反义内皮素转换酶核酸对尘螨过敏哮喘患者外周血单个核细胞释放白细胞介素5的抑制作用 被引量:2

目的 探讨转导反义内皮素转换酶 (ECE)核酸的气道上皮细胞对尘螨过敏性患者外周血单个核细胞释放Th1/Th2相关细胞因子的影响。方法 尘螨过敏患者 2 1例 ,分离其外周血单个核细胞 (PBMC) ,根据实验要求分为 :未刺激组 (A组 )、反义ECE... 目的 探讨转导反义内皮素转换酶 (ECE)核酸的气道上皮细胞对尘螨过敏性患者外周血单个核细胞释放Th1/Th2相关细胞因子的影响。方法 尘螨过敏患者 2 1例 ,分离其外周血单个核细胞 (PBMC) ,根据实验要求分为 :未刺激组 (A组 )、反义ECE上皮细胞 +PBMCs组 (A1组 )、对照组上皮细胞 +PBMCs组 (A2 组 )及螨刺激组 (B组 )、反义ECE上皮细胞 +PBMCs+尘螨提取液组 (B1组 )、对照组上皮细胞 +PBMCs+尘螨提取液组 (B2 组 ) ,共培养 72h ,螨刺激组同时加入尘螨提取液 2 0 μg/ml。培养上清用酶联免疫吸附 (ELISA)法检测白细胞介素 5 (IL 5 )、γ干扰素 (IFN γ)。结果2 1例患者中有 12例患者经尘螨刺激后IL 5释放明显增加。将 12例患者进行统计学分析发现 ,未经尘螨刺激时 ,A1组IL 5和IFN γ水平分别为 [(6 0± 1 3)× 10 -9g/L]和 [(6 3± 2 6 )× 10 -9g/L],A2 组为 [(7 5± 1 1)× 10 -9g/L]、[(70± 5 2 )× 10 -9g/L],两组比较差异均无显著性 (P >0 0 5 ) ;尘螨刺激后 ,B1组的IL 5水平和IFN γ分别为 [(8 2± 1 6 )× 10 -9g/L]、[(10 0± 4 1)× 10 -9g/L],B2 组分别为[(12 0± 1 8)× 10 -9g/L]、[(15 3± 71)× 10 -9g/L],两组IL 5比较差异有显著性 (P =0 0 4 7) ,而IFN γ水平比较差异无显著性 (P > 展开更多

Two stable transformed lines containing antisense LeERT1 or LeERT2 sequences and their hybridized line were investigated to determine the effect of LeERT1 and LeERT2 specificity in the ethylene receptor family in tomato (Lycopersicon esculenturn Mill.) on ethylene signaling. The transgenic line alel containing antisense LeER T1 displayed shorter length of seedling grown in the dark and adult plant in the light, severe epinastic petiole, and accelerated abscission of petiole explant and senescence of flower explant, compared with its wild type B 1. The transgenic line ale2 containing antisense LeERT2 also exhibited shorter hypocotyls and slightly accelerated abscission. The phenotypes of cross line dale of LeERT1 and LeERT2 were close to alel in many aspects. These results suggested that LeERT1 probably plays a relatively important role in ethylene signaling of tomato growth and development. 展开更多
