.Therefore,these tests depend on the capacity of the host to develop an immune response,which usually is heterogeneous.In the last 20 years,special attention has been given to the design of multiantigenic diagnostic methods to improve the levels of sensitivity and specificity.In this review,we summarize the state of the art in the study and use of mycobacterium biomolecules with the potential to support novel tuberculosis control strategies.

文摘Two comparative simulation experiments(a normal atmospheric-pressure opening system and a 20 MPa closed system)were conducted to study the geochemical evolution of n-alkane,sterane,and terpane biomarkers in the process of oil cracking into gas under different pressures.With an initial experimental temperature set at 300°C,the temperature was increased to 650°C at a heating rate of 30°C/h.The products were tested every 50°C starting at 300°C,and a pressure of 20 MPa was achieved using a water column.The low-maturity crude oil sample was from the Paleogene system in the Dongying sag in eastern China.The threshold temperature obtained for the primary oil cracking process in both pressure systems was 450°C.Before the oil was cracked into gas,some components,including macromolecular n-alkanes,were cracked into medium-or small-sized n-alkanes.The secondary oil cracking of heavy hydrocarbon gases of C2–5to methane mainly occurred between 550°C to 650°C,and the parameters Ln(C1/C2)and Ln(C1/C3),as well as the dry coefficients,increased.Overpressure inhibited the oil cracking process.In the 20 MPa system,the oil conversion rate decreased,the temperature threshold for gas generation rose,and oil cracking was inhibited.Compared with the normal pressure system,high-carbon n-alkanes and other compounds in the 20 MPa pressure system were reserved.Furthermore,the parameters∑C21-/∑22+,Ln(C1/C2),and Ln(C1/C3),as well as the dry coefficients,decreased within the main temperature range.During secondary oil cracking(550°C to 600°C),the Ph/nC18and Pr/nC17decreased.High pressure influenced the evolution of the biomarkers Ts and Tm,C31homohopane,C29sterane,and their related maturity parameters to different extents during oil cracking under different temperature ranges.

文摘OBJECTIVE:To analyze the distinctive pathological characteristics in the spectrums of spontaneous infrared radiation at the Taiyuan(LU 9) acupoint in patients with asthma.METHODS:A highly sensitive infrared spectrum detecting device was used to detect the spectrums of spontaneous infrared radiation at Taiyuan(LU 9) in 37 asthma patients and 34 healthy volunteers.RESULTS:Asthma patients had significantly lower infrared intensity than that of the healthy volunteers(P>0.01).Asthma patients had significantly lower overall infrared radiation intensity at the left Taiyuan(LU 9) than that of healthy volunteers(P>0.05),but there was no significant difference between healthy volunteers and asthma patients at the right Taiyuan(LU 9)(P>0.05).The infrared radiation intensity of 17 wavelength spots at the left Taiyuan(LU 9) and 4 wavelength spots at the right Taiyuan(LU 9) in asthma patients were significantly lower than those of healthy volunteers(P>0.05).At 2 μm,the infrared radiation intensity of asthma patients was significantly stronger than that of healthy volunteers(P>0.05).At 19 wavelength spots in the healthy volunteers and at 4 wavelength spots in the asthma patients,the left Taiyuan(LU 9) showed a significantly stronger intensity than that of the right Taiyuan(LU 9)(P>0.05).By Pearson's χ2 test,healthy volunteers had more wavelength spots that were significantly different between the left and right Taiyuan(LU 9) than the asthma patients(P>0.01).CONCLUSION:Changes in the infrared spectrum at the Taiyuan(LU 9) acupoint in asthma patients may reflect distinct pathological changes.Certain acupuncture points may be related to specific organs.

文摘Induced pluripotent stem cells (iPSCs) are an attractive cell source for regenerative medicine through cell therapy or drug screening. But application of iPSCs in regenerative medicine requires rapid and accurate charac- terization of iPSCs. Here, we demonstrate the detection of multiple antigens present in iPSC lysate using rapid, label- free surface plasmon resonance imaging (SPRi) assay. Validation of pluripotency is an important aspect of iPSC research. In this study, we fabricated antibody array against pluripotency biomarkers and found that our array suc- cessfully detect corresponding antigens in stem cell lysate. Each antibody recognized its specific antigens presented in iPSC lysates and a certain degree of variability was observed in comparison with other cell lysates. The results suggested that SPRi is a versatile technology feasible for the detection of multiple antigens presented in iPSC lysate. Further extension of this method may be applied in the characterization and high-throughput biomarker profiling of iPSCs.
