目的 调查星形细胞瘤间期核17p13.1的变异,探讨它与星形细胞瘤发生与演进的关系。方法 选择D17Z1和17p13.1为探针,采用双标荧光原位杂交技术,检测了43例着丝点序列正常的星形细胞瘤17p13.1的丢失。结果 4/14的星形细胞瘤、5/13间变型星... 目的 调查星形细胞瘤间期核17p13.1的变异,探讨它与星形细胞瘤发生与演进的关系。方法 选择D17Z1和17p13.1为探针,采用双标荧光原位杂交技术,检测了43例着丝点序列正常的星形细胞瘤17p13.1的丢失。结果 4/14的星形细胞瘤、5/13间变型星形细胞瘤和5/16多形胶质母细胞瘤出现了17p13.1或17p的等位或杂合性丢失。结论 星形细胞瘤染色体17p13.1的丢失是频繁发生的。这种变化在一部分星形细胞瘤病例的发生中是早期的遗传学变异,操纵肿瘤的起始阶段,不参与星形细胞瘤的演进。而另一些无17p13.1或17p异常的病例可能不是从低级向高级别演进而是源于原位。 展开更多

背景:多项体外细胞学实验显示,含氧化涂层钛金属较普通钛金属表面表现出更好的生物相容性和生物活性。目的:进一步观察等离子体电解氧化涂层钛棒在动物体内的骨整合能力。方法:取36只16周龄雄性SD大鼠(购自上海西普尔-必凯实验动物有限... 背景:多项体外细胞学实验显示,含氧化涂层钛金属较普通钛金属表面表现出更好的生物相容性和生物活性。目的:进一步观察等离子体电解氧化涂层钛棒在动物体内的骨整合能力。方法:取36只16周龄雄性SD大鼠(购自上海西普尔-必凯实验动物有限公司),建立双侧股骨远端髓内钉植入模型,左侧植入多孔纳米等离子体电解氧化涂层钛棒,作为实验组;右侧植入无涂层钛棒,作为对照组。植入2,4,6周时,处死大鼠并收获股骨远端带有金属植入物的骨组织标本,进行Van Gieson染色、双标荧光染色及生物力学评价。结果与结论:(1)Van Gieson染色显示,实验组与对照组植入物-骨组织界面均未见纤维组织长入;植入2周时,两组均可见植入物表面部分骨组织接触,实验组于植入4周时形成植入物周围骨整合,对照组于植入6周时形成植入物周围骨整合;(2)双标荧光染色显示,植入2周时,两组植入物周围均产生大量新生骨组织,与钛棒结合紧密;植入4周时,实验组新生骨与植入物表面呈现连续性接触,对照组新生骨与植入物表面呈不连续区块接触;植入6周时,两组均显示出了连续的植入物-骨组织接触,但新生骨骨量明显较4周时减少;(3)生物力学检测显示,实验组植入4周时的最大压缩载荷高于对照组(P <0.001),两组植入2,6周时的最大压缩载荷比较无差异(P> 0.05);实验组植入4周时的最大压缩载荷高于植入2,6周时(P <0.05),对照组植入6周时的最大压缩载荷高于植入2,4周时(P <0.05);(4)结果表明,等离子体电解氧化涂层钛棒植入早期的骨整合能力优于普通钛棒。 展开更多

Comparative genetic studies have shown that there are widespread synteny and colinearity of the genes among different species within grass family. Rice ( Oryza sativa L.) is a model plant, and analysis of its genome allows us to reveal the common features and the evolutionary rules of the gramineous genomes and accumulate the data for establishment of a common genetic system in the Poaceae. In this study, a rice gene Pib ( 10.3 kb), a map-based cloned gene, and RFLP markers linked with it are used as the tested probes to investigate their homology and physical location among the tested species. Southern blotting analysis showed that there were sequences homologous to Pib in maize genome. Further, Pib was localized onto the chromosomes of O. sativa ssp. indica cv. Guangluai 4, O. officinalis Wall ex Watt and the inbred line of Zea mays cv. Huangzao 4. The results of fluorescence in situ hybridization (FISH) and double-color FISH indicated that a synteny of Pib and RFLP markers linked with Pib existed among the genomes of the three tested species. 展开更多

A rapid and sensitive fluorescence labeling method was developed and validated for the microanalysis of a sulfated polysaccharide drug,namely propylene glycol alginate sodium sulfate(PSS), in rat plasma. Fluorescein isothiocyanate(FITC) was selected to label PSS, and 1, 6-diaminohexane was used to link PSS and FITC in order to prepare FITC-labeled PSS(F-PSS) through a reductive amination reaction. F-PSS was identified by UV-Vis, FT-IR and 1H-NMR spectrum. The cell stability and cytotoxicity of F-PSS were tested in Madin-Darby canine kidney(MDCK) cells. The results indicated that the labeling efficiency of F-PSS was 0.522% ± 0.0248% and the absolute bioavailability was 8.39%. F-PSS was stable in MDCK cells without obvious cytotoxicity. The method was sensitive and reliable; it showed a good linearity, precision, recovery and stability. The FITC labeling method can be applied to investigating the absorption and metabolism of PSS and other polysaccharides in biological samples. 展开更多
