参灵方防治佐剂性关节炎大鼠药源性胃黏膜损伤的实验研究 被引量:1

目的探讨参灵方对佐剂性关节炎(adjuvant arthritis,AA)大鼠药源性胃黏膜损伤的防治作用。方法建立大鼠AA模型,造模后将出现二次反应的大鼠随机取48只分为模型组、复合模型组、雷尼替丁组、参灵方小剂量组、参灵方中剂量组、参灵方大剂... 目的探讨参灵方对佐剂性关节炎(adjuvant arthritis,AA)大鼠药源性胃黏膜损伤的防治作用。方法建立大鼠AA模型,造模后将出现二次反应的大鼠随机取48只分为模型组、复合模型组、雷尼替丁组、参灵方小剂量组、参灵方中剂量组、参灵方大剂量组,加上空白对照组,每组8只,共计7组。雷尼替丁组予雷尼替丁30 mg/(kg.d)灌胃,参灵方小剂量组、中剂量组、大剂量组分别予参灵方3.5 g、7.0 g、14.0 g/(kg.d)灌胃,空白对照组、模型组和复合模型组分别予等容积0.9%氯化钠注射液灌胃,均为14 d。计算治疗前后大鼠足爪肿胀率、关节炎指数及胃黏膜损伤指数、各灌药组对胃黏膜损伤的抑制率、胃黏膜的病理学变化,并进行组间比较。结果给药前后模型组、复合模型组、雷尼替丁组、参灵方各剂量组组间关节炎指数、足爪肿胀率差异均无统计学意义(P>0.05);给药前后各组与空白对照组比较,足爪肿胀率差异均有统计学意义(P<0.05);参灵方治疗后大鼠足爪容积、关节炎指数较模型组、复合模型组均有所下降,但差异无统计学意义(P>0.05)。雷尼替丁组、参灵方小剂量、中剂量、大剂量组胃黏膜损伤指数与复合模型组比较差异均有统计学意义(P<0.01),参灵方小剂量、中剂量组与雷尼替丁组比较胃黏膜损伤指数差异无统计学意义(P>0.05),而参灵方大剂量组与雷尼替丁组比较差异有统计学意义(P<0.05)。雷尼替丁组、参灵方小剂量、中剂量、大剂量组胃黏膜损伤抑制率分别为74.05%、66.81%、74.41%、55.70%,参灵方小剂量组、大剂量组与中剂量组及雷尼替丁组比较差异有统计学意义(P<0.05)。结论参灵方对AA大鼠关节无明显影响,但对药源性胃黏膜损伤有保护作用。 展开更多

目的研究预先给予参灵方对佐剂性关节炎(adjuvant arthritis,AA)大鼠药源性胃黏膜损伤的保护作用。方法建立AA大鼠模型,造模成功后随机分为参灵方小、大剂量组、模型组、雷尼替丁组,每组10只,分别灌胃治疗(雷尼替丁组给予30mg·kg-1... 目的研究预先给予参灵方对佐剂性关节炎(adjuvant arthritis,AA)大鼠药源性胃黏膜损伤的保护作用。方法建立AA大鼠模型,造模成功后随机分为参灵方小、大剂量组、模型组、雷尼替丁组,每组10只,分别灌胃治疗(雷尼替丁组给予30mg·kg-1·d-1;参灵方2组分别给予参灵方3.5g·kg-1·d-1、7g·kg-1·d-1,28d后采用皮下注射消炎痛进一步诱导药源性胃黏膜损伤的动物模型,光学显微镜观察胃黏膜组织病理变化。免疫组织化学ABC法对大鼠胃黏膜组织凋亡相关蛋白Bcl-2、Bax的表达进行细胞计算机医学图像分析及统计学处理。结果参灵方可明显抑制消炎痛致AA大鼠损伤胃黏膜的细胞凋亡,诱导损伤的胃黏膜细胞增殖,上调Bcl-2表达、下调Bax表达,与模型组比较,差异有统计学意义(P<0.05)。结论参灵方抑制细胞凋亡,对AA大鼠药源性胃黏膜损伤有保护作用。 展开更多

龙砂膏滋方对无症状HIV感染者疗效及生活质量影响研究 被引量:2

目的探究龙砂膏滋方对无症状人类免疫缺陷病毒(HIV)感染者疗效及生活质量影响。方法将符合纳入标准的60例无症状HIV感染者随机分为观察组和对照组,每组30例,对照组经辨证接受抗病毒联合参灵扶正胶囊方案治疗,观察组于冬至日开始停服参... 目的探究龙砂膏滋方对无症状人类免疫缺陷病毒(HIV)感染者疗效及生活质量影响。方法将符合纳入标准的60例无症状HIV感染者随机分为观察组和对照组,每组30例,对照组经辨证接受抗病毒联合参灵扶正胶囊方案治疗,观察组于冬至日开始停服参灵扶正胶囊并改用龙砂膏滋方调治,服药时间约为30 d,服完膏方后继续给予抗病毒联合参灵扶正胶囊方案治疗,观察半年,比较各组患者临床治疗效果、治疗前后患者中医证候(发热、咳嗽、乏力、纳呆、腹泻、呕吐等)积分变化、T细胞亚群指标(CD_(4)^(+)、CD_(4)^(+)/CD_(8)^(+))变化、生存质量评分、不良反应情况、患者满意度,随访6个月,比较两组患者机会性感染发生率。结果较对照组患者(76.67%,23/30),观察组患者治疗总有效率(96.67%,29/30)更高(P<0.05);治疗后各组患者中医证候(发热、咳嗽、乏力、纳呆、腹泻、呕吐等)积分、生存质量评分、T细胞亚群指标(CD_(4)^(+)、CD_(4)^(+)/CD_(8)^(+))均改善,观察组患者治疗后中医证候(发热、咳嗽、乏力、纳呆、腹泻、呕吐等)积分、生存质量评分均优于对照组患者(P<0.05),但两组患者治疗后T细胞亚群指标(CD_(4)^(+)、CD_(4)^(+)/CD_(8)^(+))指标无差异(P>0.05);两组患者治疗过程中均未发生不良反应(P>0.05);观察组患者治疗满意度提升率(96.67%,29/30)高于对照组患者(76.67%,23/30)(P<0.05);观察组患者治疗后6个月内机会性感染发生率低于对照组患者(P<0.05)。结论龙砂膏滋方可提升HIV感染者临床治疗效果,改善患者症状,患者恢复较好,无不良反应,安全可靠,且患者生活质量、满意度显著提升,机会性感染发生率较低,值得推广应用。 展开更多

目的观察参灵抗癌方(SLKAF)逆转胃癌细胞失巢凋亡抵抗作用,并从磷脂酰肌醇-3-激酶/丝苏氨酸蛋白激酶(PI3K/AKT)调控Bcl-2细胞死亡调解子抗体(Bim)表达为切入点探讨其作用机制。方法采用梯度剂量的SLKAF(0,1,2,4,8,16 mg/mL)与SGC-7901... 目的观察参灵抗癌方(SLKAF)逆转胃癌细胞失巢凋亡抵抗作用,并从磷脂酰肌醇-3-激酶/丝苏氨酸蛋白激酶(PI3K/AKT)调控Bcl-2细胞死亡调解子抗体(Bim)表达为切入点探讨其作用机制。方法采用梯度剂量的SLKAF(0,1,2,4,8,16 mg/mL)与SGC-7901胃癌细胞共培养,MTT法检测胃癌细胞活力,GraphPad Prism7.0计算半数抑制浓度(IC50),并以1×IC50、2×IC50分别作为SLKAF干预SGC-7901细胞的低剂量和高剂量。运用RNA干扰技术将Bim-siRNA转染SGC-7901细胞,qPCR技术检测Bim表达,观察对Bim表达的抑制效率;以SLKAF低、高剂量分别与SGC-7901细胞和经Bim-siRNA转染的SGC-7901细胞共培养,分别采用qPCR和Western Blot检测PI3K、AKT的表达,流式细胞技术检测细胞凋亡率,软琼脂克隆形成实验检测细胞克隆形成能力。结果SLKAF对SGC-7901细胞增殖有显著的抑制作用(P<0.01),并呈明显的量—效应关系,IC50为7.626 mg/mL,故确定8、16 mg/mL分别作为低剂量和高剂量。Bim-siRNA转染SGC-7901细胞可抑制Bim表达(P<0.05)。SLKAF高、低剂量均降低SGC-7901细胞PI3K、AKT表达(P<0.05,P<0.01),提高胃癌细胞凋亡率(P<0.05,P<0.01),降低胃癌细胞克隆形成能力(P<0.01);与低剂量比较,高剂量SLKAF作用下的SGC-7901细胞/经Bim-siRNA转染的SGC-7901细胞表达PI3K、AKT均降低(P<0.01),胃癌细胞凋亡率升高(P<0.05),细胞克隆形成数降低(P<0.05);在SLKAF相同剂量下,SGC-7901细胞和经Bim-siRNA转染的SGC-7901细胞表达PI3K、AKT比较,差异无统计学意义(P>0.05),但细胞凋亡率和细胞克隆形成能力差异均有统计学意义(P<0.01)。结论SLKAF经PI3K/AKT调控Bim表达逆转胃癌细胞失巢凋亡抵抗。 展开更多

To overcome the deficiencies addressed in the conventional PID control and improve the dynamic performance and robustness of the system, a simple design and parameters tuning approach of internal model control-PID (IMC-PID) controller was proposed for the first order plus time-delay (FOPTD) process and the second order plus time-delay (SOPTD) process. By approximating the time-delay term of the process model with the first-order Taylor series, the expressions for IMC-PID controller parameters were derived, and they had only one adjustable parameter 2 which was directly related to the dynamic performance and robustness of the system. Moreover, an analytical approach of selecting 2 was given based on the maximum sensitivity Ms. Then, the robust tuning of the system could be achieved according to the value of Ms. In addition, the proposed method could be extended to the integrator plus time-delay (IPTD) process and the first order delay integrating (FODI) process. Simulation studies were carried out on various processes with time-delay, and the results show that the proposed method could provide a better dynamic performance of both the set-point tracking and disturbance rejection and robustness against parameters perturbation. 展开更多

The sensitivity of simulated tropical intraseasonal oscillations (ISO) to different cumulus parameterization schemes was analyzed using an atmospheric general circulation model (latest version-SAMIL2.2.3) developed at the Laboratory for Numerical Modeling for Atmospheric Sciences and Geophysical Fluid Dynamics (LASG) at the Institute of Atmospheric Physics (IAP) of the Chinese Academy of Sciences.Results show that the basic features of tropical climatological intraseasonal oscillations (CISO) can be captured using all three cumulus schemes.The CISO simulated by the Tiedtke scheme was found to be more realistic than that of the Manabe and Zhang-McFarlane schemes.The results of simulated transient intraseasonal oscillations (TISO) indicate that although the Tiedtke and the Zhang-McFarlane schemes in the new version SAMIL2.2.3 have been adjusted according to different problems,only the latter can simulate the eastward propagation of the 27-50-day TISO mode.It may be associated with the more realistic diabatic heating profile simulated by the Zhang-McFarlane scheme.In addition,the Manabe scheme in SAMIL2.2.3 is the same as that in the prior version SAMIL2.08.However,some aspects of the physical process,such as the radiation scheme and aerosol condition,have been changed.Conversely the eastward propagation from 100°E to the west of the tropical 27-50-day TISO mode only can be simulated using the Manabe scheme of SAMIL 2.08.Consequently,not all the improvements of physical parameterization schemes work well in every respect.The coordinated developments between dynamic frame and physical processes,and among different physical processes,are important methods that may be used to improve the model. 展开更多

A novel random walk coefficient(RWC) model of the interferometric fiber optic gyroscope(IFOG) to decompose fundamental noise sources, namely the shot noise, the excess noise, the thermal noise, and the detection circuit noise, from the overall noise was developed. The coefficients of the model were extracted from the measured RWC instead of by calculating the accurate IFOG parameters, which is simpler and more accurate. The correctness and the accuracy of the model were verified by experiments. Using this model, the RWC of the experimental IFOG was predicted and the quantitative contributions of the noise sources were determined. According to the predicted results, the parameters of the IFOG were optimized. Finally, based on the model, a noise decomposition and parameter optimization method was proposed for high sensitivity IFOG design. 展开更多
