[ Results] It was showed by transmission electron microscopy that the initial callus cells had a large central vacuole, which squeezed its cytoplasm to be a thin layer around the brim of cell, Meanwhile the nuclear was also squeezed to distribute in the corner of cell, but its nucleolus could be still observed; Compared embryogenic callus with initial callus, its cell wall became thick, and many starch grains and chloroplasts including starch grains could be observed in the cytoplasm area of cell membrane; In non-embryoenic callus, no organelles except for the vacuole could be observed; In browning callus, there was almost no organelles in cells. Like other mGluR subtypes, mGluR5s contain a phylogenetically c...Metabotropic glutamate receptor subtype 5 (mGluR5) is a Group I mGlu subfamily of receptors coupled to the inositol trisphosphate/diacylglycerol pathway. Like other mGluR subtypes, mGluR5s contain a phylogenetically conserved, extracellular orthosteric binding site and a more variable allosteric binding site, located on the heptahelical transmembrane domain. The mGluR5 receptor has proved to be a key pharmacological target in conditions affecting the central nervous system (CNS) but its presence outside the CNS underscores its potential role in pathologies affecting peripheral organs such as the gastrointestinal (GI) tract and accessory digestive organs such as the tongue, liver and pancreas. Following identification of mGluR5s in the mouth, various studies have subsequently demonstrated its involvement in mechanical allodynia, inflammation, pain and oral cancer. mGluR5 expression has also been identified in gastroesophageal vagal pathways. Indeed, experimental and human studies have demonstrated that mGluR5 blockade reduces transient lower sphincter relaxation and reflux episodes. In the intestine, mGluR5s have been shown to be involved in the control of intestinal inflammation, visceral pain and the epithelial barrier function. In the liver, mGluR5s have a permissive role in the onset of ischemic injury in rat and mice hepatocytes. Conversely, livers from mice treated with selective negative allosteric modulators and mGluR5 knockout mice are protected against ischemic injury. Similar results have been observed in experimental models of free-radical injury and in vivo mouse models of acetaminophen intoxication. Finally, mGluR5s in the pancreas are associated with insulin secretion control. The picture is, however, far from complete as the review attempts to establish in particular as regards identifying specific targets and innovative therapeutic approaches for the treatment of GI disorders.展开更多

A positive expression of like-muscarinic acetylcholine receptor M2 was observed in the mushroom body, central body and antennal lobes of the ant brain; but there is great diversity in their location and intensity among worker, queen and male ants. It is speculated that like-muscafinic acetylcholine receptor M2 plays a critical role in the central nervous system, in terms of projecting visual information and olfactory information into the protocerebrum and integrating many inputs.展开更多

Mutations of γc result inX-linked severe combined immunodeficiency syndrome inhumans, indicating that rye is essential for normal development and function of the immune system. We demonstratethat human hematopoietic cells express two γc transcriptsdiffering in their carboxyl terminal coding region. Onetranscript is the previously reported sequence (γc-long),whereas the newly identified sequence exhibits a deletion of72 nucleotides close to the 3’-end of the open reading frame(γc-short). This alteration predicts a loss of 24 amino acidsincluding a conserved tyrosine residue which is shared byseveral members of the cytokine receptor family. Thepresence of these two distinct forms of rye transcripts wasdemonstrated by sequencing of reversely transcribed andpolymerase chain reaction (RT-PCR) amplified mRNA, restriction digestion of the RT-PCR products, RNAse protection, and Northern blotting from human cell lines andhuman peripheral blood lymphocytes. Furthermore, thetwo variants were present in peripheral blood lymphocytesfrom both female and male donors, which rules out allelicvariants since rye is a single copy gene located on the Xchromosome. A truncation mutant at a site near the observed changes in γc-short has been reported by othersto alter biochemical events activated by cytokines. In the field of L2 (second language) reading, scholars generally agree that ER (extensive reading) improves L2 learners' reading speed and comprehension and enriches their L2 vocabulary (Grabe & Stoller, 1997)...In the field of L2 (second language) reading, scholars generally agree that ER (extensive reading) improves L2 learners' reading speed and comprehension and enriches their L2 vocabulary (Grabe & Stoller, 1997). This teacher-inquiry type of paper presents a practical suggestion for supplementing ER activity with the element of CL (cooperative learning). ER, theoretically speaking, focuses on the solitary task of silent reading. The CL technique used in this study was a book-talk activity. Forty-five freshmen from a course of children and young adult literatures were required to read at least 20 English books throughout a semester. CL was added to facilitate students' ER in young adult literature. After a semester, short-answer questions were asked regarding students' comments on ER as well as CL. Students overall agreed that when ER is supplemented with CL, reading in an L2 seems to be less intimidating.展开更多
Differentially expressed genes(DEGs)between individuals with high(HC)and low(LC)total carotenoid content(TCC)were sampled from a selected line of Pinctada fucata martensii with black shell in the prismatic layer.The e...Differentially expressed genes(DEGs)between individuals with high(HC)and low(LC)total carotenoid content(TCC)were sampled from a selected line of Pinctada fucata martensii with black shell in the prismatic layer.The expression levels of candidate genes were verified by qRT-PCR.Targeted resequencing was used to detect SNPs in a candidate gene,PmSR-BI.The association of TCC with SNPs in PmSR-BI was determined.Results showed that a total of 1025 DEGs were identified between HC and LC.The expression levels of the candidate gene PmSR-BI in HC were higher than those in LC.Seven SNPs in the exon and eight SNPs in the 5′regulatory regions of PmSR-BI were found.Association analysis showed that one SNP in the exon and two SNPs in the 5′regulatory regions of PmSR-BI were significantly associated with the TCC(P<0.05).All SNPs of PmSR-BI were divided into four blocks.CC haplotype in Block 1 and AG haplotype in Block 3 were significantly higher than other haplotypes.These results help elucidate the mechanism underlying carotenoid metabolism and develop marker-assisted breeding design in the species.展开更多
Objective: To investigate the significance of extra-nodal spread in special histological sub-types of breast cancer and the relationship of such spread with prognostic parameters.Methods: A total of 303 breast cancer ...Objective: To investigate the significance of extra-nodal spread in special histological sub-types of breast cancer and the relationship of such spread with prognostic parameters.Methods: A total of 303 breast cancer cases were classified according to tumor type, and each tumor group was subdivided according to age, tumor diameter, lymph node metastasis, extra-nodal spread, vein invasion in the adjacent soft tissue, distant metastasis, and immunohistochemical characteristics [estrogen receptor(ER), progesterone receptor(PR) existence, p53, c-erbB-2, and proliferative rate(Ki-67)]. The 122 cases with extra-nodal spread were clinically followed up.Results: An extra-nodal spread was observed in 40%(122 cases) of the 303 breast cancer cases. The spread most frequently presented in micro papillary carcinoma histological sub-type(40 cases, 75%), but least frequently presents in mucinous carcinoma(2 cases, 8%). Patients with extra-nodal spread had a high average number of metastatic lymph nodes(8.3) and a high distant metastasis rate(38 cases, 31%) compared with patients without extra-nodal spread.Conclusion: The existence of extra-nodal spread in the examined breast cancer sub-types has predictive value in forecasting the number of metastatic lymph nodes and the disease prognosis.展开更多
Anemia is the most common complication of inflammatory bowel disease (IBD). Control and inadequate treatment leads to a worse quality of life and increased morbidity and hospitalization. Blood loss, and to a lesser ex...Anemia is the most common complication of inflammatory bowel disease (IBD). Control and inadequate treatment leads to a worse quality of life and increased morbidity and hospitalization. Blood loss, and to a lesser extent, malabsorption of iron are the main causes of iron def iciency in IBD. There is also a variable component of anemia related to chronic inflammation. The anemia of chronic renal failure has been treated for many years with recombinant human erythropoietin (rHuEPO), which significantly improves quality of life and survival. Subsequently, rHuEPO has been used progressively in other conditions that occur with anemia of chronic processes such as cancer, rheumatoid arthritis or IBD, and anemia associated with the treatment of hepatitis C virus. Erythropoietic agents complete the range of available therapeutic options for treatment of anemia associated with IBD, which begins by treating the basis of the inflammatory disease, along with intravenous iron therapy as f irst choice. In cases of resistance to treatment with iron, combined therapy with erythropoietic agents aims to achieve near-normal levels of hemoglobin/hematocrit (11-12 g/dL). New formulations of intravenous iron (iron carboxymaltose) and the new generation of erythropoietic agents (darbepoetin and continuous erythropoietin receptor activator) will allow better dosing with the same eff icacy and safety.展开更多

Retrospective analysis of the case reports of 284 pat...The objective of this study is to investigate hormonal receptor status of MOT (malignant ovarian tumor) and to evaluate its clinical and prognostic significance. Retrospective analysis of the case reports of 284 patients with MOT of different histogenesis, stages I-IV, and immunohistochemical study of paraffin-embedded tissues were performed. Hormonal receptor status of tumors with different morphology genesis was studied and hormonal receptor phenotype of serous OC (ovarian cancer) was determined. The analysis of correlation between the expression of steroid hormone receptors (receptors to estrogens (ER), progesterone (PR) and testosterone (TR)) in ovarian tumors, histological type of tumors and clinical morphological parameters were performed. Overall and relapse-free survival rates of the patients with serous OC depending on the hormonal receptor phenotype of the tumor were assessed. Presence of positive expression of steroid hormone receptors in serous OC (ER-66.4%, PR^53.4%, TR-53.0%), mucinous OC (ER-88.0%, PR-84.0%, TR-60.0%) and in sex cord stromal tumors (ER-74.1%, PR and TR-77.8%) is proved by correlation of all steroid receptors expression with morphology type of ovarian tumors (ER - r = 0.4; PR - r = 0.4; TR - r = 0.3; p 〈 0.05). Direct correlation between hormonal receptor phenotype of serous OC and the age period of the patients was established (r = 0.5; p = 0.002): postmenopausal women patients reported the most increased frequency of serous OC with positive hormonal receptor tumor phenotypes (52.4%), in particular during their late post-menopausal period (39.0%). Significantly low overall survival among the patients with positive hormonal receptor phenotype of serous OC was recorded (29.5±3.4%) in comparison with the same score in the patients with negative phenotype of tumors (44.5±3.7%) (p 〈 0.05). Multifactor analysis of Cox-regression model has defined that positive hormonal receptor phenotype of serous OC increases the risk of disease relapse (HR 1.4; 95.0% CI 1.1-1.7), significantly decreases overall survival rates in the patients (HR 1.4; 95.0% CI 1.1-1.8). Positive hormonal receptor status of MOT is an independent factor of unfavorable clinical progress of tumor process which can be regarded as the criterion for development of the methods of hormonal therapy application in complex treatment of the patients, and demands further large-scale multi-center studies in that direction.展开更多

The study is motivated by the wide applications of hydrogen bonding fluids in the field of colloid science, and the effects of relevant factors on the depletion potential and depletion force between colloid particles have been studied. These factors include the size ratio of the colloid particle to the fluid molecule, the bulk density of the fluid, the functionality (the number of proton acceptors a and proton donors d) and hydrogen bonding strength as well as the colloid-fluid interaction energy. By comparing the depletion potential calculated under various conditions, it is shown that the effects of these factors on the depletion potential are very significant, and in particular in regulating the depletion force and its range.展开更多

The 122 cases with extra-nodal spread were clinically followed up.Results: An extra-nodal spread was observed in 40%(122 cases) of the 303 breast cancer cases. The spread most frequently presented in micro papillary carcinoma histological sub-type(40 cases, 75%), but least frequently presents in mucinous carcinoma(2 cases, 8%). Patients with extra-nodal spread had a high average number of metastatic lymph nodes(8.3) and a high distant metastasis rate(38 cases, 31%) compared with patients without extra-nodal spread.Conclusion: The existence of extra-nodal spread in the examined breast cancer sub-types has predictive value in forecasting the number of metastatic lymph nodes and the disease prognosis.
文摘Anemia is the most common complication of inflammatory bowel disease (IBD). Control and inadequate treatment leads to a worse quality of life and increased morbidity and hospitalization. Blood loss, and to a lesser extent, malabsorption of iron are the main causes of iron def iciency in IBD. There is also a variable component of anemia related to chronic inflammation. The anemia of chronic renal failure has been treated for many years with recombinant human erythropoietin (rHuEPO), which significantly improves quality of life and survival. Subsequently, rHuEPO has been used progressively in other conditions that occur with anemia of chronic processes such as cancer, rheumatoid arthritis or IBD, and anemia associated with the treatment of hepatitis C virus. Erythropoietic agents complete the range of available therapeutic options for treatment of anemia associated with IBD, which begins by treating the basis of the inflammatory disease, along with intravenous iron therapy as f irst choice. In cases of resistance to treatment with iron, combined therapy with erythropoietic agents aims to achieve near-normal levels of hemoglobin/hematocrit (11-12 g/dL). New formulations of intravenous iron (iron carboxymaltose) and the new generation of erythropoietic agents (darbepoetin and continuous erythropoietin receptor activator) will allow better dosing with the same eff icacy and safety.
文摘The objective of this study is to investigate hormonal receptor status of MOT (malignant ovarian tumor) and to evaluate its clinical and prognostic significance. Retrospective analysis of the case reports of 284 patients with MOT of different histogenesis, stages I-IV, and immunohistochemical study of paraffin-embedded tissues were performed. Hormonal receptor status of tumors with different morphology genesis was studied and hormonal receptor phenotype of serous OC (ovarian cancer) was determined. The analysis of correlation between the expression of steroid hormone receptors (receptors to estrogens (ER), progesterone (PR) and testosterone (TR)) in ovarian tumors, histological type of tumors and clinical morphological parameters were performed. Overall and relapse-free survival rates of the patients with serous OC depending on the hormonal receptor phenotype of the tumor were assessed. Presence of positive expression of steroid hormone receptors in serous OC (ER-66.4%, PR^53.4%, TR-53.0%), mucinous OC (ER-88.0%, PR-84.0%, TR-60.0%) and in sex cord stromal tumors (ER-74.1%, PR and TR-77.8%) is proved by correlation of all steroid receptors expression with morphology type of ovarian tumors (ER - r = 0.4; PR - r = 0.4; TR - r = 0.3; p 〈 0.05). Direct correlation between hormonal receptor phenotype of serous OC and the age period of the patients was established (r = 0.5; p = 0.002): postmenopausal women patients reported the most increased frequency of serous OC with positive hormonal receptor tumor phenotypes (52.4%), in particular during their late post-menopausal period (39.0%). Significantly low overall survival among the patients with positive hormonal receptor phenotype of serous OC was recorded (29.5±3.4%) in comparison with the same score in the patients with negative phenotype of tumors (44.5±3.7%) (p 〈 0.05). Multifactor analysis of Cox-regression model has defined that positive hormonal receptor phenotype of serous OC increases the risk of disease relapse (HR 1.4; 95.0% CI 1.1-1.7), significantly decreases overall survival rates in the patients (HR 1.4; 95.0% CI 1.1-1.8). Positive hormonal receptor status of MOT is an independent factor of unfavorable clinical progress of tumor process which can be regarded as the criterion for development of the methods of hormonal therapy application in complex treatment of the patients, and demands further large-scale multi-center studies in that direction.

文摘The depletion potential between two colloid particles immersed in a hydrogen bonding fluid has been investigated by density functional theory. The study is motivated by the wide applications of hydrogen bonding fluids in the field of colloid science, and the effects of relevant factors on the depletion potential and depletion force between colloid particles have been studied. These factors include the size ratio of the colloid particle to the fluid molecule, the bulk density of the fluid, the functionality (the number of proton acceptors a and proton donors d) and hydrogen bonding strength as well as the colloid-fluid interaction energy. By comparing the depletion potential calculated under various conditions, it is shown that the effects of these factors on the depletion potential are very significant, and in particular in regulating the depletion force and its range.
