废水中无机磷的双波长分光光度研究 被引量:5

引入赤霉素-二氯化锡混合液作为钼蓝法的新显色剂.在1.0mol/LHNO3介质和有机相中,赤霉素-二氯化锡混合物与无机磷、硅杂多酸反应生成杂多蓝,经萃取分离测定有机相,以Λ=690nm为测定波长,Λ=780nm为参比波长,获得线性回归方程为△A=0.113... 引入赤霉素-二氯化锡混合液作为钼蓝法的新显色剂.在1.0mol/LHNO3介质和有机相中,赤霉素-二氯化锡混合物与无机磷、硅杂多酸反应生成杂多蓝,经萃取分离测定有机相,以Λ=690nm为测定波长,Λ=780nm为参比波长,获得线性回归方程为△A=0.1138C+0.0153,校正因子r=0.9993.表观摩尔吸光系数为1.2×104L·mol-1·cm-1.PO43-质量浓度位于0.1~3mg/L,符合朗伯-比耳定律.赤霉素-二氯化锡混合还原剂还原速度快,产物稳定性好,操作简便,结果令人满意. 展开更多

建立了一种基于96孔板-酶标仪的双波长紫外分光光度法高通量筛选6-羟基烟酸转化菌的方法。实验以251nm为测定波长、231nm为参比波长测定转化样品的6-羟基烟酸含量,6-羟基烟酸与ΔA251-231在0.5~11μg/mL浓度范围内有良好的线性关系,服... 建立了一种基于96孔板-酶标仪的双波长紫外分光光度法高通量筛选6-羟基烟酸转化菌的方法。实验以251nm为测定波长、231nm为参比波长测定转化样品的6-羟基烟酸含量,6-羟基烟酸与ΔA251-231在0.5~11μg/mL浓度范围内有良好的线性关系,服从朗伯-比尔定律,平均回收率为99.11%~100.81%。利用96孔板-酶标仪,每天筛选量可达到2000~5000个反应,达到高通量筛选要求。 展开更多

Iin this paper an isosbestic dual--wavelength spectrophotometric method for the simultaneous determination of zinc and cadmium using meso--tetrakis [4-(trimethylammonium) phenyl] porphine has been developed. The interference from excess of reagent was masked by adding lead ions solution.The molar absorptivities (Ae) for zinc and cadmium were 6. 54 × 105 and 4. 27 × 105 L. mol-1. cm-1,respectively. The method could be used to determine zinc and cadmium in synthetic mixtures, and the satisfied results were obtained. 展开更多

Determination of Cholesterol Content in Waste Fat by Dual-wavelength Spectrophotometry

[Objective]The aim was to find a way to measure the cholesterol content in deserted oil.[Method]Dual-wavelength spectrophotometry method was used for the determination of cholesterol in edible oil.630 nm and 720 nm were set as target band and reference band in this method,respectively.The result was compared to liquid chromatography method.[Result]The relative standard deviation(RSD)was at 1.26% and recovery was between 101.9% and 110.7%.Linear range was 8.0-40.0 μg/ml.The tested result was close to the result of liquid chromatography.[Conclusion]It was proved that this method was easy,fast and accurate.The disturbance of sitosterol was eliminated without isolation.It can be used for fast identification of the authenticity of waste fat. 展开更多
