RT_PCR and Southern blotting using 3′_untranslated region (3′_UTR) as specific probe revealed that pea isoactin genes were expressed in roots, stems, leaves, tendrils, pollen and juvenile siliques, but displayed different patterns of transcript accumulation. Two_fold serial dilution electrophoresis showed PEAcⅠ mRNA preferentially accumulated in rapidly developing tissues: it peaked in seven days' stem; remained at a rather high level in leaves within a month but decreased significantly later; varied a little in tendrils and reached a median and a very low level respectively in juvenile siliques and in pollen. PEAcⅡ displayed somewhat similar expression pattern to PEAcⅠ. The observed differences in sequences and transcript accumulation patterns suggest that the individual pea actin genes may differ in their transcriptional regulation and cellular function. Phylogenetic tree of actins showed that pea actin isoforms are as diverged from each other as they are from other plant actins, and pea actins might have originated from a common ancestor before the divergence of the dicot and monocot plants.展开更多
Through a simulation test carried out with soil columns (61.8 cm in diameter), the effect of precipitation on salt-water dynamics in soils was studied by in-situ monitoring of salt-water dynamics using soil salinity s...Through a simulation test carried out with soil columns (61.8 cm in diameter), the effect of precipitation on salt-water dynamics in soils was studied by in-situ monitoring of salt-water dynamics using soil salinity sensors and tensiometers. The results show that in the profile of whole silty loam soil, the surface runoff volume due to precipitation and the salt-leaching role of infiltrated precipitation increased with the depth of ground water; and in the profile with an intercalated bed of clay or with a thick upper layer of clay, the amount of surface runoff was greater but the salt-leaching role of precipitation was smaller than those in the profile of whole silty loam soil. In case of soil water being supplemented by precipitation, the evaporation of groundwater in the soil columns reduced, resulting in a great decline of salt accumulation from soil profile to surface soil. The effect of precipitation on the water regime of soil profile was performed via both water infiltration and water pressure transfer. The direct infiltration depth of precipitation was less than 1 m in general, but water pressure transfer could go up to groundwater surface directly.展开更多
Laminaria and Saccharina have recently been recognized as two independent clades from the former genus Laminaria. Traditional morphological taxonomy is being challenged by molecular evidence from both nucleus and plas...Laminaria and Saccharina have recently been recognized as two independent clades from the former genus Laminaria. Traditional morphological taxonomy is being challenged by molecular evidence from both nucleus and plastid. Intensive work is in great demand from the perspective of genome colinearity. In this study, 118 sequence-tagged site(STS) markers were screened for phylogenetic analyses, 29 based on genome sequences, while 89 were based on expressed sequence tag(EST) sequences. EST-based STS marker development(29.37%) had an effi ciency twice as high as genome-sequence-based development(9.48%) as a result of high conservation of gene transcripts among the relative species. S. ochotensis, S. religiosa, S. japonica, and L. hyperborea showed great homogeneity in all 118 STS markers. Our result supports the view that the diversifi cation between the genera Saccharina and Laminaria was a more recent event and that Saccharina and Laminaria shared high phylogenetic affi nity. However, when it came to the single nucleotide polymorphism(SNP) level among the 41 SNPs, L. hyperborea owned 29 unique SNPs against 12 within the left three Saccharina species and 12 of the 13 indels were supposedly unique for L. hyperborea, indicated by its high variability. Originating from homologous ancestors, species between the recently diverged genera Laminaria and Saccharina may have taken in enough mutations at the SNP level only, in spite of different evolutionary strategies for better adaptation to the environment. Our study lays a solid foundation from a new perspective, although more accurate phylogenetic analysis is still needed to clarify the evolutionary traces between the genera Saccharina and Laminaria.展开更多

Here, we report the isolation of IGF-Ⅱ and expression analysis of IGFs in turbot Scoph...The insulin-like growth factors Ⅰ and Ⅱ (IGF-Ⅰ and IGF-Ⅱ) are important proteins involved in fish growth and develop- ment. Here, we report the isolation of IGF-Ⅱ and expression analysis of IGFs in turbot Scophthalmus maximus, aiming to clarify their function in embryonic and larval development of fish. The deduced IGF-Ⅱ gene is 808 bp in full length, which encodes a protein of 219 amino acids and is 93% similar with that ofParalichthys olicaceus in amino acid sequence. The tissue abundance and the ex- pression pattern of IGFs in a turbot at early development stages were investigated via reverse transcription-polymer chain reaction. Result showed that the IGF-Ⅰ and IGF-Ⅱ genes were widely expressed in tissues of S. maximus. IGF-Ⅰ was detected in all tissues ex- cept intestines with the highest level in liver, while IGF-Ⅱ transcript presented in all tissues except muscle. At the stages of embry- onic and larval development, the mRNA levels of IGFs sharply increased from the stage of unfertilized egg to post larva, followed by a decrease with larval development. However, there was an increase in IGF-Ⅰ at the embryonic stage and IGF-Ⅱ at the gastrula stage, respectively. These results suggested that IGFs play important roles in cell growth and division of the turbot. Our study provides reference data for further investigation of growth regulation in turbot, which can guarantee better understanding of the physiological role that IGFs play in fish.展开更多

Part of the amplified F protein DNA product (nucleotide sequence 47-422) and the deduced amino acid sequences were compared phylogenetically with those from strains previously reported in other geographic regions. Phylogenetic analysis indicated that the Kazakhstanian pigeon paramyxovirus type 1 (PPMV-1) isolates belong to genotype VI or 4bii. To our knowledge, this is the first reported VI isolates that possess the sequences of 112 GKRQKR116* F117 within the F0 protein. The information is fundamental to improving the efficiency of control strategies and vaccine development for NDV.展开更多

Objective:To explore the mechanisms of fulminant hepatitis(FH) in the early stages,and to determine the critical pathways in its initiation and progression.Methods:Twelve BALB/c mice were divided into four groups:one ...Objective:To explore the mechanisms of fulminant hepatitis(FH) in the early stages,and to determine the critical pathways in its initiation and progression.Methods:Twelve BALB/c mice were divided into four groups:one group left as negative control and sacrificed immediately after injection of phosphate-buffered saline(PBS),and another three groups with concanavalin A(Con A) administration sacrificed at 1,3,and 6 h after injection.Affymetrix GeneChip Mouse 430 2.0 Array was employed to evaluate the expression profile of each of the 12 samples.Further analysis was done on the microarray data to extract the genes that were differentially expressed.Enrichment analysis was carried out to determine relevant pathways within which regulated genes were significantly enriched.Results:A total of 393,8354 and 11 344 differentially expressed genes were found,respectively,at three time points.During 0-1 h and 1-3 h,most of the pathways enriched with regulated genes were related to immune response and inflammation,among which Toll-like receptor(TLR) signaling and mitogen-activated protein kinase(MAPK) signaling appeared during both phases,while cytokine-cytokine receptor interaction,apoptosis,T cell receptor signaling,and natural killer(NK) cell-mediated cytotoxicity pathways emerged during the second phase.Pathways found to be significant during 3-6 h were mostly related to metabolic processes.Conclusion:The TLR signaling pathway dominates the early responses of Con A-induced FH in mice.It stimulates the production of type I cytokines,therefore recruiting and activating T/NK cells.Activated T/NK cells exert their cytotoxicity on hepatocytes through inducing death receptorintermediated apoptosis,resulting in liver injury.展开更多

It can be produced by microbial fermentation. In this study, the microbial flora of mangrove sediment was screened to identify strains with high production of PDO by fermentation of glycerol. The PDO productivities of the isolated strains were tested, and the strain with highest PDO productivity was characterized using the API20E and 16-s rRNA sequence analysis. The physiological and phylogenetic analysis indicated that the strain was closed related to K. pneumoniae species and was named as K. pneumoniae HSL4. The structure of the dha cluster which was responsible for the biosynthesis of PDO was analyzed. It is observed that K. pneumoniae HSL4 was tolerant to salt and partly tolerant to acetate and lactate, which will favor industrial applications. Fed-batch fermentation experiments revealed K. pneumoniae HSL4 exhibited an excellent ability to produce PDO with high concentration (80.08 g L^-1), productivity (2.22 g L^-1h^-1) and conversion (0.435 g g^-1 or 0.53 mol mol^-1). The metabolic flux profile illuminated that glycerol was consumed rapidly and PDO was accumulated quickly to a high level during the exponential growth phase. This study provided important information for further fermentation and metabolic engineering of PDO production by K. pneumoniae HSL4.展开更多
