The material T240_6 derived from SC 2 young embryo of the combination CA9211/RW15 (6D/6V alien substitution) was telosomic substitution line of 6VS identified by GISH (genomic in situ hybridization) analysis. The 6VS was microdissected with a needle and transferred into a 0.5 mL Ep tube. In the 'single tube', all the subsequence steps were conducted. After two round of LA (Linker adaptor)_PCR amplification, the size of PCR bands ranged from 100 to 3 000 bp, with predominate bands 600-1 500 bp. The products were confirmed by Southern blotting analysis using Haynaldia villosa (L.) Schur. genomic DNA labeled with 32 P as probe. The PCR products were purified and ligated into clone vector-pGEM_T easy vector. Then, the plasmids were transformed into competence E. coli JM109 with cool CaCl 2. It was estimated that there were more than 17 000 white clones in the library. The size of insert fragments distributed from 100-1 500 bp, with average of 600 bp. Using H. villosa genomic DNA as probe, dot blotting results showed that 37% clones displayed strong and medium positive signals, and 63% clones had faint or no signals. It is demonstrated that there were about 37% repeat sequence clones and 67% single/unique sequence clones in the library. Eight H. villosa_specific clones were screened from the library, and two clones pHVMK22 and pHVMK134 were used for RFLP analysis and sequencing. Both of them were H. villosa specific clones. The pHVMK22 was a unique sequence clone, and the pHVMK134 was a repeat sequence clone. When the pHVMK22 was used as a probe for Southern hybridization, all the powdery mildew resistance materials showed a special band of 2 kb, while all the susceptible ones not. The molecular biological mechanism of the biosynthe... Blue-grained wheat derived from the hybrid Triticum aestivum L. X Thinopyrum ponticum (Podp.) Barkworth et D. R. Dewey (Agropyron elongatum (Host) P. Beauv., 2n=70). The molecular biological mechanism of the biosynthetic pathway of blue pigments in the blue grain remains unclear yet. Dihydroflavonol 4-reductase (DFR) is one of the key enzymes controlling flavonoid synthesis in anthocyanin biosynthetic pathway, and may directly participate in the formation of blue pigment in the aleurone layer of blue-grained wheat. Here we cloned a DFR cDNA (TaDFR) from the developing seeds of blue-grained wheat, and four DFR genomic DNAs from Th. ponticum (ThpDFR.t), blue-grained wheat (TaDFR.bg), white-grained offspring of light blue-grained wheat (TaDFR.wg) and Chinese Spring (2n=42) (TaDFR.csg), respectively. TaDFR cDNA encodes a 354 amino-acids polypeptide with high identity to DFR from Hordeum vulgare L. (94%), Oryza sativa L. (83%), Zea mays L.(84%). The result of cluster analysis showed that TaDFR cDNA nucleotide sequence has 100% identity with that of TaDFR.csg. The four DFR genomic DNAs have extraordinary high homology and each has three introns. The differences of the four DFR genomic DNAs mainly exist in introns. Southern blotting analysis showed that there are at least 3-5 DFR copies in wheat, the copy numbers in different color grain wheats are not significantly different. The hybridization band patterns were the same, but different from that of Th. ponticum. DFR in blue-grained wheat belongs to a DFR superfamily. Northern blotting analysis indicated that the DFR expressed in the developing seeds of both blue- and white-grained wheat at 15 d after flowering (DAF), the mRNA levels of DFR reached the highest at 18 DAF, then declined quickly and disappeared at 33 DAF But the expression levels in blue-grained seeds were higher than that in white grain at the same seed developing stages. DFR transcripts accumulated in young leaves, and leaf sheaths of blue- and white-grained wheat and Th ponticum, but not detected in roots from different color wheats and developing seeds of Th. ponticum. Results indicated that there may exist some regulatory gene(s) which can increase the expression of DFR in the aleurone layer of blue-grained wheat, and thus resulting in the formation of blue pigments. 展开更多

'Chinese Spring') X barley (Hordeum vulgare L. cv. 'Betzes') was analyzed, and six disomic alien substitution lines were screened by GISH. The chromosome configurations in pollen mother cells at meiotic metaphase I (PMCs M I) of F, from each disomic substitution line respectively crossed with double ditelocentric lines 2A, 2B and 2D of 'Chinese Spring' were observed, and a set of wheat-barley disomic alien substitution lines 2H(A), 2H(B) and 2H(D) were obtained. The RFLP analysis with the probe psr131 on the short arm of wheat homeologous group 2 combining with four restriction enzymes were carried out. The results indicated that the probe psr131 could be used as molecular marker to tag the barley chromosome 2H. The barley chromosome 2H had good genetic compensation ability for wheat chromosomes 2B and 2D in vitality and other agronomic characters. The result of testing seed was that the wheat appearance starch quality had been changed from the half-farinaceous of 'Chinese Spring' to the half-cutin of substitution lines by transferring the barley chromosome 2H to wheat. 展开更多

The time course of the initiation of different types of apical primordia on the stem of wheat (Triticum aestivum L.) was investigated to establish a comprehensive pattern of apical primordium development and quantitative dynamic relationships among different primordium initiation. The results showed that the initiation of leaf and bracteal primordia took the S shape, and that of spikelet and floret took the rise stage of parabolic shape in relation to GDD (growing degree days after sowing) in wheat. The bracteal was much like vegetative rather than reproductive organ in terms of the dynamic pattern of primordium initiation. The progress of spikelet and floret primordium initiation could well reflect the characteristics of different genotypes and the impact of different ecological environments on wheat apical development. The integrated model of different primordium initiation in shoot of wheat was an S_shaped curvilinear, and could be divided into three sub_models. The number of leaf primordia was affected by both genotype and environment, while the numbers of bracteal, spikelet and floret primordia were affected mainly by environmental factors. Measured with mean thermochron, the rates of initiation of leaf, bracteal and spikelet primordia in normal planting were the highest among all the sowing date treatments. In contrast to the relationships among leaf, bracteal, spikelet and their initiation duration, the correlation between the number of floret and its initiation duration was highly significant. 展开更多

The genomic composition of 1911289, a wheat ( Tritium aestivum L.) maintainer of K-CMS, was examined by several methods, such as genomic in situ hybridization (GISH), biochemical marking, and DNA molecular marking. The results got by GISH and PCR amplification of dispersed rye-specific repetitive DNA sequence suggested that the alien chromatin in T911289 derived from rye. Specifically PCR amplification of the rye-specific microsatellite primers (SCM9) and seed storage protein analysis indicated that the alien chromatin in T911289 had developed from the short arm of 1R chromosome of rye (1RS). PCR amplification by using microsatellite primers locating on 1BS and seed storage protein analysis also revealed that 1911289 had lost the arm of 1BS or a small distal segment of it. We conclude that T911289 is a heterogeneous population which displays two distinct different types of translocation, i.e. the Robertsonian translocation and small segment translocation. The Robertsonian translocation type observed in our study is different from the 1BL/1RS translocation which is widely used in wheat production; it may be a novel and complex translocation form. Though the linkage between the desirable agronomic traits and the deleterious genes expressed as sticky dough has not got broken in T911289, the recovery of small segment translocation will still benefit the genetic study of wheat and rye. 展开更多

[Method] Through measuring the minimum inhibitory concentra-tion, the in vitro antibacterial activity of golden buckwheat water extract was deter-mined; meanwhile, the therapeutic effect of golden buckwheat oral solution on my-coplasma infection was determined by artificial y infecting chickens with Mycoplasma gal isepticum culture. [Results] The golden buckwheat water extract had obvious in-hibitory effects against Pseudomonas aeruginosa and Escherichia coli, and a certain inhibitory effect on Salmonel a and Staphylococcus aureus; administration of golden buckwheat oral solution at the dose of 0.5%-1.0% continuously for 5 d had a good therapeutic effect against mycoplasma infection. Green fluorescent protein (GFP) gene was successfully transferred into the isolated zygotes and early proembryos of wheat (Triticum aestivum L.) by electroporation. A frequency, as high as 46.7% of GFP gene transient expression in early proembryos, was achieved under 150 V/cm electric field strength, 25 muF capacitor, 200 mug/mL of linear plasmid DNA and an electroporation buffer at pH 7.2. Compared with five-day-old proembryos, the zygotes and early proembryos needed lower optimum strength of electric field. After culturing in KM8p medium, the electroporated early proembryos divided and GFP gene expression was observed in daughter cells and subsequent divisions. There was no mosaicism of gene expression in the zygotes and 2-, 4- and 8-celled proembryos. 展开更多

The North China Plain,where summer corn(Zea mays L.)and winter wheat(Triticum aestivum L.)are the major crops grown,is a major agricultural area in China.Permeable soils make the region susceptible to groundwater pollution by NO_3-N,which is applied to fields in large amounts of more than 400 kg NO_3-N ha^(-1)as fertilizer.A field experiment was established in 2002 to examine the relationship among N fertilization rate,soil NO_3-N,and NO_3-N groundwater contamination.Two adjacent fields were fertilized with local farmers' N fertilization rate(LN)and double the normal application rate(HN),respectively,and managed under otherwise identical conditions.The fields were under a traditional summer corn/winter wheat rotation.Over a 22-month period,we monitored NO_3-N concentrations in both bulk soil and soil pore water in 20-40 cm increments up to 180 cm depth.We also monitored NO_3-N concentrations in groundwater and the depth of the groundwater table.No significant differences in soil NO_3-N were observed between the LN and HN treatment.We identified NO_3-N plumes moving downward through the soil profile.The HN treatment resulted in significantly higher groundwater NO_3-N,relative to the LN treatment,with groundwater NO_3-N consistently exceeding the maximum safe level of 10 mg L^(-1),but groundwater NO_3-N above the maximum safe level was also observed in the LN treatment after heavy rain.Heavy rain in June,July,and August 2003 caused increased NO_3-N leaching through the soil and elevated NO_3-N concentrations in the groundwater.Concurrent rise of the groundwater table into NO_3-N- rich soil layers also contributed to the increased NO_3-N concentrations in the groundwater.Our results indicate that under conditions of average rainfall,soil NO_3-N was accumulated in the soil profile.The subsequent significantly higher- than-average rainfalls continuously flushed the soil NO_3-N into deeper layers and raised the groundwater table,which caused continuous groundwater contamination with NO_3-N.The results suggest that under common farming practices in the North China Plain,groundwater contamination with NO_3-N was likely,especially during heavy rainfalls,and the degree of groundwater contamination appeared to be proportional to the N application rates.Decreasing fertilization rates, splitting fertilizer inputs,and optimizing irrigation scheduling had potential to reduce groundwater NO_3-N contamination. 展开更多

Soil moisture content per 20 cm was measured vertically within 0-300 cm soil layers in anexperiment with five treatments: deep-loosening tillage (DLT), traditional tillage (TT), plastic mulch (PM),straw mulch (SM) and plastic plus straw mulch (PSM). All mulch treatments were under no tillage conditions.Total storage of precipitation in soil from 0 to 300 cm was determined before sowing. Results showed thatthe new methods of tillage and mulch were the basic ways to improve water condition in dryland wheat fields.In a rainy year, PM with no tillage played a significant role in storing and conserving precipitation, while ina rain-deficient year, the role was not significant. Due to evaporation, DLT did not promote the storage ofsoil moisture. SM was the best way to store and conserve soil moisture. In SM treatment the wheat yieldsincreased by more than 20%. 展开更多

Fate of ^(15)N-Labeled Urea Under a Winter Wheat-Summer Maize Rotation on the North China Plain 被引量:44

A field experiment was conducted to investigate the fate of ^15N-labeled urea and its residual effect under the winter wheat (Triticum aestivum L.) and summer maize (Zea mays L.) rotation system on the North China Plain. Compared to a conventional application rate of 360 kg N ha^-1 (N360), a reduced rate of 120 kg N ha^-1 (N120) led to a significant increase (P 〈 0.05) in wheat yield and no significant differences were found for maize. However, in the 0-100 cm soil profile at harvest, compared with N360, N120 led to significant decreases (P 〈 0.05) of percent residual N and percent unaccounted-for N, which possibly reflected losses from the managed system. Of the residual fertilizer N in the soil profile, 25.6%-44.7% and 20.7%-38.2% for N120 and N360, respectively, were in the organic N pool, whereas 0.3%-3.0% and 11.2%-24.4%, correspondingly, were in the nitrate pool, indicating a higher potential for leaching loss associated with application at the conventional rate. Recovery of residual N in the soil profile by succeeding crops was less than 7.5% of the applied N. For N120, total soil N balance was negative; however, there was still considerable mineral N (NH4^+-N and NO3^--N) in the soil profile after harvest. Therefore, N120 could be considered ngronomically acceptable in the short run, but for long-term sustainability, the N rate should be recommended based on a soil mineral N test and a plant tissue nitrate test to maintain the soil fertility. 展开更多

The fate of urea-and ammonium bicarbonate (ABC)-nitrogen (N) applied by prevailing traditional techniques to winter wheat (Triticum aestivum L.) or maize (Zea mays L.) grown in the fields of Fluvo-aquic soil was investigated using 15N tracer-micro-plot technique. Results show that:(1) at maturity of wheat, N recovery in plants and N losses of urea and ABC applied at seeding in autumn were 31-39%, and 34-46%, respectively, while the corresponding figures for side-banding at 10 cm depth in early spring were 51-57%, and 5-12%; surface-broadcast of urea followed by irrigation at early spring was as efficient as the side-banding in improving N recovery in plants and reducing N loss, however, such technique was found less satisfactory with ABC. (2) At the maturity of maize, N recovery in the plants and N loss of urea and ABC sidebanded at seedling stage or prior to tasseling ranged from 23% to 57%, and 9% to 26%, respectively. (3) Either in Wheat or in maize experiment, the majority of residual fertilizer N in soil profile (0-60 cm) was in the form of biologically immobilized organic N, however, the contribution of ammonium fixation by clay minerals increased markedly with depth in soil profile. (4) Though the proportion of residual fertilizer N was generally highest in the top 20 cm soil layer, considerable reaidual N (mostly 6-11 % of the N applied) was found in 60-100 cm soil layers. 展开更多

Nicholas aspires to develop a substantive form of reason to base a critical theory of society aimed at human emancipation in the spirit of the Frankfurt School. The most interesting part of this effort is that although Nicholas thinks that Habermas's theory of procedural and situated communicative rationality is an initial step towards a substantive conception of reason, he recognizes the limitations of formalism in this conception and argues that the requisite terms provides Maclntyre's theory of tradition-constituted and tradition-constitutive reason. The mediating figure in this project is Charles Taylor, whose critique, first, is used against Habermas and, then, his Gadamerian idea of the "fusion of horizons" is adopted in order to expand Maclntyre's theory. 展开更多

To provide a scientific basis for sustainable land management, a 20-year fertility experiment was conducted in Changwu County, Shaanxi Province, China to investigate the effects of long-term application of chemical fertilizers on wheat grain yield and yield stability on the Loess Plateau using regression and stability analysis. The experiment consisted of 17 fertilizer treatments, containing the combinations of different N and P levels, with three replications arranged in a randomized complete block design. Nitrogen fertilizer was applied as urea, and P was applied as calcium superphosphate. Fertilizer rates had a large effect on the response of wheat yield to fertilization. Phosphorus, combined with N, increased yield significantly (P 〈 0.01). In the unfertilized control and the N or P sole application treatments, wheat yield had a declining trend although it was not statistically significant. Stability analysis combined with the trend analysis indicated that integrated use of fertilizer N and P was better than their sole application in increasing and sustaining the productivity of rainfed winter wheat. 展开更多

[Resulti The fluctuation of NRA in wheat changed greatly from 20:00 pm to 11:00 am. The enzyme activity remained constant, but at 14:00 the enzyme activity was the high- est, higher than all the other time points except the enzyme activity measured at11:00. The enzyme activity was the lowest of 17:00, which was lower than all the other time points except the enzyme activity measured at 2:00. Concentrations of atmospheric nitrous oxide (N2O), a potent greenhouse gas, have been continuously increasing, and cropland soils are one of the largest sources of N2O. Variations in environmental and anthropogenic factors have substantial impacts on both the frequency and magnitude of N2O emissions. Based on measurements from a wheat-maize system in the North China Plain, the authors parameterized the Agricultural Production Systems Simulator (APSIM) model, which was initially developed in Australia, for simulating N2O emissions under different agricultural management practices. After calibrating one of the key parameters -- the fraction of N2O lost in nitrification (k2) -- the results showed that the model successfully captured the daily N2O fluxes under different nitrogen fertilization treatments, but underestimated some large peak fluxes. By pooling all data together, the calibrated APSIM model also performed well in representing cumulative N2O emissions under various treatments at annual and finer (monthly and daily) time scales. 展开更多
