[Method] A rice double mutant with stripes on stems, leaves and spikelets were taken as experimental materials to study the enzyme activity changes in different growth stages and amino acid variation in rice. [ Result] The SOD activity in mutant was higher than that in wild plant at tillering metaphase, but lower than that in wild type before heading stage and late flowering; the POD activity in three stages increased firstly then declined and the activity showed highest maximal activity at before heading stage. However, the POD activity in wild type showed the opposite change trend; the CAT activity presented degression at three stages, especially high at tillering metaphase, but reverse changes in wild type; the MDA activity decreased at three stages, but it was still higher than that in wild type, besides, the soluble sugar content of mutant was lower, but total amino acid content was increased. [ Conclusion] The expression of mutant characteristics was correlated with SOD, POD, CAT and MDA activity Changes and these changes made the mutant survive and rice quality change at last.展开更多

A paddy field with unknown anthro- pogenic contamination in Deqing County, Zhejiang Province, China was selected to characterize the spatial variability and distribution of micronutrients in rice grain and soil. A total of 96 paired soil and rice grain samples were collected at harvest. The micronutrients in the soil samples were extracted by diethylenetriamine pentaacetic acid (DTPA). The mean micronutrient concentrations in rice grain were 3.85 μg Cu g^-1, 11.6 μg Fe g^-1, 39.7 μg Mn g^-1 and 26.0 μg Zn g^-1 The mean concentrations were 2.54μg g^-1 for DTPA-Cu, 133.5μg g^-1 for DTPA-Fe, 30.6μg g^-1 for DTPA-Mn, and 0.84μg g^-1 for DTPA-Zn. Semivariograms showed that measured micronutrients in rice grain were moderately dependent, with a range distance of about 110 m. The concentrations of the DTPA-extractable micronutrients all displayed strong spatial dependency, with a range distance of about 60 m. There was some resemblance of spatial structure between soil pH and the grain Cu, Fe, Mn, and Zn. By analogy, similar spatial variation was observed between soil organic matter (SOM) and DTPA-extractable table micronutrients in the soil. Kriging estimated maps of the attributes showed the spatial distributions of the variables in the field, which is beneficial for better understanding the spatial variation of micronutrients and for potentially refining agricultural management practices at a field scale.展开更多

Mor...Disparity in the root morphology of six rice(Oryza sativa L.) genotypes varying in potassium(K) efficiency was studied with three K levels:5 mg/L(low),10 mg/L(moderate) and 40 mg/L(adequate) in hydroponic culture. Morphological parameters included root length,surface area,volume and count of lateral roots,as well as fine(diameter<0.2 mm) and thick(diameter>0.2 mm) roots. The results indicate that the root growth of all genotypes was reduced under low K,but moderate K deficiency increased the root length of the efficient genotypes. At deficient and moderate K levels,all the efficient rice genotypes developed more fine roots(diameter<0.2 mm) than the inefficient ones. Both fine root count and root surface area were found to be the best parameters to portray K stress in rice. In accordance with the root morphology,higher K concentrations were noted in shoots of the efficient genotypes when grown at moderate and deficient K levels,indicating that root morphology parameters are involved in root uptake for K and in the translocation of K up to shoots. K deficiency affected not only the root morphology,but also the root ultra-structure. The roots of high-efficient genotypes had stronger tolerance to K deficient stress for root membrane damage,and could maintain the developed root architecture to adapt to the low K growth medium.展开更多

The fine mapping of the gene(s) responsible for low glutelin content will provide information regarding the distribution of glutelin related genes in rice genome and will generate markers for the selection of low glutelin rice varieties. Following an SDS-PAGE screen of rice germplasm from Taihu Valley of China, Japonica selection W3660 is identified to be a novel mutant characterized with low glutelin content. For fine mapping the mutant gene for low glutelin content, F2 and F3 populations were derived from a cross between W3660 and Jingrennuo. SDS-PAGE analysis of the total endosperm protein showed that the low glutelin content trait was controlled by a single dominant nuclear gene. Genetic mapping, using SSRs, located this gene to chromosome 2, in the region between SSR2-001/SSR2-004 and RM1358. The dis- tances of the two markers to the target gene were 1.1 cM and 3.8 cM respectively. By semi-quantitative RT-PCR analysis, the transcripts of GluB4/GluB5 genes located within the region do not change. However, GluB5 gene located proximal to SSR2-001/SSR2-004 was specifically reduced. SSR profiles of seven Japonica varieties were compared with that of W3660 for loci in the relevant genetic region. The markers SSR2-004 and RM1358 were used for marker- assisted selection. The selection efficiencies of SSR2-004 and RM1358 were 96.8% and 92.7% respectively. This provides a standard starting point for the breeding of low glutelin content rice varieties in China.展开更多

This study examined the effect of seasonal precipitation, maximum and minimum temperatures and extreme climatic variables on yield levels and variability of rice, maize and wheat yields. The authors applied a stochastic production function approach as suggested by Just-Pope for panel data at the district level. The estimation results indicated that climate trends in Nepal had a significant influence on crop yield levels and variances in various magnitudes and directions. The results showed an increase in precipitation negatively influenced maize yield levels and positively influenced wheat yield levels; however, a positive influence was found to reduce yield variability in rice and wheat. Similarly, an increase in maximum temperature apparently led to decreased maize and increased wheat yield levels, respectively, and also led to increased yield variability in rice and maize. Likewise, the minimum temperature was helpful to increase yield levels for all crops and to decrease the yield variability in rice and maize. Moreover, the extreme climates such as low precipitation and high maximum temperatures significantly influenced the reduction in yield levels of rice and maize, respectively that could be used to design an index insurance product for Nepal.展开更多

To better understand genetic variations among...Rice blast caused by Magnaporthe oryzae (M. oryzae) is one of the most destructive diseases, which causes significant rice yield losses and affects global food security. To better understand genetic variations among different isolates of M. oryzae in nature, we re-sequenced the genomes of two field isolates, CH43 and Zhong-10-8-14, which showed distinct pathogenecity on most of the rice cultivars. Genome-wide genetic variation analysis reveals that ZHONG-10-8-14 exhibits higher sequence variations than CH43. Structural variations (SVs) detection shows that the sequence variations primarily occur in exons and intergenic regions. Bioinformatics analysis for gene variations reveals that many pathogenecity-related pathways are enriched. In addition, 193 candidate effectors with various DNA polymorphisms were identified, including two known effectors AVR-Pik and AVR-Pital. Comparative polymorphism analysis of thirteen randomly selected effectors suggests that the genetic variations of effectors are under positive selection. The expression pattern analysis of several pathogenecity-related variant genes indicates that these genes are differentially regulated in two isolates, with much higher expression levels in Zhong-10-8-14 than CH43. Our data demonstrate that the genetic variations of effectors and pathogenecity-related genes are under positive selection, resulting in the distinct pathogeuicities of CH43 and Zhong- 10-8-14 on rice.展开更多
