目的 :研究卡氏肺孢子虫发育增殖过程的超微结构。方法 :用地塞米松诱导大鼠卡氏肺孢子虫肺炎 ,取肺组织制成透射电镜标本 ,观察肺内虫体的超微结构和发育增殖过程。结果 :肺内卡氏肺孢子虫有滋养体、囊前期和包囊三个发育阶段。滋养体... 目的 :研究卡氏肺孢子虫发育增殖过程的超微结构。方法 :用地塞米松诱导大鼠卡氏肺孢子虫肺炎 ,取肺组织制成透射电镜标本 ,观察肺内虫体的超微结构和发育增殖过程。结果 :肺内卡氏肺孢子虫有滋养体、囊前期和包囊三个发育阶段。滋养体的生殖方式有二分裂、外出芽、内出芽及接合生殖 ,并见到囊前期和包囊内的囊内小体的分裂增殖过程。此外 ,虫体内有丰富的线粒体 ,且随虫体的发育和分裂 ,线粒体的数量、大小和形状也发生明显的变化。严重感染时虫体以滋养体为主。结论 :卡氏肺孢子虫的生活史由滋养体、囊前期和包囊三个发育阶段构成 。 展开更多

目的:研究PLZF调控小鼠早期T细胞发育与自我更新的功能,以及PLZF对胸腺细胞发育和自我更新的影响。方法:利用新生小鼠进行胸腺移植,以Rag2/γc−/−小鼠为受体。首先比较PLZF−/−与野生型(PLZF+/+)小鼠胸腺细胞总数的差异,再通过流式细胞... 目的:研究PLZF调控小鼠早期T细胞发育与自我更新的功能,以及PLZF对胸腺细胞发育和自我更新的影响。方法:利用新生小鼠进行胸腺移植,以Rag2/γc−/−小鼠为受体。首先比较PLZF−/−与野生型(PLZF+/+)小鼠胸腺细胞总数的差异,再通过流式细胞术和细胞分选,筛选小鼠的骨髓造血干细胞和胸腺的DN2a细胞以及PLZF−/−小鼠的早期T细胞系前体(ETP),以及PLZF−/−或野生型小鼠在新生小鼠胸腺移植模型的移植物DN1细胞中的表达情况。结果:PLZF−/−小鼠的胸腺细胞总数和早期T细胞系前体数目与野生型小鼠相比显著下降,并且细胞总数的减少不是由细胞内源性引起的。通过研究PLZFEGFP报告基因小鼠和新生小鼠胸腺的肾移植模型,发现PLZF在Rag2/γc−/−受体小鼠胸腺移植物的DN1(Lineage−CD44+CD25−)细胞中高表达,且供体细胞的PLZF缺失会显著影响来源胸腺移植物的DN1细胞的比例。结论:PLZF可能在T细胞的正常增殖发育过程中不是必需的,但很可能参与维持早期T细胞系前体的自我更新。 展开更多

Identification of iron-loaded ferritin as an essential mitogen for cell proliferation and postembryonic development in Drosophila 被引量:1

Animal cells require extrinsic cues for growth, proliferation and survival. The propagation of Drosophila imaginal disc cells in vitro, for example, requires the supplementation of fly extract, the composition of which remains largely undefined. Here I report the biochemical purification of iron-loaded ferritin as an active ingredient of fly extract that is required for promoting the growth of clone 8 imaginal disc cells. Consistent with an essential role for iron- loaded ferritin in cultured cells, overexpression of ferritin or addition of iron in a nutrient-poor diet increases animal viability and body weight, promotes cell proliferation, and shortens the duration of postembryonic development. Conversely, overexpression of dominant-negative ferritin or addition of iron chelator causes the opposite effects. Fer- ritin mutant flies arrest development at the first-instar larval stage with a severe starvation phenotype reminiscent of that seen in starved larvae. I conclude that iron-loaded ferritin acts as an essential mitogen for cell proliferation and postembryonic development in Drosophila by maintaining iron homeostasis and antagonizing starvation response. 展开更多

The development, survivorship and reproduction of Bemisia tabaci B bi otype on eggplant at seven constant temperatures (17, 20, 23, 26, 29, 32 and 35℃) were studied. The developmental periods from egg to adult varied from 48.7 days at 17℃ to 13.9 days at 29℃ and the develo pmental threshold estimated for a generation by linear regression was 12.4℃. The optimum temperature for B. tabaci population growth was 26℃, both extremely low (<17℃) and high temperature (>32℃) delayed the de velopment. Survivorships from egg to adult was 67.3 % at 26℃, 27.6 % and 29.0 % at 35℃ and 17℃ resp ectively. The average longevity of females ranged from 39.6 days at 20℃ to 12.8 days at 35℃. Ovipo sition per female varied from 164.8 eggs at 20℃ to 78.5 eggs at 32℃. Both the longevity and oviposition of B. tabaci females at different temperatures were significantly different ( P < 0.05), and the intrinsic rate of natural increase ( r m ) for B. tabaci at 29℃ was the highest. 展开更多

Erythropoiesis is a process during which multipotential hematopoietic stem cells proliferate, differentiate and eventually form mature erythrocytes. Interestingly, unlike most cell types, an important feature of erythropoiesis is that following each mitosis the daughter cells are morphologically and functionally different from the parent cell from which they are derived, demonstrating the need to study erythropoiesis in a stage-specific manner. This has been impossible until recently due to lack of methods for isolating erythroid cells at each distinct developmental stage. This review summarizes recent advances in the development of methods for isolating both murine and human erythroid cells and their applications. These methods provide powerful means for studying normal and impaired erythropoiesis associated with hematological disorders. 展开更多
